The Study on Function and Localization of Nup97 in Fission Yeast (분열효모에서 Nup97의 기능과 세포 내 위치에 대한 연구)

  • We studied on the function and localization of fission yeast Schizosaccharomyces pombe Nup97p, which is homologous to nucleoporin Nic96p in budding yeast Saccharomyces cerevisiae. There was no effect on growth and $poly(A)^{+}$ RNA distribution of cells when nup97 gene was overexpressed. However, the haploid ${\Delta}nup97::kan^{r}$ null mutants confirmed extensive $poly(A)^{+}$ RNA accumulation in the nucleus, abnormal DNA distribution, and cessation of growth when nup97 expression was repressed. We determined the subcellular localization of Nup97 tagged at the N terminus or the C terminus with GFP. Both fusions complemented growth defect of ${\Delta}nup97::kan^{r}$ null mutants. An integrated version of the nup97-GFP fusion was constructed at the nup97 locus. Nup97-GFP fusions expressed from its own promoter was localized at the nuclear periphery with a punctate appearance. These results suggest that Nup97p in fission yeast is also nucleoporin, which is involved in mRNA export.

Analysis of Factors Affecting the Smoking Rates Gap between Regions and Evaluation of Relative Efficiency of Smoking Cessation Projects (지역 간 흡연율 격차 영향요인 분석 및 금연사업 상대적 효율성 평가: Clustering Analysis와 Data Envelopment Analysis를 활용하여)

  • Background: Based on the importance of ceasing smoking programs to control the regional disparity of smoking behavior in Korea, this study aims to reveal the variation of smoke rate and determinants of it for 229 provinces. An evaluation of the relative efficiency of the cease smoking program under the consideration of regional characteristics was followed. Methods: The main sources of data are the Korean Statistical Information Service and a national survey on the expenditure of public health centers. Multivariate regression is performed to figure the determinants of regional variation of smoking rate. Based on the result of the regression model, clustering analysis was conducted to group 229 regions by their characteristics. Three clusters were generated. Using data envelopment analysis (DEA), relative efficiency scores are calculated. Results from the pooled model which put 229 provinces in one model to score relative efficiency were compared with the cluster-separated model of each cluster. Results: First, the maximum variation of the smoking rate was 16.9%p. Second, sex ration, the proportion of the elder, and high risk drinking alcohol behavior have a significant role in the regional variation of smoking. Third, the population and proportion of the elder are the main variables for clustering. Fourth, dissimilarity on the results of relative efficiency was found between the pooled model and cluster-separated model, especially for cluster 2. Conclusion: This study figured regional variation of smoking rate and its determinants on the regional level. Unconformity of the DEA results between different models implies the issues on regional features when the regional evaluation performed especially on the programs of public health centers.

Influencing factors to dental caries and periodontal diseases in Korean adults (성인의 치아우식증 및 치주질환에 영향을 미치는 요인)

  • Objectives: The aim of this study was to investigate the factors related to dental caries and periodontal disease in Korean adults. Methods: The subjects were 5,149 adults over 19 years old who participated in the 5th Korea National Health and Nutrition Examination Survey 2012. Data were analyzed by chi-square test and logistic regression analysis using SPSS 18.0 statistical package program. The questionnaire consisted of 17 questions of independent variables and 2 kinds of dependent variables including dental caries and periodontal diseases by direct interview. The independent variables included 5 questions of general characteristics, 3 questions of health behavior, 5 questions of oral health behavior, and 4 questions of chronic diseases diagnosed by the doctors. Results: The prevalence rate of dental caries was higher in those who did not regularly work out and did not receive regular checkups including dental floss and dental brush use. The prevalence rate of periodontal diseases was higher in male adults, smokers, and those who were obese and had diabetes mellitus, poor oral health care habit without using dental floss and inter-dental brush. Conclusions: In order to prevent the dental caries in the adults, it is very important to do regular exercise and regular dental checkup. The best ways of dental caries prevention include tooth brushing after meal with use of dental floss and inter-dental brush. This study suggests that dental health promotion can be enhanced by smoking cessation, ideal body weight maintenance, timely tooth brushing after meal, and use of dental floss and inter-dental brush.

Health-related lifestyle and self-reported symptoms in dental hygiene students (일부 치위생과 학생의 건강관련 생활습관과 자각증상)

  • Objectives : The study investigated and analyzed the relationship between health-related lifestyle and psychosomatic self-reported symptom in dental hygiene students. Methods : A self-reported questionnaire was filled out by 478 dental hygiene students in Gyeonggi-do and Gangwon-do from March 7 to June 21, 2012 by random sampling method after informed consent. Results : The health-related lifestyle showed the results as follows. Nonsmokers accounted for 89.1%. Those who never exercise accounted for 67.9% and 37.9% of the students sleep for 5-6 hours. Those who take alcohols twice per month accounted for 58.2%. The correlation between the symptoms and lifestyle included multiple subjective symptoms(I), respiratory(A), eyes and skin(B) and digestive organs(C) symptoms and smoking status(p<.01), mouth and anal(D), depression(K), nervousness(E). There existed the correlation in average sleeping time(p<.01) and impulsivess(H) and smoking status(p<.05) and lie scale(L) and regular exercise(p<.05) and aggressiveness(F) and drinking habits and irregular and life(G) and breakfast habits. The smoking habit, sleeping time, and snack intake had an influence on psychosomatic self-reported oral health-related symptoms. Conclusions : Cessation of smoking, adequate sleeping time, and reduction of snack intake can improve the oral health-related lifestyle and reduce the self-reported symptoms in the dental hygiene students.

Effects of d-Amphetamine and Chlorpromazine on the Orienting Response and General Behavioral Activity in Rats (d-Amphetamine 및 Chlorpromazine이 흰쥐의 정위반응(定位反應) 및 일반행동에 미치는 영향)

  • To see if the treatments of d-amphetamine and chlorpromazine have any influence upon the orienting response and general behavioral activity, 3 groups of male Holtzman rats were prepared, namely d-amphetamine animals (1.0 mg/kg.i.p.), chlorpromazine rats (1.0 mg/kg.i.p.) and the physiological saline control animals. The general behavioral activity was examined by visual scanning using the time-sample method in the adaptation period of orienting response. The occurence of orienting response and its rate of habituation were evaluated by observing cessation of ongoing activity in response to a sound stimulus (1,000 Hz, 70 db & 0.1 sec), or turning of head toward the source of stimulus in 20 trials. Attention shift from sound to light stimulus was also tested in 10 trials. The results obtained were as follows; 1. The general behavioral activity of d-amphetamine group was significantly greater than that of control, however, the chlorpromazine animals showed the tendency to decrease in activity. 2. The d-amphetamine group showed the occurence of orienting response to sound significantly more often than that of placebo controls. However, the chlorpromazine group exhibited significantly fewer orienting response than the placebo group did. 3. The d-amphetamine group displayed no clear out habituation to the orienting response following the repetition of trials, though the placebo and the chlorpromazine groups demonstrated apparent habituation to the response. 4. The three animal groups did not differ significantly from each other with regard to shift of attention from sound to light stimulus. It is inferred that the d-amphetamine tends to increase general activity has a definite facilitative action of orienting response and a inhibitory influence upon the habituation of the latter response. On the contrary, the chlorpromazine tends to decrease general activity, has a inhibitory action of orienting response and facilitatory action of habituation of the response.

  • Amiodarone is widely used to control fatal arrhythmia. However, amiodarone therapy is associated with a relatively high incidence of pulmonary toxicity, up to 5 to 10%. Typical symptoms are nonspecific and often manifest as nonproductive cough, dyspnea and interstitial infiltrates in patients with acute pneumonitis or chronic fibrosis. However, hemoptysis is a very rare symptom of amiodarone pulmonary toxicity. We report a case of amiodarone pulmonary toxicity, who presented with hemoptysis and was successfully treated with the cessation of amiodarone, with review of the relevant literature.

A Study on Substance Abuse, Knowledge and Attitude about Substances in the Elementary Students (일부지역 초등학생의 약물남용과 약물에 관한 지식 및 태도)

  • Purpose: To provide basic data for strategy of substance abuse through the investigation of the status of substance abuse and knowledge and attitude about drugs in the elementary students. Method: Subjects were 565 6th grade of elementary students. Data were collected from June to July 2001 by using structured questionnaire. Result: 1) Among the students of respondents, 10.2% of them had the experience in smoking, 26.7% and 08% of them in drinking and inhalants, respectively. 2) The degree of students' knowledge about substance was 20.9 points on average with the highest score being 30. Students' knowledge about substances, smoking came on top, next came drinking and inhalants followed them. There were no significant differences in the knowledge of substances by general and drug related characteristics of the students. 3) The students' attitude about substances was negative with 30 points on average with the highest score being 108. As for the attitude about substances, they revealed the most generous in drinking next in smoking and inhalants. Those of boys, bored in school life and experienced in substance abuse were significantly generous in the attitude about substances. 4) There was a significant weak inverse relationship between the student's knowledge and attitude about substances (r=-0.13). As for the relationship between the student's knowledge and attitude about substances, the strongest relationship was shown in inhalants (r=-0.18) next in drinking (r=-0.16) and smoking (r=-0.06). Conclusion: The results of the finding suggest that the developing of programs in substance abuse for the elementary students are needed not only for the prevention but also for the cessation.

Smoking, Meat Intake and Exercise related to Alcohol Use

  • This study was conducted to investigate the relationship between drinking, smoking, meat intake, and exercise. The participants were 1,060 males and females who voluntarily responded to the survey by visiting the health promotion booth at the H Festival held in C city in 2014. Research data were collected in interview form using questionnaires on alcohol use, smoking, exercise, and meat intake. The data were analyzed by descriptive analysis, cross tabulation analysis, multinominal logistic regression after stratification of gender using the SPSS 24 program. The results of the study are as follows: Smokers had a significantly higher frequency of drinking ($x^2=163.33$, p<.001) than non-smokers. Meat intake was high when alcohol use was high ($x^2=35.13$, p<.001). The risk rates for smoking, meat consumption, and exercise related to alcohol use are as follows. The smoking was higher as the frequency of drinking increased. For men, smoking OR(odd ratio) was 6.26 (p=.001) and for women, smoking risk OR was 16.82 (p=.002). Meats intake showed a higher OR as the alcohol use increased. The OR of almost daily ingestion of meat at frequent drinking days was 4.40 (p=.002) for males and 4.52(p=.009) for females. As the frequency of drinking increased, the OR of days of exercise tended to decrease. In men, the OR was 0.36 (p=.003), the lowest in the probability of exercising more than 5 days when drinking high. In the case of women drinking less than once a week, the OR was 0.43 (p=.027), which was the lowest for exercise for 5 days or more. In conclusion, the higher the frequency of drinking, the higher the smoking and meat intake, and the less exercise. If drinking, smoking, and meat intake are high but there is a lack of exercise, a chronic disease can occur and cause premature death unless there is a change in the new health behavior. The results of this study suggest researches to understand the motives related to alcohol abstinence, smoking cessation, dietary control and exercise, and to develop programs.

Genes involved in leaf senescence and regulation of their expression

  • We have isolated more than a dozen cDNA clones corresponding to genes that were expressed in Arabidopsis leaves when they were kept in the dark. The nucleotide sequence analysis showed that some of the clones encoded proteins with significant homology to $\beta$-glucosidase (din2), branched-chain $\alpha$-keto acid dehydrogenase subunit E1$\beta$(din3), and another subunit E2 (din4), yeast RAD23 (din5), asparagine synthetase (din6), pre-mRNA splicing factor SRp35 (din7), phosphomannose isomerase (din9), seed imbibition protein (din10), and 2-oxoacid-dependent oxidase (din11). Accumulation of transcripts from din3,4,6 and 10 occurred rapidly after the plants were transferred to darkness. Transcripts from din2,9, and 11 could be detected only after 24 h of dark treatment. Inhibition of photo-synthesis by DCMU strongly induced the accumulation of transcripts from those genes, and application of sucrose to detached leaves suppressed the accumulation both in the dark and by DCMU. These observations indicate that expression of the genes is caused by sugar starvation resulted from the cessation of photosynthesis. We further showed that din2-encoded protein also accumulated in senescing leaves. Given these results, possible roles of din genes in leaves in the dark and senescing leaves are discussed.

Residue Depletion of the Sulfaquinoxaline and Trimethoprim Combination in Broilers (육계에서 설파퀴녹살린 및 트리메토프림 합제의 잔류분석)

  • Sulfaquinoxaline (SQX) and trimethoprim (TMP) are chemotherapeutics that are extensively used in various animal species for the treatment or prevention of coccidia and coccidia-like parasites. Little information about the depletion kinetics of these compounds in chickens exists in the literature. In this study, a new commercial liquid concentrate of SQX in combination with TMP (100 g/l of SQX and 33.4 g/l of TMP) was administered with drinking water at a dose of 0.75 ml/l or 1.5 ml/l. The edible tissue concentrations of the drugs were determined by the validated high-performance liquid chromatography/mass spectrometric method. Residue concentrations of SQX and TMP were lower than their maximum residual limits (MRLs) in all tissues from both dose groups at 5 days after the treatment. The optimal withdrawal time of SQX/TMP combination was suggested to be over 5 days after cessation of medication in broilers.
