The antiproliferative activity of cannabidiol ethyl ethers against human ora epitheloid carcinoma cells

  • Cannabidiol derivatives (1, 2 and 3), and 5-fluorouracil (4, 5-FU) were tested for their growth inhibitory effects against human oral epitheloid carcinoma cell lines (KB) using two different 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and sulforhodamine B protein (SRB) assay. These compounds showed a potent inhibitory activity in vitro in the micromolar range against KB cell lines. In general, the antitumor activity of these compounds (1, 2, 3 and 4) was in a dose-dependent over the micromolar concentration ranges from $1\;{\mu}M\;to\;100\;{\mu}M$. The comparison of $IC_{50}$ values of these compounds in tumor cell lines shows that their susceptibility to these compounds decreases in the following order: CBD > 5-FU > CBDME > CBDDE by the MTT assay and SRB assay. Cannabidiol derivatives (1, 2 and 3), and 5-FU were tested for their cytotoxic effects on NIH 3T3 fibroblasts using two different MTT assay and SRB assay. These compounds exhibited potent cytotoxic activities in vitro in the micromolar range against NIH 3T3 fibroblasts. In general, the cytotoxic activities of these compounds (1, 2, 3 and 4) were in a dose-dependent over the micromolar concentration range $1\;{\mu}M\;to\;100\;{\mu}M$. The comparison of $CD_{50}$ values of these compounds on NIH 3T3 fibroblasts shows that their susceptibility to these compounds decreases in the following order; CBD > 5-FU > CBDDE > CBDME by MTT assay, CBD > 5-FU > CBDME > CBDDE by SRB assay. These results suggest that cannabidiol (1, CBD) retains the most growth-inhibitory activity against KB cell lines.

Gibberellic acid에 의한 chlorella의 생장과 호흡 및 투과성과의 관계 (Relationship between growth, respiration and permeability of chlorella cell treated with gibberellic acid)

  • Effect pf gibberellic acid (GA) on the growth rate, respiratory activity and solute uptake of Chlorella cells were measured and their correlation were discussed. Growth rate and respiratory activity of the algal cells are enhanced considerably by very samll amount (50 ppm) of GA treatment although they are suppressed by relatively higher concentration more than 100 ppm. Phosphate uptake of the algal cells, however, decreased even though lower concentration of GA is applied. Thereforem it is inferred that the growth enhancement of the algae by GA is not due to the increase of the permeability of the algal cells but expansion growth owing to the increase of osmotic pressure caused by the increase of hydrolase activity of the algae.

Effect of 2-NBDG, a Fluorescent Derivative of Glucose, on Microbial Cell Growth

  • A fluorescent glucose analogue,2-[N-(7-nitrobenz-2-ox a-1,3-diazol-4-yl) amino] -2- deoxy-D-glucose (2-NBDG), which had previously been developed for the analysis of glucose uptake in living cells, was investigated to determine its biological activity on microorganisms.2-NBDG did not show any inhibitory effect on growth of yeast cells and bacteria. In contrast, 2-NBDG exhibited strong inhibitory effects on filamentous fungal growth. The growth of filamentous fungi was completely inhibited, when 2-NBDG was supplemented as sole carbon source. The inhibitory effect was decreased by the addition of glucose in the test medium. Furthermore, 2-NBDC inhibited chitinase activity of Trichoderma sp. These results suggested that the inhibitory effects of 2-NBDG on filamentous fungi might be partially due to the inhibition of chitinase.

재조합효모 배양에서 비이온성 계면활성제가 외래 Glucoamylase 생산 및 분비에 미치는 영향

  • The effects of non-ionic surfactants (Triton X-100 and Tween 80) on cloned glucoamylase production and secretion in recombinant Saccharomyces cerevisiae culture were studied. Even though the extracellular glucoamylase activity was increased by addition of Tween 80 due to the increase of the cell mass, Tween 80 did not play a role in the increase of glucoamylase secretion. On the addition of Triton X-100 addition, the secretion efficiency was increased while the cell growth was inhibited. Triton X-100 was added to the culture broth after 24 hr of culture to minimize the inhibition of the cell growth, and consequently the glucoamylase activity in the culture broth was increased by 12%.

  • In a continuous effort to develop novel anticancer agents we newly synthesized and evaluated the antitumor activity of nucleoside analogues. One analogue, 4 - [2-Chlor-6- (3-iodo- benzy lamino) -purin -9-yl]- 2,3-dihydroxy-cyclopentanecarbo xylic acid methylamide (LJ-331), has been shown to exert a potent inhibition of human cancer cell growth in vitro including human lung (A549), stomach (SNU-638) and leukemia (HL-60) cancer cells. Following mechanism of action study revealed that LJ - 331induces cell cycle arrest at the G1 phase in HL-60 cells and evokes apoptotic phenomena such as an increase in DNA ladder intensity and chromatin condensation by a dose- and time-dependent manner. LJ-331 also activated the caspase-3 activity in HL-60. This result suggests that the growth inhibition of human cancer cells by LJ-331 might be related to the cell cycle arrest and induction of apoptosis.

  • 토양에서 분리한 알칼리내성 Bacillus sp.의 자체 chromosomal DNA에서 유래된 strong promoter를 지닌 plasmid p-12B1을 공여균주에 삽입시키고 이 promoter의 활성을 최적화할 수 있는 배양조건을 검토하였다. 글루코오스가 고갈된 시점부터 세포의 활성은 유지되나 포자는 형성되지 않을 정도로 낮은 글루코오스 소비속도를 유지해 제한된 글루코오스를 연속적으로 공급해줌으로써 promoter의 활성을 최대로 높일 수 있었다. 또한 배지조성의 변화로 균체를 생육시킨 후 유전자 발현을 유도하는 것이 가능하였으며, 이 점에 대해서는 보다 구체적인 연구가 진행되어야 할 것이다.

Enhancement of Biocontrol Activity of Serratia plymuthica A21 -4 Toward Phytophthora Blight of Pepper by Amendment of Nutritional Condition

  • Serratia plymuthim A21-4 strongly inhibits the mycelial growth, zoospore formation, and cystospore germination of Phytophthor spp and Pythium species. The bacterial isolate produced antifungal substance and chitinase. The bacteria also enhanced to plant growth remarkably in low nutritional condition. The application of cell suspension of A21-4 to pepper seedlings in greenhouse experiments and soil drenching in farmer's field was proved successfully to control the phythophthora blight of pepper. For the effective control, however, relatively high density of cell number(10$\^$9/cfu/$m\ell$) is required. Density effect was similar in plant growth promoting activity of A21-4. Though this investigation we improved the problem with changes of culture condition of bacteria and some nutritional amendment.

  • Spermidine, spermine and IAA promoted an increase in $\beta$-1, 4-endoglucanase activity in hypocotyls of Glycine max. The optimal concentration for the increase of the enzyme activity was 10-6 M for spermidine, 10-8 M for spermine and 10-6 M for IAA. However, IAA had innocuous effect on arginine decarboxylase and ornithine decarboxylase activites, and the content of polyamine. Such cumulative results suggest that the increase in $\beta$-1, 4-endoglucanase activity by IAA is not attributed by the effect on the biosynthesis of polyamine by IAA but spermidine, spermine and IAA induce cell wall loosening and therefore extension growth of cells.

Enoylpyruvate Transferase Isozymes in Bacillus megaterium

  • UDP_GlcNAc is metabolized to form vegetative cell wall, cortical peptidoglycans, and outermost layer consisting of galactosamine-6-phosphate ploysaccharide in life cycle of Bacillus megaterium. To obtain a better understanding of the UDP-GlcNAc regulation, we examined the activity of the common first enzyme for the synthesis of nucleotide precursors of peptidoglycans, enoylpyruvate transferase by newly developed method. Both the specific and the total activity decreased after the end of exponential growth followed by and increase from t5 but decreased again parallel to the appearance of the activity of UDP_GlcNAc-4-epimerase. Antibody specificity to anti-transferase IgG and the elution profile on DEAE-Sepharose revealed that B. megaterium has at least two enoylpyruvate transferase isozymes, and UDP_GlcNAc was metabolized to vegetative cell wall and cortical peptidoglycan by each isozme in exponential growth and in sporulation, respectively in life cycle.

키틴과 파추출액 반응물의 항암 작용 (Antitumor Activity of Reaction Mixture of Chitin and Green Onion Extract)

  • Antitumor activity was tested by administration of reaction mixture of green onion extract and chitin to mice bearing sarcoma-180 cells. An intraperitoneal injection of mixture(20 mg/kg/day) to mice Have an 52% inhibition of tumor growth. Inhibition of tumor growth was found to be dose-dependent. When eighty miligrams of the mixture were administered, the weight of tumor was reduced significantly. HPLC analysis indicated the mixture was composed of N-acetyl-D-glucosamine, N-acetylchitobiose and N-acetylchitotriose.
