The purpose of this research was to investigate the effects of Calendula officinalis extract (CE) on the activation of murine lymphocytes and macrophages. CE was administered p.o. once a day for 7 days at the concentration of 500 mg/kg. The administration of CE increased the viability of thymocytes, but decreased the viability of splenocytes. Also, CE increased the viability of thymocytes and splenocytes at the concentration of $5{\mu}g$/ml in vitro. The administration of CE did not affect the population of thymic $CD4^+$/$CD8^+$ cells and splenic $CD4^+$/$CD8^+$ cells. Furthermore, the administration of CE enhanced the phagocytic activity of peritoneal macrophages, but decreased the phagocytic activity in vitro. CE decreased the production of nitric oxide from peritoneal macrophages in vivo and in vitro system. These results suggest that CE enhance of cell viability by a direct influence on thymocytes and phagocytic activity by an indirect influence on peritoneal macrophages.
Laboratory column experiments for simultaneous removal of Cd and Cr(VI) are conducted using newly developed material, referred to as Fe-loaded zeolite, which has both reduction ability of iron and ion exchange ability of zeolite. Breakthrough curves were obtained from each column experiment, and described with advection-dispersion equation. Apparent parameters including $K_{app}\;and\;D_{app}$ were newly introduced for effectively describing the Cr(VI) breakthrough curve. $K_{app}$ decreased with increasing initial contaminant concentration and with decreasing flow rates. Whereas, $D_{app}$ were not significantly affected by initial contaminant concentration or flow rate.
In order to investigate the temporal variability of dissolved and particulate trace metals in the Han River, water samples were collected intermittently at two sites for 3 years (August 91 to December 94). Surface seawaters covering the range of salinity were also collected at the estuarine region to evaluate the role of estuary for the riverine fluxes of trace metals within the estuary during October 95 and 96. During the study period, dissolved metal concentrations in riverwaters varied by a factor of 5-10 for Fe, Ni, Co and Cu and 50-100 for Mn, Cd and Pb depending upon the water level; high concentration during the low water and low concentration in high water period except for Fe. The concentration of dissolved Fe increased with increasing water discharge. These concentration-discharge relationships of the studied trace metals are explained by the successive dilution of waters from two different origins, which can be presumably identified as anthropogenic discharges and watershed flushing. Although estuarine waters at early mixing region were not collected due to the difficulty of sampling, mixing behaviors of metals were inferred from the concentration-salinity relationships through the laboratory mixing experiment and field sampling, and distribution coefficients between dissolved and labile particulate phases. It is suggested that the Han River estuary plays a role of accumulating Fe, Mn, Co and Pb from riverine sources due to high turbidity caused by strong tidal current, whereas this system serves as a source of dissolved Cd due to release caused by extended residence time of riverine particles.
Direct exposure of renal tubular brush-border membranes (BBM) to free cadmium (Cd) causes a reduction in phosphate (Pi) transport capacity. Biochemical mechanism of this reduction was investigated in the present study. Renal proximal tubular brush-border membrane vesicles (BBMV) were isolated from rabbit kidney outer cortex by Mg precipitation method. Vesicles were exposed to $50{\sim}200\;{\mu}M\;CdCl_2$ for 30 min, then the phosphate transporter activity was determined. The range of Cd concentration employed in this study was comparable to that of the unbound Cd documented in renal cortical tissues of Cd-exposed animals at the time of onset of renal dysfunction. The rate of sodium-dependent phosphate transport $(Na^+-Pi\;cotransport)$ by BBMV was determined by $^{32}P-Iabeled$ inorganic phosphate uptake, and the number of $Na^+-Pi$ cotransporters in the BBM was assessed by Pi-protectable $^{14}C-labeled$ phosphonoformic acid $([^{14}C]PFA)$ binding. The exposure of BBMV to Cd decreased the $Na^+-Pi$ cotransport activity in proportion to the Cd concentration in the preincubation medium, but it showed no apparent effect on the Pi-protectable PFA binding. These results indicate that an interaction of renal BBM with free Cd induces a reduction in $Na^+-Pi$ cotransport activity without altering the carrier density in the membrane. This, in turn, suggest that the suppression of phosphate transport capacity $(V_{max})$ observed in Cd-treated renal BBM is due to a reduction in $Na^+-Pi$ translocation by existing carriers, possibly by Cd-induced fall in membrane fluidity.
The purpose of this study was to investigate the metallothionein of acute cadmium poisoning mice as a Cadmium index. Forty male ICR mice were injected with cadmium chloride solution from 1/8LD50 to 1/2LD50 dose. At 24 hours after exposed Cd, I examined Cd and metallothionein (MT) intissues(liver and kidney) and fluids(whole blood and urine) and also measured low molecular proteins, N-acetyl-D-glucosaminidase(NAG) and 2- microglobuline (2-MG) in urine. The concentration of Cd and MT of liver kidney whole blood and urine were increased with dose dependent manner. Urinary Cd and urinary MT and very good significance (p<0.01) and urinary MT had good significance with kidney Cd and NAG but not 2-MG Conclusionally MT in urine was very correlated with kidney Cd and urine Cd, So MT maybe useful as a Cd poisoning index.
This study was performed to investigate the effect of chlorella on cadmium (Cd) toxicity in Cd-administered rats. Sixty male Sprague-Dawley rats (14 week-old) were blocked into 6 groups. Cadmium chloride was given at levels of 0 or 325 mg (Cd: 0, 160 ppm), and chlorella powder at levels of 0, 3 and 5%. Cadmium was accumulated in blood and tissues (liver, kidney and small intestine) in the Cd-exposed groups, while the accumulation of Cd was decreased in the Cd-exposed chlorella groups. Fecal and urinary Cd excretions were remarkably increased in Cd-exposed chlorella groups. Thus, cadmium retention ratio and absorption rate were decreased in the Cd exposed chlorella groups. Urinary and serum creatinine, and creatinine clearance were not changed in experimental animals. In addition, metallothionein (MT) synthesis in tissues was increased by Cd administration. The Cd-exposed chlorella groups indicated lower MT concentration compared to the Cd-exposed groups. Moreover, glomerular filtration rate (GFR) was not changed by dietary chI orella and Cd administration. According to the results above, this study could suggest that Cd toxicity can be alleviated by increasing Cd excretion through feces. Therefore, when exposed to Cd, chlorella is an appropriate source which counteracts heavy metal poisoning, to decrease the damage of tissues by decreasing cadmium absorption.
The publications on dietary intake, respiratory intake and blood concentration of cadmium and lead in some Korean women are reviewed. Reported values of dietary Cd intake as geometric mean were $17.1{\mu}g/day$ in 1986, $21.2{\mu}g/day$ in 1994, $16.7{\mu}g/day$ in 2000, and blood concentration were $1.45{\mu}g$ in 1986, $1.27{\mu}g/l$ in 1994 and $2.74{\mu}g/l$ in 2000, respectively. In case of Pb, the dietary intake were $33.1{\mu}g/day$ in 1986, $20.5{\mu}g/day$ in 1994, and $18.4{\mu}g/day$ in 2000 and the blood concentration were $51.5{\mu}g/l$ in 1986, $44.3{\mu}g/l$ in 1994, $37.3{\mu}g/l$ in 2000, respectively. Dietary intake is an almost exclusive route of Cd exposure, however respiratory intake in case of Pb is thought to affective exposure route in Korean women. When compared with the values reported in the literature, both of dietary Cd and Pb intake levels appear to be similar to or somewhat higher than the levels in east and south-east Asia.
One of the methods to increase the solubility of a drug is to use complexation with a cyclodextrin. Due to the hydrophobic nature of the interior cavity of the cyclodextrin, it has been known that undissociated lipophilic drugs can be included within the cyclodextrin by hydrophobic interaction. Recently, inclusion of hydrophilic or dissociated form of a drug has been investigated. In this study, the synergism of pH and complexation with $hydroxypropy-{\beta}-cyclodextrin\;(HP\;{\beta}\;CD)$ to increase the solubility of two oxicam derivatives was investigated. In addition, the effect of partition coefficient of dissociated and undissociated form of the drug on the extent of complexation with HP ${\beta}$ CD was studied. The solubility was measured by equilibrium solubility method. The solubility of tenoxicam and piroxicam increased exponentially with an increase in solution pH above the pKa of the drug in the presence and absence of HP ${\beta}$ CD. The solubility of the drugs increased linearly as a function of HP ${\beta}CD$ concentration at fixed pH. Although the stability constant of ionized species is less than that of the unionized species, the concentration of the ionized drug complex is greater than that of the unionized drug complex due to higher concentration of ionized species at pH 7.3.
The concentrations of heavy metals such as cadmium (Cd), chromium (Cr), copper (Cu), lead (Pb), zinc (Zn), nickel (Ni), and arsenic (As) in sand samples collected from selected children's playground and their correlation with environmental parameters, such as concentration of particulate matter in the atmosphere ($PM_{10}$), apartment age (> 5 years), clay and organic matter contents in sand samples, were analyzed. The average heavy metal concentration in samples was 0.040 mg $kg^{-1}$ for Cd, 0.200 mg $kg^{-1}$ for Cr, 1.75 mg $kg^{-1}$ for Cu, 15.1 mg $kg^{-1}$ for Ni, 3.42 mg $kg^{-1}$ for Pb, 66.7 mg $kg^{-1}$ for Zn and 0.750 mg $kg^{-1}$ for As, all of which were below the environmental regulatory level established by Korea Ministry of Environment. However, in the consideration of direct and oral exposure by children to playground sand, the risk of the concentration range in the samples might be greatly enhanced. Heavy metal concentration in samples collected from high $PM_{10}$ (> $70{\mu}gm^{-3}$) area was slightly greater than in samples from low $PM_{10}$ (< $70{\mu}gm^{-3}$), indicating the contribution of particulate matter in air phase to heavy metal concentration in playground sand samples. The concentrations of both Cd and Pb were the highest in apartments older than 21 years (0.050 mg $kg^{-1}$ and 5.28 mg $kg^{-1}$ for Cd and Pb respectively) and showed positive correlation with apartment age (p<0.01 and p<0.001 for Cd and Pb, respectively). Clay content in playground sands ranged 3.8~11.2% and was positively correlated with heavy metal concentration. Organic matter content was negligible (mostly < 0.1%) and showed poor correlation with heavy metal concentration. In conclusion, concentration of heavy metals in playground sand was found to be predominantly influenced by the apartment age and clay content in sand samples and supplemented by dust deposition of particulate matter ($PM_{10}$) from atmosphere.
To investigate the protein binding characteristics of ibuprofenlysine, the effects of drub conentration, pH, ionic strength and protein concentration on the binding of drug to protein concentration on the binding of drug to protein were studied by fluorescence probe method. The conformational change of protein was investigated by circular dichroism (CD) measurement. As the concentration of drug increases, the association constant decreases. These may be due to complex formation of the probe and drug, or the interaction of the protein-probe complex and drug. The association constant for ibuprofenlysine increased with increasing protein concentration. These finding suggest a sharing of one ibuprofenlysine molecule by more than one protein molecule in the binding. The binding between ibuprofenlysine and protein was dependent on pH and ionic strength. It seems that both hydrophobic binding and some electrostatic forces are involved in the binding of ibuprofenlysing to protein.
