Ammonia Production from Yeast Extract and Its Effect on Growth of the Hyperthermophilic Archaeon Sulfolobus solfataricus

  • Utilization of yeast extract and formation of byproduct metabolite were investigated for hyperthermophilic archaeon Sulfolobus solfataricus (DSH 1617). In both batch and fed-batch cultivations of S. solfataricus, maximal cell density, {{{{ { NH}`_{4 } ^{ +} }}} ion production and pH change were highly dependent on the ratio of yeast extract to glucose in the medium. Variation of {{{{ { NH}`_{4 } ^{ +} }}}} ion level was identified as a major cause of pH change during cultivation, and acidification of culture broth was attributed to consumption of {{{{ { NH}`_{4 } ^{ +} }}}} ions rather than formation of acid byproducts. It was also observed that increase of {{{{ { NH}`_{4 } ^{ +} }}}} ion concentrations in the medium resulted in greater degree of growth inhibition.

  • A simple, but effective on-line method for estimating the mycelial cell mass concentration from agitation speed data, a most readily-available process variable, has been developed for DO-stat cultures of Agaricus blazei. The dynamic change of dissolved oxygen concentration (DOC) in the initial transient period and the change in yield were considered in the development of the estimation algorithm or estimator. Parameters in the estimation algorithm were calculated from the agitation speed data at 20% of DOC. The proposed estimator could accurately predict the cell mass concentration regardless of DOC levels in the tested range of $10{\sim}40%$, showing a good extrapolation capability.

  • We investigated the speed change of the diamond spherical abrasive during the substrate surface polishing under the pad compression by using classical molecular dynamics modeling. We performed three-dimensional molecular dynamics simulations using the Morse potential functions for the copper substrate and the Tersoff potential function for the diamond abrasive. As the compressive pressure increased, the indented depth of the diamond abrasive increased and then, the speed of the diamond abrasive along the direction of the pad moving was decreased. Molecular simulation result such as the abrasive speed decreasing due to the pad pressure increasing gave important information for the chemical mechanical polishing including the mechanical removal rate with both the pad speed and the pad compressive pressure.

  • Thin films of SnO2, SnO2-ZnO and ZnO were prepared by the spray. chemical vapor deposition and vacuum evaporation method. They had good sensitivity to various gases involving toxic gases(i. e. SO2, CO). The change in conductivity of thin film guts sensors prepared was considered as the change in carrier concentration caused by gas absorption. And also the conductivity of the thin film elements had great dependence on atmospheric pressuie around them.

  • On this study, $CO_2$ gas, net of Ar gas, and mixed gas in solid wire(AWS ER 70S-6) and flux cored wire(AWS E71T-1) were used to weld on Mild steel(SS400). After the progress, the status changes of the welds in Mild steel(SS400) were investigated with compositional changes. For stable experiments, welding was conducted using the automatic feeder. Radiation testing, hardness testing, chemical composition analysis and penetrated cross-section were measured. Through these experiments, shapes of penetrated cross-section, chemical composition changes, and weld defects according to the variation of welding gas were known. Weld defects and weld cross-sectional shapes by the variation of the welding voltage were also detected.

  • Fluorescence of green fluorescent protein mutant, 2-5 GFP is observed during denaturation by guanidine. The fluorescence intensity decreases exponentially but the fluorescence lifetime does not change during denaturation. The fluorescence lifetime of the denatured protein is shorter than that of native form. As the protein structure is modified by guanidine, solvent water molecules penetrate into the protein barrel and protonate the chromophore to quench fluorescence. Most fluorescence quenchers do not affect the fluorescence of native form but accelerate the fluorescence intensity decay during denaturation. Based on the observations, a simple model is suggested for the structural change of the protein molecule during denaturation.

  • It was attempted to investigate the reaction of LB biological membrane in the gas surrounding by the use of LB method. The performance of the experiment was based on the idea that the adhesion of gas molecular on the surface of LB membrane, which induces the change of electrical properties, may make it possible to develop various sensor system modified from the sensor organs. The experimental results showed that for the acetone gas, the frequency changes in the proportional to the concentration of the gas. The reappearance of the gas reaction vs. frequency change was also able to be obtained properly.

  • This study is about cure kinetics of DGEBA/MDA/MN(malononitrile) system by Barrett method and Integral method using DSC dynamic run. Curing behavior was shown through DSC and the heat change involved in a reaction could be measured directly with DSC. The kinetic parameters such as activation energy, pre-exponential factor and reaction order were given by Barrett method and Integral method obtained in an assumption that the area of DSC enthalpic analysis curve was propotional to the enthalpic change.

  • The core-shell latex particles were prepared by sequential emulsion polymerization of alkyl methacrylate and styrene(ST) by using an water-soluble initiator(APS) after preparing monomer pre-emulsion in the presence of an anionic surfactant(SDBS). In organic/organic core-shell polymerization, the pre-emulsion method, which minimized required quantity of sulfactant, has been used to increase the conversion rate and the stability of core-shell latex particles as well as to reduce the formation of secondary particle that cause problems of soap-free emulsion during shell polymerization. We used several methods to observe the core-shell structure. The core-shell structure was studied by measuring pH change during hydrolysis by NaOH, glass transition temperature($T_g$) by differential scanning calorimeter(DSC), morphology of latex by transmission electron microscope(TEM) and change of particle size and distribution by a particle analyzer.

Environmental Effects Analysis by the Fertilizer Change with Wastewater Reuse in Paddy Fields (하수처리수의 농업용수 재이용시 시비량 변화에 따른 환경영향 분석)

  • The objective of this paper is to analyze the environmental effects by the fertilizer change with wastewater reuse for agriculture. For this research, Lysimeter tests are being implemented to cultivate rice with different levels of fertilizer applications with wastewater irrigation., and to analyze the nutrient loading by wastwater reuse in paddy fields was examined the CREAMS-PADDY model. CREAMS-PADDY model is modified from CREAMS model for considering the hydrologic cycles in paddy field. As a result, in the lysimeter treated by irrigation with wastewater and chemical fertilizer with half of the conventional amount showed generally similar tendency to the control plot. This may require the modifications of standard cultural practices for rice in terms of fertilizer and pesticide applications. However, high concentration of sodium in wastewater might cause damage to physico-chemical properties of paddy soil. And the wastewater reuse effects on nutrient loads were quantitatively analyzed and this results provide the reasonable management for agricultural reuse.
