This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 2-year-old female catfishes (Silurus asotus). Female Korean catfishes collected in the vicinity of Chollabukdo have synchronous ovaries which discontinously ovulate eggs during the breeding season (from late May until early July). The fishes were killed by severing the spinal cord just posterior to the head after immobilization with tricaine methanesulfonate (MS 222). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were examined by means of light and transmission electron microscopy. The size of the nucleoli and number of the yolk granules increased as the oocyte groved. Yolk granules were deposited in the oocyte as fluid Due to tile presence of large early and late maturing oocytes, their ovaries were enlarged, transparent, granular and greenish in color. As the percentages of fish in late maturing oocyte (LMO) and ripe oocyte (RO) stage increased from March to April, mean gonadosomatic index (GSI) values (19.95%) increased. Zona radiata changed from squamous into cuboid in shape in the early maturing oocyte (EMO) stage. Processes, microvilli, from the zona radiata and from the oocyte grow, and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radials becomes more compact and devoid of pore canals during oocyte maturation. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in both gonadosomatic index and macroscopic appearance. The main cytological changes such as increase in size of cell, nucleus, nucleolus, and increase in number of nucleoli and mitochondria demonstrated with electron microscopy in the previtellogenic oocytes of Korean catfish, provided evidences for important synthetic processes in an early preparatory phase of oocyte development. The electrophoretic pattern of major band in mature stage was much thicker (24 k, 66 k, 90-110k dalton) than that in previtellogenic phase.
Immobilized cells of Aspergillus niger was employed to produce citric acid by fermentation of milk factory waste water. A. niger ATCC 9142 as a citric acid production strain was cultured for 3 days and was entrapped with Ca-alginate bead about 2.5∼3.5 mm. The optimal pH and temperature were estimated to be 3.0 and $30^{\circ}C$, respectively. Dilution rate for fermentation was calculated to be $0.025 h^{-1}$ . Maximum amount of citric acid was obtained at 4.5 g/$\ell$ with the optimized fermentation condition. The yield of citric acid produced by immobilized A. niger ATCC 9143 was 70.3%. The yield was increased by 20% with immobilized cell, compared to that of the shake flask culture. Hence, the milk factory waste water is worthy to be used for the substrate of citric acid fermentation.
Objective: We compared the regenerative effects of microcurrent therapy (MT) according to the type of electric current, which were direct current microcurrent therapy (DCMT) and alternating current microcurrent therapy (ACMT) on atrophied calf muscle in cast-immobilized rabbit. Method: Rabbits were allocated into control group (sham MT), ACMT group, and DCMT group. Before starting treatment, right gastrocnemius (GCM) muscle was immobilized by cast for 2 weeks. Compound muscle action potential of tibial nerve in nerve conduction study, circumference of calf muscle using a ruler, and thickness of medial and lateral GCM muscle measured by ultrasound, cross sectional area (CSA), and proliferating cell nuclear antigen (PCNA) ratios (%) of muscle fibers were measured on the immunohistochemical analysis. Results: The mean atrophic changes (%) in right medial and lateral GCM muscle thickness, right calf circumference, and amplitude of CMAP of the right tibial nerve in ACMT group and DCMT group were significantly lower than those in control group, respectively (p<0.05). The mean CSA (μm2) of type I and type II and PCNA ratios (%) of medial and lateral GCM muscle fibers in ACMT group and DCMT group were significantly greater than those in control group, respectively (p<0.05). There were no significant differences between the ACMT group and DCMT group at all parameters. Conclusion: This study demonstrated that ACMT and DCMT showed better regeneration effect than sham MT. Microcurrent may be effective in regeneration of atrophied muscle regardless of the type of current.
This study was aimed to enhance the Fe(II) oxidation rate using immobilized cells of Thiobacillus ferroxidans. For this purpose, a medium for the minimization of jarosite formation was developed first. Secondly, cell immobilization in celite beads was carried out. And then, repeated-batch and continuous operatons of Fe(II) oxidation by using immobilization cells were performed. In a series of flask cultures, three types of media were tested: media with a much lower salt concentration than that of the 9K medium; media which contained different nitrogen sources from that of the 9K medium, that is $(NH_4)_2HPO_4$, $NH_4Cl and HNO$_3$; media which contained $(NH_4)_2HPO_4$ as nitrogen and phosphate source, but without $K_2HPO_4$ as nitrogen and phosphate source in the 9K medium. As a result, the M16 medium which contained 3 g/L of $(NH_4)_2HPO_4$ as nitrogen and phosphate source was found to be the optimal one. It sustained good cell growth allowing no jarosite formation. In the repeated-batch operations, the rate of Fe(II) oxidation gradually increased to reach a maximum value as the batch was repeated. As a result of repeated-batch operations. a maximum Fe(II) oxidation rate was 2.33 g/L . h. In the continuous operations, the iron oxidation rate could be increased to 2.14 g/L .h at a dilution rate of 0.25 $h^{-1}$ which is greater than the maximum specific growth rate (0.12 $h^{-1}$) of the bacteria.
A phosphate solubilizig bacterium, Pantoea agglomerans, was isolated from rhizosphere soils collected from Chungbuk area. A greenhouse experiment was conducted to study the effect of combined application of rock phosphate and P. agglomerans inoculation on plant growth and phosphate accumulation of rice (Oryza sativa L.). Apart from control that received no inputs, six treatments were planned as follows; 1) seed bacterization, 2) free cell inoculation and 3) bacteria immobilized beads inoculation, individually and in combination with 1 and 2.5 g of rock phosphate per pot. The results showed that plant growth and phosphate uptake were significantly enhanced as a result of bacterial inoculation. Bacterial inoculation in the form of immobilized beads and 1 g of rock phosphate was found to affect positively the rice plant growth and phosphorus accumulation than other treatments. The available phosphate concentration of the pot mixture also found improved as a result of P. agglomerans inoculation. A positive correlation was observed between the phosphate concentration in the pot mixture and phosphate accumulation in plant.
In order to reduce energy input in direct ethanol production from raw starch by co-immobilized Aspergillus awamori(A) and Zymomonas mobilis(Z), A-Z 36 culture system which was changed to anaerobic after 36 h of aerobic fermentation without sterilization was investigated. This immobilized cell system can not be carried out under unsterile conditions because of growth of microbial contaminants from original medium. Among some food additives such as sorbic acid, benzoic acid, dehydroacetic acid, p-hydroxybenzoic acid, Vantocil IB and Neupectin-L, Vantocil IB and Neupectin-L were a potent antibacterial agent in A-Z 36 culture cell system and were not affected in hydrolysis of substrate as compared with the case of control. Ethanol yield(6.9 g/l) in system of addition of 0.1% Neupectin-L was slightly higher than that in control(6.4 g/l). When 2% starch was fed five times in fed-batch culture with 0.1% Neupectin-L, ethanol yield and productivity were 34 g/l and 2.0 g/l/day, respectively.

As the occurrence of harmful algal blooms (HABs) have become severe in precious water resources, the development of efficient harmful algae treatment methods is considering as an important environmental issue for sustainable conservation of water resources. To treat HABs in water resources, various conventional physical and chemical methods have been utilized and showed treatment efficiency, However, these methods can lead to discharging of cyanotoxins into the water bodies by chemical or physical algal cell lysis or destruction. Thus, to overcome this limitation, the development of safe HABs treatment methods is required. In the present study, adsorption technology was investigated for the removal of harmful algal species, Microcystis aeruginosa from aqueous phases. Industrial waste biomass, Corynebacterium glutamicum biomass was valorized as biosorbent (PEI-modified alginate/biomass composite fiber; PEI-AlgBF) for M. aeruginosa through immobilization with alginate matrix and cationic polymer (polyethylenimine; PEI) coating. The functional groups characteristic of PEI-Alg was determined using FT-IR analysis. By adsorption process used PEI-AlgBF, 52 and 67% of M. aeruginosa could be removed under the initial density of M. aeruginosa 200×104 cells mL-1 and 50×104 cells mL-1, respectively. As the increasing surface area of PEI-AlgBF, the removal efficiency was increased. In addition, we could find that adsorptive removal of M. aeruginosa has occurred without any M. aeruginosa cell lysis and destruction.
An organic solvent stable lipase from solvent-tolerant Pseudomonas sp. BCNU 171 had an optimal pH of 8 and an optimal temperature of 37℃. This crude extracellular lipase from BCNU 171 exhibited increased stability in the presence of various types of solvents at high concentrations (25%, v/v). The lipase stability was found to be highest in the presence of xylene (137%), followed by toluene (131%), octane (130%), and butanol (104%). Overall, BCNU 171 lipase tended to be more stable than immobilized commercial lipase (Novozyme435) in the presence of organic solvents. Furthermore, BCNU 171 lipase maintained about 90% of its enzyme original activity in the presence of NH4+, Na+, Ba2+, Hg2+, Ni2+, Cu2+, and Ca2+ion and significantly increased its enzyme activity in the presence of various emulsifying agents. Thus, the organic solvent stable lipase from Pseudomonas sp. BCNU 171 could be usable as a potential whole cell biocatalyst and for synthetic applications of enzymes for industrial chemical processes in organic solvents without using immobilization.
Structural sites which cations can occupy in garnet structure are centers of the tetrahedron, octahedron, and distorted cube sharing edges with the tetrahedron and octahedron. Among them, the size of cation occuping at tetrahedral site (the center of tetrahedron) is closely related with the size of a unit cell of garnet. Accordingly, garnet containing iron with relative large ionic radii in tetrahedral site can be considered as a promising matrix for the immobilization of the elements with large ionic radii, such as actinides in radioactive wastes. We synthesized several garnets with the batch composition of $Ca_{1.5}GdCe_{0.5}ZrFeFe_3O_{12}$, and studied their properties and phase relations under various conditions. Mixed samples were fabricated in a pellet form under a pressure of $200{\~}400{\cal}kg/{\cal}cm^2$ and were sintered in the temperature range of $1100\~1400^{\circ}C$ in air and under oxygen atmospheres. Phase identification and chemical analysis of synthesized samples were conducted by XRD and SEM/EDS. In results, garnet was obtained as the main phase at $1300^{\circ}C$, an optimum condition in this system, even though some minor phases like perovskite and unknown phase were included. The compositions of garnet and perovskite synthesized from the batch composition of $Ca_{1.5}GdCe_{0.5}ZrFeFe_3O_{12}$ were ranged $[Ca_{l.2-1.8}Gd_{0.9-1.4}Ce_{0.3-0.5}]^{VIII}[Zr_{0.8-1.3}Fe_{0.7-1.2}]^{VI}[Fe_{2.9-3.1}]^{IV}O_{12}$ and $Ca_{0.1-0.5}Gd_{0.0-0.8}Ce_{0.1-0.5}\;Zr_{0.0-0.2}Fe_{0.9-1.1}O_3$, respectively. Ca content was exceeded and Ce content was depleted in the 8-coordinated site, comparing to the initial batch composition. This phenomena was closely related to the content of Zr and Fe in the 6-coordinated site.
Objectives : This is the experimental paper to study the curative and preventive effects of Dansambohyultang, Methods : Oriental herb medicine for gastro-intestinal disease caused by stress, on the rats put under the stress through starvation, heating and immoblization, the author used four different group; the normal group, which was not put under stress and well fed, and the control group, to which the drug was not administered and put under the stress through starvation, heating and immoblization, the Sample I, to which the drug was administered before they were put under stress, the Sample lI, to which the drug was administered after they were put under stress. Results : When a Masson's trichrome stain method was applied to the control group, a small size of ulcer was found in the fundus and the pylorus, and atrophy was observed in the neck region of mucous membrane. When the drug was administered to the Sample I and the Sample II, the former recovered from the ulcer and atrophy to almost normal and showed better results than the latter. When a Masson's trichrome stain method was applied to the control group, atrophy of mucous membrane was found all over the intestines. When the drug was administered to the Sample I and the Sample ll, the curative and recovered effects was seen in both, but the former showed better results than the latter. After application of Mayer's hematoxylin stain method, the observation of numerical changes of goblet cells in the small intestines showed the most significant decrease in the control group. In the Sample I, the number of the cells in the duodenum, the jejunum and the ileum returned to an almost normal level. In the Sample II, a slight increase was observed only in the ileum. After application of Mayer's hematoxylin stain method, the observation of numerical changes of goblet cell in the large intestines showed a significant decreased in the control group. Both the Sample I and the Sample II showed an increase in the number of goblet cells in comparison with the control group. When a serotonin-immunoreaction test was applied, the control group showed a general decrease in the number of gastro-entero-endocrine cells, The Sample I showed the effect in all over gastro.intestinal tract as compared with control group and the Sample II showed the effect in all over gastro-intestinal tract except the duodenum and the former showed better results than the latter. Conclusion : According to the above results, the regions which are most sensitive to stress were observed to the ileum and the colon. It was concluded that Dansambohyultang has a remedical value particularly on the abnormalities caused by stress in all over gastro-intestinal tract and it was found to be more effective in the Sample I than in the Sample ll. Judging from the fact that the Sample I showed better results than the Sample ll, Dansambohyultang can be said to have a preventive effect more than a curative effect on gastro-intestinal tract disease.
