Cervus elaphus 종의 sequencing과 BLAST search에 의한 감별 (Identification of Cervus elaphus Species by Sequencing Analysis and BLAST Search)

  • Objectives : Cervus elaphus species are some of the most medicinally important genera in the Oriental medicine. This study was performed to determine if Cenvus elaphus species could be identified by sequencing analysis and to verify Basic Local Alignment Search Tool (BLAST) search, which was used to assess genetic identification. Methods : The DNAs of Cervus elaphus species were extracted, amplified by PCR, and sequenced. The DNAs of Cervus species were identified by BLAST search in website. Results : By BLAST search one of Cervus elaphus species was identified as Cervus elaphussibericus but the other was identified as Cervus elaphus nelsoni. This work showed that identification can efficiently be performed by BLAST search. Conclusion : These results suggest that sequencing following BLAST search might be able to provide the identification of Cervus elaphus species.

  • Velvet antlers from Cervus elaphus species are one of most famous, expensive and commonly used medicinal materials in traditional oriental medicine. Some distributor had illegal practice of disguising the origin of antlers in Korea market. Therefore, a test to distinguish antler essential to ensure the healthy development of the herbal industry. In this study, the variation in DNA sequences of the mitochondrial ATPase8 and cytochrome-coxidaseI (COI) genes of Cervus elaphus from China, the Republic of Altai, and Canada were evaluated. In addition, the sequence variation among, Rein deer and Cervus elaphus species was also evaluated. Although the sequences of deer from the Republic of Altai and Canada were very similar, polymorphisms that were conserved in each species were observed in the ATPase8 and COI genes. Therefore, these polymorphic markers could be used to distinguish Cervus elaphus antlers from different locations.

Cervus 종의 Phylogenetic analysis에 의한 판별 (Authentication of Cervus Species by Phylogenetic analysis)

  • Objectives : This study was performed to determine if an antler could be identified as one of the Cervus species by phylogenetic analysis, which was used to assess genetic authentication. Methods : The DNAs of an antler were extracted, amplified by PCR, and sequenced. The DNAs of an antler were identified by Phylogenetic analysis. Phylogenetic analysis was made using MEGA software (Molecular Evolutionary Genetics Analysis, 3.1) Results : By phylogenetic analysis an antler was identified as Cervus elaphus nelsoni not as Cervus elaphus sibericus. This work showed that authentication can efficiently be performed by phylogenetic analysis. Conclusion : These results suggest that phylogenetic analysis might be able to provide the authentication of Cervus species.

사슴 미토콘드리아 DNA의 염기서열 및 PCR-RFLP분석에 의한 녹용의 종 감별 (Identification of Deer Antler Species Using Sequence Analysis and PCR-RFLP of Mitochondrial DNA)

  • 우리나라는 전 세계 녹용의 약 80% 이상을 소비하고 있는 양록 대국이나 최근 국내 녹용시장에서의 녹용 둔갑판매 및 불법유통 현상이 문제점으로 대두되고 있다. 따라서 본 연구는 녹용의 종 감별 기술을 개발하고자 현재 국내에서 유통되고 있는 러시아산 원용, 북미산 대록, 국산화용, 중국산 깔깔이 및 알래스카산 순록 등 5종의 대표적인 녹용들을 대상으로 종간 염기서열 변이성이 매우 높은 유전자로 알려져 있는 mt DNA내 cytochrome b 및 D-loop 유전자 영역의 염기서열 분석 및 종간 변이성 비교분석을 수행하였다. 각 녹용시료에서 mt DNA를 분리하고 cytochrome b와 D-loop유전자의 특정 영역을 포함하는 primer를 설계 합성하고 PCR로 증폭한 후 DNA 증폭산물의 염기서열을 분석하여 종간 유전정보의 동일성 여부를 비교한 결과 녹용 종간에 명확한 차이를 보이는 염기서열 부위가 검출되었고 이러한 종간 염기배열 차이에 근거하여 녹용의 종 감별이 가능하였다. 또한, mt DNA cytochrome b유전자에서 종간 특이적 염기서열을 인지하는 두 종류의 제한효소(NlaIV 및 TaqI)을 이용한 PCR-RFLP 기법으로 녹용으로 인정되지 않는 순록의 종 특이적 RFLP 분자표지를 검출하였고 이를 이용하여 녹용과 순록간의 종 판별이 가능하였다. 한편, D-loop 유전자의 특정 영역 염기서열 분석기법을 이용하여 시중에서 러시아산 원용으로 유통되고 있는 녹용 절편 32개를 무작위표본 추출하여 녹용의 종 감별을 조사한 결과 러시아산 원용으로 인정되는 것은 62.5%에 불과하였고 나머지는 중국산 마록(25.0%)과 엘크 및 순록의 아종으로 추정되는 시료도 일부 검출되었다. 따라서 본 연구를 통해 사슴 녹용 mt DNA 유전자의 염기서열 유전정보 변이 차이를 이용한 염기서열 분석법과 특정 제한효소(NlaIV 및 TaqI)를 이용한 PCR-RFLP 기법은 녹용의 과학적인 종 감별과 이를 바탕으로 녹용 원산지의 추정도 가능할 것으로 기대된다.

Genetic Distance Methods for the Identification of Cervus Species

  • Objectives : This study was performed to determine if unknown species of antler samples could be identified by genetic distance methods. Methods : The DNAs of 4 antler samples were extracted, amplified by PCR, and sequenced. The DNAs of antlers were identified by genetic distance. Genetic distance method was made using MEGA software (Molecular Evolutionary Genetics Analysis, 3.1). Results : By genetic distance methods, all 4 antler samples were closest to Cervus elaphus nelsoni among Cervus species. Conclusion : These results suggest that genetic distance methods might be used as a tool for the identification of Cervus species.

First Molecular Characterization of Hypoderma actaeon in Cattle and Red Deer (Cervus elaphus) in Portugal

  • Hypoderma spp. larvae cause subcutaneous myiasis in several animal species. The objective of the present investigation was to identify and characterize morphologically and molecularly the larvae of Hypoderma spp. collected from cattle (Bos taurus taurus) and red deer (Cervus elaphus) in the district of Castelo Branco, Portugal. For this purpose, a total of 8 larvae were collected from cattle (n=2) and red deer (n=6). After morphological identification of Hypoderma spp. larvae, molecular characterization was based on PCR-RFLP and mitochondrial CO1 gene sequence analysis. All larvae were morphologically characterized as the third instar larvae (L3) of H. actaeon. Two restriction enzymes were used for molecular identification of the larvae. TaqI restriction enzyme was not able to cut H. actaeon. However, MboII restriction enzyme differentiated Hypoderma species showing 210 and 450 bp bands in H. actaeon. Furthermore, according to the alignment of the mt-CO1 gene sequences of Hypoderma species and to PCR-RFLP findings, all the identified Hypoderma larvae were confirmed as H. actaeon. This is the first report of identification of Hypoderma spp. (Diptera; Oestridae) from cattle and red deer in Portugal, based on morphological and molecular analyses.

  • The article is devoted to the study of the density dependence of the horns tissue on the species and the age of the representatives of the deer family. In scientific literature this problem was covered partially. Basic physical parameters (mass, volume, and density) of horns from red deer (Cervus elaphus), fallow deer (Dama dama), European (Capreolus capreolus) and Siberian roe deer (Capreolus pygargus) were estimated. The present study was designed to analyze the results for making identification of species and age of the animal possible.

엘크의 구포자충 감염사례 (A case of elk (Cervus elaphus) coccidiosis in Korea)

  • A thirty-five week old elk was referred for Chronic Wasting Disease (CWD) confirmation from Chungnam province in Korea. Necropsy revealed much bloody contents in the lumen of small and large intestines, and reddening of its mucosa. Microscopically, few coccidia showing various developmental stages were infiltrated in lamina propria of small intestine with granulomatous inflammation and congestion. They were identified as Eimeria spp. according to their location and morphological characteristics. Because fecal examination and oocyst culture were not available, the species of the Eimeria could not be confirmed. There were no detection of pathogenic bacteria such as clostridia and virus in intestinal contents. CWD was shown to be negative in immunohistochemistry test. In conclusion, it was the first Eimeria case detected in small intestine of Korean domestic elk as far as we know although Eimeria spp. were not the main cause of death.
