Deformation Measurement of Polymer Scaffold Using Particle Image Analysis (입자 영상 해석을 이용한 고분자 지지체 변형 측정)

  • Polydimethylsiloxane (PDMS) is used as a scaffold for cell culture. Because both the stress and strain acting on the substrate and the hemodynamic environment are important for studying mechano-transduction of cellular function, the traction force of the surface of a substrate has been measured using fluorescence images of particle distribution. In this study, deformation of the cross-sectional plane of a PDMS block was measured by correlating particle image distributions to validate the particle image strain measurement technique. Deformation was induced by a cone indentor and a shearing parallel plate. Measured deformations from particle image distributions were in agreement with the results of a computational structure analysis using the finite-element method. This study demonstrates that the particle image correlation method facilitates measurement of deformation of a polymer scaffold in the cross-sectional plane.

  • A numerical analysis of the micro-discharge in an AC pplasma dispplay cell has been made using time-deppendent, 2-dimensional multi-fluid equations to understand the discharge pphysics of He-Xe discharge. The time deppendent distribution of the electron tempperature, densities of electrons, various ions and excited sppecies, and the effects fo the wall charge accumulated on the dielectric surface are obtained and comppared with the results of direct observation of time deppendent behavior of VUV and visible sppectra from single discharge cell observed using a gated, image intensified CCD to elucidate the discharge physics.

Pyramidal Deep Neural Networks for the Accurate Segmentation and Counting of Cells in Microscopy Data

  • Cell segmentation and counting represent one of the most important tasks required in order to provide an exhaustive understanding of biological images. Conventional features suffer the lack of spatial consistency by causing the joining of the cells and, thus, complicating the cell counting task. We propose, in this work, a cascade of networks that take as inputs different versions of the original image. After constructing a Gaussian pyramid representation of the microscopy data, the inputs of different size and spatial resolution are given to a cascade of deep convolutional autoencoders whose task is to reconstruct the segmentation mask. The coarse masks obtained from the different networks are summed up in order to provide the final mask. The principal and main contribution of this work is to propose a novel method for the cell counting. Unlike the majority of the methods that use the obtained segmentation mask as the prior information for counting, we propose to utilize the hidden latent representations, often called the high-level features, as the inputs of a neural network based regressor. While the segmentation part of our method performs as good as the conventional deep learning methods, the proposed cell counting approach outperforms the state-of-the-art methods.

  • LiDAR(Light Detection and Ranging) with digital aerial photograph can be used to measure tree growth factors like total height, height of clear-length, dbh(diameter at breast height) and crown projection area. Delineating crown is an important process for identifying and numbering individual trees. Crown delineation can be done by watershed method to segment basin according to elevation values of DSMmax produced by LiDAR. Digital aerial photograph can be used to validate the crown projection area using LiDAR. And tree height can be acquired by image processing using window filter$(3cell\times3cell\;or\;5cell\times5cell)$ that compares grid elevation values of individual crown segmented by watershed.

  • Cell type packaging line is more suitable for the products with various models and small quantities like mobile phone or mp3 player than conveyor type packaging line. Cell type packaging line is applicable to package various product models, but it can cause wrong product compositions and missing of items. So, automatic missing item detection system is needed. We designed an missing item detection system with a bar code reader, infrared sensors, and s digital camera. and also developed the programs for sensor data acquisition, image data processing, GUI, and data management.

  • Recently, great advance is achieved in the field of genome-wide functional screening using cell-based assay. Here, we briefly introduce well-established and typical cell-based assays of GPCR and some parameters which should be considered for genome-wide functional screening. Because of characters and importance of GPCR as drug targets, several ways of assay systems were devised. Among them, high-content screening (HCS) that is based on the analysis of image by confocal microscope is becoming favorite choice. The advances in this technology have been driven exclusively by industry for their convenience. Now, it is turn for academy to define more detail signaling networks via HCS using cDNA or siRNA libraries at genome-wide level. By isolating novel signaling mediators using cDNA or siRNA library, and postulating them as new candidates for therapeutic target, more understanding about life science and more increased chances to develop therapeutics against human disease will be achieved.

  • In gravure printing, the amount of ink from cells has a great effect on the qualities of final printed products. And printability of final products is determined by every kinds variables. Ink transfer process is not verified scientifically because gravure cell is of small size and print speed is rapid. Therefore in order to study of ink transfer mechanism, this study is using the Computational Fluid Dynamics Evaluation. Polyflow 3-10 simulation software is used for considering of non-Newtonian flow. Among the various factors, this study have dealt with gravure cell types used computer simulation in order to define distinctive features in ink flow and transfer. The results of simulation, it defined the distribution of pressure, speed, stream function, viscosity, shear rate during the gravure printing. It is fined out the difficulties and characteristics according to the shape of cell types. Through this study, the condition of gravure printing is depending on the print condition and characteristic of cells.

Cytotoxicity of Multipurpose Contact Lens Solutions on the Cultured Corneal Epithelial Cells Evaluated by Image Analysis (이미지 분석법을 이용한 소프트 콘택트렌즈용 다목적용액의 각막상피세포 독성 평가)

  • Purpose: To determine the effect of marketed multipurpose contact lens solutions (MPSs) on human corneal epithelial cells (HCEpiCs) toxicity by using image analysis. Methods: HCEpiCs were exposed six MPSs (product A-F) at 0.05~50% for 2h, 12h, 24h, and 48h respectively. HCEpiCs were fixed and stained with Draq5 after exposure with MPSs, and the cell viability and apoptosis were evaluated by using confocal microscope and ImageXpress UltraTM. Results: Viabilities of HCEpiCs exposed to MPS A-F for a 2h were not affected, while reductions (52~75%) in cell viability over a 12h exposure of MPS B, MPS C, MPS D and MPS F, and significant more reductions (29~73%) over a 24h and 48h-exposure. Apoptosis of HCEpiC was not affect over a 12h MPS exposure, however was significantly increased (199~526%) over 24h and 48h MPS exposure. Among the products MPS D, E and F reduced viability of HCEpiCs and apoptosis increased more than MPS A (p<0.05). Conclusions: Lower concentration of MPSs have not an cytotoxic effect on HCEpiCs, however higher concentration of MPSs induce apoptosis and reduce viability of HCEpiCs. Therefore, it need to develop MPS having antimicrobial effectiveness with low cytotoxicity.

Simultaneous velocity and temperature measurement of thermo-fluid flows by using particle imaging technique (화상처리기법을 이용한 온도장 및 속도장 동시 측정기법 개발)

  • A quantitative flow visualization technique was developed to measure velocity and temperature fields simultaneously in a two-dimensional cross section of thermo-fluid flows. Thermochromic liquid crystal(TLC) particles are used as temperature sensor and velocity tracers. Illuminating a thermo-fluid flow with a thin sheet of white light, the reflected colors from the TLC particles in the flow were captured simultaneously by two CCD cameras; a 3-chip CCD color camera for temperature field measurement and a black and white CCD camera for velocity field measurement. Variations of temperature field were measured by using a HSI true color image processing system and TLC solution. The relationship between the hue values of TLC color image and real temperature was obtained and this calibration curve was used to measure the true temperature under the same camera and illumination condition. The velocity field was obtained by using a 2-frame PTV technique using the concept of match-probability to track true velocity vectors from two consecutive image frames. These two techniques were applied at the same time to the unsteady thermal-fluid flow in a Hele-Shaw cell to measure the temperature and velocity field simultaneously and some results are discussed.

  • As the modern techniques have improved, people intend to store and manage the information on the web. Especially, it is the image data that is given a great deal of weight of the information because of the development of the scan and popularization of the digital camera and the cell-phone's camera. However, most image retrieval systems are still based on the text annotations while many images are creating everyday on the web. In this paper, we suggest the new approach for the semantic image retrieval using the RDF metadata based on the representation of the spatial relationships. For the semantic image retrieval, firstly we define the new vocabularies to represent the spatial relationships between the objects in the image. Secondly, we write the metadata about the image using RDF and new vocabularies. Finally. we could expect more correct result in our image retrieval system.
