Microstructural Characteristics of the Ordered and Disordered Leaves in Citrus junos Sieb.

  • We compared microstructural features of the ordered cell and disordered leaves in Citrus junos Sieb. by electron microscopy. In the cell of the ordered leaves, many chloroplasts and large vacuoles were particularly observed. Also a lot of vessel, companion cell and big nucleus were presented in vascular bundle regions. The mitochondria and the other organelles were interspersed among the chloroplasts in a thin, peripheral layer of cytoplasm. The chloroplast possessed typical grana and intergranal lamellae, numerous starch grains and a few small osmophilic globules. Besides, microbodies were closely associated with the mitochondria and the chloroplast. The process of the formation of the secondary cell wall from primary cell wall was observed the vessel elements, the tonoplast wall and the secondary cell wall. It was observed that the oil sac with the unique perfume distributed the adjacent cell wall. In the cell of disordered leaves, the all of the organelles were thrust toward the cell wall due to the fusion of vacuoles in the cells. It was observed that a lot of the very small particles spreaded in the cytoplasm. The loss of unique perfume of the leaves was resulted in the destruction of the oil sac. Also, there was not observed grana, lamellae, starch and osmophillic globules in the chloroplast. The small distributed organelles was not observed but the elongation of the cell wall was proceed no longer. Therefore, the plasma membrane diverged from the cell wall. All of organelles in the cell had poor function and deformation. A massive vacuole was fulfilled in single cell and the vacuole contains a lot of large and small particles. The organelles were presented on the side of the cell wall according to the enlargement of vacuole and they were observed to be breakdown.

Influence of Plant Growth Substances on Cytolysome-like Organelles in the Mesophyll Cells of Soybean (식물생장소가 대두 엽육세포의 Cytolysome-like Organelle에 미치는 영향)

  • Leaf tissue of Glycine max Merr. was fixed in para-formaldehyde-glutarldehyde and postfixed in osmium tetroxide or postassium permanganate for electron microscopy. The origin of cytolysome-like organelles of mesophyll cell was studied and changes of fine structure of the organelles according to treating solutions such as gibberellin (GA), kinethin (KI), 2,4-dichlorophenoxy acetic acid(2, 4-D) or 2, 4-D+GA(1mg/l, respectively) were observed. The cylolysome-like organelles differentiate in endoplasmic reticulum and plasmalemma, and they drop into vacuoles being isolated from the formers. They seem to change into myelin-like structure and to be degenerated by autodigestion. Cytolysome-like organelles involved in cell walls and vacuoles showed activity of acid phosphatase. In the group of GA and KI treatment, cytolysome-like organelles were similar to that of the control group. But in the treatmental groups of 2,4-D and 2,4-D+GA, myelin-like structures increased in size and autodigestion of this organelles were similar to that of the control group. But in the treatmental groups of 2,4-D and 2,4-D+GA, myelin-like structures increased in size and autodigestion of this organelle seemed to be accelerated. In the treatmental group of 2,4-D+GA, myelin-like structures shown high electron density were observed in cytoplasm and vacuoles together.

  • Label-free optical diffraction tomography (ODT), an imaging technology that does not require fluorescent labeling or other pre-processing, can overcome the limitations of conventional cell imaging technologies, such as fluorescence and electron microscopy. In this study, we used ODT to characterize the cellular organelles of three different stem cells-namely, human liver derived stem cell, human umbilical cord matrix derived mesenchymal stem cell, and human induced pluripotent stem cell-based on their refractive index and volume of organelles. The physical property of each stem cell was compared with that of fibroblast. Based on our findings, the characteristic physical properties of specific stem cells can be quantitatively distinguished based on their refractive index and volume of cellular organelles. Altogether, the method employed herein could aid in the distinction of living stem cells from normal cells without the use of fluorescence or specific biomarkers.

  • The hemocytes of Drosophila melanogaster were observed with electron microscope, and five types of the cells were identified; prohemocyte, plasmatocyte, granular cell, crystal cell and oenocytoid, accounting for about 5%, 35%, 45%, 10%, 5% respectively of total cell numbers. Prohemocytes are characterized by a low concentration of intracellular organelles. Plasmatocytes are spindle or oval in shape and have relatively plenty of organelles and lysosomes. Granullar cells are the most polymorphic. They have numerous pseudopod-like projections and contain various granules and inclusions. In this cell type, intracellular organelles are fully developed. Crystal cells are characterized by numerous crystals composed of fine granules arranged regularly. Oenocytoids are the largest one among all cell types and contain relatively developed organelles.

  • A sequential sub-cellular study of the epicarp of Nerium indicum has been performed. Outer epidermis of the ovary wall is covered externally with a thin cuticle. Cytoplasm possesses most of the cell organelles in the ovary stage itself. Outermost zone of the pericarp is the epicarp, developing from the outer epidermis. In the developing fruit, cell organelles are found with its maximum intensity. In mature fruit, the epicarp becomes multilayered due to additional development of few collenchymatous cells close to the outermost layer. Epicarpic cell possesses large central vacuole, around which a thin layer of cytoplasm is present. Number of cell organelles are considerably reduced in the mature fruit. In the ovary stage starch grains are electron transparent, while in the mature fruit it is fruit it is electron transluscent.

  • Everything in the surrounding universe can be attributed into five elements. Human organs can be also linked to the five elements. Cells, the smallest unit of the human body, consist of cellular organelles as little organs. Here, we extended the concept of the five elements to a cellular level via the human organs, theoretically re-evaluating the overall association of cellular organelles in maintaining the homeostasis of cellular functions.

Ultrastructural Study on Induced Resistance of Cucumber Plants against Sphaerotheca fuliginea by Oligochitosan

  • The induced resistance of cucumber leaves treated with oligochitosan to the infection of the cucumber powdery mildew, Sphaerotheca fuliginea, was investigated using transmission electron microscopy. The results showed that when the plants were treated with oligochitosan and challenged with inoculum, a significant decrease of the disease occurred. The mycelial development in the treated leaves was markedly inhibited. The cytoplasm of the powdery mildew mycelium was aggregated, with its organelles disintegrated and the cytoplasm collapsed. The protoplasm in haustoria became electron-dense. Haustoria became malformed, their organelles disintegrated, the hausterial wall thickened and eventually the whole complex necrotized. The host cells produced defence structures and materials associated with infection and a hypersensitive response. The host cell wall was thickened and deeply stained; several layers of papilla structure were produced under the cell wall; dark materials were deposited between the cell wall and plasmalemma; extrahaustorial plasmalemma was deeply stained and extrahaustorial matrix appositions had large deposits of electron-dense material; the cytoplasm was disordered, host organelles disintegrated and eventually the whole host cell disintegrated and necrotized.

Effects of Moschus, Bovis Calculus, Ursi Fel Aqua-Acupuncture on Liver Damage (사향(麝香).우황(牛黃).웅담(熊膽) 약침(藥鍼)이 간손상(肝損傷)에 미치는 영향(影響)에 관한 연구(硏究))

  • In order to investigate experimentally that effect of Moschus, Bovis Calculus, Ursi Fel aqua-axupuncture on acutely damaged liver of rats induced by radix aconiti. The author admininisterated aqua-acupuncture according to method of manufacture stimulation to corresponding points, Kan-su$(BL_8)$ and Ki-mun($LR_{14}$), and carried out hematological, serological examination and electromicroscopical obseration. WBC level was decreased significantly in the experimental groups at 24 and 48 hours as compared with the control group. ${\gamma}-GTP$ level was decreased significantly in the experimental groups at 24 and 48 hours as compared with the control group. GPT level was decreased significantly in the experimental groups at 24 and 96 hours as compared with the control group, but GOT level did not reeveal any statistical difference. At 24-hours group, the cell organelles of the hepatic cells are destructed. The cisternae of the smooth endoplasmic reticulum are particulary dilated in the 24-hours control group compared with experimental group. At 48-hours group, the destructed cell organelles are recovered in the experimental group compared with control group. At 96-hours group, the cell organelles of hepatic cells are showing feture of normal group. According to the above findings, it is considered that Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture has effects of recovery of acutely damaged liver.

  • In order to investigate experimentally that effect of Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture on acutely damaged liver of rats induced by radix aconiti, the author gave Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture according to method of manufacture stimulation to corresponding points, Kan-su (B18) and Ki-mun (Liv14), and carried out hematological, serological examination and electromicroscopical observation. The following results were obstained : 1. At 6 hours, cell organelles induced by radix aconiti toxicity prominently destructed in control and aqua-acupuncture treatment groups. 2. At 12 hours, cell organelles, destruction was some similarities between control group and aqua-acupuncture treatment group. 3. At 24 hours, rough endoplasmic reticulum and Golgi complex comparatively developed in aqua-acupuncture group treatment as compared with control group. 4. At 48 hours, cell organelles, destruction was recovered in aqua-acupuncture treatment group as compared with the control group and it was similiar normal findings. According to the above findings, it is considered that Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture has effects of recovery of acutely damaged liver.

A Morphologic Study on the Differentiation of Chicken Embryo Lens (계배(鷄胚) lens의 분화(分化)에 관(關)한 형태학적(形態學的) 연구(硏究))

  • Embryonic and postembryonic chicken lenses have been analyzed morphologically to investigate the differentiation of the lens fibers by light and electron microscopes. Morphogenesis of the chick lens was initiated as lens epithelial cells were proliferated and proceeded to elongate the cells characteristically at posterior side, by which the disintegrations of nuclei were accompanied during the early developmental stages. Primary and secondary lens fibers were identified at the late developmental stages, while interconnections between neigh-boring cells well developed and denucleation commenced. On day of hatching, the chicken lens fibers contained few cell organelles within the cytoplasm and showed the homogeneity of cytoplasmic appearance. On day 10 of hatching, the lens were fully differentiated; fiber cells, in which most cell organelles except polysomes were disappeared, showed a slender and elongated prismatic shape. At that stage gap junctions were particularly developed or cytoplasmic ridges are closely interlocked between adjoining cells. In conclusion, differentiation of chick lens involves the division of epithelial cells, the elongation into fiber cells, the loss of cell organelles and the increase of gap junction.
