Study on the Excited Energy Transfer in Light-harvesting Complex (LH2) of Rhodobacter sphaeroides

  • A green mutant of Rhodobacter sphaeroides 60I was acquired by chemical induction. The blue-shifted of the carotenoid absorption was found in the Light-harvesting complex II (LH2) of the mutant. With the excitation at different wavelength, we observed that the evolution of excited-state dynamics in LH2 of Rhodobacter sphaeroides 60I. The dynamical traces demonstrate a dominant absorption followed concomitantly by an ultrafast transmission increase and decay with 818nm excitation.

  • A new species of Gram-negative halophilic cartenoid producing bacterium was isolated from the Haeundae Coast, Korea. This strain is non-motile, aerobic, orange-pigmented, rod-shaped, and produced carotenoids, mainly astaxanthin. All the type strains of the genus Paracoccus were compared with this strain using 16S rDNA sequence analysis, fatty acid patterns, and physiological reaction profiles. From the results obtained, this strain is classified as a new species, Paracoccus sp..

  • More than ten cartenoid pigments were separated from the dark red extracts of abdominal skin of Bombina Orientalis by TLC, and column chromatography, and the visible and infrared spectrum of purified carotenoids were determined. Two major pigments were identified as ${\beta}$-carotene and 4-hydroxy-echinenone respectively through physical and chemical properties, chromatographic, visible and infrared spectral characteristics.

Excessive Water Tolerance Test by Waterlogging Treatments on the Indian Corn Accessions (침수처리를 통한 인도 수집 옥수수의 내습성 검정)

  • This research was carried out to excessive water tolerance test among corn accessions collected from India to breed corn cultivars targeting India market. The corn accessions were 20 Inbred lines and cultivars from India as well as Korean cultivars Gwangpyungok and Chaloksusu. Excessive water tolerance test was done in the green house by immerging the pots containing corn seedlings for two weeks. Then, the plant heights were measured to compare the control plants that were not grown in the immerging state. The results showed that seven accessions of high tolerance in flooding; H2(92.9%), H18(88.8%), CN114A(98.1%), CN351A(94.3%), Super900M(95.3%), P3394(98.8%), 31N27(96.7%) in which the percent is comparison to the control plants. Whereas nine accessions showed high damage by immerging; H1(78.9%), H8(73.4%), H10(77.1%), H19(79.0%), H26(74.1%), H31(75.7%), H34(77.5%), H36(77.4%), H40(74.6%). However, the reduction on the contents of chlorophyll a, chlorophyll b, carotenoids in leaf revealed contrast results to the flooding tolerance. Particularly, H36($7.249{\mu}g{\cdot}mg^{-1}$) and H40($7.642{\mu}g{\cdot}mg^{-1}$) showed rapid reduction in the chlorophyll a content during the flooding treatment. Whereas two Indian commercial varieties 37N27($0.630{\mu}g{\cdot}mg^{-1}$) and P3394($1.208{\mu}g{\cdot}mg^{-1}$) showed slight reduction. The reduction of chlorophyll a and carotenoid contents was positively correlated during the excessive water stress.

  • The old-gold-crimson ($og^c$) fruit color mutation produces deep red tomato fruit with high lycopene content. age is a null mutation allele of lycopene-${\beta}$-cyclase (Crt-b) gene (B locus) that converts lycopene to ${\beta}$-carotene in the cartenoid biosynthesis pathway in tomato. Breeding of high lycopene tomato cultivars can be accelerated by marker-assisted selection (MAS) for introgression of $og^c$ allele by using a gene-based DNA marker. In order to develop a marker, single nucleotide deletion of adenine(A) with. in a poly-A repeat that has been known to be responsible for frame-shift mutation of $og^c$ was confirmed by resequencing mutant allele and wild-type allele at B locus of several tomato lines. For allele discrimination and detection of $og^c$, derived CAPS (dCAPS) approach was used by designing a primer that artificially introduced restriction enzyme recognition site of Hin fI in PCR products from $og^c$ allele. This dCAPS marker is co-dominant gene-based PCR marker that can be efficiently used for MAS breeding program aiming the development of high lycopene tomato.

  • Carotenoid pigments from muscle of mussel, Mytilus coruscus, and blue mussel edulis, were separated by thin layer and column chromatography. The isolated carotenoids were identified by comparative test with reference carotenoids, reduction with sodium borohydride, isomerization with iodine and absorption spectrophotometry. The carotenoid content in the muscle of mussel were 0.4mg% in male and 2.7mg% in female, and the carotenoids were composed of 23.4%, 33.4% mytiloxanthin, 26.3%, 22.5% 3, 4, 3'-trihydroxy-7', $8'-didehydro-{\beta}-carotene$, 24.8%, 22.8% pectenoxanthin, 14.0%, 9.9% pectenolone and 5.1%, 6.1% diatoxanthin in male and female, respectively. While, the carotenoid contents in the muscle of blue mussel were 1.1mg% in male and 3.2mg% in female, and the carotenoids were composed of 33.8%, 35.6% mytiloxanthin, 28.4%, 44.7% pectenoxanthin, 18.1%, 5.0% diatoxanthin, 9.7%, 8.7% pectenolone and 5.5%, 3.1%, 3, 4, 3'-trihydroxy-7', $8'-didehydro-{\beta}-carotene$ in male and female, respectively.

  • According to Yoshihisa Kuroki's report that the total amount of carotenoid was more in the susceptible to pine bark beetle, than in the resistant ones, carotenoids were extracted from needle leaves of one year old seedlings of Pinus thunbergii, Pinus desiflora, Pinus rigida, Pinus koraiensis, Pinus rigitaeda and Pinus taeda which are all important pines in Korea, to find their resistant ratio to the insect. The carotenoids were analyzed and compared using the spectra of them by spectrophotometer. The results were as follows: 1. The visible absorption spectra of carotenoids in those pine trees were proved to be very similar. 2. The total amount of carotenoids in needles differed with the tree species and the contents were arranged in decreasing order P. koraiensis>P. rigida>P. thunbergii>P. rigitaeda>P. taeda, it seemed that there was corelation between the cartenoid content and the extent of harm caused by the pine bark beetles except P. koraiensis. 3. But carotenoids were contained in Robinia pseudoacacia, Castanea crenata, Chamaecyparis obtusa and Cedrus deodra leaves too. 4. The total amounts of carotenoids in these pines of 9 species were arranged in decreasing order Robinia pseudoacacia>Pinus koraiensis>Pinus rigida>Pinus thunbergii>Castanea crenata>Pinus rigitaeda>Pinus taeda>Chamaecyparis obtusa>Cedrus deodara. Therefore, it was proved that there was no correlation between carotenoid cotent and extent of resistance to the insect. 5. In the thin-layer chromatography of these carotenoids, 13 kinds of components in P. densiftora, P. koraiensis and P. rigida and 12 kinds of spots in other pines, were detected respectively, under ultra-violet fluorescent lamp $3,600{\AA}$ and $2,537{\AA}$. 6. The eighth spots from the bottom in P. densiflora, P. koraiensis and P. rigida were not found in other pines and other 4 species (Robinia pseudoacacia, Castanea crenata, Chamacyparis obtusa, Cedrus deodra). Especially the spot in P. densiflora fluoresced strong cobalt blue-fluorescence under ultra-violet fluorescent lamp $2,537{\AA}$.

Variation in Cartenoid Pigment and lipids of the Arkshell, (Anadara broughtonii) according to the Environmental Factors of the Growing Area (서식환경요인에 따른 피조개육의 Carotenoid색소와 지질성분의 변화)

  • The seasonal variations of environmental sea water, arid carotenoid and lipid composition of reddish muscle tissue of arkshell, Anadara broughtonii in Chungmu and $Y{\check{o}}su-area$ were investigated. In the sea water, pH value, salinity, electrical conductivity of Chungmu and $Y{\check{o}}su-area$ were similar tendency, but water temperature in annual average at Chungmu-area was about $2^{\circ}C$ higher than that of $Y{\check{o}}su-area$. The concentration of nitrite-nitrogen, ammonia-nitrogen, phosphate-phosphours at Chungmu-area were slightly higher than those at $Y{\check{o}}su-area$. Meanwhile the concentration of silicate-silicious at $Y{\check{o}}su-area$ was about 2 times higher than that of Chungmu-area. The bacterial density of the sea water was ranged from 3.6 to 93/100ml for coliform and 3.0 to 15/100ml for fecal colifrom at Chungmu-area, but the coliform was ranged 7.3 to 150/100ml and the fecal coliform was ranged 3.6 to 20/100ml at $Y{\check{o}}su-area$. Among the coliform, 8.9% Escherichia coli, 33.9% Citrobacter freundii, 41.1% Enterobacter aerogenes groups were classified and 16.1% was not identified. In the hemoglobin content of reddish muscle tissue of arkshell, one from Chungmu-area was ranged from 1.1 to 2.5 g /dl and one from $Y{\check{o}}su-area$ was ranged from 1.7 to 4.4 g /dl. In total carotenoid content, 0.80 to 1.28mg/100 g muscle was in Chungmu-area and 0.45 to 0.99mg/100 g muscle was in $Y{\check{o}}su-area$. Among the carotenoid compositions, pectenolone content was 63.0% in annual average from Chungmu area and 59.6% from $Y{\check{o}}su-area$, 6.5% from Chungmu area and 18.9% from $Y{\check{o}}su-area$ in ${\beta}-carotene$, 9.9% from Chungmu-area and 9.1% from Yosu-area in pectenoxanthin, 11.2% from Chungmu-area and 5.2% from $Y{\check{o}}su-area$ in diatoxanthin monoester were observed. In the seasonal variation for the major carotenoids, the content of pectenolone was continuously incresedbefore spawning period and showed the maximum in July, but decresed after spawning period. Diatoxanthin monoester was gradually incresed during all growing period from March to next January. And ${\beta}-carotene$ was decresed before spawning period and showed the maximum in July but incresed after spawning period. The major fatty acids of total lipid and fractionated lipid classes from total lipid namely neutral-, glyco- and phospholipid were $C_{16:0},\;C_{18:1},\;C_{22:6},\;C_{20:2},\;C_{18:2},\;C_{20:5}\;and\;C_{18:3}$ acid in the muscle tissue of arkshell. The content of neutral lipid showed about 2 times higher than polar lipid in the muscle tissue of arkshell from both Chungmu and, $Y{\check{o}}su-area$. The content of total and neutral lipid was decresed during spawning period. In the seasonal variation for fatty acid compositions, the content of polyenenoic acid in total and neutral lipid was decresed and saturated acid was incresed. On contrary, polyenoic acid in glyco and phospholipid was incresed and saturated acid was decresed during spawning period from July to September. The sterol composition in total lipid were mainly consisted of cholesterol, campesterol, brassicasterol and ${\beta}-sitosterol$. Especially, the rate of content in cholesterol showed decrese during spawning period butcampesterol showed increse from March to November.
