Dynamics of Water Environmental Factors and Phytoplankton Before and After Inflow of Seawater in Shingwa Reservoir (시화호에서 해수유입 전.후의 수환경 요인과 식물플랑크톤 동태)

  • The dynamics of water quality and phytoplankton population had examined by monthly sampling from the upper to the lower part of watergate in an artificial Shihwa Reservoir in which situated near newly cities and incustrial complex on the west coast of Korea from January 1997 to December 1998. Among environmental factors, yearly average concentration of chl-a, TN and TP seemed to eutrophic or hypertrophic conditions that ranged 146.4~245.8 $\mu\textrm{g}$/$\ell$, 1.6~2.7 mg N/$\ell$, 258~448 $\mu\textrm{g}$ P/$\ell$, 26.9~80.7 $\mu\textrm{g}$/$\ell$, 1.0~2.4 mgN/$\ell$ and 74~239 $\mu\textrm{g}$P/$\ell$ respectively. Water quality was extremely deteriorated to consistently accumulation into inner reservoir by load of pollutants from autochthonous and allochthonous until early July 1997 after embankment. Water pollution of Shihwa Reservoir was remarkble on the biological condition with largely persistent bloom of phytoplankton and increase rate of standing crops was 2.4/yr. The development trend of phytoplankton in water ecosystem were closely related to increse and decrease of physico-chemical factors and those scale seemed to control by nutrient contents. Inflow of seawater into reservoir to object of repair of water quality. As to see dominant species, composition of those composed to mostly freshwater algae before inflow of seawater such as Selenastrum capricornutum of green algae, cyclotella atomus, C. meneghiniana of diatom and Microcystis spp. of blue-green algae and the other hand brackish algae were dominated after inflow of seawater such as Chaetoceros dicipiens, Skeletonema costatum of diatom, Dinophysis acuminata, Gymnodinium mikimotoi, G. sanguineum, Gyrodinium spirale, Prorocentrum minmum of dinoflagellate and Eutreptiella gymnastica of euglenoid. Moreover, small flagellates including Chroomonas spp. of cryptomonad were abundant throughout the year. The cause of water deterioration during fill of the freshwater were complexly supported with extra and intra parameters. The variation pattern of phytoplankton were related to water temperature and salinity by inflow of seawater based to plentiful nutrients. The dynamics of phytoplankton were assessed to ecosystem that clearly condition of dominant by unique or a few angel species seasonally.

Characteristics of the Algal Growth inhibition Substances Produced by Alteromonas sp. SR-14 (Alteromonas sp. SR-14가 생산하는 조류증식 저해 물질의 특성)

  • In previous reports, the authors isolated the algicidal marine bacterium, Alteromonas sp. SR-14 and demonstrated its growth inhibition of diatom, Chaetoceros calcitrans (C. calcitrans). In this paper, we studied the effects of cell free culture filtrate of Alteromonas sp. SR-14 on the growth of C. calcitrans, and the characteristics of the algal growth inhibition substance. The culture filtrate of Alteromonas sp. SR-14 grown in peptone broth showed growth inhibition activity against C. calcitrans. The reasonable culture conditions of the bacterium for producing of algal growth inhibition substances were $15~20^{\circ}$ in temperature, 7.0-9.0 in pH and $23~30{\textperthousand}$ in salinity, respectively. The algal growth inhibition activity of culture filtrate was increased from stationary phase in growth curve of Alteromonas sp. SR-14. The molecular weights of algal growth inhibition substances produced by Alteromonas sp. SR-14 were ranged about from 3 KDa to 12 KDa. Among the substances, less than 10 KDa fraction were stable by heating at $100^{\circ}$ for 10 minutes, while more than 10 KDa fraction were heat labile. According to the experimental results, the algal growth inhibition substance produced by the bacterium was not a single compound.

Growth Performance of the Rotifer Brachionus plicatilus and the Larvae of Two Bivalves Fed on the Cryptophyte Teleaulax amphioxeia (은편모류 Teleaulax amphioxeia의 윤충류 Brachionus plicatilus 및 이매패류 유생 2종에 대한 먹이 기능성 평가)

  • The aim of this study was to verify the performance of Teleaulax amphioxeia as prey for Brachionus plicatilis and for the larvae of the bivalves (Pacific oyster Crassostrea gigas and Manila clam Ruditapes philippinarum) to select the best of five T. amphioxeia strains. The five strains of T. amphioxeia were collected from the coasts of Korea, purecultured and then fed to the rotifer and bivalve larvae. The density and fecundity rate of the rotifer fed on strain 01 were the highest. The instantaneous growth rate of the rotifer fed strain 08-2 was the highest. The maximum density and fecundity rate of the rotifer fed strain 08-2 were significantly higher than those fed Chlorella ellipsoidea, while instantaneous growth rate showed the opposite pattern. Survival rates of the Manila clam larvae fed each strain of Teleaulax showed no significant difference between strains. Survival rates of the Pacific oyster fed Teleaulax were significantly higher than those fed Chaetoceros sp., but lower than those fed I. galbana. This study indicates that strain 08-2 showed better growth for B. plicatilis and better survival for the bivalve larvae than did the other strains, and that T. amphioxeia can also be used as a prey organism for rotifers or bivalve larvae.

Optimal Enrichment Temperature for Rotifer and Microalgae Selection for Cold-Water Species Culture (한해성 품종 종묘생산을 위한 로티퍼의 적정 영양강화 수온 및 미세조류 선택)

  • The purpose of this study is to establish methods for enhancing the survival and growth of cold-water fish and crustacean larvae based on the nutritional components of zoo and phyto live foods. Rotifers, Brachionus rotundiformis, were cultured with a supplement of freshwater condensed Chlorella vulgaris at $28^{\circ}C$ and enriched with Algamac $2000^{(R)}$ at 16, 20 and $26^{\circ}C$, respectively. Isochrysis galbana, Chaetoceros calcitrans and Chlorella ellipsoidea were centrifuged for component analysis after being cultured for approximately one week with conway medium at $20^{\circ}C$. The crude protein and lipid contents of the rotifers were 58.4% and 10.9%, respectively, before enrichment. After enrichment at each temperature, total protein and essential amino acid contents were increased by reducing the enrichment temperature. However, unsaturated fatty acids and multiple fatty acid index (UI) showed their highest values at $20^{\circ}C$. Mono-unsaturated fatty acid content was highest (72.6%) at $16^{\circ}C$. The total protein contents of C. calcitrans and C. ellipsoidea were higher, 33.0% and 35.2%, respectively, than that of I. galbana, 27.8%. Methionin, leusine and histidine, essential amino acids of C. ellipsoidea, had considerably higher values, 50.2, 287.2 and 68.1 mg/g dry matter, respectively, compared to other microalgae. Total lipids, UI, DHA and n-3 PUFA of I. galbana had higher values, 23.6, 272.0, 12.9% and 45.2%, respectively, than other microalgae. Therefore, for cold-water fish and crustacean larvae that require high n-3 PUFA and DHA contents, enrichment of rotifers is desirable at $20^{\circ}C$. Fish larvae would also need more I. galbana than other microalgae.

  • Phytoplankton samples, collected about twenty day interval on May of 1967, at same twenty-one stations located in the north-eastern Korea Strait were examined in an attempt to determine the specific distribution and abundance of phytoplankton. Water bottle samples preserved with neutralized formula were used for the phytoplankton study. In May, water temperature and salinity show both lower at the sea of Korea side than that of Japan side, and both indicate gradually higher to the sea of Japan side. Fourty-six species of diatoms and three species of dinoflagellates in early May, and thirty-six species of diatoms and six species of dinoflagellates in the May were detected. Among them thirty species of diatoms and two species of dinoflagellates were appeared commonly in both cruise. Chaetoceros curvisetus and Rhizosolenia castracanei were wide distributed species, and Leptocylindrus danicus was the most numerous species at this sea in May. In generally, the sea of Korea side was more rich in species and numbers of phytoplankton.

Seasonal Dynamics of Phytoplankton and Environmental Factors around the Chagwi-do off the West Coast of Jeju Island, Korea

  • The dynamics of phytoplankton abundance with seasonal variation in physicochemical conditions were investigated monthly at 10 stations around the Chagwi-do off the west coast of Jeju Island, Korea, including inshore, middle shore, and offshore in the marine ranching are a from September 2004 to November 2005. Water temperature varied from 12.1 to $28.9^{\circ}C$ (average $18.8^{\circ}C$), and salinity from 28.9 to 34.9 psu (average 33.7 psu). The chlorophyll a concentration was $0.02-2.05\;{\mu}g\;L^{-1}$ (average $0.70\;{\mu}g\;L^{-1}$), and the maximum concentration occurred in the bottom layer in April. A total of 294 phytoplankton species belonging to 10 families was identified: 182 Bacillariophyceae, 52 Dinophyceae, 9 Chlorophyceae, 12 Cryptophyceae, 6 Chrysophyceae, 4 Dictyophyceae, 13 Euglenophyceae, 6 Prymnesiophyceae, 5 Prasinophyceae, and 5 Raphidophyceae. The standing crop was $2.21-48.69\times10^4\;cells\;L^{-1}$ (average $9.23\times10^4\;cells\;L^{-1}$), and the maximum occurred in the bottom layer in April. Diatoms were most abundant throughout the year, followed by dinoflagellates and phytoflagellates. A phytoplankton bloom occurred twice: once in spring, peaking in April, and once in autumn, peaking in November. The spring bloom was represented by four Chaetoceros species and Skeletonema costatum; each contributed 10-20% of the total phytoplankton abundance. The autumn bloom comprised dinoflagellates, diatoms, and phytoflagellates, of which dinoflagellates were predominant. Gymnodinium conicum, Prorocentrum micans, and P. triestinum each contributed over 10% of the total phytoplankton abundance.

The Influence of Nutrients Concentration and the Ratio on Phytoplankton Community Structure during Late Spring and Early Summer in Sagami Bay, Japan (춘계와 하계의 영양염 농도와 그 구성비가 식물플랑크톤의 군집구조에 미치는 영향평가)

  • The relationship between nutrients and phytoplankton dynamics was investigated daily from 12 April to 22 July 2003 in Sagami Bay, Japan. According to multidimensional scaling (MDS) and cluster analysis, phytoplankton community was divided into four distinct groups. The first group was consisted of centric diatom species, such as Guinardia spp., Detonula spp., Letocylindrus danicus, Skeletonema costatum, Eucampia zodiacus and Chaetoceros spp.. The second and third clusters comprised mainly diatoms and dinoflagellates, respectively. The other cluster was restricted to the samples collected during the last sampling period when the rainfall and river discharge was frequently recorded. Canonical correspondence analysis (CCA) was applied to analyze four groups respectively, which focused on the effects of nutrients concentration and ratio on phytoplankton variations. Based on CCA analysis, most species of centric diatom were negatively correlated with DSi concentrations and Si/N ratio. Nutrients were strongly limited phytoplankton growth during the summer when the rainfall was not observed, whereas river discharge by rainfall and counterclockwise coastal currents (although the surface circulation pattern is often altered by Kuroshio Current, the counterclockwise coastal currents are generally dominant) has brought phytoplankton population accumulation and triggered the micoalgae growth in western part of the bay. Phosphorous (P) was strongly limited after significantly increases in the phytoplankton abundances. However, silicate (Si) was not a major limiting factor for phytoplankton production, since Si/DIN and Si/P ratio did not create any potential stoichiometric limitation. This indicates that high Si availability contributes favorably to the maintenance of diatom ecosystems in Sagami Bay.

Heterologous Gene Expression System Using the Cold-Inducible CnAFP Promoter in Chlamydomonas reinhardtii

  • To increase the availability of microalgae as producers of valuable compounds, it is necessary to develop novel systems for gene expression regulation. Among the diverse expression systems available in microalgae, none are designed to induce expression by low temperature. In this study, we explored a cold-inducible system using the antifreeze protein (AFP) promoter from a polar diatom, Chaetoceros neogracile. A vector containing the CnAFP promoter (pCnAFP) was generated to regulate nuclear gene expression, and reporter genes (Gaussia luciferase (GLuc) and mVenus fluorescent protein (mVenus)) were successfully expressed in the model microalga, Chlamydomonas reinhardtii. In particular, under the control of pCnAFP, the expression of these genes was increased at low temperature, unlike pAR1, a promoter that is widely used for gene expression in C. reinhardtii. Promoter truncation assays showed that cold inducibility was still present even when pCnAFP was shortened to 600 bp, indicating the presence of a low-temperature response element between -600 and -477 bp. Our results show the availability of new heterologous gene expression systems with cold-inducible promoters and the possibility to find novel low-temperature response factors in microalgae. Through further improvement, this cold-inducible promoter could be used to develop more efficient expression tools.

Variation of Phytoplankton and Nutrients in the Namdaechon Estuary, Korea (남대천 하구역의 식물플랑크톤과 영양염 변동)

  • Spatio-temporal variations of phytoplankton abundance and nutrient concentrations were investigated in the Namdaechon estuary, Yangyang, from April to December, 2003. A total of 51 phytoplankton species were identified with 32 diatom species in the study area. Phytoplankton abundance ranged from 14 cells/mL to 3,798 cells/mL. Small sized benthic and pennate-type diatoms like Cymbella spp., Fragilaria spp., Navicula spp., Synedra spp. were dominant at throughout the whole study area. Various planktonic species like Chlamydomonas sp., Peridinium spp., Euglena spp., Cryptomonas spp. etc. were abundant especially at the estuary of the Namdaechon, from May to August. Phytoplankton bloom (>3,000 \;cells/mL) occurred from the late May to mid June. After September when the sandbank of estuary was broken by the typhoon 'Maemi', planktonic species disappeared. These planktonic species were followed by oceanic diatoms, Skeletonema costatum and Chaetoceros spp.. Concentrations of dissolved inorganic nitrogen (DIN) and silicate were relatively high at the upstream in summer. In May, before phytoplankton bloom, high concentrations of phosphate $(>3.5\;{\mu}mol/L)$ were observed at all the study area. These results suggested that spatio-temporal variation of phytoplankton in the Namdaechon estuary was related to formation of sandbank at the mouth of the estuary, fluid speed affected by sandbank and nutrients supplied in spring. High correlation $(r^2=0.928)$ between chlorophyll a and biological oxygen demand (BOD) implies that BOD was related to phytoplankton abundance in the Namdaechon estuary.

Phytoplankton composition in intensive shrimp ponds in Bac Lieu province, Vietnam

  • Algal overgrowth in shrimp culture ponds can affect the quality of the aquatic environment, thereby adversely affecting the shrimp and causing economic losses. The objective of this study was to evaluate the variation in phytoplankton composition in intensive shrimp ponds in Bac Lieu province, Vietnam. Phytoplankton samples were collected in three black tiger shrimp (Penaeus monodon) ponds and three whiteleg shrimp (Litopenaeus vannamei) ponds. The collected data were analyzed using SPSS and canonical correlation analysis softwares. In total, 75 species of phytoplankton were recorded in black tiger shrimp ponds and 64 species in whiteleg shrimp ponds. Diatoms had the highest species diversity with 29-30 species (39%-47%), followed by green algae with 9-19 species (14%-25%); species numbers of other phyla varied from 5-12 (8%-16%). The total number of phytoplankton species throughout the study varied from 34-50 species. Algal density was relatively high and ranged from 497,091-2,229,500 ind./L and 1,301,134-2,237,758 ind./L in black tiger shrimp and whiteleg shrimp ponds, respectively. The diatom density tended to increase during the final stage of the production cycle in black tiger shrimp ponds. Blue-green algae and dinoflagellates also increased in abundance at the end of the cycle, which can affect shrimp growth. Diatoms were significantly positively correlated with pH, salinity, total ammonia nitrogen, and nitrate (NO3-) concentrations (p < 0.05). Blue-green algae and dinoflagellates were positively correlated with salinity, phosphate (PO43-), and NO3-. Algal species diversity was lower in the whiteleg shrimp ponds than in the black tiger shrimp ponds. Several dominant algal genera were recorded in the shrimp ponds, including Nannochloropsis, Gyrosigma, Chaetoceros, Alexandrium, and Microcystis. The results of this study provide basic data for further investigations, and they contribute to the management of algae in brackish-water shrimp ponds.
