CD44v3 and VEGF-C Expression and its Relationship with Lymph Node Metastasis in Squamous Cell Carcinomas of the Uterine Cervix

  • Background: To investigate the expression of CD44v3 and vascular endothelial growth factor-C (VEGF-C) and their relationship with lymph node metastasis in squamous cell carcinomas (SCC) of the uterine cervix. Materials and Methods: Expression of CD44v3 and VEGF-C was analyzed in 109 cases of cervical SCC by immunohistochemistry (IHC). The relationship was analyzed between expression and the patient age, histological differentiation, formation of tumor emboli in lymphoid vessels, lymph node metastasis, FIGO staging, and TNM classification. Results: Expression rates for both CD44v3 and VEGF-C were 43.1% in cervical SCC. The cells with positive immunohistochemical staining of CD44v3 were distributed mainly around the keratin pearls in well differentiated carcinomas, but distributed diffusely in the moderately and poorly differentiated lesions. VEGF-C was found stained positively in most of the tumor cells. There were differences in expression between normal epithelium and atypical hyperplasia as well as carcinoma. Both CD44v3 and VEGF-C were found to be associated positively with lymph node metastasis and TNM classification (both p=0.000). Neither CD44v3 nor VEGF-C was found to be associated with patient age, histological differentiation, formation of tumor emboli in lymphoid vessels and FIGO staging. CD44v3 was found to be associated with VEGF-C positively (p=0.000). Conclusions: Abnormal expression of CD44v3 and VEGF-C is associated closely with the lymph node metastasis in cervical SCC, and these agents may cooperate in carcinogenesis and development of metastatic lesions.

Role of Th17 Cell and Autoimmunity in Chronic Obstructive Pulmonary Disease

  • The molecular mechanisms involved in the pathogenesis of chronic obstructive pulmonary disease (COPD) are poorly defined. Accumulating evidences indicate that chronic inflammatory responses and adaptive immunity play important roles in the development and progression of the disease. Recently, it has been shown that IL-17 producing CD4 T cells, named Th17 cells, which have been implicated in the pathogenesis of several inflammatory and autoimmune diseases, are involved in airway inflammation and COPD. In addition, we and others suggest that autoimmunity may play a critical role in the pathogenesis of COPD. Here, we will review the current understanding of roles of Th17 cells and autoimmune responses in COPD.

  • Recombinant E. coli JM109(pZH3-5/pMT) harboring manganese transport gene(mntA) and metal sequestering protein, metallothionein(MT), was cultivated to accumulate cadmium in aqueous phase. Bioaccumulation followed Michaelis-Menten type kinetics. Equilibrium isotherm showed Langmuir type isotherm. The optimum pH for $Cd^{2+}$ uptake was 7-7.5.

서울시내 수산시장에서 유통되고 있는 수산물의 비소(As), 카드뮴(Cd) 및 납(Plb)의 함량 (Heavy Metals in Fishery Products, Sold at Fish Markets in Seoul)

  • 우리나라 연안에서 어획, 또는 수입된 어류 33종 256건, 이매패류 15종 312건, 갑각류 12종 91건(총 782건), 기타 수산물 10종 123건의 시료를 대상으로 As, Cd 그리고 Pb의 함량을 A.A.S로 분석한 결과, 어류에서는 0.021mg/kg(N.D.~0.162 mg/kg), 0.0l4mg/kg(0.001~0.120 mg/kg), 0.111 mg/kg(0.015~0.499 mg/kg), 이매패류의 경우 0.052 mg/kg(0.003~0.311 mg/kg), 0.182 mg/kg(N.D.~1.905 mg/kg), 0.138 mg/kg(0.013~0.462 mg/kgg), 갑각류의 경우 0.042 mg/kg (N.D.~0.328 mg/kg), 0.079 mg/kg(0.002~1.113 mg/kg), 0.109 mg/kg(0.006-0.510 mg/kg) 그리고 기타 수산물의 경우 0.024 mg/kg(N.D.~0.181 mg/kg),0.033 mg/kg(0.001~0.214 mg/kg),0.118 mg/kg(0.010~0.877 mg/kg)으로 각각 나타나, 이매패류가 다른 종류에 비해 유의하게 높은 결과를 보였고(p<0.0001), 국내의 최대허용농도 이하였으며, 수산물을 통해 섭취되는 비소, 납, 카드뮴의 주간섭취량을 분석한 결과 FAO/WHO에서 권장하는 PTWI에 비해 1.4~26.4%정도인 것으로 나타났다.

Limited in vitro differentiation of porcine induced pluripotent stem cells into endothelial cells

  • Background: Pluripotent stem cells (PSCs) including embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) offer the immense therapeutic potential in stem cell-based therapy of degenerative disorders. However, clinical trials of human ESCs cause heavy ethical concerns. With the derivation of iPSCs established by reprogramming from adult somatic cells through the transgenic expression of transcription factors, this problems would be able to overcome. In the present study, we tried to differentiate porcine iPSCs (piPSCs) into endothelial cells (ECs) for stem cell-based therapy of vascular diseases. Methods: piPSCs (OSKMNL) were induced to differentiation into ECs in four differentiation media (APEL-2, APEL-2 + 50 ng/mL of VEGF, EBM-2, EBM-2 + 50 ng/mL of VEGF) on cultured plates coated with matrigel® (1:40 dilution with DMEM/F-12 medium) for 8 days. Differentiation efficiency of these cells were exanimated using qRT-PCR, Immunocytochemistry, Western blotting and FACS. Results: As results, expressions of pluripotency-associated markers (OCT-3/4, SOX2 and NANOG) were higher observed in all porcine differentiated cells derived from piPSCs (OSKMNL) cultured in four differentiation media than piPSCs as the control, whereas endothelial-associated marker (CD-31) in the differentiated cells was not expressed. Conclusions: It can be seen that piPSCs (OSKMNL) were not suitable to differentiate into ECs in the four differentiation media unlike porcine epiblast stem cells (pEpiSCs). Therefore, it would be required to establish a suitable PSCs for differentiating into ECs for the treatment of cardiovascular diseases.

RAW264.7세포주와 염증생쥐모델에서 항염증(抗炎症) 작용(作用)에 대한 청열활혈탕가계혈등(淸熱活血湯加鷄血藤)의 효과(效果) (The Anti-inflammatory Effect of Cheongyeolhawlhyeoltanggagyehyeoldeung (CYHHT) in cultured RAW264.7 cells and murine models of inflammation)

  • Purpose : The Purpose of this research was to investigate the effects of Cheongyeolhawlhyeoltanggagyehyeoldeung (CYHHT) on anti-inflammatory effects. Methods : As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators were determined in mouse lung fibroblast cells(mLFC) and RAW264.7 cells. Also, changes in pathological features by drug treatment were investigated in the in vivo edema-induced rats by carrageenin /arachidonic acid or in the colitis-induced mice by DSS treatment. Results : The cytotoxicity of CYHHT on mLFC and RAW264.7 cells was not observed at 100, 50, 10, and $1{\mu}g/ml$ of CYHHT treatments. $IL-1{\beta}$, IL-6 and NOS-IImRNA expression of RAW264.7 cells was inhibited by CYHHT treatments in a dose-dependent manner. CYHHT treatment of RAW264.7 cells inhibited $TNF-{\alpha}$ and COX-2 mRNA expression. CYHHT treatment of RAW264.7 cells significantly inhibited IL-6 and NO production. CYHHT treatment of RAW264.7 cells inhibited ROS production. CYHHT inhibited rat's paw edema induced by carrageenin or arachidonate treatment in all concentrations examined. The body weight and colon length of colitis-induced mice were recovered to a normal level by DSS treatment. Clinical disease levels were significantly improved compared to the control animals. CYHHT treatment of colitis-induced mice significantly increased hematological values such as WBC and RBC counts, Hgb and HCT levels, but decreased PLT values. CYHHT treatment of colitis-induced mice decreased IL-6 and $TNF-{\alpha}$ production significantly CYHHT treatment of colitis-induced mice significantly increased CD3+(T) cell counts. In contrast, CYHHT treatment decreased CD19+ B cell counts and CD3+/CD69+ significantly, and also decreased B/T ratio (%) though not significant. Conclusion : These results indicated that CYHHT could be used for treating diverse female diseases caused by the inflammation.

광전도체의 CdS 단결정 성장과 물리적 특성 (Growth of CdS Single Crystal as Photoconductor and Its Physical Characteristics)

  • 승화방법으로 광전도체의 CdS 단결정을 성장하였고 외삽법으로 구한 $a_{o}$$c_{o}$의 격자상수 값은 각각 $4.131{\underline{8}}{\AA}$$6.712{\underline{2}}{\AA}$임을 알았다. Hall 측정값으로 부터 상온에서의 CdS 단결정의 운반자 농도와 이동도는 각각${\sim}10^{23}m^{-3}$$2.93{\times}10^{-2}m^{2}$/V sec이였으며 온도에 따른 이동도 변화는 33 K에서 150 K까지는 $T^{1/2}$에 따라 증가하는 경향이 있고 180 K에서 상온까지는 $T^{-2}$에 따라 감소한 경향이 나타났다. 광전류 측정으로부터 나타난 단파장대의 봉우리는 진성전이에 기인하는 봉우리였으며 이 봉우리의 에너지 값은 CdS 광전도체에 에너지 밴드 갭과 동일한 값을 나타냄을 알았다.

Cu계 $I-III-VI_2$ 화합물 박막 태양전지 연구 (A Study on the Cu-based $I-III-VI_2$ Compound Thin Film Solar Cells)

  • Cu계$I-III-VI_2$화합물은 직접천이형 반도체로 광흡수계수가 매우 높아 박막형 태양전지 제조에 매우 유리하다 또한 화학적으로 안정하며 Ga, A1등을 첨가하면 에너지 금지대폭을 조절할 수 있어 Wide Bandgap 태양전지 및 탠덤구조 태양전지를 제조하기에도 용이하다. $CuInSe_2(CIS)$ 물질에서 In을 $20-30\%$ 정도 치환한 $Cu(In,\;Ga)Se_2(CIGS)$ 태양전지의 경우 $19.5\%$의 세계 최고 효율을 보고하고 있으며 이는 다결정 실리콘 태양전지의 효율과 비슷한 수준이다. 본 연구에서는 동시 진공증발법을 이용하여 증착한 CIGS 박막 및 $CuGaSe_2(CGS)$ 박막을 이용하여 태양전지를 제조하였다. 공정의 재현성 및 결정립계가 큰 광흡수층 제조를 위하여 실시간 기판 온도 모니터링 시스템을 도입하였으며 버퍼층으로는 용액성장한 CdS 박막을 사용하였다. SLG/MO/CIGS(CGS)/CdS/ZnO/Al구조의 태양전지를 제조하여 면적 $0.5cm^2$에서 각각 $15\%$(CIGS)와 $7\%(CGS)$의 효율을 얻었다.

교류전원 구동방식에 의한 형광 OLED의 발광 특성 (Emission Characteristics of Fluorescent OLED with Alternating Current Power Source Driving Method)

  • To operate organic light emitting device (OLED) with alternating current (AC) power source without AC/DC(direct current) converter, we fabricated the fluorescent OLED and measured the emission characteristics with AC and DC. The OLED operated by AC showed higher maximum current efficiency of 8.2 cd/A and maximum power efficiency of 8.3 lm/W. But current efficiency and power efficiency of AC driven OLED showed worse than DC driven OLED at high voltage above 10 V. This result can be explained by the peak voltage of AC was $\sqrt{2}$ times than DC, In case of low driving voltage the emission characteristics were improved by the peak voltage of AC, but in case of high driving voltage the emission efficiencies were decreased by the roll off phenomena. Finally, serial OLED arrays using twelve OLEDs driven by AC 110 V showed average voltage of 9.17 V, voltage uniformity of 99.0%, average luminance of $1,175cd/m^2$, luminance uniformity of 94.4%.

CGL 층으로 MoOx를 사용한 다중 적층구조 OLED의 발광 특성 (Emission Characteristics of Multi-Tandem OLED using MoOx with CGL)

  • We studied emission characteristics of blue fluorescent multi-tandem OLEDs using $Al/MoO_x$ as charge generation layer(CGL). Threshold voltage for 2, 3, 4, and 5 units tandem OLEDs was 8, 11, 14 and 18 V, respectively. The threshold voltage in multi-tandem OLEDs was lower than multiple of 4 V for the single OLED. Maximum current efficiency and maximum quantum efficiency of single OLED were 7.6 cd/A and 5.5%. Maximum current efficiency for 2, 3, 4, and 5 units tandem OLEDs was 22.6, 31.4, 41.2, and 46.6 cd/A, respectively. Maximum quantum efficiency for 2, 3, 4, and 5 units tandem OLEDs was 11.8, 15.8, 21.8, and 25.6%, respectively. The maximum current efficiency and maximum quantum efficiency in multi-tandem OLEDs were higher than multiple of those for the single OLED. The intensity for 508 nm peak was changed and the peak wavelength was red shift by increase of tandem unit in electroluminescent emission spectra. These phenomena can be caused by micro-cavity effect with increasing of organic layer thickness.
