Reduction of Microflora in the Manufacture of Saengshik by Hygienic Processing

  • This study was conducted to determine the effect of hygienic processing (HP) on the reduction of microorganisms during manufacturing of saengshik with two vegetables (carrots and cabbage) and two grains (barely and glutinous rice) compared to general processing (GP). For GP, distilled water was used for washing raw materials and equipment. For HP, aqueous ozone (3 ppm) in combination with 1% citric acid and 70% alcohol were used for washing raw materials and the equipment, respectively. In carrots, after cutting, total aerobic bacteria (TAB), yeast and mold (YM) and coliforms were significantly increased to 5.19, 8.04 and 2.08 ($log_{10}$ CFU/g), respectively (p<0.05). Washing effectively reduced the increased microorganisms from cross contamination during cutting, but cross contamination increased with subsequent GP drying and milling procedures to 8.56, 8.27 and 3.71 ($log_{10}$ CFU/g) for TAB, YM and coliforms, respectively (p<0.05). On the other hand, HP washing of carrots with 3 ppm ozone in combination with 1% citric acid showed higher antimicrobial effect than GP washing, significantly decreasing the number of microorganisms (p<0.05). Further cross contamination did not occur through drying and milling due to cleaning the equipments with 70% alcohol prior to processing. After milling, the number of TAB, YM and coliforms were significantly decreased to 3.89, 4.47 and not detectable level ($log_{10}$ CFU/g), respectively (p<0.05). Similar results were observed in cabbage and grains. During storage for two months at different temperatures (22 or $4^{\circ}C$), there were no changes in numbers of spoilage microorganisms in the packaged saengsik after either processing. This suggests the importance of HP for the reduction of microorganisms during saengsik production, and demonstrates the effectiveness of disinfection at each processing stage in minimizing contamination levels to enhance microbial safety of saengshik products.

Changes in Physicochemical Properties and by the Aging Conditions of Garlic (숙성 조건 변화에 의한 흑마늘의 이화학적 특성변화)

  • When garlic is aged for a certain period of time while maintaining proper humidity, it causes a browning reaction by its own ingredients and enzymes. In this study, the physicochemical properties were analyzed by varying the temperature conditions for aging garlic. It was found that about 20% of the weight of garlic was reduced through the aging process. During the aging process, both samples showed the highest water content on the 10th day of aging, and the water content of the two samples was similar after the aging was completed. In the analysis of total sugar content change, AG1 (22.0±0.7 vs 21.4±0.6) showed a significantly higher sugar content on the 10th day of aging, but showed a tendency to gradually decrease thereafter, whereas AG2 showed a steady increase in sugar content during the aging process (18.7 < 21.4 < 21.7 < 22.3 < 23.7). The pH change that occurred during the aging process showed the most pH change on the 10th day. In AG1, the pH gradually decreased as the aging period increased, whereas in AG2, the pH increased significantly after the 10th day. In the analysis of browning, AG1 increased sharply on the 10th day and maintained the state, and AG2 showed a significant increase in browning according to the aging process. In conclusion, based on the results of this study, if various ripening temperature conditions are set, it is judged that it will play a positive role in improving the productivity of black garlic.

Prediction of the Environmental Conditions in Underground Tunnelling Spaces (地下터널 굴착作業場內 作業環境豫測)

  • A comprehensive, nonsteady state, computer simulation program for the environmental conditions in advancing tunnels (the HEADSIM simulation program) is constructed and successfully validated with heat balance amongst all heat sources, and with mass conservation amongst various airflows including the leakage air from ducts, under timedependent variations of inlet air conditions. which include sudden, diurnal and seasonal changes. Heat conduction in the wall strata and face strata is simulated with most complicated boundary conditions using the finite difference method, and the climatic conditions in roadway sections which contain air ducts, booster fan, spray cooler, compressed air pipes, cold water pipes, return water pipes, machinery and broken rock are simulated taking into account the variations of face operation and the heat storage mechanism in the strata. The limitations of simulation time steps and roadway section lengths are defined according to the stability criteria satisfying the principles of thermodynamics. Variations of heat transfer coefficients, which are newly set, and those of wetness factors are taken into account according to the variations of other parameters and the stepwise advance of the face. Newly-derived formulae are used for computing the air duct leakage and the pressure inside of the duct. A new concept of an 'imaginary duct' is introduced to simulate the climatic conditions in tunnels during holiday periods, which directly affect conditions on subsequent working days under the consideration of natural convection. A subsidiary program (the WALLSIM simulation program) is made to compute the dimensionless tunnel surface temperatures and to compare the results with those from analytical approaches, and to demonstrate the stability, convergence and accuracy of the strata heat conduction simulation, adopting the finite difference method. The WALLSIM also has wide applications, including those for the computation of age coefficients.

The Spreading Characteristics of the (Polymer/18-Crown-6) Composite Solution for Water Cast-Membranes ((Polymer/18-Crown-6)복합박막 제조용액의 수면전개 특성)

  • The (polymer/carrier) composite membranes for separating the metallic ion have been prepared by the water cast-method. The morphology of these membranes was affected by the physical properties of the spreading solution. The surface tension of the (polymer/18-crown-6) composite solution was decreased with increasing the concentration of 18-crown-6 compound and the surface tension of polymer solution decreased the following order PVC>PS>CA. The viscosity of CA solution decreased with increasing the contents of the 18-crown-6 compound, but PVC and PS solutions showed no significance changes according as the concentration of 18-crown-6 compound. In the composite solutions, the spreading ability was improved by' the cyclic 18-crown-6 molecules which acted as an electric buffer and diminished the intermolecular force between the polymer chains. The (polymer/18-crown-6) composite membrane was more uniform than that of the mono polymer membrane on the coagulation state of polymer, and the top and bottom sides of membrane showed also the more smooth structure according as the concentration increment of 18-crown-6 molecule.

Structure and Succession of Zooplankton Community in Several Artificial Lakes in the Han River System (한강 수계 주요 댐호에서의 동물플랑크톤 군집 구조와 천이)

  • Structure and succession of zooplankton community studied by hydraulic and ecological characteristics targeting the five lakes in the Han river system from March to December 2008. Results separated by river-type lake and lake-type lake depending on the type of hydraulic, Paldang lake and Cheongpyeong lake were river-type lake, while Chungju lake, Hoengseong lake and Doam lake was lake-type lake. The Paldang lake was a eutrophic lake, zooplankton community density and species number were the most among the five lakes. Relative dominance of the rotifera was the largest and the yearly first dominant species was a small cladocera Bosmina longirostris. The Cheongpyeong lake was a mesotrophic-eutrophic lake, hydraulic characteristics and zooplankton community changes were similar the Paldang lake. Relative dominance of the cladocera was the largest and the yearly first dominant species was a small cladocera Bosmina longirostris. The Chungju lake was a oligotrophic-mesotrophic lake, zooplankton community density was the least among the five lakes. Relative dominance of the copepoda was the largest and the yearly first dominant species was a large cladocera Daphnia galeata. The Hoengseong lake was a oligotrophic-mesotrophic lake, relative dominance of the rotifera was the largest and the yearly first dominant species was a small cladocera Bosmina longirostris. The Doam lake was a mesotrophic-eutrophic lake, zooplankton community density showed dramatic difference at the investigation time. Relative dominance of the rotifera was the largest and the yearly first dominant species was the copepoda Nauplius.

Analysis of the Changes in Rainfall Quantile according to the Increase of Data Period (자료기간 증가에 따른 확률강우량의 거동특성 분석)

  • To account for the influence of heavy storm in Korea by climate change like global warming, the frequency analyses for annual maximum rainfall sequence in 12 rainfall gauge stations are carried out. In order to analyze the temporal change, the rainfall quantile of each station is estimated by the 30-yr data period being moved from 1954 to 1998 with 1-yr lag. Through the analysis for l00-yr rainfall quantile it has been shown that the recent heavy storms increase comparing with storms in the past. From the additional estimating of the rainfall quantile of each station by the 30-yr data period being cumulated from 1954 to 1998 with 1-yr, the change of the probable rainfall by including the heavy storm duration is realized. When the hydraulic structures are determined, it is important to select the data size and necessary to reestimate the flood prevention capacity in existing river systems.ystems.

Monolayer Characteristics of Bilayer Forming Phosphate Amphiphiles (이분자막 형성능을 가지는 인산형 양친매성 화합물의 단분자막 특성)

  • The monolayer characteristics of phosphate amphiphiles with azobenzene at air/water interface were studied by the measurment of $\pi-A$ curves and absorption spectra. Immediately after being spread on the water surface, these amphiphiles having strong intermolecular hydrogen bonding interactions showed the typical absorption spectra which resulted from domain formation. But the aggregated domains could be controlled by changing the subphase conditions (adding bulky salt and rasing pH). Addition of metal ions in subphase changes the molecular orientation of monolayer. As the metal ion charge increases ($1\leq2$ < 3 < 4 valence), the absorption maximum (310nm) of the amphiphile with azobenzene shifts to a longer wavelength (350nm) which means that the orientation of the amphiphile is tilted. These results suggest that the molecular orientation, and furthermore the aggregation state of monolayer can be possibly controlled by the interaction of metal ions with different charge types.

Optimal Treatment of Molasses Wastewater Using UASB Process (UASB 공정에 의한 당밀폐수의 최적처리 방안)

  • The purpose of this study is to get optimum operating factors of Upflow Anaerobic Sludge Blanket (UASB) reactor by introducing methods that make it to reduce inhibition possible in each process wastewater treatment. The used substrates, concentrated corn starch liquid (CSL) wastewater, modified starch, filtering and decoloring wastewater, ion refining wastewater, and mixed wastewater including modified starch and not including modified starch, are generated from molasses process. The seeding sludge is the digested sludge that had been applied to molasses wastewater. Batch test to reduce the inhibition factors that might be existed in each wastewater was examined. Based on the this test, the optimum operating factors according to alkalinity and pH variation was studied through the continuous test using three 5.5 L UASB reactor. The first reactor added $NaHCO_3$ to control alkalinity. The hydraulic retention time (HRT) reduced to 8 hours and the organic loading rate increased gradually. The second reactor changed the pH of influent from 7.0 to 6.0 using NaOH. The third reactor was operated without changes to compare the above two reactors. As the result, the inhibition in concentrated CSL wastewater was removed by adding iron (II). When trace metals were added to mixed wastewater not including modified starch, the digestability by gas production rate increased to more fifty percentage than mixed wastewater that was not adding the trace metals. The reason that the inhibition did not decreased in spite of adding trace metals and nutrients was influenced by high concentration generated during the acid fermentation. The UASB reactors using the mixed wastewater with the most effective performance were operated as 500 mg/L as $CaCO_3$ alkalinity and 6.0 pH at steady state, and at this time, the gas production rates were 283 and 311mL gas/g $COD_{added}$. The COD removal rates were 84.7 and 86.3%, respectively.

Effects of Soaansintang(SOAT) on the hemodynamics and electrocardiogram of isolated rat hearts induced by electrical stimulation (소아안신탕(小兒安神湯)이 STRESS를 유발한 흰쥐의 적출심장(摘出心臟)에 미치는 영향)

  • It has long been known that SOAT is effective for sudden palpitation occurring unexpectedly in Oriental Medicine. However, effect of SOAT on the isolated heart has not been studied yet. The purpose of this study is to investigate the effect of SOAT on hemodynamics and ECG of isolated rat hearts induced by electrical stimulation using Langendorff perfusion apparatus for nonworking heart. SOAT extract was manufactured by water-alcohol precipitated method. Sprague-Dawley rats weighting $120{\sim}150g$ were used for the experiments, Subject animals were divided into four groups, which are consisted of 1) control(Group orally administered by normal saline 1ml for 14days), 2) sample A(Group orally administered by SOAT extract 1ml for 14days), 3) sample C(Group injected by SOAT extract 0.5ml after stimulation, 4) sample C(Group injected by SOAT extract 1ml after stimulation. To evluate the effects of SOAT on hemodynamics and ECG of isolated rat heart induced by stimulation, heart rate, left ventricular pressure, systolic power, diastolic power, coronary artery perfusion volume and ECG were measured using Langendorff apparatus in both stimulation mode(5 volts, 450 beats/min) and arrythmic mode(5 volts, 420 beats/min including 60 beats/min) The results obtained are as follows : 1. After receiving stressful electrical stimuli, isolated heart showed the heart rate, left ventricular pressure, systolic power, diastolic power, coronary artery perfusion volume were all decreased temporarily, but perfusion continued longer recovery to the control state appeared. However, the coronary artery perfusion volume diminished continuously. 2. The heart rates did not change significantly with both stimulation mode and arrhythmic mode, among experimental groups. 3. The left ventricular pressure showed with both stimulation mode and arrhythmic mode, the significant changes(p<0.05) especially in the injection sample group. In case of stimulation mode, low concentration injection group(0.5ml) was more significantly increased rather than high concentration group(1ml) and in case of arrhythmic mode, high density group(1ml) was so increased than the other(0.5ml). 4. For the systolic power and diastolic power, no significant changes were noticed in the stimulation mode, but in the arrhythmic mode of injection sample groups, significant change(p<0.05) was noticed in both systolic power and diastolic power. Specially the high concentration group(1ml) showed more significant increase than the low concentration group. 5. For the coronary artery perfusion volume, no significant change difference among sample groups was observed in both the stimulation mode and the arrhythmic mode. 6. For the ECG recordings, arrhythmia was induced by electrical stimulus of arrythmia mode and after the stimulus was removed, irregular wave appeared temporarily, but as perpusion continued, recovery to the control state was abtained like the stimulation mode. According to the above results, SOAT significantly changed the hemodynamic data from the electrically stressed, isolated hearts of connected Langendorff perfusion apparatus and we propose SOAT has the direct effects on the muscular function of heart.

Experimental Studies on the Compressive Strength of the Frozen Soils (동결토의 압축강도에 관한 실험적 연구)

  • Upon freezing a soil swells due to phase change and its compression stress increase a lot. As the soil undergo thawing, however, it becomes a soft soil layer because the 'soil changes from a solid state to a plastic state. These changes are largely dependent on freezing temperature and repeated freezing-thawing cycle as well as the density of the soil and applied loading condition. This study was initiated to describe the effect of the freezing temperature and repeated freezing-thawing cycle on the unconfined compressive strength. Soil samples were collected at about 20 sites where soil structures were installed in Kangwon provincial area and necessary laboratory tests were conducted. The results could be used to help manage effectively the field structures and can be used as a basic data for designing and constructing new projects in the future. The results were as follows ; 1. Unconfined compressive strength decreased as the number of freezing and thawing cycle went up. But the strength increased as compression speed, water content and temperature decreased. The largest effect on the strength was observed at the first freezing and thawing cycle. 2. Compression strain went up with the increase of deformation speed, and was largely influenced by the number of the freezing-thawing cycle. 3. Secant modulus was responded sensitivefy to the material of the loading plates, increased with decrease of temperature down to - -10$^{\circ}$C, but was nearly constant below the temperature. Thixotropic ratio characteristic became large as compression strain got smaller and was significantly larger in the controlled soil than in the soil treated with freezing and thawing processes 4. Vertical compression strength of ice crystal(development direction) was 3 to 4 times larger than that of perpendicular to the crystal. The vertical compression strength was agreed well with Clausius-Clapeyrons equation when temperature were between 0 to 5C$^{\circ}$, but the strength below - 5$^{\circ}$C were different from the equation and showed a strong dependency on temperature and deformation speed. When the skew was less then 20 degrees, the vertical compression strength was gradually decreased but when the skew was higher than that, the strength became nearly constant. Almost all samples showed ductile failure. As considered above, strength reduction of the soil due to cyclic freezing-thawing prosses must be considered when trenching and cutting the soil to construct soil structures if the soil is likely subject to the processes. Especially, if a soil no freezing-thawing history, cares for the strength reduction must be given before any design or construction works begin. It is suggested that special design and construction techniques for the strength reduction be developed.
