Effect of Red ginseng extract on immune function of Israeli carp, Cyprinus carpio. (홍삼추출물이 이스라엘잉어의 면역반응에 미치는 영향)

  • Israeli carps were intraperitoneally injected with an aqueous extract of red ginseng (RG) at five different concentrations (0, 1, 10, 25 and 50 mg/kg of body weight). The fish were sacrificed after 3 days and measured the neutrophil activity and plasma components. The activity of lysozyme, Ht, TIC and NBT reduction were significantly enhanced in the concentrations of more than 10 mg/kg. Further attempts were made to measure the effects of RG on leucocyte function at five different concentrations (1-100 ${\mu}g/m{\ell}$), in vitro. The proliferation of lymphocytes co-incubated with phytohaemagglutin (PHA) and concanavalin A (Con A) was significantly increased at 10 ${\mu}g/m{\ell}$ concentration, but suppressed at the highest concentration (100 ${\mu}g/m{\ell}$). RG alone or with lipopolysaccharide (LPS) had no effect on proliferation at any concentrations tested. Migration and reactive oxygen intermediates (ROI) production of leucocytes were enhanced in all concentration higher than 10 ${\mu}g/m{\ell}$. These results suggest that the RG activates the leucocyte function and may be able to be used as a stimulant of fish innate defences.

Immunohistochemical studies of the pancreatic endocrine cells of the various animals (각종 동물의 췌장 내분비세포의 면역조직화학적 연구)

  • This study was attempted to comparative investigate the types and regional distribution of the endocrine cells in several vertebrates immunohistochemically using seven antisera. From carp pancreas could be observed 4 types which are insulin-, glucagon-, som- and BPP-immunoreactive cells. Insulin-immunoreactive cells were mainly distributed at the periphery and a few cells occupied the central region of the islets. Glucagon-immunoreactive cells were distributed at the periphery of the islets, and som - and BPP-immunoreactive cells were located at the central region. From frog pancreas could be observed 4 types which are insulin-, glucagon-, som- and BPP-immunoreactive cells. Insulin-immunoreactive cells were distributed throughout the islets. Som-immunoreactive cells were distributed at the periphery of the islets, and glucagon- and BPP-immunoreactive cells were found as single cell or as small groups located between the pancreatic acini. From snake pancreas could be observed 3 types which are insulin-, glucagon- and som -immunoreactive cells. Insulin-immunoreactive cells were distributed throughout the small islets, and they also were scattered at the periphery of the large islets. Glucagon-immunoreactive cells were distributed at the periphery of the islets, whereas som-immunoreactive cells were occupied the central region. From Ogolgae pancreas could be observed 4 types which are insulin-, glucagon-, som-and BPP-immunoreactive cells. Insulin-immunoreactive cells were distributed throughout the small islets, but at the periphery of the large one. Glucagon- immunoreactive cells were distributed at the periphery of the small islets and in the large islets showed scattering entired. Som-immunoreactive cells were distributed at the periphery of the small islets and in the large islets were located at the central region. A small numbers of BPP-immunoreactive cells were located at the periphery of the small islets and the exocrine regions. From the pancreas of the Korean native goat could be observed 6 types which are insulin-, glucagon-, som-, BPP-, 5-HT- and porcine-CG-immunoreactive cells. Insulin-immunoreactive cells were distributed throughout the islets. Som-immunoreactive cells were located at the periphery of the islets, but a tew were scattered at the central region of islets and in the epithelium of the secretory duct. Glucagon-, BPP-, 5-HT- and porcine CG-immunoreactive cells were distributed at the periphery of the islets. These findings indicated that the regional distribution patterns and cell types of pancreatic endocrine cells in vertebrates varies considerably among phylogenetically different vertebrates.

Analysis of Residual Fluoroquinolones Contents in Fish (어류 중 Fluoroquinolone계 항균제의 분석 및 잔류량 조사)

  • The residual contents of fluoroquinolones in fish and shrimp were analyzed by using HPLC-FLD and LC-MS. The limit of detection (LOD) and limit of quantification (LOQ) for fluoroquinolones by HPLC-FLD were 0.0030-0.0083 and 0.0090-0.0252 mg/kg, respectively. Those by LC-MS were 0.0019-0.0040 and 0.0059-0.0122 mg/kg, respectively. Recoveries regarding ofloxacin, norfloxacin, pefloxacin, ciprofloxacin and enrofloxacin ranged from 76.4-104.6%. The detection rate of fluoroquinolones was 7.5% from a total of 268 samples. In only one Korean bullhead sample, the sum of ciprofloxacin and enrofloxacin contents was detected at a higher level than its maximum residue limits (MRL) of 0.1 mg/kg. Ofloxacin, norfloxacin and pefloxacin were not detected in any samples and detection rates of ciprofloxacin and enrofloxacin were 3.4% and 6.7%, respectively. In detection rate by species eel (2.6%), loach (2.2%), Korean bullhead (1.5%), carp (0.7%) and snakehead (0.4%) were in order.

Measurement of Backscattering Cross Sections of Live Fish (활어의 후방산란 단면적 측정)

  • The backscattering cross sections of live fish and the probability density function of fish echoes were measured at side aspect. The measurements were made at 40 and 170 in KHz. The specimen fishes on the experiments were two catfish-total length of 16.8cm(L/λ is about 4.5, where the L is length, the λ is the sound wavelength) and 23cm(L/λ, ~6.1), a carp(Cyprinus carpio) of 17.5cm(L/λ, ~4.7), a Telapia(Tilapia mossambica)of 19.5cm(L/λ, ~5.2) and several fishes. Those lengths ranged from 10.7 to 24.0cm. The results of the measured maximum backscattering cross sections were 5.62$\times$10 super(-5) to 7.23$\times$10 super(-4) m super(2). Huang and clay reported that, when the fish was moving gently, the probaiblity density function of the fish echces was approximately the same with the Rayleigh probability density function. Thereafter Ehrenberg et al. also reported that the Rayleigh probability density function on fish was performed at the critical acoustic length ratio or more in theoretical base. Recently Dahl and Mathisen tested the Rayleigh probability density function on fish when the fish-length-to-wavelength ratios were greater than 100. In this paper the experimental result was also not accorded with the Rayleigh probability density function when the fish-length-to-wavelength rations were lower than the critical ratio.

Chilodonella hexasticha(Protozoa, Ciliata) from Korean freshwater fish (한국산 담수 어류에 기생하는 Chilodonella hexaticha에 관하여)

  • During a study to document more fully the parasitic ciliates of Korean freshwater fishes, we found numerous small ciliates in the gills and skin of crucian carp, Carassius carassius. Cursory examination indicated that the ciliate were very similar with Chilodonella sp. In this study, we described the morphometrics of this ciliate in detail and compared with other species of Chilodonella in the world. Numerous small ciliates were observed in the body surface, fins and gills of infected fish and excessive mucus production were seen in those fish. Sometimes ulcer was observed in the body of moribund fish. From the scrutinized observation of the parasite, it was identified as Chilodonella hexasticha. The parasite was dorsoventrally flattened body, without a notch in the posterior margin, and it measured 30-$45{\mu}m$ long and 15-$30{\mu}m$ wide. In number of kineties, the right band ciliature was 3-5 and the left band ciliature was 3-5. The right ventral ciliary band shaped arch and was longer than the left, straight band. It had a single oval macronucleus, 8-$15{\mu}m$ in diameter, a single micronucleus, 2-$4{\mu}m$ in diameter. The cytopharynx was reinforced by 10-16 conspicuous nematodesmata, which shaped like a tube and curved at its inner end. Two contractile vacuoles, one anteriorly at right and the second posteriorly at left were observed in wet mounts. This parasite reproduced by a simple division at $22{\pm}1^{\circ}C$.

Ecological Study on the Seaweed Porphyra pseudolinearis Originated from the East Sea, Korea (동해안 고유종 긴잎돌김(Porphyra pseudolinearis)의 생태학적 연구)

  • Growth and sexual differentiation of the seaweed Porphyra pseudolinearis Ueda have been investigated monthly in the intertidal zone of the East Sea, Korea. Young blades of P. pseudolinearis appeared at the beginning of October. Carp os pores were released at the end of November immediately after carposporangia formation. Then the thalli of P. pseudolinearis were extinguished at the end of March. Young thalli were budded through the stages of conchocelis and conchospore. Thalli showed lanceolate type in shape, cordate type in holdfast, absence of microscopic spinulate process and sexual generation. Ratios of length to width in female thalli ranged from 5.6 to 7.4 at the maturation in December and slightly decreased 3.3 to 4.8 in January and 4.9 to 7.3 in December while the ratios of male thalli ranged from 4.2 to 4.8 in January. On October 12, average five individuals were obsered in a quadrate (30 cm ${\times}$ 30 cm), 238$\pm$18 individuals for the maturation stage in December and then it was reduced to 150 individuals in February and 15 individuals in March. Average sex ratios for female, male and vegetative thalli were 31.3% 46.9% and 21.9% respectively in early December, the beginning time of sex maturation. The sex ratio of female and male thalli in December 17, changed to 69.4%, 30.6% respectively.

Ideological Background of Paving Patterns of Classical Gardens in Suzhou, China (중국 쑤저우 고전원림 포장문양의 사상적 배경 고찰)

  • Compared to pavement of Chinese palace gardens, those of private gardens in south part of the Yangtze River(長江) in China shows variety of patterns. In order to figure out what causes the difference in paving pattern, this article focuses on the traditional Chinese ideologies during the all time. An analysis on 'Classical Gardens of Suzhou', which has been designated as UNESCO World Heritage, was used to determine how ideologies have influence on paving pattern of Classical Gardens. As a result, various Chinese ideologies such as Nature worship, Totemism, Confucianism, Taoism, Buddhism, and Folk Culture were found in paving patterns of the private gardens. Pattern of sun in the Retreat & Reflection Garden(Toisawon) is an example of Nature worship among primitive beliefs of ancient China. Phoenix pattern of the Couple's Retreat Garden(Ouyuan, Liuyuan) reflects Totemistic beliefs. Confucianism is the underlying philosophy of Eight Square pattern, Six Square pattern, and Cross Square pattern. These patterns were mainly used to pave large area and easily found in many gardens. Patterns reflect Taoism are "Wufu holding life" pattern(the Five blessing holding life, 五福捧壽), Flat peach pattern, and Alluding Eight Immortals pattern. Paving patterns related to Buddhism are patterns of 'Eight treasures' and Endless knot pattern appears most frequently since it expresses concept of reincarnation well. Paving patterns shows folk culture and beliefs of the time in China are bat, butterfly, dragonfly, frog, carp, and coin(ingot).

Vertebral Anomalies of Five Different Juvenile of Cyprinid Fishes from Kumho River (금호강(낙동강)산 잉어과 어류 5종에서 치어의 척추골 변형)

  • Frequencies and the types of abnormal vertebrae in the juvenile of five cyprinid fishes collected from the Kumho River during 1995 and 1996 were examined. Types of vertebral anomalies in investigated species were spinal curvature, fused vertebrae, helical sutures of cetera and abnormal vertebrae with one or two additional spines. The frequencies and the types of vertebral anomalies were different among the species. Of all the examined species, the type with one or two additional spines showed the highest frequencies, 11.72~12.11%. The frequencies of fused vertebrae was 4.45~7.68%. Thes two types of vertebral anomalies were observed in all species. Among the several types of fused vertebrae, the frequencies of double fused vertebrae were higher than those of other types. Also, the incidence of fused vertebrae located in the caudal region of vertebral column was much higher than that in other regions. The percentages of spinal curvature and helical sutures of vertebrae in the investigated species were 0.02~0.15% and 0.02%, respectively. Among the examined specimens, vertebral anomalies include fused vertebrae and one or two additional spines were shown in the three species, Korean slender gudgeon (Squalidus gracilis majimae), False dace (Pseudorasbora parva) and Crucian carp (Carassius auratus). In addition to the two vertebral anomalies, spinal curvature was shown in the Korean gudgeon (Squalidus chankaensis tsuchigae). Dark chub (Zacco temmincki) had fused vertebrae, one or two additional spines, spinal curvature, and helical sutures of vertebrae. This species has the most variable vertebral anomalies. Frequencies of fused vertebrae and one or two additional spines in the all tested fishes were not related with their standard lengths measured. However, spinal curvature and helical sutures of vertebrae were shown only in the specimens smaller than 20mm in standard length.

Relationships between the Nutritional Status for Lunch-Box, the Taste of Food and the Environmental Factors of Middle School Girls in Kwangju City (광주시내 여중학생의 도시락 영양실태와 식품 기호 및 환경요인과의 관계)

  • This article concerned with the nutritional status and the taste of the lunch box of 311 middle school girls in Kwangju City from May 18 to June 12 in 1987. The purpose of this research was making materials to show direction of the education about nutrition by checking relationships among the nutritional status for the lunch box, the taste of food and the environmental factors. The results observed in the study were as follows: 1. The intake of nutrition from the lunch box and the ratio between the recommended dietary allowance and the contained nutrients in the lunch box as follows. Calorie(603 Kcal, 78.6%), protein (21.7g, 93.1%) Animal protein (10.3g, 132.1%) calcium (151.7 g, 56.9%) Ferrum 93.1 mg, 51.7%), Vitamin a (129.3 RE, 55.4%) Vitamin B1 (0.29 mg, 72.5%), Vitamin B (0.26 mg, 55.3%) Niacin (4.7 mg, 94.0%), Vitamin C 913.2 mg, 79.0%). Except animal protein, all the recommended dietary allowance. 2. According to the intake of calorie, the ratio of taken carbohydrate, protein and fat was 77.7 : 15.5 : 6.8. The intake of protein was desirable but most of calorie depended on carbohydrate. Among the total intake of protein, the ratio of animal protein was 47.5%, which was a high rate. 3. Most of calorie (75%), was taken from staples, protein (41.5%), vitamin B1 (48.3%) were taken at the same rate from staples and side dishes. Most of fat, calcium, ferrum, vitamin A, vitamin B, and vitamin C, was taken from side dishes. 4. In taking among the five basic food groups, proportion of the grain fourth food group was the highest, light color vegetable the third food groups, proportion of the grain fourth food group was the highest, light color vegetable the third food group was second, and Calcium food the second food group was the lowest. 5. As the staples, students liked tchajangmyon, mandu and ttokkuk as written order. They disliked Kongbap and Patpap. As the side dishes for the lunch box, they liked kimchi, ham, sausage, cuttlefish, dried slices of filefish, eggs and green seaweed as written order. As a side dishes they liked Laver, Cucumber, Squid, lettuce, Potatoes. They disliked pork fat, cow's intestines, cow's liver, Crussian carp, pickled fish. The favorite snack was fruits, ice cream, hamburger, Chocolate and milk. 6. In taking condition of the principal food, rice rate (65.6%) was the most, and mixed food was 5 or 10%. 7. Favorite cooking was frying, roasting and kimch. But disagreeable cooking was pickling. 8. Favorite food was what was pungent but disagreeable food was what was salty. 9. the higher parents educational background was, the higher their income was. Also the more various the information about the nutrition was, the better the intake of nutrition was. 10. The preference tendency influenced in choosing the side dish of the Lunch box. The higher the preference tendency of the girl students became, the better their nutrition condition became.

Evaluation of Habitat Improvement Using Two-Dimensional Fish Habitat Modeling after the Connectivity Restoration in an Isolated Former Channel (2차원 어류 서식처 모의를 이용한 격리된 구하도의 연결성 복원에 따른 서식지 개선 평가)

  • Lateral connectivity between a main channel and a former channel plays an integral role in maintaining ecological functions of stream-floodplain ecosystems. This study virtually restored the connectivity of the former channel, which is currently isolated by channelization, in the Mangyeong River, Korea. Fish habitat improvement after the connectivity restoration was evaluated using River2D, two-dimensional depth-averaged hydraulic modeling, depending on normal and flood flow conditions. Target fish species were crucian carp (Carassius auratus), which are known as lentic species, and pale chub (Zacco platypus), known as lotic species. The weighted usable area (WUA) of the two species was increased after the connectivity restoration: the two-way connection between the main and formal channels was more effective than the one-way connection. The result of the physical habitat simulation at a flood flow condition demonstrated an increased rate of the WUA than during a normal flow condition. In particular, the WUA of pale chub increased about four times on the two-way connectivity restoration. This result suggests that habitat availability of both lentic and lotic fish species will increase after a connectivity restoration, and a two-way connectivity restoration may be more effective. In addition, the restored formal channel would function as a shelter for fish during the flood season.
