Optical Resolution of Dabsyl Amino Acids in Reversed-Phase Liquid Chromatography

  • The dabsylation of amino acids has been applied to resolve their optical isomers with the use of chiral mobile phase in high performance liquid chromatography. The dabsyl amino acids were successfully separated on reversed phase column($C_{18}$) by adding a chiral L-benzylproline-Cu(II) chelate to the mobile phase. The separation selectivity of the dabsyl amino acid enantiomers was not less than that of dansyl amino acids. The retention order of the dabsyl amino acid enantiomers was as those of the dansyl amino acid enantiomers except dabsyl threonine. The optical selectivity of the dabsyl amino acids increase with pH of the mobile phase and concentration of the chelate, but slightly decreases with concentration of buffer and organic solvent composition. However serine, methionine, valine, and leucine showed a slight decrease in the optical selectivity with increase in pH. The retention times of the dabsyl amino acids decreases with increasing pH and acetonitrile concentration but increases with the concentration of the chiral chelate added. The mechanism of the optical resolution is based on a stereospecific interaction including a intramolecular hydrophobic effect and SN-2 reactivity of the ligand exchange chromatography.It is advantageous to detect absorption at 436 nm, which is less interferent them the other detection systems. The derivatized dabsyl amino acids are stable for a month.

Characteristics Evaluation of Radiation Shielding Materials Used Waste Glass and Chelate Resins (폐유리와 킬레이트 수지를 사용한 방사선 차폐재의 재료특성 평가)

  • Various approaches have been attempted to develop recycling technologies related to industrial waste resources containing metals. Among them, glass is not decomposed into microorganisms, so landfill is not suitable, and interest in the recycling of waste glass is increasing. In this paper, by incorporating chelate resin to suppress the elution of heavy metals in waste glass and using waste glass as a fine aggregate and we want to evaluate the strength, drying shrinkage, alkali-silica reaction and heavy metal leaching of shielded filler materials and to provide basic data for utilizing waste glass as an economical and environmentally friendly shielding filler. As a result of the test, it was found that the use of waste glass as a fine aggregate was effective in the development of strength, but the incorporation of chelate resin had an influence on the strength development. In addition, the addition of chelate resin was effective in improving drying shrinkage but it was found to affect the alkali - silica reaction. As a result of the heavy metal leaching test, the KSLP test method satisfies all the criteria for heavy metal leaching. However, in case of lead, the limit of US ANSI 67-2007a was exceeded and further study should be done.

Estimation of Rumen By-pass Rate of Chromium-methionine Chelates by Ruminal Bacteria Analysis (반추미생물 분석에 의한 Chromium-methionine Chelate의 반추위 By-pass율 추정)

  • The study was designed to estimate the in vitro rumen by-pass rate of both chromium methionine chelate as an organic supplement and $ClCl_3$ as an inorganic supplement. Rumen by-pass rates of the supplements were evaluted by comparing ruminal metabolites in rumen fluid and Cr and methionine contents in the body of ruminal microorganism. For in vitro digestion examination, basic nutrients for ruminal microbes were supplied with 7g(DM) of feed, 2g of rice straw, and 2g of corn silage per each incubation jar. Three treatments including Control(no supplementation of Cr), T1(1000ppb supplementation of $ClCl_3$) and T2(chromium methionine chelate supplementation equivalent to 1000ppb of Cr content) were prepared with five replications per each treatment. pH of T2 was lower than that of Control and T1 regardless of incubation time. Ammonia content was higher in T2 than in Control and T1 during first 6 hours of incubation. However, the ammonia content in Control was remained low after 6 hours. Total volatile fatty acids(VFA) content in control was increased constantly as incubation time was extended. Therefore, VFA content in T1 and T2 were significantly lower (P<0.05) than those of Control. Dry matter recovery rate by ruminal microorganism was the lowest in T1, however ruminal microbial population was increased most efficiently in T2 during 12 hours of in vitro incubation. Cr concentrations in the body of ruminal microbes were not different(P>0.05) between Control and T2, but it was significantly high in T1(P<0.05). Contents of methionine and cystine in ruminal microbes also were not different between Control and T2(P>0.05), but it was relatively low in T1. Based on the above results, the chromium methionine chelate was believed to by-pass rumen and could remain intact until it reaches small intestine compared to inorganic chromium. This results implies that chromium methionine chelate could be more effective to function in the small intestine of ruminant animals.

  • Cotton xanthate, which was obtained by treating cotton with carbon disulfide in alkaline solution, was treated with the solution of polyvalent metal ions to produce cotton xanthate-metal chelates. This chelation reaction was readily and simply achieved, and antimicrobial agents with suitable structures could subsequently be coupled to the chelate with ease at moderate pH values and in aqueous solution. Metal ions used in present work include Cu(II), Zn(II) and Fe(III). Tetracycline, streptomycin, neomycin and pyrithion were used as antimicrobial/antifungal agents. Antibacterial activities were measured employing ditch plate method against G(+) Staphylococcus aureus, Streptococus faecalis, and G(-) Escherichia coli, Enterobacter aerogenes, and the fungus, Aspergillus niger. All the cotton xanthate-metal-antimicrobial agent chelates exhibited activities whereas the cotton xanthate-metal chelates themselves were inactive. Considering the extensive washing procedures and results from control experiments, possibility of the involvement of physical adsorption for the binding of drugs could be excluded.

  • We studied on the synthesis of chloromethylated polystyrene as a precursor of optically active polymers for direct resolution of optical isomers. Changing the degree of crosslinking and the kind of crosslinking agents, several polystyrene resin matrices were synthesized. The matrices were chloromethylated with methylal and chlorosulfonic acid as chloromethylating agents. The effects of solvents of various dielectric constants on the chloromethylation were quantitavely examined. We also synthesized chloromethylated polystyrene of macroreticular type that retained large surface area and good physical stability. The differences between the macroreticular type and macroporous type were investigated.

  • We examined effects of crosslinking agents, i.e., ethyleneglycol dimethacrylate (EGD) and butanediol dimethacrylate (BDD) containing ester groups on chloromethylation of crosslinked polystyrene resin matrices. It was proved that the ester group in methylmethacrylate (MMA) accelerates the chloromethylation of the divinylbenzene (DVB)-crosslinked styrene-MMA copolymer. As the MMA content increased in the styrene-MMA copolymers, the chloromethylation was enhanced. Complete chloromethylation was obtained at about 25% MMA content.

  • The purpose of this study is to investigate the feasibility of applying the chitosan for a natural mordant. The chitosan, a natural chelate macromolecule, is acquired from the crustacean. Investigation was focused on the change of the dyeability according to the chitosan treatment based on the Caesalpinia sappan and the color change of the dyed fabric according to the application of heavy metal mordant. The change of air permeability of the fabric with the dyeing condition was also investigated systematically. The effect of dyeing with/without mordant on the air permeability of the fabric after the chitosan treatment. It seems that the increase in the dye-uptake is attributable to the fact that chitosan forms a complex with the Caesalpinia sappan and Al during mordant dyeing of cotton fabric. For nylon fabric, the darkest color was achieved on the occasion of non-mordant dyeing of the fabric. The mordant treatment or chitosan treatment, however, reduced the dyeability. While the dyed nylon fabric could maintain the same air permeability as the grey nylon fabric, the cotton fabric lowered the air permeability after dyeing.

Performance of Synthesized Metal Chelate Complex of Tris- (8-hydroxyquinoline) Aluminum $(Alq_3)$ in Fabrication of OLEDs

  • The metal chelate complex of aluminum 8-hydroxyquinolines $(Alq_3)$ is an important electro-luminescent (EL) material used in fabricating organic light emitting (OLEDs). In this work, diodes / displays using in house synthesized $Alq_3$ and simple structure have been fabricated. It is demonstrated that luminance of $12460\;cd/m^2$ and efficiency of 3.1 cd/A are achievable in these fluorescent devices. Possible degradation effects are analyzed.
