Effects of SosokMyeongTang extract on the Regional Cerebral Blood Flow and Blood Pressure in Rats (소속명탕(小續命湯)이 백서(白鼠)의 국소뇌혈류량(局所腦血流量)과 혈압(血壓)에 미치는 영향(影響))

  • SosokMyeongTang(SMT) have been used in oriental medicine for many centuries as a therapeutic agent for cerebral disease. The effects of SMT on the vascular system is not known. The purpose of this study was to investigate the effects of SMT on the changes in blood pressure(BP) and regional cerebral blood flow(rCBF) of rats. SMT consists of the following components : Radix Ledebouriellae(防風), Radix Cocculi or Stephaniae(防己), Semen Armeniacae(杏仁), Cortex Cinnamomi(肉桂), Radix Scutellariae(黃芩), Radix Paeoniae Lactiflorae(白芍藥), Radix Ginseng(人蔘), Rhizoma Cnidll(川芎), Herba Ephedrae(麻黃), Radix Glycyrrhizae(甘草), Radix Aconiti(附子?), Fructus Zizyphi Jujubae(大棗), Rhizoma Zingiberis(生薑) and the changes of BP and rCBF were tested Leser-Doppler Flowmetry(LDF) The experimental results were as follows ; BP was not affected by SMT in rats, but rCBF was increased significantly by SMT in a dose dependent manner. SMT increased previous decreasd rCBF due to pretreatment methylene blue, but did not increased previous decreasd rCBF due to pretreatment L-NNA, indomethacin. Pretreatment with indomethacin decreased BP compaired with control group. These results suggest that SMT causes a diverse response of blood pressure and regional cerebral blood flow(rCBF). The increased rCBF is mediated by nitric oxide synthease.

  • Objectives : The purpose of this study is to investigate the effects of $GB_{41}$ supplementation and $BL_{66}$ draining on changes in cerebral blood flow in normal rats. Methods : Regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in normal rats are observed, and those mechanisms were also investigated with pre-treatment of indomethacin(IDM) and methylene blue(MTB) each. Results : In this study, $GB_{41}$ supplementation and $BL_{66}$ draining elevated rCBF in time-dependent manner. Pre-treatment with indomethacin(IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. But pre-treatment with methylene blue(MTB), an inhibitor of guanylate cyclase, didn't affect rCBF levels. In addition, pre-treatment with IDM also decreased MABP levels. Conclusions : In conclusion, these results suggest that $GB_{41}$ supplementation and $BL_{66}$ draining is effective to increase rCBF, and the mechanisms are thought to be related to cyclooxygenase pathways.

  • The study was designed to investigate the effects of Palmul-Tang(PMT) on the change of cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD) and mean arterial blood pressure(MABP)] in normal and cerebral ischemic rats. The change of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was determinated by video-microscopy. The results in normal rats were as follows ; PMT significantly increased rCBF and PAD in a dose-dependent, and PMT increased MABP in a dose-dependent. This results were suggested that PMT significantly increased rCBF by dilating PAD. The results in cerebral ischemic rats were as follows ; Both rCBF and PAD were significantly and stably increased by PMT(10㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. The present authors thought that PMT had an anti-ischemic effect through the improvement of cerebral hemodynamics.

Effects of Acanthopanax Sessiliflorus $S_{EEM}$ Ethyl Alcohol Extract on the Cerebral Blood Flow and Mean Arterial Blood Pressure in Normal & Ischemic Rats (오갈피 에탄올 추출물이 뇌혈류 및 혈압에 미치는 효과)

  • This study was designed to investigate the effects of Acanthopanax sessiliflorus 5.,M (ASS) on the regional cerebral blood flow (rCBF) and mean arterial blood pressure (BP) in normal rats and ischemic rats. Experimental groups of rCBF and BP in normal and ischemic rats as follows ; ALE was injected ASS leaves 45% ethyl alcohol extract, ASE was injected ASS stems 45% ethyl alcohol extract, ARE was injected ASS roots 45% ethyl alcohol extract. The results were as follows ; In normal rats, ALE and ARE significantly increased rCBF in a dose-dependent manner, but ASE significantly decreased rCBF and MABP in a dose-dependent manner, ALE increased MABP In ischemic rats, rCBF was significantly and staDly improved by ARE (10 mg/kg, i.p.) during the period of cerebral reperfusion, which was contrasted with the findings of rapid and marked increase in control group. From the above results, it was thought that all part of Acanthopanax sessiliflorus $S_{EEM}$ was effective for hemodynamics and especially ARE was more effective than other parts.

Experimental Study of CheonghunHwadam-tang on the Cerebral Blood Flow in Rats (청훈화담탕이 국소뇌혈류량에 미치는 실험적 연구)

  • CheonghunHwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CheonghunHwadam-tangGamypang(CHTG) was CHT adding Aurantii Fructus(AF), Gastrodae Rhizoma(GR). The effects of CHT on the cerebral blood flow and cardiacvascular system is not known. The purpose of this Study was to investigate effects of CHT and CHTG on the regional cerebral blood flow(rCBF), mean arterial blood pressure(BP), cardiac muscle contractile force(CMF), heart rate(HR). The changes of cerebral blood flow and movement of cardiacvascular system(BP, CMF, HR) was determinated by Laser-Doppler Rowmetry(LDF). The results were as follows; 1. CHT extract increased rCBF, but decreased BP, HR in a dose-dependent manner. 2. AF extract accelerated rCBF and movement of cardiacvascular system in a dose-dependent manner. 3. GR extract increased significantly rCBF(10.0mg/kg, p<0.05) as well as accelerated BP and rCBF in a dose-dependent manner. 4. CHTG extract increased significantly rCBF(10.0mg/kg, p<0.01) in a dose-dependent, but was not changed movement of cardiacvascular system. This results suggest that CHTG is significantly treated more than CHT in vertigo by wind, fire and phlegm.

Effect of Kyungisan in on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (균기산(勻氣散)이 흰쥐의 국소뇌혈류량 및 평균혈압에 미치는 효과)

  • Kyungisan (KGS) has been used in oriental medicine for many centuries as a therapeutic agent for treatment of stroke caused by deficiency of qi(氣虛). This study was performed to evaluate effects of KGS extract on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in rats. The result of this study were as follow ; 1. KGS significantly increased rCBF irrelevant to MABP in normal rats, 2. To prescribe KGS after pretreatment with indomethacin(IDN) decreased rCBF as compared with control group to administered only KGS in normal rats. But the change of MABP is not significantly as compared with control group. 3. To prescribe KGS after pretreatment with methylen blue( MTB) decreased MABP and rCBF as compared with control group to administered only KGS in normal rats. Especially, it significantly decreased rCBF. These results suggest that KGS increase rCBF by enlargement diameter of pial artery in brain. The active mechanism of KGS is related with prostaglandin activated by cyclooxygenase. So, I suggest that KGS has an anti-ischemic effect through the improvement of cerebral blood flow and can be used for stroke.

Effects of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the Regional Cerebral Blood Flow and Mean Blood Pressure in Rats (대나무 추출액과 죽력의 혼합물이 국소 뇌혈류량 및 평균 혈압에 미치는 영향)

  • This Study was designed to investigate the effects of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal and cerebral ischemic rats. Experimental materials were as follows ; BE- 1 was Bamboo Extract (BE) extracted with 70% ethyl alcohol, BE-11 was BE extracted with distilled water at $121^{\circ}C$ for 30 min, BE-111 was BE extracted with distilled water at $121^{\circ}C$ for 3 hrs, MLC was mixture of Bambusae Caulis in Liquamen (BCL) and BE-111 mixed at the ratio of 1 to 100 (MLC100), 1 to 50 (MLC50), 1 to 20 (MLC20), 1 to 10 (MLC10), 1 to 5 (MLC5). The results were as follows , The Changes of BE- 1 on the rCBF and MABP in normal rats were not showed, BE- 11 significantly decreased rCBF in a dose-dependent manner Dut increased MABP in a dose-dependent manner. BE-111 increased rCBF in a dose-dependent manner, MLC significantly increased rCBF in a dose-dependent manner and increased MABP in a dose-dependent manner. rCBF was significantly and stably increased by MLC5 (1 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. As results above ; The present author thought that BE- 111 and MLC increased rCBF by dilating pial arterial diameter.

  • Experimental Study of Taegye($KI_3$) Taebaek($SP_3$) Draining Acupuncture on the Improvement of Cerebral Blood flow and Arterial Blood Pressure. Objectives : This study was designed to investigate the effects of acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining and determine the mechanism of action of PCBL by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. Methods : This study was designed to investigate whether acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining affect Regional cerebral blood flow(rCBF), Mean arterial blood pressure(MABP) in normal rats. To make manifest whether acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining was mediated by inhibitor of cyclooxygenase or guanylate cyclase which diate arterial diameter. Results 1. Acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining significantly increased rCBF but decreased MABP. This result suggests that Acupuncturing on Taegye($KI_3$) Taebaek($SP_3$) draining might significantly increase rCBF by dilating arterial diameter. 2. Acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining induced increase of rCBF was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. 3. This result suggests that the action of Acupuncture on Taegye($KI_3$) Taebaek($SP_3$) draining might be mediated by cyclooxygenase.

Effects of Xingjian(LR2),Shaofu(HT8) by Reinforcing and Reducing on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Normal Rats (행간(行間)·소부(少府) 보사침법(補瀉鍼法)이 국소뇌혈류량(局所腦血流量) 및 평균혈압(平均血壓)에 미치는 영향(影響))

  • Objective: The purpose of this study is to examine influences by the order, Reduction and Reinforcement in Acupuncture on cerebral hemodynamics [regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP)] in normal rats. Methods: This experiments was to to investigate eath other changes of rCBF and MABP at Xingjian(LR2)(1st) Shaofu(HT8)(2nd) Reduction, Xingjian(LR2)(1st) Shaofu(HT8)(2nd) Reinforcement, Shaofu(HT8)(1st) Xingjian(LR2)(2st) Reduction and Shaofu(HT8)(1st) Xingjian(LR2)(2st) Reduction in Acupuncture. Results: 1. LR2(1st) HT8(2nd) Reduction in Acupuncture was decreased rCBF and MABP in compared with normal condition. 2. LR2(1st) HT8(2nd) Reinforcement in Acupuncture was significantly decreased rCBF, and was decreased MABP in compared with normal condition. 3. HT8(1st) LR2(2st) Reduction in Acupuncture was decreased rCBF during acupuncture but was recovered rCBF after with-drawing of the neddle. 4. HT8(1st) LR2(2st) Reduction in Acupuncture was decreased MABP during acupuncture and after withdrawing of the neddle. 5. HT8(1st) LR2(2st) Reinforcement in Acupuncture was significantly increased rCBF during acupuncture and 30min after withdrawing of the neddle. 6. HT8(1st) LR2(2st) Reinforcement in Acupuncture was decreased MABP during acupuncture, but was recovered MABP after withdrawing of the neddle. Conclusions: I suggested that LR2 HT8 Reduction in Acupuncture and LR2 HT8 Reinforcement in Acupuncture cause a diverse response of cerebral hemodynas.

Effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley Extract on the Changes of Cerebral Flow (당귀와 백수오 추출물이 뇌혈류 변화에 미치는 영향)

  • This study was designed to investigate the effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley extract (AAC) on the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. The results were as follows. AAC significantly increased rCBF but significantly decreased MABP in a dose-dependent manner in normal rats. The increase of AAC-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and AAC-induced MABP was decreased by pretreatment with methylene blue. In cerebral ischemics, rCBF was stably improved by AAC (10 mg/kg, i.p.) during the period of cerebral reperfusion, which was contrasted with the findings of rapid and marked increase in the control group. These results suggest that AAC can increase rCBF in the normal state, as well as improve the stability of rCBF in cerebral ischemic state.
