Experimental Studied on the Second Intermediate HoBts of Clonorchis sinensis IV. Observations on the fate of metacercariae of Clonorchis sinensie in the fish hast, CuffricuEus eigenmunni (간흡충 제이중간숙주에 관한 실험적 연구 IV. 치리체내에 있어서 간흡충 피낭유충의 운명)

  • In order to judge appropriately the suitability as the second intermediate host of Clonorchis sinensis, the present experiments were proposed to survey the metacercaria of C. sinensis in the fish host, Cuztriculus eigenmanni, in the 6eld, and to observe, by experimental infection of the fish with Clonorchis cercaria, the penetrating ability of the cercaria, maturity, the process of degeneration and extinction, and infectivity of the metacercaria. The following is a brie summary of the leading facts gained through the experiments. 1. All the ten fish (mean body weight $13.4{\pm}3.439$) which were caught in Ok-ku irrigation reservoir were in acted with the metacercariae and the average number of the metacercariae found in the fish body/gram was 4.55. 2. The Bish was easily subject to invasion of the cercaria, and the examination of the cercariae after having the fish infected disclosed that 5:l hours after the infection, all of the cercariae formed their cysts in muscle and the metacercariae kept growing, that 7 days later the metacercariae were found folding their bodies twice, and that in 15 days the characteristic structure of the metacercariae was complete and made a vigorous rotary movement intermittently. 3. In the case of control fish, Pseudorasbora parka, both penetrating ability of the cercariae and the infective degree of the metacercariae to control host were stronger as compared with the 5sh host because that the control fish had higher suceptibility to C. sinensis. 4. Afterwards, the metacercariae in the fish host came to a state of maturity and beyond this stage some metacercariae started the Process of degeneration and extinction in as early as 30 days, and as more days elapsed, mortality of the metacercariae was not uniformly increased but difference of mortality appeared to be according to the individual of Ssh host. That is, in 390, 540 and 586 days, all of the metacercariae in the fish hosts became dead, meanwhile almost all of those found in the fish host of 596 days after the infection survived (mortality; 8, 51%) , Percentage of orally administered metacercariae developing to maturity in rabbit is 10.465 in the last case. 5. As the days were progressed the shapes of the metacercariae in the fish host tended to change from ellipsoid to almost round shapes, the length became shorter while the width was wider. The results shown above revealed that C. eigenmanni was, though it could not be proper host than p. larva. suitable as the second intermediate host in addition to our previous observation results.

  • 5-Substituted 4-oxo-2-thioxo-1,,2,3,4-tetrahydropyrimidine were synthesized by interaction of 4­oxo-2-thioxo-1,2,3,4-tetrahydropyrimidine-5-sulfonylhydrazide with some aldehydes to give the corresponding Schiff-bases, which after cyclization gave corresponding thiazolidinones. For some of the thiazolidinones, Mannich bases reaction was carried out. All the derivatives were tested for their possible inhibitory effect on Schistosoma mansoni cercarial elastase (CE). Only, N'-(4-methylbenzyledine)-4-oxo-2-thioxo-1,2 ,3,4-tetrahydropyrimidine-5-sulfonylhydrazide was found to have potent inhibitory effect on the CE activity with $IC_{50} = 264{\mu}M.$ Upon its use as a paint for mice tails before infection with S. mansoni cercariae, the compound formulated in jojoba oil caused a significant reduction ($93\%$; P-value = 0.0002) in the worm burden. IgG & IgM in mice sera were measured by using several S. mansoni antigens by ELISA. Sera from treated infected mice (TIM) 2, 4, and 6 weeks (W) post infection (PI) showed 1.2 folds lower, 1.2 folds higher, 1.7 folds lower IgM reactivity against soluble cercarial antigenic preparation (CAP), respectively, when compared with sera collected from infected untreated mice (IUM). Sera from TIM 2, 4, and 6WPI showed 1.3, 1.6, and 1.7 folds higher IgG reactivity, respectively against CAP than the IgG reactivity from IUM. Sera from TIM 2, 4 and 6WPI showed 1.5, 1.2 folds lower and 1.4 folds higher IgM reactivity, respectively against soluble worm antigenic preparation (SWAP) when compared with sera collected from IUM. Sera from TIM 2, 4, and 6WPI showed 1.4, 1 folds lower and 1 fold higher IgG reactivity, respectivley to SWAP when compared with sera from IUM. Sera from TIM 2, 4, and 6WPI had generaly lower IgM and IgG reactivities against soluble egg antigen (SEA) when compared with sera from IUM.

Life history of Echinopowphium recurvatum (Trematoda: Echinostomatidae) in Korea (한국에서의 오리극구흡충 (Echinoparyphium recurvatum)의 생활사)

  • The present study was performed to observe characteristics of the lifE history of Echinoparvphium recunpatum under both natural and laboratory conditions in Korea. A batch of Rcnix aunculnria corecna was collected from Sunamchon, one of the stream of West Naktonggang (River), in Kangso-gu, Pusan during August and September 1992. Out of 106 snails examined by crushing,52 (49.0%) were infected with larval E. recurvaum i.e. rediae, cercariae and metacercariae. Cercariae naturally shed from snails encysted in the snails of same species and loaches, but not in mud-snails. Adult worms were detected from chicks and ducks experimentally infected with metacercariae, but not from rats and mice. The average recovery rate of adults from chicks was 13.1%. Rediae were sac-like, $2.437{\;}{\times}{\;}0.317{\;}mm$ in average size, with a muscular pharynx and a brownish cecum which reached the anterior half of the body. Cercariae consisted of a spindle-shaped body ($0.262{\;}{\times}{\;}0.129{\;}mm$ in average) and a rod-like tail ($0.528{\;}{\times}{\;}0.056{\;}mm$ in average). In the cercarial body, 45 collar spines were observed on the head crown, and double rows of excretory ducts with fine granules were laterally arranged between the pharynnx and the ventral sucker. Metacercariae were spherical, $0.144{\;}{\times}{\;}0.142{\;}mm in average size, with thick hyaline outer and thin elastic inner walls, and many excretory granules. Adults were slender and more attenuated in the anterior end, $0.2760{\;}{\times}{\;}0.550{\;}mm$ in average size, and had 45 collar spines including four end group spines on both ventral corners. From the above results, it was confirmed that R. auriculnna corennc plays a pivotal role in the life cycle of 5. recuwatum as the first and/or second intermediate hosts in Korea.

  • Life cycle stages, including daughter sporocysts, cercariae, and metacercariae, of Parvatrema duboisi (Dollfus, 1923) Bartoli, 1974 (Digenea: Gymnophallidae) have been found in the Manila clam Ruditapes philippinarum from Aphae-do (Island), Shinan-gun, Jeollanam-do, Korea. The daughter sporocysts were elongated sac-like and 307-570 (av. 395) ㎛ long and 101-213 (av. 157) ㎛ wide. Most of the daughter sporocysts contained 15-20 furcocercous cercariae each. The cercariae measured 112-146 (av. 134) ㎛ in total length and 35-46 (av. 40) ㎛ in width, with 69-92 (av. 85) ㎛ long body and 39-54 (av. 49) ㎛ long tail. The metacercariae were 210-250 (av. 231) ㎛ in length and 170-195 (av. 185) ㎛ in width, and characterized by having a large oral sucker, genital pore some distance anterior to the ventral sucker, no ventral pit, and 1 compact or slightly lobed vitellarium, strongly suggesting P. duboisi. The metacercariae were experimentally infected to ICR mice, and adults were recovered at day 7 post-infection. The adult flukes were morphologically similar to the metacercariae except in the presence of up to 20 eggs in the uterus. The daughter sporocysts and metacercariae were molecularly (ITS1-5.8S rDNA-ITS2) analyzed to confirm the species, and the results showed 99.8-99.9% identity with P. duboisi reported from Kyushu, Japan and Gochang, Korea. These results confirmed the presence of various life cycle stages of P. duboisi in the Manila clam, R. philippinarum, playing the role of the first as well as the second intermediate host, on Aphae-do (Island), Shinan-gun, Korea.

STUDIES ON THE TREMATODE LERVAE INFECTED IN THE HARD CLAM, MERETRIX LUSORIA (대합에 기생하는 흡충유 유충의 연구)

  • Larval morphogenesis and infection rates of Cercaria pectinata Huet were investigated in regards to histological changes and mortality of the hard clam, Meretrix lusoria $(R\ddot{o}ding)$, in Buan located on the Western Coast of Korea. The trichocerous cercariae, Cercaria pectinata, and its sporocysts parasitize mainly in the tissues of goand, digestive gland and gills of the hard clams. One branched sporocyst grows into a great number of cylindrical sporocysts, and a heavily infected clam possesses 2,000 to 5,000 cylindrical spo ro cysts. Each of them produces approximately 30 to 70 germinal cells and cercariae. Of 2,639 clams examined 347 individuals $(13.2\%)$ were found to be infected. The infection rates showed seasonal variations, the rate being highest $(29.6\%)$ in July, and lowest $(2.0\%)$ in March. Whereas, the infection rates did not change significantly with the changes of size class of the shell. No infection was observed in Meretrix petachialis (Lamarck) collected from Myeong-ji, Kimhae Gun located near Busan. In the iefected clams, the gametogenesis (oogenesis and apermatogenesis) did not undergo completely, therefore the heavily infected clam seemed to be castrated. Also the tissues of the digestive gland and gills were compressed by the multiplication of the parasites. When they were kept in aquariums in the laboratory, high mortality has occurred in the infected clams. All of the 60 infected clams died within 35 days.

Population Dynamics of Intermediate-Host Snails in the White Nile River, Sudan: A Year-Round Observational Descriptive Study

  • We aimed to explore the population dynamics of snail in 3 sites of the White Nile in Sudan. More specifically, we aimed to investigate the annual patterns of snail populations that act as intermediate hosts of schistosomes and monthly snail infection rates and ecological characteristics presumably related to snail populations. We collected snails for 1 year monthly at 3 different shore sites in the vicinity of El Shajara along the White Nile river in Khartoum State, Sudan. In addition, we measured air and water temperatures, water turbidities, vegetation coverages, and water depths and current speeds. Most of the collected snails were Biomphalaria pfeifferi and Bulinus truncatus. The population densities of snails and their infection rates varied across survey sites. The collected snails liberated S. mansoni and S. haematobium cercariae as well as Amphistome and Echinostome cercariae. Infected snails were found during March-June. The ecological characteristics found to be associated with the absence of snails population were: high turbidity, deep water, low vegetation coverage (near absence of vegetation), high water temperature, and high current speed. To our knowledge, this is the first longitudinal study of the snail population and ecological characteristics in the main basin of the White Nile river.

A Study on the Intermediate Hosts of Paragonimus westermani in Bogil Islet, Chonra-Nam-Do, Korea (전라남도 완도군 보길도에서의 폐흡충(paragonimus westermani) 중간숙주 조사)

  • Collections of Semisulcospira snails and crayfishes were made to detect out larval trematodes in Bogil Islet, Wando-Gun, Chonra-Nam-Do, Korea in August, 1985. In addition, intradermal test to figure out the prevalence rate of paragonimiasis in the islanders was also carried out. The results are summarized as follows: 1) Nineteen out of 186 examinees(10.2%) showed positive reactions by intradermal test with Paragonimus antigen. 2) Forty four snails out of 182 snails collected were infected with trematode cercariae. These larvae were Metagonimus sp., Cercaria yoshidae, Cercaria incerta and Cercarianipponensis. 3) Metacercarial incidence of Paragonimus westermani in crayfish was 65.4%. Mean number of metacercariae per crayfish was 6.5.

Growth and development of Fibricola seoulensis metacercariae in tadpoles (Fibricola seoulensis 피낭유충의 실험감염 올챙이내 성장 및 발육)

  • In order to observe the growth and development of Fibricola seoulensis metacercariae, the tadpoles of Rana nigromaculata were experimentally infected with the cercariae. The meta cercariae of various developmental stages were recovered from the tadpoles after 2 to 65 days of infection. They were prepared for morphological observation, and were given orally to mice to observe their infectivity. The following results were obtained. 1. All of the tadpoles exposed to the cercariae were observed to harbour the larvae in their abdominal cavity. 2. The young metacercariae of 2 days after infection were $121.1{\mu}m$ long and $63.3{\mu}m$ wide. They grew linearly for the first 14 days to be $262.0{\mu}m$ long and $166.4{\mu}m$ wide. Thereafter, no more growth recognized until 65 days. 3. The larvae of 2 days old were similar with cercarial body and had 2 suckers, a pharynx, 2 ceca and a primordium of germ cells but no tribocytic organ. On the 8th day, they had tribocytic organ, and their morphology resembled that of mature metacercariae. 4. The metacercariae younger than 10 days could not infect the mice. Only the metacercariae older than 14 days had infectivity. The recovery rates increased by the age of metacercariae from 19.0% in 14 days old to 70.0% in 40 days old. Above findings indicate that the tadpole is indispensable for metacercarial development and it needs at least 2 weeks for maturation. The tadpole is a pivotal host in the life cycle of F. seoulensis for connection between the snail and the frog.

Studios on the Metagonimus fluke in the Daecheong Reservoir and the upper stream of Geum River, Borea (대청호 및 그 상류의 Metagonimus 흡충에 관한 연구)

  • The prevalences of the cuke belonging to genus Metagonimus hove been reported along the upper stream of inhabitants by several workers since 1980, however the taxonomical problems of the fluke was not yet settled. The larval flukes; cercaria and metacercaria as well as their intermediate hosts, and adult were studied in order to identify the Mepagonimus in the areas. The results obtained are summarized as follows: 1. The snails, Semisulcospira globus were collected (rom the three different localities along the upper stream of the River. The cercariae were found from 125(7.2%) out of 1,730 snails by natural emerging method, and were identified into 5 species including Metagenimus sp. (3.7%), Pseudexorchis major(1.4%), Cercaria nipponensis (0.9%), Cercaria incerpa(0.6%), and Cercaria yoshidae(0.6%). Cercariae of Metagonimus species had four to dye oral spines on its anterior of the first line. 2. The cercariae of Metagonimus were experimentally exposed to goldfish. nfection rate was 22.9% out of 105 goldfish, and the encysted metacercariae were found in fins(86.7%) and on scales (13.7%) of the fishes, but not in their muscle, head or visceral organs. 3. Seven species of ask were caught in the Daecheong Reservoir and the upper stream. Infestations with metacercaria of Metagonimus were found 100% in Opsariichtys widens and the parasitized numbers of the metacercariae were observed from 250 to 2,400 per fish. In the upper stream, Zacco termmincki, Z. platypus and Pseudogobio esocinus were infected 100% with the metacercaria, on the other hand, the fishes caught in the reservoir showed the lower infestation rates, and a few metacercariae found in the fishes Carassius carassius and Cyprinus carpio in the reservoir and the stream. The majority of metacercariae was detected only on the scales of fishes. 4. In order to know the infectivity and the distribution patterns in the intestine of hosts, rats and dogs were infected with the metacercariae obtained from O. bidens and Z. platypus. In addition the metacercariae obtained from Z. temmincki, P. esocinus and goldfish were given to the rats. The recovery rates of the worms in the small intestine of dogs were higher (63.3~65.8%) than those of the rats (3.5~31.6%). The flukes were found mostly in the middle and the lower part of small intestine of the rats and the dogs, but no worm was collected in the upper part of the intestine of rats. 5. The sixte of adult flukes varied by the hosts. In the adult cukes, oral sucker was smaller than ventral sucker, and the right and left testes were located diagonally, the uterine tubules circled around the upper left testis. The average egg sixte was $29.1{\times}1.7{\mu\textrm{m}}$. According to the above results, the cukes belonging to genus Metagonimus distributed along the Geum River was concluded to be identical with Miyata type of M. yokogawai as that Saito had proposed.

Cercaria caribbea LVIII Cable, 1963 (Digenea: Cyathocotylidae) in the Republic of Korea and Its Surface Ultrastructure

  • Cercaria caribbea LVIII Cable, 1963 (Digenea: Cyathocotylidae) was detected from a brackish water gastropod species (Cerithideopsilla cingulata) in a coatal area of Shinan-gun, Jeollanam-do (Province), the Republic of Korea, and its surface ultrastructure was studied using a scanning electron microscope. The cercariae were found freely swimming or enveloped within daughter sporocysts when the snail host was mechanically broken. They were morphologically characterized by a linguiform and ventrally concave body, a long and bifurcated tail, and the presence of a holdfast (=tribocytic) organ posterior to the ventral sucker. On the whole ventral and dorsal surfaces, peg-like tegumental spines were densely distributed. Around the oral sucker, several sensory papillae, each with a short cilium, were distributed, and on the tail, sensory papillae, each with an extensively long cilium, were observed. This is the first record describing a cyathocotylid cercaria from a brackish water gastropod in the Republic of Korea.
