Effects of Quality Characteristics and Antioxidant Activity of Korean Cultivated Wild Ginseng Extract (산양삼의 품질특성 및 항산화 활성에 미치는 영향)

  • In this study, we investigated the nutritional and functional constituents as well as quality characteristics and antioxidant activity of Korean cultivated wild ginseng (KG). The chemical compositions and amino acid content of KG were 7.56% water, 73.01% carbohydrates, 12.58% protein, 1.99% lipids, and 5.54% ash as well as 16.17 mg/g of amino acids, respectively. The major ginsenoside and minor ginsenoside contents of KG were 15.94 mg/g and 0.04 mg/g, respectively. The total polyphenol and flavonoid contents of KGE (Korean cultivated wild ginseng with 70% ethanol extract) were 8.93 mg GAE/g and 3.96 mg RHE/g, respectively. KGE also showed higher antioxidant activity than the other extracts (KGW, Korean cultivated wild ginseng with water extract) with regard to DPPH and ABTS radical scavenging activities (57.75% and 70.73%, respectively), nitrite oxide scavenging activity (44.01%), SOD-like activity (78.05%), reducing power activity ($1.08OD_{700nm}$), and ferrous ion-chelating activity (65.33%). Additionally, KGE had higher elastase, collagenase, and tyrosinase inhibition activities than KGW. These results suggest that KGE can be used as a bioactive and functional material in the food industry.

  • Carboxymethyl chitin(CM-chitin) was prepared by the reaction of alkali chitin with monochloroacetic acid in isopropyl alcohol. According to the pH variation, the adsorptivity of this chelating polymer to the alkali-earth metal ions such as $Ca^{2+},\;Mg^{2+}$, $Sr^{2+}$, $Ba^{2+}$ ions was determined by batch method. The adsorption tendency of this chelating polymer to most metal ions was increased with the increase of pH. The highest degree of adsorption was observed toward $Ca^{2+}$ ion among the alkali-earth metal ions. The selectivity adsorption property toward $Ca^{2+}$ ion was examined in the solution of $Ca^{2+}$ and $Mg^{2+}$ ions, and it was observed that CM-chitin showed excellent selectivity to $Ca^{2+}$ ion than $Mg^{2+}$ ion. $Mg^{2+}$ ion bound to CM-chitin molecule in the presence of $Ca^{2+}$ ion owing to low equilibrium constant. In the adsorption experiment of $Ca^{2+}$ and $Mg^{2+}$ ions to the CM-chitin under coexistence of $Na^+$ and $K^+$ ions, it observed that adsorptivity of only $Ca^{2+}$ ions was not affected by these monovalent cations.

Inhibitory Effect of Extract from Acanthocoris sordidus on Oxidative Damage (꽈리허리노린재(Acanthocoris sordidus) 추출물이 산화적 손상에 미치는 억제 효과)

  • Here, we showed that Acanthocoris sordidus extract inhibited both cell and DNA damage caused by oxidative stress. In a radical scavenging assay, the scavenging activity of the A. sordidus extract against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radicals was 48.9% and 37.8%, respectively, that of ascorbic acid, which was used as a positive control. The ferrous iron chelating activity of the A. sordidus extract was 80.0% compared to that when ethylenediaminetetraacetic acid (EDTA) was used a control. To verify the inhibitory effect of the extract on oxidative cell damage induced by reactive oxygen species (ROS), a lipid peroxidation assay was performed. The results showed that peroxidation was completely inhibited in an extract-treated group compared to a radical-treated group. The level of p21 protein expression was 68.1% that of a control sample. The DNA cleavage-inhibiting property of the A. sordidus extract-treated group was 53.3% that of a control group. Moreover, the phosphorylation of the H2AX protein was reduced to 39.0% of that treated with radical agents, indicating that the extract might inhibit the DNA damage that causes radical oxidation. Taken together, our findings suggest that the A. sordidus extract is effective not only in repressing oxidation by free oxygen radicals and hydroxyl radicals but also in decreasing cell and DNA damage caused by oxidative stress.

Salmonella Typhimurium SL1344 Utilizing Human Transferrin-bound Iron as an Iron Source Regardless of Siderophore-mediated Uptake (Salmonella Typhimurium SL1344의 사람의 트렌스페린(hTf)에 부착된 철 이용에 관한 연구)

  • Inorganic iron is essential for various metabolic processes, including RNA synthesis, electron transport, and oxygen detoxification in microorganisms. Many bacterial pathogens compete for iron acquisition in diverse environmental condition such as host. Salmonella Typhimurium SL1344 also requires inorganic iron as a cofactor for growth. When a M9 minimal liquid medium was supplemented with ethylenediamine di-o-hydroxyphenylactic acid (EDDA) which acts as an iron-chelating agent, growth of Salmonella Typhimurium SL1344 in the supplemented medium was completely arrested by deficient of useful iron under iron-depleted condition. However, a number of siderophores, which are small, high-affinity iron chelating compounds secreted by microorganisms such as bacteria and fungi, were produced for utilization of restricted iron under iron-depleted condition. A M9 minimal liquid medium complemented with human transferrin (hTf)-iron complex turned completely off production of siderophores, but growth of Salmonella Typhimurium SL1344 maintained level similar to compare one complemented with iron (III) chloride (FeCl3). This means that human transferrin (hTf)-bound iron can utilize via directly interaction with Salmonella Typhimurium SL1344 without productions of siderophores. Through construction and analysis of negative mutant for utilization of human transferrin (hTf)-bound iron, we confirm that the bacterium can directly use human transferrin (hTf)-bound iron without extracellularly intermediated carriers such as siderophores.

Study on the Enzyme of Basidiomycetes(I) -The Effects of Iron Ions on the Light-Induced Mitochondrial $F_0F_1-ATPase$ of Lentinus edodes- (담자균류의 효소에 관한 연구(I) -표고버섯 중의 광감응성 Mitochondrial $F_0F_1-ATPase$의 철이온 효과-)

  • The effects of the iron ions for the light-induced mitochondrial $F_0F_1-ATPase$ of Lentinus edodes was studied. The enzyme activity was stimulated up to 202% by 0.1 mM $Fe^{2-}$ ion, but was inhibited by $Fe^{3+}\;and\;Mg^{2+}$. In the presence of 0.5 mM $Mg^{2+}$, the activity also increased 32% by 0.1 mM $Fe^{2+}$ ion, and decreased to a similar extent by $Fe^{3+}$ ion than by only $Fe^{3+}$ ion. Also, the activity was inhibited 53% by 5.0 mM $Fe^{2-}$ ion in the presence of 0.5 mM $Mg^{2+}$ ion and various concentration of $Fe^{3+}$ ion(mM). These results showed that $Fe^{2+}$ strongly stimulated the enzyme activity and its role for the enzyme was independent of $Mg^{2+}$ ion, but was dependent of $Fe^{3+}$ ion. From inactivation of the enzyme by addition of metal chelating agent, EDTA, it is suggested that the enzyme is to be metalloenzyme. The optimal pH and temperature of the enzyme in the presence of 0.1 mM $Fe^{2+}$ was 7.6 and $63^{\circ}C$, respectively.

Anti-Melanogenesis and Anti-Wrinkle Effects of Sargassum micracanthum Extracts (잔가시 모자반 추출물의 주름 개선 및 미백 효과)

  • Sargassum micracanthum (SM) is a member of the family Sargassaceae and commonly occurs along the coast of Korea. Extracts from SM were evaluated for their antioxidant and collagenase and tyrosinase inhibitory activities based on their total phenolic concentration (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect, and metal-chelating effect. The TPC of SM ethanol and water extracts used in the study was 11.20 and 11.70 mg/g of dry sample, respectively. Both SM ethanol and water extracts had high DPPH radical-scavenging effect. The metal-chelating effect of SM ethanol extract (40.47% at 0.5 mg/ml) was higher than that of water extract (23.28% at 0.5 mg/ml). With regard to the anti-wrinkling effect, SM ethanol extract showed collagenase inhibitory activity with an $IC_{50}$ value of $488.20{\mu}g/ml$. Lastly, regarding the anti-melanogenesis effect, SM ethanol extract showed higher tyrosinase inhibitory activity (45.08% at 5 mg/ml) than that shown by the water extract (21.29% at 5 mg/ml). These results suggest that SM has the potential to be a resource with natural anti-melanogenesis, anti-wrinkle, and anti-oxidant effects.

Extraction Characteristics and Antioxidant Activity of Ethanol Extract of Rhus javanica Bark (붉나무 껍질 에탄올 추출물의 추출특성과 항산화 활성에 관한 연구)

  • This study was conducted to investigate the several physicochemical extraction characteristics and the antioxidant activities of ethanol extract from Rhus javanica bark. The contents of soluble solid and sugar in extract was measured to 73.5 mg/100g dry basis and $17.8^{\circ}Brix$, respectively. The contents of total aromatics, total flavonoids and total phenolic compounds was measured to 0.508 in absorbance, 49.88 mg/100g and 296.6 mg/100g, respectively. The reducing power of extract was about 27.5 % of ascorbic acid with the same soluble solid contents of the extract. But the ferric reducing antioxidant power and the DPPH radical scavenging ability of extract were measured to equivalent to those of ascorbic acid. The metal ion chelating ability of the extract was 81.58 % whereas that of ascorbic acid was 74.73 %. The nitrite scavenging ability of the extract was measured to 51.76 % at pH 2. And the antioxidative effect of the extract on soybean oil was observed with Rancimat test.

$Site-Specific^{99m}$Tc-Labeling of Antibody Using Dihydrazinoph-thalazine (DHZ) Conjugation to Fc Region of Heavy Chain

  • The development of an antibody labeling method with $^{99m}$Tc is important for cancer imaging. Most bifunctional chelate methods for $^{99m}$Tc labeling of antibody incorporate a $^{99m}$Tc chelator through a linkage to lysine residue. In the present study, a novel site-specific $^{99m}$Tc labeling method at carbohydrate side chain in the Fc region of 2 antibodies (T101 and rabbit anti-human serum albumin antibody (RPAb)) using dihydrazinophthalazine (DHZ) which has 2 hydrazino groups was developed. The antibodies were oxidized with sodium periodate to pro-duce aldehyde on the Fc region. Then, one hydrazine group of DHZ was conjugated with an aldehyde group of antibody through the formation of a hydrazone. The other hydrazine group was used for labeling with $^{99m}$Tc. The number of conjugated DHZ was 1.7 per antibody. $^{99m}$Tc labeling efficiency was 46-85% for T101 and 67∼87% for RPAb. Indirect labeling with DHZ conjugated antibodies showed higher stability than direct labeling with reduced antibodies. High immunoreactivities were conserved for both indirectly and directly labeled antibodies. A biodistribution study found high blood activity related to directly labeled T1 01 at early time point as well as low liver activity due to indirectly labeled T101 at later time point. However, these findings do not affect practical use. No significantly different biodistribution was observed in the other organs. The research concluded that DHZ can be used as a site-specific bifunctional chelating agent for labeling antibody with $^{99m}$Tc. Moreover, $^{99m}$Tc labeled antibody via DHZ was found to have excellent chemical and biological properties for nuclear medicine imaging.edicine imaging.

Study on the Role of Metal ions for the Activity of the Mitochondrial $F_1-ATPase$ in Lentinus edodes (표고버섯의 Mitochondrial $F_1-ATPase$ 활성도에 미치는 금속이온의 역할에 관한 연구)

  • The role of metal ions for the activity of the mitochondrial $F_1-ATPase$ was studied. Removal of non-heme iron ion from the mitochondria by dialysis against chelating agents, 10 mM ethylenediaminetetraacetic acid(EDTA) and 10 mM o-phenanthroline(o-Phe), led to 56% and 49% inactivation of the enzyme, respectively. The enzyme dialyzed against EDTA was reactivated 81% by the addition of 0.5 mM $Fe^{3+}$ and 70% by 0.5 mM $Mg^{2+}$. But, $Fe^{2+}$ did not reactivate the enzyme. Coexistence of 0.5 mM $Fe^{2+}$ and 0.5 mM $Mg^{2+}$ resulted in 95% reactivation of the enzyme, while $Fe^{3+}$ with 0.5 mM $Mg^{2+}$ did not reactivate the enzyme like the effect of $Fe^{2+}$ alone. The enzyme dialyzed against o-Phe showed the similar results. These data showed that $Fe^{3+}$ is predominantly required for the activity of the mitochondrial $F_1-ATPase$ in Lentinus edodes and stimulated the activity of it by $Mg^{2+}$. $Fe^{3+}$ and $Mg^{2+}$ increased enzyme's affinity for substrate, decreasing the Km value 1.67 mM to 0.65 mM.

Effect of Harvest Date on Antioxidant of Dendranthema zawadskii var. latilobum (Maxim.) Kitam and D. zawadskii var. yezoense (Maek.) Y.M. Lee & H.J. Choi (구절초와 남구절초의 항산화 효과에 미치는 수확시기의 영향)

  • This study was conducted to compare the antioxidative effects of Dendranthema zawadskii var. latilobum (Maxim.) Kitam and D. zawadskii var. yezoense (Maek.) Y.M. Lee & H.J. Choi due to the harvest date. Harvested samples at May 9th (early), July 17th (middle) and September 3rd (late stage) were extracted with 80% ethanol, and biological activities and antioxidant substance contents were detected. The earlier harvested samples showed the higher moisture contents and extraction yields. DPPH radical scavenging effect of early harvested D. zawadskii var. latilobum (Maxim.) Kitam ($RC_{50}$ = $0.128\;mg\;{\cdot}\;mL^{-1}$) was similar to BHT ($0.121\;mg\;{\cdot}\;mL^{-1}$). ABTS radical scavenging effects of both species harvested at early stage were higher than that of ascorbic acid which was well-known natural antioxidant. However, both species harvested at late stage showed the highest $Fe^{2+}$ chelating effect, but it was lower than that of EDTA. Total polyphenol and flavonoid contents of both species were higher when samples were harvested earlier stage. However, both species harvested at early stage contained more total polyphenols (79.93 and $75.10\;mg\;{\cdot}\;g^{-1}$) than flavonoids (57.84 and $54.91\;mg\;{\cdot}\;g^{-1}$).
