Amino acid analogs, like other inducers of stress response, induce the synthesis of stress proteins in mammalian cells. In this study, Drosophila Kc cells, in which translation is tightly controlled during stress response, was treated with proline analogs, L-azetidine-2-carboxylic acid (AzC) and 3,4-dehydro-L-proline (dh-P). Kc cells exposed to AzC or dh-P induced the synthesis of several proteins which had the same molecular weights as known heat shock proteins. However, in Kc cells, normal protein synthesis still continued in the presence of amino acids analogs unlike in heat-shocked cells. For the induction of stress response, the incorporation of dh-P into the protein was not essential, but the incorporation of AzC was. The stress protein synthesis was regulated mainly at the transcriptional level by AzC, whereas it was regulated by dh-P at the transcription level and possibly posttranscription level. During recovery, the stress protein synthesis stopped sooner in analog-treated cells than in heat-shocked cells even though the accumulated amount of Hsp70 was much less in proline analogstreated cells. It could be concluded that the proline analogs, AzC and dh-P, induced stress response through a different mechanism from heat shock.

뛰어난 혈당강하작용을 가지고 있는 oxirane carboxilic acid의 analogue 합성과 관련하여 oxirane carboxilic acid의 기본골격을 용이하게 형성할 수 있고 동시에 다앙한 관능기를 갖는 side chain을 도입할 수 있는 방법을 개발함으로써 다양한 유도체들을 합성하는데 이용하고자 함. 방법 및 결과 $\alpha$,$\beta$-unsaturated ester로부터 dioxirane을 이용한 직접적인 epoxidation을 통해서 보통의 방법으로는 얻기 어려운 oxirane carboxilic acid의 ester를 높은 수율로 합성할 수 있는 방법이 개발되었으며 특히 분자내에 cpoxide 존재하에서도 Mitsnobu 방법을 이용한 O-alkylation에 의해 aryl ether 결합을 형성할 수 있는 방법이 개발되었으며 이들 방법을 이용하여 다양한 유도체들을 합성하였다.
Objective: In the present study, an liquid chromatography/mass spectrometry (LC/MS) metabolomics approach was performed to investigate potential biomarkers of milk production in high- and low-milk-yield dairy cows and to establish correlations among rumen fluid metabolites. Methods: Sixteen lactating dairy cows with similar parity and days in milk were divided into high-yield (HY) and low-yield (LY) groups based on milk yield. On day 21, rumen fluid metabolites were quantified applying LC/MS. Results: The principal component analysis and orthogonal correction partial least squares discriminant analysis showed significantly separated clusters of the ruminal metabolite profiles of HY and LY groups. Compared with HY group, a total of 24 ruminal metabolites were significantly greater in LY group, such as 3-hydroxyanthranilic acid, carboxylic acids, carboxylic acid derivatives (L-isoleucine, L-valine, L-tyrosine, etc.), diazines (uracil, thymine, cytosine), and palmitic acid, while the concentrations of 30 metabolites were dramatically decreased in LY group compared to HY group, included gentisic acid, caprylic acid, and myristic acid. The metabolite enrichment analysis indicated that protein digestion and absorption, ABC transporters and unsaturated fatty acid biosynthesis were significantly different between the two groups. Correlation analysis between the ruminal microbiome and metabolites revealed that certain typical metabolites were exceedingly associated with definite ruminal bacteria; Firmicutes, Actinobacteria, and Synergistetes phyla were highly correlated with most metabolites. Conclusion: These findings revealed that the ruminal metabolite profiles were significantly different between HY and LY groups, and these results may provide novel insights to evaluate biomarkers for a better feed digestion and may reveal the potential mechanism underlying the difference in milk yield in dairy cows.
The enzymatic isolation of residual lignins obtained from spruce and beech pulps (obtained by sulfite, kraft, ASAM and soda/AQ/MeOH pulping processes) and their characterization was described in previous publications. Here, the residual lignins have been submitted to potassium permanganate oxidation (KMnO4 degradation), and 9 aromatic carboxylic acids (3 of them are dimeric) were identified after methylation with diazomethane by GC/MS. The analytical challenge during qantification by the internal standard methods was the partly high protein content of the samples, which resulted in elevated anisic acid yields in the degradation mixture of sulfite residual lignins. The results are compared with the KMnO4 degradation of the corresponding MWLs and discussed in terms of S/G ratios and degrees of condensation. The latter was calculated as a quotient between the aromatic carboxylic acids derived from condensed and non-condensed lignin structures. Typical degradation patterns for the various processes have been observed. Among other parameter, the relative compositions between iso-hemipinic acid (which is for condensation in pos. 5 of the aromatic ring) and meta-hemipinic acid and 3,4,5-trimethoxyphthalic acid (both are for condensation in pos. 6 of the aromatic ring) was found to be process specific. Kraft and soda/AQ/MeOH residual lignins yielded higher amounts of iso-hemipinic acid. In contrast, the relative yields of meta-hemipinic acid and 3,4,5-trimethoxyphthalic acid (the latter in beech lignins) are higher in sulfite and particularly in ASAM residual lignin. In case of beech residual lignins the amount of acids originated from non-condensed syringyl type lignin units was surprisingly high. The condensation degree of residual lignins was shown to be generally higher than that of MWLs. This was especially true for the G units. ASAM residual lignin exhibited very high S/G ratios and degrees of polymerization. Causality between condensation degree and total yield of degradation products was demonstrated.
Polyester was prepared through the esterification reaction between watsepaper liquefied by ethylene glycol and carboxylic acid. Liquefaction was carried out at the previously determined condition of 100 minutes, $160^{\circ}C$, and 3% sulfuric acid, and the hydroxyl value of the liquefied product was 411 mg KOH/g. In order to remove bubbles produced during the curing step, the method to introduce a slight nitrogen stream into reaction vessel and/or the method to preheat a polyester film at $85^{\circ}C$ before curing step were used alone or in combination. But if curing temperature was $130^{\circ}C$, simple method to cure a film for 5 hours at $130^{\circ}C$ without using both methods was found to be most effective. The polyesters prepared with various carboxylic acids showed significant different physical properties, and maleic acid was best among them. Also, the effect of reaction time and temperature, C/H (carboxyl group/hydroxyl group) ratio, and type of additive on the crosslinkage of polyester was investigated. Lithium hydroxide or citric acid as additive was used to enhance the crosslinkage of polyester and citric acid was proved to be much more effective than lithium hydroxide. The effect of reaction temperature on the crosslinkage was marginal, but the crosslinkage decreased above $130^{\circ}C$. The crosslinkage was 86% when the polyester was prepared at an optimum condition such as $130^{\circ}C$ and 15 minutes of reaction condition, 1.5 of C/H ratio, $130^{\circ}C$ and 5 hours of curing condition, and 10% addition of citric acid.
Cinnamyl derivatives are abundant secondary metabolite in biomass, and they have been studied on their biological activities. However, little information was available for plant growth regulation of the cinnamyl derivatives. In here, the acid, amide, alcohol, aldehyde and ester of cinnamyl derivatives were screened for their root growth inhibition properties including germination. The aldehyde, amide and ester derivatives showed better the root growth inhibition than the carboxylic acid, and the meta-positioned electron withdrawing group on cinnamyl derivatives enhanced the inhibition activity. 3-Chlorocinnamic acid, cinnamaimde and 4-methoxycinnamaldehyde were highlighted with the early stage root development inhibition ($GR_{50}$ < 100 mg/L) on Brassica campestris.
A gas chromatography-mass spectrometric (GC/MS) procedure for the determination of amineptine (dihydro-10, 11-dibenzo[a, d] cycloheptenyl-5-amino-7-heptanoic acid) and its main metabolites in human urine was described. Amineptine has been known to be extensively metabolized by the ${\beta}$-oxidation of the heptanoic side chain with formation of pentanoic side chain metabolite ($C_5$-metabolite), and lactamizarion by internal dehydration of (${\beta}$-oxidized metabolite (${\delta}$-lactam). The detection of these compounds was based on acid hydrolysis, liquid-liquid extraction and trimethylsilylated derivatization of the carboxylic acid group. For the determination of amineptine and its metabolites in biological fluids, selected ions at the m/ 192, molecular ion and one of the characteristic ions were monitored by GC/MS. On the excretion study of amineptine in human urine, 70-90% of amineptine, ${\delta}$-lactam, and $C_5$-metabolite were found to be excreted within 4 hours and their excretion completed within 20 hours.

Dowex 1-X8 resin ion exchanged with calcon carboxylic acid (CCA-Dowex 1-X8) and 2-methyl-8-hydroxyquinoline(MHQ) impregnated-Amberlite XAD-4 resin (MHQ-XAD-4) were examined for the separation and preconcentration of ferric ion from the various matrices. The stabilities of these resins were investigated, and their capacities on ferric ion were also measured. The effect of pH on the adsorption of ferric ion and matrix ions, such as Al(Ⅲ) and Ca(Ⅱ), was investigated to determine the optimun pH ranges. Separation and preconcentration of iron in aluminium foil and mineral water samples were studied by elution method with these resin columns. The recovered ions by 10ml of 2F nitric acid was determined by flame atomic spectrophotometry. SP-Sephadex C-25 column was used to separate ferrous and ferric ion in mineral water by stepwise elution with ferrozine and 1 % ascorbic acid-ferrozine solution. The concentrated and separated each ion could be determined spectrophotometrically at the analytical wavelenth of Fe(Ⅱ)-ferrozine complex (562nm).
To exploit plant growth promoting bacteria in the roots of Miscanthus sacchariflorus, a biomass energy crop, total 64 bacteria were isolated. For the investigation of plant growth promoting effects from the isolated bacteria, production of indole acetic acid (IAA) and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activities were tested and other cultural conditions were examined. As results, 8 isolates showed plant growth promoting effects on the M. sacchariflorus and an isolate designated Agrobacterium sp. BE516 has the highest activity by enhancing the shoot elongation over 2-fold than the control. Agrobacterium sp. BE516 produced 64 ${\mu}g$ IAA per mL and showed ACC deaminase activity which is involved in the resistance to environmental stress such as high salt and drought. It could grow at low temperature in the range from 4 to $15^{\circ}C$, at pH 4.0 and at 4% NaCl. These results indicate that the Agrobacterium sp. BE516 can be useful as a bio-fertilizer for M. sacchariflorus under the stressed conditions.
