Hatchery-reared seeds provides a key source of animal protein for human consumption and restocking for fishery management. For stock enhancement program, we have inspected the hatchery-reared seeds of 33 species in 2009, 44 species in 2010, 43 species in 2011 and 46 species in 2012 for legally designated diseases. Results showed that abalone was the most abundant in the marine species group and then sea cucumber, olive flounder, rockfish and swimming crab were followed. Crucian carp was the most abundant and then mandarin fish, Korean bullhead, melanian snail and Chinese mitten crab were followed in the freshwater species group. The number of inspection for black sea bream, rock bream, scorpionfish, black scraper, and eel has continuously decreased for four years. The inspection for flathead mullet has been carried out only in 2009. The total number of inspection cases for eight pathogens in this study were 8,476 and disqualification cases were 56 (0.67%) by detection of aquatic animals pathogens such as koi herpesvirus, white spot syndrome virus, red sea bream iridovirus or viral haemorrhagic septicemia virus.
We developed and subsequently characterized mouse antibodies (MAbs) against viral hemorrhagic septicemia virus (VHSV, genotype IVa), the causative agent of VHS. Five hybridoma clones secreting MAbs against VHSV were established. The MAbs recognized the glycoprotein (MAbs 2C10, 18H4, 23H6, and 30B7) and nucleocapsid protein (15E10) of VHSV by western blot analysis. All five MAbs reacted with VHSV-infected cells and tissue homogenates of VHSV-infected olive flounder (Paralichthys olivaceus) by western blot analysis. Whereas, no reactivity was observed in normal cells and tissue homogenates of normal olive flounder. Moreover, these MAbs reacted with VHSV, but did not react with other fish viruses (infectious hematopoietic necrosis virus, hirame rhabdovirus, spring viraemia of carp virus, infectious pancreatic necrosis virus, marine birnavirus, and nervous necrosis virus) by enzyme linked immunosorbent assay (ELISA). These results indicate that the MAbs are specific to VHSV and can be of value in VHSV detection.
The purpose of this study was to obtain reference data of parameters for hematological health diagnosis in cultured freshwater fish and also evaluate application of automatic dry-type chemistry analyzer (FUJI DRI-CHEM 3000) used to those blood tests. A blood profile of total 200 fish for rainbow trout (Onchorhynchus mykiss), israel carp (Cyprinus carpio), tilapia (Oreochromis niloticus) and eel (Anguilla japonica) cultured in Inland Fisheries Research Institute of NFRDI was determined by hemoglobin (Hb) and plasma chemistry tests: total protein (TP), albumin (ALB), alkaline phosphatase (ALP), blood urea nitrogen (BUN), lactate dehydrogenase (LDH), triglyceride (TG), total cholesterol (TCHO), creatinine (CRE), aspartate aminotransferase (AST), alanine aminotransferase (ALT), glucose (GLU). The values of ALT, TG, LDH, ALB, TCHO, AST and ALP were outside from the minimum and/or maximum of the established detectable range of the analyzer. ALT and TG were not detectable in the range of 67%~61.5%. LDH, ALB and TCHO were not detectable in the range of 36~17%. AST and ALP were not detectable in the range of 5.5~0.5%. However, the values of BUN, CRE, GLU, Hb and TP were below the detectable limits of the analyzer.

Ministry of Agriculture, Fisheries and Food (MAFF) and National Fisheries Research and Development Institute (NFRDI) have inspected the hatchery-reared seeds of 22 marine species and 11 freshwater species for aquatic animal diseases in stock enhancement program in 2009. Results showed that total 12 local selfgovernments have been restocking the sea with cultured juveniles. Gyeongsangnam-do, Jeollanam-do, Jejudo and Chungcheongnam-do have a preference for marine species seeds to freshwater species. On the contrary, freshwater species were released mostly in Gyeonggi-do, Jeollabuk-do and Chungcheongbuk-do. In the marine species group, abalone was the most abundant as 24.5%, and then sea cucumber (15.2%), olive flounder (11.5%), swimming crab (5.6%), black sea bream and rockfish (6.8%), rock bream (5.1%), black rockfish (4.6%) and scorpionfish (4.5%) were followed. Crucian carp was the most abundant as 19.4%, and then eel (17.0%), Korean bullhead (12.3%), melanian snail (12.0%), catfish (8.4%) were followed in the freshwater species group. The total number of inspection cases in this study were 1,080 and disqualification cases were 19 by detection of aquatic animals pathogens such as red sea bream iridovirus (RSIV), koi herpesvirus (KHV) or white spot syndrome virus (WSSV).

The estuary of the Han River constantly suffers from pollutants and pathogenic microorganisms which could cause serious damage to aquatic organisms living there. Despite of this potential risk, it is hard to find any reliable scientific reports on the status of reportable disease infection to the organisms living in this area. In this study, cyprinid fish and crustaceans in Jeonryu-ri, a region of the Han River estuary, were investigated for the infection by representative reportable communicable diseases(SVC, spring viraemia of carp; KHVD, koi herpesvirus disease; EUS, epizootic ulcerative syndrome; WSD, white spot disease) and parasites. Peripheral fish and primary freshwater fish were observed in Jeonryu-ri with cyprinid caught most frequently. Crustaceans were mostly marine species. No positive bands to any of the reportable diseases were produced in any of the fish and crustacean examined in this study by PCR. No trace of Clonorchis sinensis, a liver fluke potential threat to human health, was detected in any of fish samples. However, many fish were infected by metacecaria of other flukes, and other various parasites such as nematode, cestode, copepod, monosite and acanthocephalan. These results suggest that important aquatic organisms in the Han River estuary is not seriously polluted yet. However, it is important to keep monitoring the diseases since the water quality in this region is constantly changing, and devastating influence of infectious diseases is unpredictable. Further, it is required to expand monitoring area toward upstream and increase the number of fish for examination.

This study evaluated in vitro cytotoxicity of 5 pesticides, including 2 herbicides, 2 germicides, and an insecticide, as an alternative to the fish acute toxicity test. The in vitro cytotoxicity was tested using a neutral red uptake (NRU) assay with epithelioma papulosum cyprini (EPC) cells that originated from the epidermal tissue of Cyprinus carpio (common carp). An in vivo fish acute toxicity test was conducted according to OECD Test Guideline No. 203 using Aphyocypris chinensis (Chinese bleak), Oryzias latipes (Japanese medaka), and C. carpio. The results showed that the sensitivity of the cell viability assay for the pesticides was similar to the fish acute test in ranking order despite having approximately 10 times less absolute sensitivity. The $r^2$ correlation values were calculated as 0.38 (p = 0.26), 0.76 (p = 0.05) and 0.90 (p = 0.01) for A. chinensis, O. latipes, and C. carpio, respectively. These results suggested that the potential of EPC cell viability assay as an alternative to the fish acute toxicity test due to their good correlation and NRU assay is expected to serve as a useful tool for predicting acute fish lethality for pesticides if further studies with a large set of pesticides are conducted.
Fish were grown in a water recycling rearing system consisting of a filter system, of which the area is about $30\;m^2$ and the volume of filter gravels is about $15\;m^3$, 6 rearing and 2 supplementary tans about $7\;m^2$ of surface area each. The results for 180 days from April 18 to October 14,1980 are summarized as following: One rearing tank was used for main experiment stocked with an equal number, and weight of each Israeli strain and $F_1$, hybrid between this strain and Korean local strain of common carp. Gross total production was 461 kg with a net production of 395 kg, and the Israeli strain grew to about 8 times initial weight and the hybrid about 6 times. Based on an analysis of results of the above mentioned main experiment and the other miscellaneous fish grown in the remaining tanks, it was considered to be feasible to implement a commercial production in this system by improving parts of the system and adding a number of the units. A special problem seems to be an adoption of more effective aeration device when fish loading increases. A solid waste removal system was deviced by use of by-pass flow ana this system worked very efficiently in view of supply water economy. A serious problem encountered in this experiment was heavy outbreaks of the gill-fluke, Dactylogyrus sp. which was relatively easily eradicated with Dipterex or DDVP treatment in the first trials, but later on strains which were resistant to any of these chemicals appeared.

BtPlus is a group of biopesticides that are made of Bacillus thuringiensis and immunosuppressant. A new BtPlus that exhibits high insecticidal activity against mosquito larvae has been investigated in control efficacy in field conditions and its environmental safety against aquatic system. This study assessed the control efficacy of BtPlus against mosquito larvae with two different application methods. In aerial spraying application (100 mL per $3.3m^2$), BtPlus was effective at 50% or above formulation concentrations to control mosquito larvae. For a direct application to aqueous mosquito habitat, a semi-field mimicking paddy rice field was constructed. In this condition, BtPlus showed 80% and 100% control efficacies at 0.1% and 0.2% concentrations, respectively. BtPlus also showed 40% mortality against adults at 0.1% concentration in 10% sugar bait. However, its control efficacies against adults were much less than against larvae. Safety assessment of BtPlus against ecosystem was evaluated using young carp (Cyprinus carpio), a water flea (Daphnia magna), and a honey bee (Apis mellifera). BtPlus did not give any adverse effects on these nontarget organisms. Based on these results, BtPlus can be applied to control mosquitoes by direct aqueous application to paddy rice field.
The solubility in water of granular pesticides is not equal because the difference of methods of producing a granular pesticide. This study was conducted to investigate fish acute toxicity of butachlor 5% GR, fipronil 0.4% GR by impregnation and carbosulfan 3% GR, diazinon 0.4% GR by coating, using carp (C. carpio), medaka (O. tatipes) and loach (M. anguillicaudatus). The test solution of each pesticide was prepared by direct addition of granular pesticide and add after grinding granular pesticide to powder in test water, We also investigated $LC_{50}$ and residual concentration until 96 hours. Test results were appeared that the treatment of powder was 1.2~4 time higher than granular and toxicity increased clearly according to elapsed time, And the residual amounts by time were detected much at early time in the powder treatment of butachlor GR and diazinon GR. Conclusively, fish acute toxicity and residual concentration in test solution appeared higher in the powder treatment than treat granular form directly in water. Also, 96 hours toxicity values were stable comparatively and the error is less than 48 hours.
Ergasilus sieboldi, a parasitic Copepoda, was detected from the gill of Silurus asotus fingerling(7~10 cm long) cultured at a fish farm in Chungnam Province (Fig. 1 and 2). The infected fish did not show any particular external symptoms except a feeble swimming around the boarder of the pond. The isolated parasite was of 1.07 mm in body length with two egg sacks of 1.05 mm and one pair of hook-shaped secondary tentacle. Bychowsky(1963) described that this parasite infected the gill of Salmonidae, Serranidae, Cyprinidae and Siluridae and that the fish in the Baltic Sea, Black Sea. Caspian Sea. Japanese sea zones, Siberian rivers and Lake Bikal were infectd. Bychowsky further pointed out that heavily infected fish with this parasite become emaciated and slow in growth. and often die. As this parasite infected on the peri-branchial area. the infected fish showed a severe disturbance of respiration. Urawa et ai.(1980) studied on the development of Neoergasilus japonicus. an Ergasilidae, which infects Japanese freshwater fish. In Korea, Chun(1985) reported the identification of Ergasilus sieboldi from imported carp. and Neoergasilus and Pseudogasilus japonicus from bluegills. respectively. Chun also identified Pseudoergasilus zacconis in cultured ayu(Plecoglossus altivelis). However. this report deals with the detection of Ergasilus sieboldi from cultured catfish(Silurus asotus) for the first time in Korea.
