Gallic acid caused cultured mice TM4 Sertoli cells apoptosis and necrosis

  • Objective: The study was designed to determine the cytotoxic effect of gallic acid (GA), obtained by the hydrolysis of tannins, on mice TM4 Sertoli cells apoptosis. Methods: In the present study, non-tumorigenic mice TM4 Sertoli cells were treated with different concentrations of GA for 24 h. After treatment, cell viability was evaluated using WST-1, mitochondrial dysfunction, cells apoptosis and necrosis was detected using JC-1, Hoechst 33342 and propidium iodide staining. The expression levels of Cyclin B1, proliferating cell nuclear antigen (PCNA), Bcl-2-associated X protein (BAX), and Caspase-3 were also detected by quantitative real-time polymerase chain reaction and Western-blotting. Results: The results showed that 20 to $400{\mu}M$ GA inhibited viability of TM4 Sertoli cells in a dose-dependent manner. Treatment with $400{\mu}M$ GA significantly inhibited PCNA and Cyclin B1 expression, however up-regulated BAX and Caspase-3 expression, caused mitochondrial membrane depolarization, activated Caspase-3, and induced DNA damage, thus, markedly increased the numbers of dead cells. Conclusion: Our findings showed that GA could disrupt mitochondrial function and caused TM4 cells to undergo apoptosis and necrosis.

  • Platelet-derived growth factor (PDGF) is a critical regulator of mesenchymal cell migration and proliferation. The vital functions of PDGFs for angiogenesis, as well as development of kidney, brain, cardiovascular system and pulmonary alveoli during embryogenesis, have been well demonstrated by gene knock-out approaches. Clinical studies reveal that aberrant expression of PDGF and its receptor is often associated with a variety of disorders including atherosclerosis, fibroproliferative diseases of lungs, kidneys and joints, and neoplasia. PDGF contributes to cancer development and progression by both autocrine and paracrine signaling mechanisms. In this review article, important features of the PDGF isoforms and their cell surface receptor subunits are discussed, with regards to signal transduction, PDGF-isoform specific cellular response, and involvement in angiogenesis, and tumorstromal interactions.

  • The purpose of this study is to research and develop PEO/PVDF electrolytes and $LiMn_2O_4$ composite cathode for all-solid state lithium rechargeable battery. We investigated AC impedance response and charge/discharge cycling of $LiMn_2O_4$/SPE/Li cells. The cell resistance was decreased so much initial charge process from 0% SOC to 100% SOC. The radius of semicircle of $LiMn_2O_4$/SPE/Li cell was so much from initial state to 20th cycling. The discharge capacity of the $LiMn_2O_4$ composite cathode was 144mAh/g based on $LiMn_2O_4$.

High-brightness and wide-view transflective liquid crystal display with two in-cell imprinted optical films in an inverse-twisted-nematic geometry

  • An inverse-twisted-nematic (ITN) transflective (TRF) liquid crystal (LC) display, where two imprinted optical films (IOFs) with surface microstructures are embedded was developed. One of the IOFs serves as an in-cell patterned retarder with multioptic axes, and the other behaves as a viewing-angle enhancement film. In the presence of an applied voltage, the surface microstructures on the IOFs provide the spontaneous twist of the LC from a vertically aligned state to a $90^{\circ}$ twisted-nematic (TN) state in the transmissive part, and to a $45^{\circ}$ TN state in the reflective part. The developed ITN TRF LC display exhibits high transmission and reflectance, fast response, and wide-viewing characteristics, along with achromaticity.

Synergistic Effect of Ethaselen and Selenite Treatment against A549 Human Non-small Cell Lung Cancer Cells

  • Background: In this study, we aimed to evaluate the growth inhibitory effect of the combination of ethaselen (BBSKE) and low fixed dose of selenite against A549 human non-small cell lung cancer cells in vitro. Materials and Methods: Growth inhibitory effects against A549 cells were determined by SRB assay. Combination index (CI) values were calculated based on Chou-Talalay median-effect analyses. Dose reduction index (DRI) values were applied to calculate dose reduction of selenite. Contents of free thiols and GSH were determined by DTNB assay and intracellular ROS levels by DCFH-DA fluorescence labeling. Results: Compared with BBSKE or selenite single treatment, the combined application of ethaselen and a low fixed dose of selenite shortened the onset time of sodium selenite, reduced $IC_{50}$ values, and increased the maximum inhibition rates, suggesting a possible molecular mechanism of the synergism. Obvious synergistic effects were observed after different times of combination treatment, especially after 24 h. Compared with selenite single treatment, dosage of selenite could be remarkably reduced in combination therapy to gain the same inhibitory effect on cell proliferation. Compared with BBSKE single treatment, the content of free thiols and GSH were significantly reduced and ROS levels greatly elevated in the combination group. For the combination treatment, cell viability increased as greater concentrations of GSH were added. Conclusions: All these results indicate that the combination treatment of BBSKE and selenite showed synergism to inhibit A549 cell proliferation in vitro, and also reduced the selenite dosage to mitigate its toxicity which is very meaningful for combination chemotherapy of lung cancer. The synergism was probably caused by the accelerated exhaustion of intracellular reductive substances, such as free thiols and GSH, which ultimately leads to enhanced oxidative stress and apoptosis.

Fabrication and Electrical Properties of Local Damascene FinFET Cell Array in Sub-60nm Feature Sized DRAM

  • We fabricate local damascene FinFET cell array in sub-60nm feature sized DRAM. The local damascene structure can remove passing-gate-effects in FinFET cell array. p+ boron in-situ doped polysilicon is chosen for the gate material, and we obtain a uniform distribution of threshold voltages at around 0.7V. Sub-threshold swing of 75mV/d and extrapolated off-state leakage current of 0.03fA are obtained, which are much suppressed values against those of recessed channel array transistors. We also obtain a few times higher on-state current. Based on the improved on- and off-state current characteristics, we expect that the FinFET cell array could be a new mainstream structure in sub-60nm DRAM devices, satisfying high density, low power, and high-speed device requirements.

TIMP-1 in the regulation of ECM and apoptosis

  • The importance of apoptosis in normal development and pathogenesis has been well recognized, and explosive progress towards dissecting its commitment step has been made during the past decade. Mitochondria, Apaf-1, caspase, and bcl-2 family members play central roles in the commitment step. However, it is still unclear how upstream cell survival pathways regulate apoptosis. It is also unknown whether the bcl-2 family members have any effect on the upstream survival pathways. We have demonstrated that the anti-apoptotic gene product bcl-2 greatly induces expression of the tissue inhibitor of metalloproteinase-1 (TIMP-1) in human breast epithelial cells. Surprisingly, we found that TIMP-1, like bcl-2, is a potent inhibitor of apoptosis induced by a variety of stimuli. Functional studies indicate that TIMP-1 inhibits a classical apoptotic pathway mediated by caspases, and that focal adhesion kinase (FAK)/Pl 3-kinase and mitogen activated protein kinase (MAPK) are critical for TIMP- 1 -mediated cell survival. We also showed specific association of TIMP-1 with the cell surface. Consistently, a 150-H)a surface protein was identified in MCF10A cells that specifically binds TIMP-1. Taken together, we hypothesize that TIMP-I binding on the cell surface induces a cell survival pathway that regulates the common apoptosis commitment step. The results of these studies will address a new paradigm in the regulation of apoptosis by an extracellular molecule TIMP-1, and also greatly enhance our understanding of TIMP-1's pleiotropic activity in many physiological and pathological processes. This information may also be useful in designing more rational therapeutic interventions aimed at modulating the anti-apoptotic activity of TIMP-1 .

Analysis of the Effects of CO Poisoning and Air Bleeding on the Performance of a PEM Fuel Cell Stack using First-Order System Model (일차계 모델을 이용한 고분자전해질 연료전지 스택의 CO Poisoning 및 Air Bleeding 효과 분석)

  • We analyze the effects of CO poisoning and air bleeding on the performance of a PEM (polymer electrolyte membrane) fuel cell stack fabricated using commercial MEA (membrane electrode assembly). Dynamic response data from the experiments on the performance of a stack are identified by obtaining steady-state gains and time-constants of the first-order system model expressed as a first-order differential equation. It is found that the cell voltage of the stack decreases by 1.3-1.6 mV as the CO concentration rises by 1 ppm. The time elapsed to reach a new steady state after a change in the CO concentration is shortened as the magnitude of the change in the CO concentration increases. In general, the steady-state gain becomes bigger and the time-constant gets smaller with increasing the air concentration (air-bleeding level) in the reformate gas to restore the cell voltage. However, it is possible to recover 87%-96% of the original cell voltages, which are measured with free of CO, within 1-30 min by introducing the bleed air as much as 1% of the reformate gas into the stack.

Electro-Optic Characteristics of Polymer Dispersed Liquid Crystal Cell with Transparent State Initially (초기에 투명한 상태인 고분자 분산형 액정셀의 전기 광학 특성)

  • We fabricated a polymer dispersed liquid crystal (PDLC) cell using LC with negative dielectric anisotropy and UV curable monomer exhibiting transparent state initially (called normally transparent (NT)) and studied the electro-optic characteristics. The NT PDLC cell made with the ratio of LC : monomer = 70/30 wt%, curing temperature of 20 $^{\circ}C$ and strong UV intensity of 198 ㎽/$\textrm{cm}^2$ had high contrast ratio and showed good electro-optic characteristics. In this condition, LC is aligned vertically on the substrate due to the vertical alignment layer and the polymer made with the UV exposure does not influence the alignment of the LC much. Therefore, the transmittance at the zero voltage is very high and the scattering state of the cell is good after applying the voltage because LC with negative dielectric anisotropy tries to align perpendicular to the field. And also, the NT PDLC cell showed better viewing angle characteristics than that in the normally scattering (NS) PDLC.

  • This work deals with fast-to-compute global control laws for time-optimal motion of strongly nonlinear dynamic systems like resolute robots. the theory of cell-to-cell mappings for dynamical systems offer the possibility of doing the vast majority of the control law computation offline in case of time optimization with constrained inputs. These cells result from a coarse discretization of likely swaths of state space into a set of nonuniform, contiguous volumes of relatively simple shapes. Once the cells have been designed, the bang-bang schedules for the inputs are determined for all likely starting cells and terminating cells. the resulting control law is an open-loop optimal control law with feedback monitoring and correction.
