This paper introduces page-level rewrite interval prediction (PRWIP). By recording and analyzing the memory access history at page-level, we are able to predict the future memory accesses to any pages. Leveraging this information, this paper proposes a faster incremental checkpoint design by overlapping checkpoint phase with computing phase and thus achieves higher performance. Experimental results show that our new incremental checkpoint design can achieve averagely 22% speedup over traditional incremental checkpoint and 14% over the previous state-of-the-art work.

For a system with multiple real-time tasks of different deadlines, it is very difficult to find the optimal checkpoint interval because of the complexity in considering the scheduling of tasks. In this paper, we determine the optimal checkpoint interval for multiple real-time tasks that are scheduled by RM(Rate Monotonic) algorithm. Faults are assumed to occur with Poisson distribution. Checkpoints are inserted in the execution of task with equal distance in the same task, but different distances in other tasks. When faults occur, rollback to the latest checkpoint and re-execute task after the checkpoint. We derive the equation of maximum slack time for each task, and determine the number of re-executable checkpoint intervals for fault recovery. The equation to check the schedulibility of tasks is also derived. Based on these equations, we find the probability of all tasks executed within their deadlines successfully. Checkpoint intervals which make the probability maximum is the optimal.

Defects in the mitotic spindle or in the attachment of chromosomes to the spindle are believed to release an activated form of spindle checkpoint complex that inhibits APC-dependent ubiquitination and subsequently arrests the cell cycle at metaphase. When the spindle assembly is disrupted, the fission yeast mitotic arrest deficient (mad) mutants fail to arrest and rapidly lose viability. To enhance our understanding of the molecular mechanisms for the pathway of checkpoint function, the functional characterizations of Mad 1 p from Schizosaccharomyces pombe involved in this process have been carried out. Yeast two-hybrid and various deletion analyses of S. pombe Mad1 p reveal that the C terminus of Mad1p is critical for the binding of Mad2p and maintenance of Mad 1 p-Mad2p interaction. In addition, it was found. that the Mad1p region (residues 206-356) is essential for Mad1p-other checkpoint components. Mad1p truncating this region is sufficient to bind Mad2p but abolishes the checkpoint function, indicating that the checkpoint function is necessary for interaction of Mad 1 p-other checkpoint components. The possible functions of S. pombe Mad1p at the cell cycle checkpoint are discussed.

It is not practically feasible to apply hardware-based fault-tolerant schemes, such as hardware replication, in mobile devices. Therefore, software-based fault-tolerance techniques, such as checkpoint and rollback schemes, are required. In checkpoint and rollback schemes, the optimal checkpoint interval should be applied to obtain the best performance. Most previous studies focused on minimizing the expected execution time or response time for completing a given task. Currently, most mobile applications run in real-time environments. Therefore, it is extremely essential for mobile devices to employ optimal checkpoint intervals as determined by the real-time constraints of tasks. In this study, we tackle the problem of determining the optimal inter-checkpoint interval of checkpoint and rollback schemes to maximize the deadline meet ratio in real-time systems and to build a probabilistic cost model. From this cost model, we can numerically find the optimal checkpoint interval using mathematical tools. The performance of the proposed solution is evaluated using analytical estimates.
Immune checkpoint blockades including monoclonal antibodies (mAbs) of cytotoxic T-lymphocyte antigen-4 (CTLA-4), programmed death-1 (PD-1), and programmed death-ligand 1 (PD-L1) have been emerged as a promising anticancer therapy. Several immune checkpoint blockades have been approved by US Food and Drug Administration (FDA), and have shown notable success in clinical trials for patients with advanced melanoma and non-small cell lung cancer. Radiotherapy is a promising combination partner of immune checkpoint blockades due to its potent pro-immune effect. This review will cover the current issue and the future perspectives for combined with radiotherapy and immune checkpoint blockades based upon the available preclinical and clinical data.
Cancer growth and progression are associated with immune suppression. Cancer cells have the ability to activate different immune checkpoint pathways that harbor immunosuppressive functions. Monoclonal antibodies that target immune checkpoints provided an immense breakthrough in cancer therapeutics. Among the immune checkpoint inhibitors, PD-1/PD-L1 and CTLA-4 inhibitors showed promising therapeutic outcomes, and some have been approved for certain cancer treatments, while others are under clinical trials. Recent reports have shown that patients with various malignancies benefit from immune checkpoint inhibitor treatment. However, mainstream initiation of immune checkpoint therapy to treat cancers is obstructed by the low response rate and immune-related adverse events in some cancer patients. This has given rise to the need for developing sets of biomarkers that predict the response to immune checkpoint blockade and immune-related adverse events. In this review, we discuss different predictive biomarkers for anti-PD-1/PD-L1 and anti-CTLA-4 inhibitors, including immune cells, PD-L1 overexpression, neoantigens, and genetic and epigenetic signatures. Potential approaches for further developing highly reliable predictive biomarkers should facilitate patient selection for and decision-making related to immune checkpoint inhibitor-based therapies.
Spindle position checkpoint monitors the orientation of mitotic spindle for proper segregation of replicated chromosomes into mother cell and the daughter, and prohibits mitotic exit when mitotic spindle is misaligned. BUB2 forms one of the key upstream element of spindle position checkpoint in budding yeast, but its functional homologues have not been identified in higher eukaryotes. Here, we analyzed the functions of two putative BUB2 homologues of C. elegans in the spindle orientation checkpoint. From the C. elegans genome database, we found that two open reading frames (ORFs), F35H12_2 and C33F10_2, showed high sequence homology with BUB2. We obtained the expressed sequence tag (EST) clones for F35H12_2 (yk221d4) and C33F10_2 (yk14e10) and verified the full cDNA for each ORF by sequencing and 5' RACE with SL1 primer. The functional complementation assays of yk221d4 and yk14e10 in ${\Delta}bub2$ of S. cerevisiae revealed that these putative BUB2 homologues of C. elegans could not replace the function of BUB2 in spindle position checkpoint and mitotic exit. Our attempt to document the component of spindle position checkpoint in metazoans using sequence homology was not successful. This suggests that structural information about its components might be required to identify functional homologues of the spindle position checkpoint in higher eukaryotes.
The DPB11 gene, which genetically interacts with DNA polymerase II ($\varepsilon$) encoded by POL2 and DPB2, is required for DNA replication and the S phase checkpoint in Saccharomyces cerevusiae. The transcripts of DPB11 did not fluctuated as evidently as DPB2 and POL2 genes during cell cycle. To identify the physical interaction between Dpb2 and Dpb11, we examined the interaction by two-hybrid system. The interaction between Dpb2 and Dpb11 was detected in a two-hybrid assay. These results suggest that the amount of the Dpb2-Dpb11 complex may peak at the G1/S boundary. Therefore, we strongly suggest the involvement of the Dpb2-Dpb11 complex in a progression of DNA replication and S-phase checkpoint.

We analyze checkpoint strategy for multiple real-time periodic tasks with hard deadlines. Real-time tasks usually have deadlines associated with them. For multiple real-time tasks, checkpoint strategy considering deadlines of all tasks is very difficult to derive. We analyze the problem of checkpoint placement for such multiple periodic tasks. In our strategy, the interval between checkpoints is determined for each task considering its deadline. An approximated failure probability over a specified interval is derived. Then the number of checkpoints for each task is selected to minimize the approximated failure probability. To show the usefulness of our strategy, error bound between the exact and the approximated failure probability is estimated, which is revealed to be quite small.
Immune checkpoint inhibitor is associated with variety of immune-related adverse events. We present a case of polyneuropathy induced by immune checkpoint inhibitor, which was refractory to steroid and immunoglobulin. While high-dose steroid and immunoglobulin were not effective, we tried rituximab which is effective in other immune-mediated polyneuropathy. After rituximab treatment, patient's clinical symptom and nerve conduction study finding was markedly improved. We suggest rituximab might be effective in polyneuropathy induced by immune checkpoint inhibitor.
