Influence of N Fertilization Level, Rainfall, and Temperature on the Emission of N2O in the Jeju Black Volcanic Ash Soil with Carrot Cultivation (당근 재배 화산회토양에서 질소시비 수준 및 강우, 온도 환경 변화에 따른 N2O 배출 특성)

  • This study was conducted to obtain investigated characteristic factors which has an influence on nitrous oxide ($N_2O$) emissions related to the environment change of nitrogen application level, rainfall and temperature. It was done by the carrot cultivation at black volcanic ashes soil in the experimental field of Jeju Special Self-governing Province Agricultural Research and Extension Services from 2010 to 2011. During the carrot cultivation period, the more amount of nitrogen fertilizer applied, the more amount of $N_2O$ emissions were released. Generally $N_2O$ emissions were so deeply released to climate as that in the first and middle of cultivation with heavy rainfall released amount is high, otherwise it was released very low at the end of cultivation and drought season. $N_2O$ emissions type was considered to relate with the rainfall pattern and soil water content. We obtained the result correlated with $N_2O$ emissions, in 2010, as the soil water and soil temperature were significant to $0.5718^{**}$ ($r$) and $0.4908^{**}$ ($r$) respectively, but soil EC was not significant to 0.2704 ($r$). In 2011, soil water was significant to $0.3394^*$ ($r$), but soil temperature and soil EC were not significant to 0.2138 ($r$) and 0.2462 ($r$) respectively. Also, $NO_3$-N and soil nitrogen ($NO_3-N+NH_4-N$) were not significant to 0.0575 ($r$) and 0.0787 ($r$) respectively. During the carrot cultivation period, the average emissions factor released by the nitrogen fertilizer application for 2 years was presumed to be 0.0025 $N_2O$-N kg / N kg. This factor was 4 times than the IPCC (0.0100 $N_2O$-N kg / N kg) factor.

  • This study was conducted to investigate the characteristic factors which have been influenced on nitrous oxide ($N_2O$) emissions related to the environment change of nitrogen application level, rainfall and temperature during the soybean cultivation at black volcanic ash soil from 2010 to 2011. During the soybean cultivation, the more amount of nitrogen fertilizer applied, $N_2O$ emissions amounts were released much. $N_2O$ emissions with the cultivation time were released much at the first and middle of cultivation with heavy rainfall, but it was released very low until the end of cultivation and drought season. $N_2O$ emissions mainly were influenced by the rainfall and soil water content. The correlation ($r$) with $N_2O$ emissions, soil water, soil temperature and soil EC in 2010 were very significant at $0.4591^{**}$, $0.6312^{**}$ and $0.3691^{**}$ respectively. In 2011, soil water was very significant at $0.4821^{**}$, but soil temperature and soil EC were not significant at 0.1646 and 0.1543 respectively. Also, $NO_3$-N and soil nitrogen ($NO_3-N+NO_4-N$) were very significant at $0.6902^{**}$ and $0.6277^*$ respectively, but $NO_4$-N was not significant at 0.1775. During the soybean cultivation, the average emissions factor of 2 years released by the nitrogen fertilizer application was presumed to be 0.0202 ($N_2O$-N kg $N^{-1}\;kg^{-1}$). This factor was higher about 2.8 and 2 times than the Japan's (0.0073 $N_2O$-N kg $N^{-1}\;kg^{-1}$) value and 2006 IPCC guideline default value (0.0100 $N_2O$-N kg $N^{-1}\;kg^{-1}$) respectively.

Study on Comparison of Ingredient Quantities and Biological Activities of Galgeun-tang according to Extraction Solvent (용매별 갈근탕 추출물의 성분 분석 및 효능 비교)

  • To investigate the difference and change of ingredient and efficacy of Galgeun-tang (GGT) according to extraction solvent, water and 70% EtOH, the quantities of index components and the several in vitro activities of two kinds of GGT extract were compared. The contents of extracts were analyzed with HPLC. The biological activities such as anti-inflammatory and anti-obesity effects were measured through cell line-based in vitro assay. The cytotoxicity was measured by CCK-8 assay in RAW 264.7 cell, BEAS-2B cell, HaCaT cell and 3T3-L1 cell. We compared effects of two kinds of extract by measuring NO, PGE2, IL-6 and TNF-${\alpha}$ in RAW264.7 cell, RANTES in BEAS-2B cell, MDC and RANTES in HaCaT cell and GPDH activity and leptin level in differentiated 3T3-L1. 70% EtOH extract of GGT contained more compositions than water extract. The inhibitory effect of water extract of GGT on NO in RAW 264.7 cell and GPDH activity in 3T3-L1 was stronger than that of 70% EtOH. 70% EtOH has a stronger inhibitory effect on PGE2 in RAW264.7 cell, RANTES in BEAS-2B cell, MDC, RANTES in HaCaT cell, leptin in 3T3-L1 cell than water extraction. These results suggest that the ingredient and efficacy from 70% EtOH extract of GGT are more effective than water extract.

Occurrence of Diatom in the Late Quaternary Sediments of the Northeastern East Sea (Sea of Japan) and its Paleoceanographic Changes (동해 북동부해역 제 4기 후기 퇴적물의 규조 산출과 고해양학적 변화)

  • A total of 50 diatom species and 1 subspecies belonging to 31 genera except Chaetoceros resting spores were identified in the 45 sediments subsampled from a gravity core GH98-1223 collected from the western Hokkaido Island located in the northeastern East Sea (Sea of Japan). The most dominant species is Thalassionema nitzschioides (Grunow) Hustedt, ranging 29 to 59% of the total assemblages, and most species including Denticulopsis seminae (Simonsen and Kanaya) Simonsen and Pseudoeunotia doliolus (Wallich) Grunow were less than 5% in average. Frequencies of cold-water species are generally higher than those of warm-water species and the vertical distribution of cold-water species was largely opposite to that of warm-water species in spite of ecological habitat difference. Frequency of cold-water species, D. seminae is reverse to that of P. doliolus, an indicator of the Tsushima Warm Current, which is consistent with diatom temperature value (T$_{d}$ value). The variation of T$_{d}$ values shows that the upper part of core with greater-than-average T$_{d}$ values represents postglacial warming trend. These T$_{d}$ values clearly demonstrate that the study area located in the northern part of the East Sea is gradually influenced by Tsushima Warm Current. In addition, the zig-zag variation in the lower part reflects the unstable seawater for diatom habitat. Chaetoceros resting spores indicating productivity and upwelling was 5.3 to 40%, with maximum peak at 80 cm. Chaetoceros resting spores/Chaetoceros vegetative cells, an indicator of relative amounts of biogenic material in the sediments was high at the upper 80 cm level, corresponding to the change of T$_{d}$ values. On the basis of diatom assemblages, the northeastern part of East Sea has experienced the effects of Tsushima Warm Current during the postglacial period of Holocene, which is similar to the modem climatic environment. However, the variation of P. doliolus reflects that the intensity of Tsushima Warm Current has been oscillated in the East Sea.

Migration of the Dokdo Cold Eddy in the East Sea (동해 독도 냉수성 소용돌이의 이동 특성)

  • The cold eddies around the Ulleung Basin in the East Sea were identified from satellite altimeter sea level data using the Winding-Angle method from 1993 to 2015. Among the cold eddies, the Dokdo Cold Eddies (DCEs), which were formed at the first meandering trough of the East Korea Warm Current (EKWC) and were pinched off to the southwest from the eastward flow, were classified and their migration patterns were analyzed. The vertical structures of water temperature, salinity, and flow velocity near the DCE center were also examined using numerical simulation and observation data provided by the Hybrid Coordinate Ocean Model and the National Institute of Fisheries Science, respectively. A total of 112 DCEs were generated for 23 years. Of these, 39 DCEs migrated westward and arrived off the east coast of Korea. The average travel distance was 250.9 km, the average lifespan was 93 days, and the average travel speed was 3.5 cm/s. The other 73 DCEs had moved to the east or had hovered around the generated location until they disappeared. At 50-100 m depth under the DCE, water temperature and salinity (T < $5^{\circ}C$, S < 34.1) were lower than those of ambient water and isotherms made a dome shape. Current faster than 10 cm/s circulates counterclockwise from the surface to 300 m depth at 38 km away from the center of DCE. After the EKWC separates from the coast, it flows eastward and starts to meander near Ulleungdo. The first trough of the meander in the east of Ulleungdo is pushed deep into the southwest and forms a cold eddy (DCE), which is shed from the meander in the south of Ulleungdo. While a DCE moves westward, it circumvents the Ulleung Warm Eddy (UWE) clockwise and follows U shape path toward the east coast of Korea. When the DCE arrives near the coast, the EKWC separates from the coast at the south of DCE and circumvents the DCE. As the DCE near the coast weakens and extinguishes about 30 days later after the arrival, the EKWC flows northward along the coast recovering its original path. The DCE steadily transports heat and salt from the north to the south, which helps to form a cold water region in the southwest of the Ulleung Basin and brings positive vorticity to change the separation latitude and path of the EKWC. Some of the DCEs moving to the west were merged into a coastal cold eddy to form a wide cold water region in the west of Ulleung Basin and to create a elongated anticlockwise circulation, which separated the UWE in the north from the EKWC in the south.

Residual Amount of Herbicide Dimepiperate and Hydroxy Dimepiperate in Surface Water and Leachate (제초제(除草劑) Dimepiperate와 그 대사물(代謝物)의 지표수(地表水) 및 침투수중(浸透水中) 잔류량(殘留量)의 경시적(經時的) 변화(變化))

  • This experiment was conducted to determine the residual amount of dimepiperate (S-1-methyl-1-phenylethyl piperidine-1-carbothioate) and dimepiperate-OH(S-1-methyl-1-phenylethyl-4-hydroxy piperidine-1-carbothioate) in surface water and leachate using a lysimeter system. In surface water, the concentration of dimepiperate was 851-897 ppb in the treatment of 3.0 Kg prod/10a. 1,755-1,781 ppb in the treatment of 6.0 Kg prod/10a 1 day after the treatment. The amount of dimepiperate gradually decreased and was infinitesimal from 35 days after the treatment. In leachate dimepiperate was detected from the 45 days, but its concentration was very low, below 1 ppb, compared to that in surface water. The metabolite of dimepiperate, dimepiperate-OH in surface water was detected after the application of dimepiperate. The highest level of 88-173 ppb occurred 2 day after the treatment of dimepiperate and then decreased gradually. The residual concentration was below 2.13 ppb during 28-63 days. In leachate, the concentration of dimepiperate-OH in the different treatments in the range of 0.5-11 ppb from the 21 days, although each treatment showed different detected date and concentration. The detected concentration showed a little change until 63 days.

Heat Shock Treatments Induce the Accumulation of Phytochemicals in Kale Sprouts (열처리에 의한 케일 새싹의 기능성물질 축적)

  • The objective of this study was to determine the effect of heat shock treatments on the phytochemicals including antioxidants and anticancer materials in kale (Brassica oleracea L. var. acephala) sprouts. In study I, kale sprouts grown under the growing system for four days were soaked at 40, 50, or $60^{\circ}C$ distilled water for 10, 30, or 60 seconds, and in study II, kale sprouts were soaked at $50^{\circ}C$ distilled water for 10, 20, 30, 45, or 60 seconds. After the heat shock treatments, the sprouts were transferred into normal growing conditions and recovered there for two days. Fresh and dry weights, electrolyte leakage, total phenolic concentration, antioxidant capacity, total flavonoid concentration, phenylalanine ammonia-lyase (PAL) activity, and glucosinolates content of the sprouts were measured before and after the heat shock treatments. As a result, there was a significant decrease in the fresh and dry weight of kale sprouts treated with heat shock compared with control at harvest in study I. Especially, heat shock at $60^{\circ}C$ lead to more pronounced growth inhibition compared with heat treatments at 40 and $50^{\circ}C$. Electrolyte leakage by cell collapse was the highest in the sprouts exposed to $60^{\circ}C$ distilled water, which agreed with the growth results. Heat shock at $50^{\circ}C$ significantly induced the accumulation of phenolic compounds. In study II, fresh weight of kale sprouts at $50^{\circ}C$ heat shock showed a significant decrease compared with the control at one and two days after the treatment. However, the decrease was minimal and dry weight of kale sprouts was not significantly different from that in control. In contrast, the heat shock-treated kale sprouts had higher level of total phenolic concentration than control at harvest. Heat shock treatments at $50^{\circ}C$ for 20 seconds or more showed at least 1.5 and 1.2 times higher total phenolic concentration and antioxidants capacity than control, respectively. The change of the total flavonoid concentration was similar with that of antioxidants. PAL activity after 24 hours of heat shock was higher in all the heat shock-treated sprouts than that in control suggesting heat shock may stimulate secondary metabolic pathway in kale sprouts. Seven glucosinolates were identified in kale sprouts and soaking the sprouts with $50^{\circ}C$ water for 20 seconds had a pronounced impact on the accumulation of total glucosinolates as well as two major glucosinolates, progoitrin and sinigrin, at harvest. In conclusion, this study suggests that heat shock using hot water would be a potential strategy to improve nutritional quality of kale sprouts by inducing the accumulation of phytochemicals with antioxidant and anticancer properties.

The Effect of Samul-tanggahyangbuja on Depression and Learning on Repeated Stress in Ovariectomized Rats (사물탕가향부자(四物湯加香附子)가 난소적출 흰쥐의 우울 및 학습에 미치는 영향)

  • Objectives: In this research, the effect of samul-tanggahyangbuja on depression and learning in ovariectomized rats subjected to repetitive stress were assessed. Samul-tanggahyangbuja is the prescription consisting of Samul-tang and Cyperi Rhizoma. Methods: Ovariectomized rats were repeatedly stressed over a 2-week period. After being orally medicated with samul-tanggahyangbuja (100 or 400 mg/kg), rats performed the Morris water maze test and forced swimming test, and social exploration was assessed in a behavior test. As well, sucrose intake was measured and measurements of blood serum corticosterone and the change of interleukin-$1{\beta}$ (IL-$1{\beta}$) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in blood samples were made. Results: 1. In the Morris water maze test, rats medicated with 100 mg samul-tanggahyangbuja mastered the maze in a shorter time on the 4th day in comparison with the control group, while rats medicated with 400 mg samul-tanggahyangbuja mastered the maze more quickly (p<0.05 on the 3rd day ; p<0.01 on the 4th day, as compared to control). 2. Immobility time in the forced swimming test was significantly decreased in rats receiving 400 mg samul-tanggahyangbuja compared with the control group (p<0.05). 3. Sucrose intake and active social behavior of rats receiving 400 mg samul-tanggahyangbuja were markedly increased in comparison with the control group (p<0.01). 4. Blood serum corticosterone measurements revealed decreased blood serum corticosterone level after medicating with samul-tanggahyangbuja. But it was not statistically significant. 5. Treatment with either dose of samul-tanggahyangbuja significantly reduced IL-$1{\beta}$ and TNF-${\alpha}$ (p<0.05). Conclusions: These results suggest that samul-tanggahyangbuja possesses the anti-depressant and cognitive-enhancing activities related to menopause.

  • Building construction trends have been changed dramatically in terms of size and mass. With the need to maximize land usage, there has been an increase in the construction of high-rise buildings. This affects not only the entire construction duration and cost, but also subsequent construction activities, such as work to increase underground facilities and in reclamation land area construction. These types of site conditions require soft ground reinforcement and the proper uplift water pressure treatment. In general, two kinds of methods have been used for uplift water pressure treatment systems. However, there have been some problems arising as the result of a lack of research and analysis on underground construction techniques, and a reliance on experiments over actual survey and analysis of site conditions. This paper focused on the problems of conventional selection procedure, by analyzing drawings and proposing a kind of modeling for a reasonable procedure. The results were applied to OO project as a sample construction case to be verified in this research. The initial plan in the case project was the Rock Anchor System. However, as there were terrible miscalculations of basic site conditions that had an extraordinary influence on the underground water level, such as the site's proximity to the Han-river, it was necessary to change the plan to include apermanent drainage system. This achieved a direct construction cost reduction \ 406,702,000 and a maximum sayings of 4% of operational cost, based on the 50-year building Life Cycle Cost.

A Study on the Nutritive Value and Utilization of Powdered Seaweeds (해조의 식용분말화에 관한 연구)

  • I. Subject of the study A study on the nutritive value and utilization of powdered seaweeds. II. Purpose and Importance of the study A. In Korea the shortage of food will be inevitable by the rapidly growing population. It will be very important study to develop a new food from the seaweeds which were not used hitherto for human consumption. B. The several kinds of seaweeds have been used by man in Korea mainly as side-dishes. However, a properly powdered seaweed will enable itself to be a good supplement or mixture to certain cereal flours. C. By adding the powdered seaweed to any cereals which have long been staple foods in this country the two fold benefits; saving of cereals and change of dietary pattern, will be secured. III. Objects and scope of the study A. Objects of the study The objects will come under four items. 1. To develop a powdered seaweed as a new food from the seaweeds which have been not used for human consumption. 2. To evaluate the nutritional quality of the products the analysis for chemical composition and animal feeding experiment will be conducted. 3. Experimental cocking and accepability test will be conducted for the powdered products to evaluate the value as food stuff. 4. Sanitary test and also economical analysis will be conducted for the powdered products. B. Scope of the study 1. Production of seaweed powders Sargassum fulvellum growing in eastern coast and Sargassum patens C.A. in southern coast were used as the material for the powders. These algae, which have been not used for human consumption, were pulverized through the processes of washing, drying, pulverization, etc. 2. Nutritional experiments a. Chemical composition Proximate components (water, protein, fat, cellulose, sugar, ash, salt), minerals (calcium, phosphorus, iron, iodine), vitamins (A, $B_1,\;B_2$ niacin, C) and amino acids were analyzed for the seaweed powders. b. Animal feeding experiment Weaning 160 rats (80 male and 80 female rats) were used as experimental animals, dividing them into 16 groups, 10 rats each group. Each group was fed for 12 weeks on cereal diet (Wheat flour, rice powder, barley powder, potato powder, corn flour) with the supplementary levels of 5%, 10%, 15%, 20% and 30% of the seaweed powder. After the feeding the growth, feed efficiency ratio, protain efficiency ratio and ,organs weights were checked and urine analysis, feces analysis and serum analysis were also conducted. 3. Experimental cooking and acceptability test a. Several basic studies were conducted to find the characteristics of the seaweed powder. b. 17 kinds of Korean dishes and 9 kinds of foreign dishes were prepared with cereal flours (wheat, rice, barley, potato, corn) with the supplementary levels of 5%, 10%, 15%, 20% and 30% of the seaweed powder. c. Acceptability test for the dishes was conducted according to plank's Form. 4. Sanitary test The heavy metals (Cd, Pb, As, Hg) in the seaweed powders were determined. 5. Economical analysis The retail price of the seaweed powder was compared with those of other cereals in the market. And also economical analysis was made from the nutritional point of view, calculating the body weight gained in grams per unit price of each feeding diet. IV. Results of the study and the suggestion for application A. Chemical composition 1. There is no any big difference in proximate components between powders of Sargassum fulvellum in eastern coast and Sargassum patens C.A. in southern coast. Seasonal difference is also not significant. Higher levels of protein, cellulose, ash and salt were found in the powders compared with common cereal foods. 2. The levels of calcium (Ca) and iron (Fe) in the powders were significantly higher than common cereal foods and also rich in iodine (I). Existence of vitamin A and vitamin C in the Powders is different point from cereal foods. Vitamin $B_1\;and\;B_2$ are also relatively rich in the powders.'Vitamin A in ·Sargassum fulvellum is high and the levels of some minerals and vitamins are seemed4 to be some influenced by seasons. 3. In the amino acid composition methionine, isoleucine, Iysine and valine are limiting amino acids. The protein qualities of Sargassum fulvellum and Sargassum patens C.A. are seemed to be .almost same and generally ·good. Seasonal difference in amino acid composition was found. B. Animal feeding experiment 1. The best growth was found at.10% supplemental level of the seaweed Powder and lower growth rate was shown at 30% level. 2. It was shown that 15% supplemental level of the Seaweed powder seems to fulfil, to some extent the mineral requirement of the animals. 3. No any changes were found in organs development except that, in kidney, there found decreasing in weight by increasing the supplemental level of the seaweed powder. 4. There is no any significant changes in nitrogen retention, serum cholesterol, serum calcium and urinary calcium in each supplemental level of the seaweed powder. 5. In animal feeding experiment it was concluded that $5%{\sim}15%$ levels supplementation of the seaweed powder are possible. C. Experimental cooking and acceptability test 1. The seaweed powder showed to be utilized more excellently in foreign cookings than in Korean cookings. Higher supplemental level of seaweed was passible in foreign cookings. 2. Hae-Jo-Kang and Jeon-Byung were more excellent than Song-Pyun, wheat cake, Soo-Je-Bee and wheat noodle. Hae-Je-Kang was excellent in its quality even as high as 5% supplemental level. 3. The higher levels of supplementation were used the more sticky cooking products were obtained. Song-Pyun and wheat cake were palatable and lustrous in 2% supplementation level. 4.
