High Productivity Ethanol Fermentation Using Flocculant Yeast (응집성 효모에 의한 고생산성 알콜 발효)

  • A tower fermentor equipped with a modified settler was used for ethanol fermentation using highly flocculating yeast, Saccharomyces uvarum. The settler was constructed of glass column divided into two chambers by a funnel shaped divider. Gas was allowed to escape from lower chamber of the settler through a small tube. This design significantly reduced the turbulence in upper chamber of the settler and made it possible to operate at high dilution rate. Using the tower fermentor, the effects of operating conditions such as initial glucose concentration, dilution rate and cell recycle ratio were studied. The maximum ethanol productivity, 64.0 g/l' h was obtained at a dilution rate 1.1 h -1 and a cell recycle ratio 5 with the corresponding ethanol concentration of 58.8 g/l, and cell mass of 88 g/l.

  • We performed process and CFD simulations of a 100 kW fuel cell system. By process simulation, we derived the input flow rate of each unit process and the recycle diesel flow rate. Through CFD simulation considering the recycle diesel flow, more efficient operational condition was found. Using 6 desulfurize reactors, a TSA process for a 100 kW fuel cell system was successfully constructed. Heat interference between reactors was found to be negligible. These results will contribute to increasing the efficiency of fuel cell system and the developed desulfurizing module design will contribute to the clean petrochemical technology as well as fuel cell systems.

  • In this paper, numerical analysis of hydrogen recycle system has been conducted in order to enhance the efficiency of automotive fuel cell. Generally, the excess hydrogen is provided in the automotive fuel cell. Since the non-reaction hydrogen reduces automotive fuel cell efficiency, reuse of the non-reaction hydrogen can be helpful to improve the fuel cell performance. In case of PEM FC, the water vapor is provided to hydrogen from the cathode so that the mixture experiences phase change depending on the changes of pressure and temperature. The internal flow of the mixture in the hydrogen recirculation system of fuel cell was investigated for real flow conditions. The variation of performance, properties and mass fractions of mixture, hydrogen and water-vapor were investigated. This study was performed based on 80KW level automotive fuel cell's recycling system.

A Study on Industrial Media for Production of Lactic acid in Batch and Continuous Fermentations (회분식 및 연속배양에 있어서 고농도 젖산의 생산을 위한 공업용 배지연구)

  • We have investigated industrial media for lactic acid fermentation to reduce the cost of nitrogen sources. Corn steep liquor (CSL) was successfully used at 5% (v/v) in batch fermentations. Use of soluble CSL improved the productivity about 20% with an advantage of clearer fermentation broth. Yeast extract-complemented CSL improved the productivity about 20% with an advantage of clearer fermentation broth. Yeast extract-complemented CSL media further increased the increased the productivity. It was found that 3.1 g/L yeast extract and 5% CSL could be an effective substitute for 15 g/L yeast extract in 10% glucose medium. Brewing yeast was also used as a sole nitrogen source equivalent to 5% CSL. A continuous culture coupled with cell-recycle by microfiltration at the dilution rate of 0.05-0.065 h-1 led to the highest lactic acid productivity. Lactic acid was recovered by electrodialysis from the cell free broth. Depleted cell free broth supplemented with 5-10 g/L of yeast extract performed reasonably in batch and continuous cultures. Reuse of the fermentation broth may reduce the cost of raw materials as well as minimize the fermentation wastes.

  • The green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. In two-phase bubble column using various organic solvents, poor recovery 08 - 32%) of hydrocarbon seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. To improve recovery efficiency, mixed-solvent of extractive solvent (octane) and biocompatible solvent (octane) was tested in two-phase column for in situ extraction. In two-phase extraction culture using mixed-solvent, the algal growth was intensely inhibited even at low concentration of polar octanol solvent. the hydrocarbon recovery in two-stage cell-recycle extraction showed a 2.9 fold increase (57%) over that in two-phase extraction. Up to 60 % of hydrocarbon could be recovered without serious cell-damage in the case of downstream separation for 6 h at the high recycle flow rate using this process after batch culture.

  • $1.96{\times}10^{-5}$(IU/cell/hr) of specific scu-PA production rate was obtained from HEK cells in maintaining ca. $8{\times}10^{5}$(cells/ml) of maximum roll density at 10(ml/hr) of perfusion rate with recycling 20% serum free conditioned media. It can be compared to $4{\times}10^{6}$(cells/ml) of maximum cell density and $4.56{\times}10^{-4}$(IU/cell/hr) of specific production rate in cultivating cells with 1% serum containing medium. Thc conversion ratio of scu-PA to tc-UK increased up to 55% as the recycling ratio increased; however, recycling the used medium seemed to have least negative effect on cell growth. It also showed that the recycling process had definitive advantage of using serum free medium in perfusion cultivation of HEK cell line.

Continuous Cultivation of Lactobacillus rhamnosus with Cell Recy-cling Using an Acoustic Cell Settler

  • Continuous production of lactic acid from glucose by Lactobacillus rhamnosus with cell recycling using an acoustic cell settler was carried out. The performance of the system, such as the concentration of cell and product were compared with the control experiment without recycling. The acoustic settler showed cell separation efficiency of 67% during the continuous operation and the cell concentration in the fermentor with recycle exceeded that of the control by 29%. Com-pared with the control, tactic acid production was increased by 40%, while glucose consumption was only increased by 8%. The higher value of lactic acid production to substrate consumption (Yp/s, product yield coefficient) achieved by cell recycling is interpreted to indicate that the recycled cell mass consumes less substrate to produce the same amount of product than the control Within system environmental changes due to the longer mean cell residence time induced the cells maintaining the metabolic pathways to produce Less by-Product but more product, lactic acid.

Effects of operating parameters on the performance of continuous flow microbial fuel cell (연속식 미생물연료전지 성능에 미치는 운전변수의 영향)

  • Effects of operating parameters such as hydraulic retention time(HRT), recycle ratio and influent COD concentration on the performance of a continuous flow microbial fuel cell(MFC) were investigated. Decrease of HRT improved mass transfer of substrate to electrogenic microorganisms, therefore resulting in increased electrode voltage and power generation of MFC. Increase of HRT promoted COD removal by elongating retention time for COD removal in MFC. Recycling of effluent increased the COD removal and coulombic efficiencies by returning suspended microorganisms into MFC. Increase of influent COD enhanced COD removal due to the improved mass transfer of substrate. Decrease of coulombic efficiency by the increase of the HRT and influent COD concentration indicated that they enhanced the activities of fermentative bacteria.

High Cell Density Culture of Bifidobacterium longum by Cross-flow Filtration (Cross-flow filtration에 의한 Bifidobacterium longum의 고농도 배양)

  • The conditions for production of high cell density of Bifidobacterium longum were investigated and the cross-flow filtration system was used to remove the inhibitory metabolites, lactic acid and acetic acid. The maximum cell growth was observed with glucose as carbon source at the concentration of 50 g/l at $37^{\circ}C$ with the initial pH 6.5. When B. longum was cultured in a cross-flow filtration system, the maximum cell growth was observed at a dilution rate(D) of $0.31\;h^{-1}$ and the dry cell weight was 16.4 g/l($3.5{\times}10^{10}\;cell/ml$), which was about four times higher than that obtained in the batch culture with pH control.

  • For developing a 100 kW MCFC power generation system, Several design parameters for a fuel cell stack and system analysis results by Cycle Tempo, a processing computer soft ware, were described. Approximately 170 cells are required to generate 100 kW at a current density of 125 mA/$cm^2$ with 6000 $cm^2$ cells. An overall heat balance was calculated to predict exit temperature. The 100 kW power is expected only under pressurized operation condition at 3 atm. Recycle of cathode gas by more than 50% is recommended to run the stack at 125 mA/$cm^2$ and 3 atm. Manifolds should be designed based on gas flow rates for the suggested operating condition. The fuel cell power generation system was designed conceptually with several choices of utilization of anode exhaust gas. Also system efficiency was calculated at various type of system and operation conditions.
