Studies on the antioxidant Effects of Carthami Flos Extract (홍화(紅花) 추출물의 항산화 효과에 대한 연구)

  • Objective : The objective of this study was to investigate the antioxidative effects of Carthami Flos extract. Methods : Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. The effect of Carthami Flos extract was examined far details of total phenolic content concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, the inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : TAC of Carthami Flos extract at the concentration of 5 mg/ml was 1.84 mM Trolox equivalent. 2. TAR of Carthami Flos extract, on the other hand, couldn't be determined due to interference from unidentified compounds. 3. Total phenolic content of Carthami Flos extract at the concentration of 5 mg/ml was 2.01 mM gallic acid equivalent. 4. Concentration of Carthami Flos extract at which DPPH radical scavenging activity was inhibited by 50% was 6.43 mg/ml as compared to 100% by Pyrogallol solution as a reference. 5. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeS04/ascorbic acid. Carthami Flos extract at the concentration of 10 ms/ml slightly but significantly decreased TBARS concentration. The extract continued to prevent lipid peroxidation in a dose-dependent manner. 6. The effect of Carthami Flos extract on reactive oxygen species(ROS) generation was examined using a cell-free system induced by hydrogen peroxide/FeS04. Addition of 1 mg/ml of Carthami Flos extract significantly reduced dichlorofluorescein(DCF) fluorescence. Carthami Flos extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the ektract significantly prevented ROS generation in vitro. Conclusion: : Antioxidant efffcts of Carffami ffor extract seem to be due, at least in part, to the prevention offree radical-induced oxidation, fellowed by inhibition of lipid peroxidation.

  • The purpose of this study was to investigate effect of water extract of Carthami Flos on the proliferation of human cancer cell-lines. The effects of Carthami Flos on the proliferation of A431, HeLa, MOLT-4, K562 cells, Balb/c 3T3 cells, mouse thymocytes, splenocytes and human lymphocytes were estimated by MTT colorimetric assay. The results were as follows; 1. Carthami Flos did not effect A431, HeLa, MOLT-4, K562 cells. 2. The cytotoxicity of mitomycin C on K562 cells was increased by the combination of Carthami Flos. 3. Carthami Flos inhibited the proliferation of Balb/c 3T3 cells. 4. Carthami Flos stimulated the proliferation of thymocytes. 5. Carthami Flos stimulated the proliferation of splenocytes. 6. Carthami Flos stimulated the proliferation of human lymphocytes.

Effect of Corydalidis Tuber.Carthami Flos.Paeoniae Radix Rubra on Anti-Inflamation and function of the Liver at Arthritis Rats (현호색(玄胡索).홍화(紅花).적작약(赤芍藥)이 관절염 흰쥐의 염증과 간(肝)에 미치는 영향)

  • This study is designed to find out the effect of Corydalidis Tuber Carthami Flos Paeoniae Radix Rubra mixed water extract on adjuvant arthritis, and to check the stability for Corydalidis Tuber Carthami Flos Paeoniae Radix Rubra mixed water extract about the liver. At first, rats were devided into four groups; Normal(Non-treated group with FCA), Control(Group administrated saline everyday for 1 week after treatment of FCA), Sample A(group administrated Corydalidis Tuber Paeoniae Radix Rubra Carthami Flos mixed water extract $9mg/200{\sim}220g$ everyday 1 week after treatment of FCA) and Sample B(Group administrated Corydalidis Tuber Paeoniae Radix Rubra Carthami Flos mixed water extract $18mg/200{\sim}220g$ everyday 1 week after treatment of FCA). After then, separated searches were made on the change of edema rate, the number of WBC, the contented quantities of total protein, total bilirubin, GOT, GPT in the serum. Result : In the right planter edema, both of Sample groups showed a decrease with statistical significance in comparison with control group. In the WBC count, Sample B group showed a decrease with statistical significance in comparison with control group and also sample A group showed a decrease, but did not show statistical significance. In the serum total protein, both of samples did not show statistically significant decrease in comparison with the control group. In the serum total bilirubin, GOT and GPT, both of samples did not show any harmful change about the liver in comparison with the control group. According to the above results, it is concluded that Corydalidis Tuber Paeoniae Radix Rubra Carthami Flos mixed water extract had the therapeutic effect of anti-inflammation in adjuvant arthritis with no injury to liver.

Carthami Flos Extract Treatment Restored Changes of Gene Expression on ICH Model in Rats (홍화(紅花) 추출물 투여에 의한 뇌출혈 흰쥐 뇌조직의 유전자 발현 조절)

  • Objectives : The pathophysiology of ICH is not fully understood, therefore, the fundamental therapeutic strategies for ICH also not well inspected either. The genetic profile for the effect of Carthami Flos extract on cerebral hemorrhage in rat brain tissue was measured using microarray technique. Genes displaying expressional change on brain damage were selected and the functional analysis on these genes was conducted. Methods : Rats were placed in a stereotaxic frame after intraperitoneal injection of chloralhydrate, and ICH was induced by injection of collagenase type IV and Carthami Flos extract was administered orally. The molecular profile of cerebral hemorrhage in rat brain tissue was measured using microarray technique to identify up- or down- regulated genes in brain tissue. Results : Upon treatment with Carthami Flos extract on the rat having brain damage, many genes show expressional change. The pattern of gene expressional change can be classified into 8 classes in which two types of classes were composed of recovered genes from up or down-regulation by brain damage, respectively. Conclusions : Further analysis using protein interaction database identified some key molecules that can be used for elucidation of therapeutical mechanism of Carthami Flos extract in future.

  • Sevral flavonoids were isolated from the ethanol extract of Carthami flos which has been used in treatment of uterin congestion and also as analgesic and antiinflammatory. They were elucidated as kaempferol, quercetin, 6-hydroxy kaempferol, kaempferol 3-glucoside (Astragalin), quercetin 3-glucoside (isoquercitrin), quercetin 7-glucoside (quercimeritrin), kaempferol 3-rutinoside and quercetin 3-rutinoside (rutin). The structures of the isolated compounds were established by spectroscopic and chemical methods.

Genome Wide Expression Analysis of the Restored Changes by Carthami Flos Extract Treatment on Rat Brain Injury (흰쥐의 손상된 뇌조직에서의 유전자 발현 변화에 대한 홍화(紅花) 추출물 투여의 작용)

  • Objectives : The source is from the flower of Carthamus tinctorius L., family Compositae. It is used in clinical medicine to promote blood circulation, remove blood stasis, promote menstruation and alleviate pain. In the present study, we investigated the genome wide analysis of Carthami Flos on the intra-cranial hemorrhage(ICH) model. Methods : ICH in rat was induced by injection of collagenase type IV and Carthami Flos extract(CFe) was administered orally. The molecular profile of cerebral hemorrhage in rat brain tissue was measured using microarray technique to identify up- or down- regulated genes in brain tissue. Results : Expression profile showed that diverse genes were up- or down-regulated by ICH induction. Administration of CFe restored the expression level of some of altered genes by ICH to normal expressional level. Interestingly, these recovered genes by CFe were involved in the same biological pathways which were significantly activated or suppressed by ICH. Conclusion : The above results might explain the therapeutic mechanism of CFe on ICH. Further, by analyzing interaction network, core genes was identified which could be key molecular targets of CFe against ICH.

  • Objectives : The purpose of this experiment is to study on the anti-cancer, anti-metastasis and immune response improvement effects of Herbal-acupuncture with Carthami Flos infusion solution(CTT-HAS). Methods : We injected Carthami Flos infusion solution into Chung-wan(CV12) of C57BL/6 mouse which is corresponding to human Chung-wan(CV12). We observed its effect on the number of $CD25^{+}/CD4^{+}$, $CD8^{+}/CD3e^{+}$, $CD69^{+}/B220^{+}$, $NK^{+}/CD3e^{+}$ cells in mouse PBMCs, the number of the pulmonary colony, and the effect on MST and ILS of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results Conclusions : 1. The spleen cells proliferation of the sample groups treated with CTT-HAS extract has increased significantly compared with that of the control group. 2. The percentage of the $CD25^{+}/CD4^{+}$, $CD8^{+}/CD3e^{+}$, $CD69^{+}/B220^{+}$, $NK^{+}/CD3e^{+}$ cells in C57BL/6 mouse PBMCs of the sample groups treated with CTT herbal-acupuncture has increased compared with that of the control group. 3. The lung colony number of the sample groups CTT Herbal-acupuncture has decreased significantly compared with that of the control group. 4. MST and ILS of the sample groups CTT herbal-acupuncture have increased significantly compared with those of the control group.

  • To evaluate the osteotoxic effect of methylmercuric chloride(MMC) on cultured mouse osteoblasts, cytotoxic effect was measured by MTT assay after cultured mouse osteoblasts were incubated with various concentrations of MMC for 20 hours. The protective effect of Flos Carthami(FC) against MMC-induced osteotoxicity was also examined in these cultures. MMC decreased cell viability of cultured mouse osteoblasts remarkably in a dose- and time-dependent manners. In protective effect of FC was remarkably effective in blocking the osteotoxicity induced by MMC. From aboved the results, it is suggested that MMC induce osteotoxicity, and the selective herba extract such as FC is very effective in blocking MMC-mediated neurotoxicity on cultured mouse osteoblasts.

  • Keokhachukeu-tang is the one of Chinese traditional prescription used for the treatment of liver disease. This prescription consists of Carthami Flos (6g), Persicae Semen (9g), Pteropi Faeces (9g), Corydalis Tuber (9g), Moutan Radicis Cortex (6g), Paeoniae Radix rubra (9g), Angelicae gigantis Radix (9g), Cnidii Rhizoma (9g), Linderae Radix (12g), Cyperi Rhizoma (12g), Aurantii Fructus (9g), and Glycyrrhizae Radix (3g). Water extract of Keokhachukeu-tang showed a moderate hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. (omitted)

  • To investigate the anti-obesitic effect of FD extract which consisted of Mori radicis Cortex, Hoelen, Pueraria radix, Schizonepetae spica, Carthami Flos, Bupleuri radix and Saposhnikoviae radix in high-fat diet-fed female lean Zucker rats. Obesity was induced by feeding high-lipid diet contained $3\%$ corn oil and $1\%$ cholesterol for 8 weeks, in which FD extract was added to the diet for treatment group. The rats fed on the high fat diet showed increased gain of body weights, leading to enhanced feed efficiency ratio. Moreover high-fat diet induced hepatic lipid peroxidation and hyperlipemia. The body weight, food efficiency ratio and lipid peroxidation in the liver was decreased as compared with the rats fed on the high fat diet by FD extract. With all such changes, the blood biochemistry about lipid was, if anything, risen. These results suggest that FD extract was supposed to have effective ingredients for improving obesity for controlling adipose tissue.
