Mode of Action and Chemical Modification of an Alkaline Xylanase (CX-III) from Alkalophilic Cephalosporium sp. RYM-202 (호알카리성 Cephalosporium sp. RYM-202로부터 분리된 alkaline xylanase (CX-III)의 작용 양상 및 화학적 변환)

  • The hydrolysis products formed from birchwood xylan by the action of an alkaline xylanase (CX-III) from alkalophilic Cephaloxporium sp. RYM-202 were xylobiose and xylooligosaccharides polymerized with more than 4 sugar molecules. This enzyme was not active on xylobiose but readily attacked xylotriose accumulating xylobiose as a major product. The predominant end-products from xylotetraose by CX-III were xylobiose and xylotriose. These results indicate that the enzyme is typically endo-type xylanase possessing transglycosidase activity. Chemical modification of CX-III with N-bromosuccinimide revealed that two tryptophan residues per molecule of CX-III were essential for its catalytic activity on xylan. On the other hand, iodoacetamide and diethylpyrocarbonate did not influence the activity of the enzyme, suggesting that cysteine and histidine residues are not involved in the active site of this alkaline xylanase.

Surface Modification Silica Nanoparticles by Aerosol Self Assembly (에어로졸 자기조립에 의한 실리카 나노분말의 표면개질)

  • Surface modification of silica nanoparticles was investigated using an aerosol self assembly. Stearic acid was used as surface treating agent. A two-fluid jet nozzle was employed to generate an aerosol of the colloidal suspension, which contained 20 nm of silica nanoparticles, surface modifier, and ethyl alcohol. Powder properties such as morphology, specific surface area and pore size distribution were analyzed by SEM, BET and BJH methods, respectively. Surface properties of the silica power were analyzed by FT-IR. The OH bond of the $SiO_2$ surface was converted to a C-H bond. It was revealed that the hydrophilic surface changed to a hydrophobic one due to the aerosol self assembly. Morphology of the surface treated powder was nanostructured with lots of pores having an average diameter of around $2\;{\mu}m$. Depending on the stearic acid concentration (0.25 to 1.0 wt%), the pore size distribution of the particles and the degree of hydrophobicity ranged from 1.5 nm to 180 nm and 29.6% to 50.2%, respectively.

  • This study was undertaken to find out the changes of chemically modified egg albumen grl after heat treatment at $95^{\circ}C$ for 30 minutes or at $120^{\circ}C$ for 30 minutes. Acetylation and succinylation increased the hardness of egg albumen gel, it was rather higher at high heat treatment($120^{\circ}C$, 30min.) than at low heat treatment($95^{\circ}C$, 30min). The cohesiveness of egg albumen gel was improved remarkably by succinylation and maleylation at both low and high heat treatment. The lightness and yellowness of egg albumen gel were decreased by chemical modification. Initial heat denaturation temperature of egg albumen was increased by 11$^{\circ}C$ by acetylation, by $12.5^{\circ}C$ by maleylation and by ,$14.5^{\circ}C$ by succinylation.

  • In the development of biomaterials as a substitute of hard tissues, the biocompatibility and osteoconductivity of the biomaterial are considered to be one of the most significant considerations. These biological properties of a material can be greatly improved by the modification of the surface properties by the depositing calcium phosphate thin films on the material since calcium phosphate films possess similar chemical compositions to hard tissues. The success of a material as a biomaterial will be determined by the interaction of the surface of the material with the adhesion molecules which induce cellular adhesion and biological responses of the adherent cells. Depending on the adsorption mechanisms and adsorbed conformation of the adhesion molecules on the surface of the biomaterial, cellular responses, such as adhesion, proliferation and differentiation of osteoblast cells, can be promoted or restricted. It has been reported that materials of which surfaces were modified with thin films of calcium phosphate appeared to be more osteoconductive. Rapid formations of bone nodule in addition to higher differentiations of osteoblast have been observed on the calcium phosphate thin films.

Improvement of Fouling Resistance with Reverse Osmosis Membrane Using Multi-layer Silane-Epoxy Surface Modification (실란-에폭시 다층 표면개질을 통한 역삼투막의 내오염성 향상)

  • In this study, to solve the major problem of reverse osmosis (RO) membrane, surface of reverse osmosis membrane was modified by silane-epoxy multi layer. Octyltrimethoxysilane (OcTES) was polymerized to membrane surface via cross-linking by Sol-gel method. n = 8 alkylgroup of OcTES formed the branch structure by self assembly. And for improve fouling resistance of RO membrane, Ether group of ethylene glycol diglycidyl ether (EGDE) was given to improve hydrophilicity of RO membrane surface by ring-opening. To analyze structure of RO membrane surface with FE-TEM and AFM. Membrane surface of the ridge and valley structure and the bridge structure was confirmed due to the multi-layer surface modification of OcTES and EGDE. And through the increase of the roughness, the branch structure was formed well on membrane surface. Through the XPS analysis was identified chemical structure of membrane surface. And confirmed that the hydrophilic surface modification is given to the surface of the film through a Contact angle analysis. In optimization of EGDE surface modification condition, was suitable 0.5 wt% EGDE concentraion and $70^{\circ}C$ ring-opening temperature. In result of fouling resistance test and MFI is SUL-H10, $PA-OcTES_{1.0}$, $PA-OcTES_{1.0}-EGDE_{0.5}$ 68.7, 60.4, 5.4 ($10E-8hr/mL^2$), multi-layer surface modified membrane improved fouling resistance.

The Influence of Plasma Surface Modification on Frictional Property of Natural Rubber Vulcanizates

  • The plasma surface modification of natural rubber vulcanizate was carried out using chlorodifluoromethane in a radio-frequency (13.56 MHz) electrodeless bell type plasma reactor. The modification was qualitatively assessed by Fourier transform infrared spectroscopy. The frictional force of the plasma-treated surface was found to decrease with the time of plasma treatment. An increase in the surface polarity, as evidenced by the decrease in contact angle of a sessile drop of water and ethylene glycol on the natural rubber vulcanizate surface, was noted with the plasma modification. In the case of similar plasma treatment of glass surface, only a reduction in the polarity was observed. The use of geometric and harmonic mean methods was found to be useful to evaluate the London dispersive and specific components of surface free energy. Irrespective of the method used for evaluation, an increasing trend in the surface free energy was noted with increasing plasma treatment time. However, the harmonic mean method yielded comparatively higher values of surface free energy than the geometric mean method. The plasma surface modification was found to vary the frictional coefficient by influencing the interfacial, hysteresis and viscous components of friction in opposing dual manners.

Lifetime Prolongation of Poly (dimethylsiloxane) Surface Modification via 2-Hydroxyethyl Methacrylate Grafting for Electroosmotic Flow

  • To use Poly-dimethylsiloxane (PDMS) for the electrokinetic flow channel, the PDMS surface must be modified to be hydrophilic. With $O_2$ plasma treatment, it is difficult to maintain hydrophilicity for more than one day. In this paper, we present the chemical modification of the PDMS surface using 2-Hydroxyethyl methacrylate (HEMA) to prolong hydrophilicity lifetime. The oxide radicals generated temporarily on the PDMS surface by $O_2$ plasma are grafted with HEMA. Once the PDMS samples have been grafted, they demonstrate improved hydrophilicity retainment and electroosmotic flow characteristics compared to the untreated PDMS and the oxidized PDMS following the $O_2$ plasma process. This phenomenon was verified by the contact angles, Fourier transform infrared (FTIR) spectra and electro osmotic flow rates observed for more than 300 hours.

  • The demand for sub-micrometer or nanometer functional ceramic powders with a better suspension behavior in aqueous media in increasing. Redispersible barium titanate (BT) nanocrystals, green light emitting Mn2+ doped Zn$_2$SiO$_4$ and ZnS nanoparticle phosphors were synthesized by a hydrothermal method or chemical precipitation with surface modification. The nanoparticle redispersibility for BT was achieved by using a polymeric surfactant. X-ray diffraction(XRD) results indicated that the BT particles are of cubic phase with 80 nm in size. XRD results of zinc silicate phosphor indicate that seeds play an important role in enhancing the nucleation and crystallization of Zn$_2$SiO$_4$ crystals in a hydrothermal condition. This paper describes and discuss the methods of surface modification, and the resulting related properties for BT, zinc silicate and zinc sulfide.

Inhibition of Purine Nucleoside Phosphorylase (PNP) in Micrococcus luteus by Phenylglyoxal

  • Micrococcus luteus purine nucleoside phosphorylase (PNP) has been purified and characterized. The physical and kinetic properties have been described previously. Chemical modification of the enzyme was attempted to gain insight on the active site. The enzyme was inactivated in a time-dependent manner by the arginine- specific modifying reagent phenylglyoxal. There was a linear relationship between the observed rate of inactivation and the phenylglyoxal concentration. At 30 $^{\circ}C$ the bimolecular rate constant for the modification was 0.015 $min^{-1}mM^{-1}$ in 50 mM $NaHCO_3$ buffer, pH 7.5. The plot of logk versus log phenylglyoxal concentration was a strainght line with a slope value of 0.9, indicating that modification of one arginine residue was needed to inactivate the enzyme. Preincubation with saturated solutions of substrates protected the enzyme from inhibition of phenylglyoxal, indicating that reactions with phenylglyoxal were directed at arginyl residues essential for the catalytic functioning of the enzyme.

Electron Beam Modification of Dual Phase Filler: Surface Characteristics and its Influence on the Properties of Styrene-Butadiene Rubber Vulcanizates

  • The present work describes modification of dual phase filler by electron beam irradiation in presence of multifunctional acrylates like trimethylol propane triacrylate (TMPTA) or silane coupling agent like bis (3-triethoxysilylpropyltetrasulphide) and in-fluence of the modified fillers on the physical properties of styrene-butadiene rubber (SBR) vulcanizates. Modulus at 300 % elongation increases whereas the tensile strength decreases with increase in radiation dose for the dual phase filler loaded styrene-butadiene rubber vulcanizates (SBR). However, modulus and tensile strength significantly increase, which is more, pronounced at higher filler loadings for TMPTA modified dual phase filler loaded SBR. These changes in properties are explained by the equilibrium swelling data and Kraus plot interpreting the polymer-filler interaction. Electron beam modification of the filler results in a reduction of tan ${\delta}$ at $70^{\circ}C$, a parameter for rolling resistance and increase in tan ${\delta}$ at $0^{\circ}C$, a parameter for wet skid resistance of the SBR vulcanizates. Finally, the influence of modified fillers on the properties like abrasion resistance, tear strength and fatigue failure and the improvement in the properties have been explained in terms of polymer-filler interaction.
