This study was performed to investigate adsorption characteristics of MTBE and Cd depending upon types of clay minerals md their physicochemical properties. The adsorption characteristics were examined by batch adsorption test on various experimental parameters such as adsorption time, ratio of solution to soil, concentration of contaminants, content of organic matter, pH, and zeta potential. The adsorption efficiency of MTBE or Cd for three types of clays decreased in response to the increase of the ratio of solution to soil whereas their adsorbed amounts increased. MTBE was greatly adsorbed in the decreasing order of vermiculite, bentonite, and CTAB-bentonite while Cd was adsorbed in the decreasing order of bentonite, vermiculite, and CTA-bentonite. An equilibrium isotherm for MTBE was well fitted to Freundlich plotting whereas that for Cd was closely corresponded to Langmuir isotherm. The adsorbed amount of MTBE on bentonite and vermiculite showed the maximum at 1% and 5% of humic acid, thereafter diminished while the adsorbed amount of MTBE on CTAB-bentonite increased in proportion to humic acid. Conversely, the adsorbed amount of Cd on the addition of humic acid continued to increase regardless of types of adsorbents. For all types of adsorbents, adsorbed quantity and adsorption efficiency of Cd have been coincidently increased at pH 8 and they were further enhanced at pH 10 showing 90% adsorption efficiency. Upon pH rose, the zeta potential on each adsorbent began to decrease, while increasing Cd concentration led to decline of zeta potential, which in turn ascribed to lowering dispersion stability that could consequently enhance adsorption capability.

Extract of Centella asiatica(ECA), which is poorly water-soluble extract from the Centella asiatica is known to express excellent wound healing properties. $ECA-{\beta}-cyclodextrin$$(asiaticoside-{\beta}-cyclodextrin\;and\;genin-{\beta}-cyc1odextrin)$ solid dispersion system, which was prepared by freezedrying method, was formulated as gels containing poloxamer 407 and propylene glycol, and evaluated with respect to their viscosity, stability, skin permeation and drug amount in the skin of hairless mouse. The average molar ratio $asiaticoside-{\beta}-CD$ and $genin-{\beta}-CD$ was 1:1.7 and 1:22, respectively. When the molar ratio of genin and ${\beta}-CD$ was 1:5, madecassic acid made 100% solid dispersion system and asiatic acid about 65%. In dissolution study, >99% of asiaticoside from $asiaticoside-{\beta}-CD$ was dissolved in 5 minutes, and >99% madecassic acid and >64% asiatic acid from $genin-{\beta}-CD$. The apparent viscosity of poloxamer 407 gels with $ECA-{\beta}-CD$ solid dispersion system increased in proportion to poloxamer 407 and propylene glycol concentration. In the accelerated stability test, all $ECA-{\beta}-CD$ poloxamer 407 gels showed that asiaticoside was most stable and madecassic acid stable and asiatic acid similar to stability of gel with free ECA. The permeation amount of asiaticoside in poloxamer gels through hairless mouse skin decreased as the concentration of poloxamer 407 increased. When propylene glycol was added at the level of 10%, the permeation amount of asiaticoside at poloxamer gels through hairless mouse skin increased but from 15% it decreased. The permeation of asiaticoside into the skin of hairless mouse was estimated to be about $0.10\;{\mu}g/cm^2$.
In this experiment we transformed the Ca-bentonite into Na-bentonite with two inorganic Na-chemicals under different temperatures. These two Na-chemicals were selected among five different Na-chemicals which carries Na as cation. The swelling capacity of the Na chemical-treated bentonite was increased with increasing Na concentration, while the maximum concentration of Na solution decreased with increasing temperature. $Na_2CO_3$ was most effective in exchanging Ca ions and resulting in the highest swelling index among the Na-chemicals. The swelling index was significantly increased with increasing temperature to $100^{\circ}C$. But the equilibration time reversely affected the swelling index due to a rapid increase in evaporation of water. Within same amount of Na treatment SI slightly decreased not only with increasing contacting time but also with increasing temperature. The adsorption for the transformed Na-bentonite was increased with increasing equilibrium concentrations of Pb and Cd ions for all the activated Na-B and indigenous Ca-B and Na-B while the adsorbability of $Pb^{2+}$ onto each Na-B sample is more than that of $Cd^{2+}$. And the maximum adsorption capacity sequence of Na-B samples for Pb and Cd has been found to be 5 % $Na_2CO_3.$ - 5 % $NaHCO_3$ > 3 % $NaHCO_3$ > 3 % $Na_2CO_3$ > 1 % $NaHCO_3$ > 1 % $Na_2CO_3$ > indigenous Na-B > indigenous Ca-B, showing that there are contradictory results about the relationship of cation adsorption to CEC.
Macrocyclic ligands have been studied as cation carriers in a supported liquid membrane system. Cd2+ has been transported using nitrogen substituted macrocycles as carriers and several divalent metal ions (M2+=Zn, Co, Ni, Cu, Pb, Mg, Ca, and Sr) have been transported using DBN3O2, DBN2O2and PolyNtnoen as carriers in a supported liquid membrane system. Competitive Cd2+-M2+ transport studies have also been carried out with the same system. Ligand structure, stability constant, membrane solvent and carrier concentration are also important parameters in the transport of metal ions.
Capillary electrophoretic direct chiral separation method is described for the determination of the absolute configuration of chiral aromatic acids, The enantiomeric separation was achieved by capillary electrophoresis using HP $\beta$-cyclodextrin (CD) as the chiral selector. The effect of CD concentration was investigated to optimize the chiral separation and resolution. When applied to microbial culture fluid. the present method allowed positive identification of chiral aromatic acids and their chirality as well.

Effects of dietary cholesterol and/or taurine supplementation on plasma and hepatic lipid peroxidation status and antioxidant enzyme activities were evaluated in rats fed one of the following semisynthetic diets for 5 weeks: control diet (CD, cholesterol-free and taurine-free diet); high cholesterol diet (HCD, CD+1.5% cholesterol): high taurine diet (HTD, CD+1.5% taurine): high cholesterol and high taurine diet (HCHTD, HCD + 1.5% taurine). Plasma malondialdehyde (MDA) level was not influenced by dietary cholesterol or taurine supplementation, while hepatic MDA level was 70% higher in rats fed HCD compared to the value for CD rats (p<0.05). Our observation that taurine supplementation significantly decreased the hepatic MDA level of rats fed HCD, but failed to decrease lipid peroxidation of rats fed CD indicates that the protective effect of taurine in the liver against lipid peroxidation is manifested only under the hypercholesterolemic environment. Plasma and hepatic glutathione peroxidase (GSH-Px) activities were not affected by dietary supplementation of cholesterol or taurine. However, hepatic superoxide dismutase (SOD) activity was significantly reduced by dietary taurine supplementation (p <0.05), and thus significantly lower in rats fed HTD compared to the value for CD (p<0.05). Plasma total cholesterol concentration was positively correlated with hepatic cholesterol concentration as expected (r=0.712, p<0.001). Plasma (r=0.399, p<0.05) and hepatic cholesterol levels (r=0.429, p<0.05) showed a significantly positive correlation with hepatic MDA concentration, respectively. Plasma taurine concentration was negatively correlated with hepatic SOD activity (r=-0.481, p<0.01), and tended to be negatively correlated with hepatic GSH-Px activity without showing statistical significance (r=-0.188, p<0.05). These results indicate an antioxidative effect of taurine in rats with elevated level of lipid Peroxidation due to high intake of dietary cholesterol. Future application of taurine as a safe candidate for a hypolipidemic agent without adversely affecting body's antioxidant defense system is speculated.

Cadmium (Cd) is of particular concern because of its widespread occurrence and high toxicity and may cause serious morpho-physiological and molecular abnormalities in in plants. The present study was performed to explore Cd-induced morpho-physiological alterations and their potentiality associated mechanisms in Sorghum bicolor leaves at the protein level. Ten-day-old sorghum seedlings were exposed to different concentrations (0, 100, and $150{\mu}M$) of $CdCl_2$, and different morpho-physiological responses were recorded. The effects of Cd exposure on protein expression patterns in S. bicolor were investigated using two-dimensional gel electrophoresis (2-DE) in samples derived from the leaves of both control and Cd-treated seedlings. The observed morphological changes revealed that the plants treated with Cd displayed dramatically altered shoot lengths, fresh weights, and relative water content. In addition, the concentration of Cd was markedly increased by treatment with Cd, and the amount of Cd taken up by the shoots was significantly and directly correlated with the applied level of Cd. Using the 2-DE method, a total of 33 differentially expressed protein spots were analyzed using MALDI-TOF/TOF MS. Of these, treatment with Cd resulted in significant increases in 15 proteins and decreases in 18 proteins. Significant changes were absorbed in the levels of proteins known to be involved in carbohydrate metabolism, transcriptional regulation, translation and stress responses. Proteomic results revealed that Cd stress had an inhibitory effect on carbon fixation, ATP production and the regulation of protein synthesis. In addition, the up-regulation of glutathione S-transferase and cytochrome P450 may play a significant role in Cd-related toxicity and stress responses. Our study provides insights into the integrated molecular mechanisms involved in response to Cd and the effects of Cd on the growth and physiological characteristics of sorghum seedlings. The upregulation of these stress-related genes may be candidates for further research and use in genetic manipulation of sorghum tolerance to Cd stress.
The sorption of Cd$^{2+}$ on calcite was studied in aqueous solutions of several electrolytes. The Cd$^{2+}$ concentration, 10$^{-8}$ M, was kept well below saturation with respect to CdCO$_3$(s). Sorption behavior of Cd$^{2+}$ in different ionic strengths of NaClO$_4$solutions shows that sorption is independent of ionic strength. This result suggests that Cd$^{2+}$ sorption on calcite surface is of a specific nature, and adsorption is controlled by an inner-sphere type of surface complex. Two stages in the sorption behavior could be identified: an initial rapid uptake, followed by slower uptake reaching a maximum steady state by 145 hrs. No evidence was observed for surface precipitation, although it can not be entirely ruled out. Desorption of Cd$^{2+}$ from the calcite surface after resuspension into Cd-free solution is initially very rapid, but depends partly on the previous sorption history. Desorption behavior of Cd$^{2+}$ show that an initial rapid desorption followed either by slow uptake reaching a maximum, as in the adsorption experiments, or slowing desorption to reach a steady state minimum. This irreversible behavior of Cd$^{2+}$ sorption and desorption may act as one of the controls for regulating the mobility of dissolved Cd$^{2+}$ natural aqueous systems. The two-stage enzyme reactor, packed with cyclodextrin glucanotransferase (CGTase) immobilized on Amberite IRA 900, coupled with ultrafiltration membrane was investigated for continuous production of cyclodextrin (CD). 5% (w/v) of soluble starch was partially cyclized, in the 0.1 l first-stage immobilized enzyme reactor, up to CD conversion yield of 10% (w/w) at retention time of 0.56hr and 1.5 units of immobilized CGTase/1g of carrier. In the second stage main immobilized enzyme reactor capacity of 1.5 l, the maximum CD conversion yield of 39% (w/v) was achieved at retention time of 2.8hr and 0.47 unit of CGTase/1 g of carrier. Unreacted residual dextrin was fractionated with ultrafiltration membrane, and then, recycled into the second-stage main bioreactor to increase the CD conversion yield. The most suitable membrane size and the volume concentration ratio (concentrate: filterate) for recycling of unreacted residual dextrin were found to be 5K dalton and 4:6, respectively. CD conversion yield was increased about 3~4% upon co-immobilization of pulluanase along with CGTase. Spent Amberite IRA 900 can be reutilized consecutively more than 3 times for immobilization of CGTase after regeneration.

The object of this study was to develop a gene therapy strategy for ovarian cancer. We have previously shown that AV with a L-plastin (LP) promoter infects breast and ovarian cancer cells and expressed ${\beta}$-galactosidase cDNA in preference to normal fibroblast cells and hematopoietic cells. We now report on the cytotoxicity of Ad.LP.CD, an AV carrying a CD cDNA which converts the pro-drug, 5-Fluorocytosine (5-FC) into the toxic drug 5-Fluorouracil (5-FU). Infection of Ad.LP.CD into either 293 cells or ovarian cancer cells generated the functional CD as measured by HPLC analysis. Using a ratio of AV to OVCAR3 cell of 100 and a 5-FC concentration of 100 ${\mu}$M, we achieve an over 95 % of cell growth inhibition. We are using flow cytometry analysis for ${\beta}$ -galactosidase and ovarian cancer associated folate receptor to screen primary ascites samples for infectivity after infection with an adenoviral vector, i.e., Ad.LP.LacZ. This vector system may be of value in the treatment of microscopic disease of ovarian cancer in the peritoneal cavity.
