Several pathotypes have been recognized in citrus bacterial canker, which causing serious damage in citrus cultivation area. To control the disease, it is important to understand the pathological diversity and reason of difference in virulence of the causal pathogen. We analyzed 124 strains of Xanthomonas causing citrus bacterial canker by southern hybridization with an internal 3.4-kb BamHI fragment from pthA gene. Assuming each band represented an intact gene, each strain of Xanthomonas was estimated to have approximately 1 to 4 copies of pthA gene. X. a. pv. citri A type had more than 3 copies of pthA gene, and the number of pthA gene in X. a. pv. citri $A^*,\;A^w$, and X. a. pv. aurantifolii B, C were different from 1 to 3 according to the strains. When the pthA gene profile was classified into 13 groups according to the number and size of hybridization bands, most of the A types belong to the 3A group, and 4A and 4B type was dominant when they had 4 bands. However, there was no general pattern of difference between the virulence and pthA gene group in this test.

Bacterial canker of Japanese apricot (Prunus mume Sieb. et Zucc.) was found in all orchards located at southern area of Korea. Typical symptoms were characterized by dark spots formed on fruits, brown lesions on leaves, and bacterial exudate oozed out of the cracked bark of diseased tree. Thirty-eight isolates from 16 different areas were identified on the basis of biochemical and physiological characteristics (LOPAT and GATTa test) and also on the basis of 165 rDNA and ITS sequences. Pathogenicity tests confirmed that bacterial canker of Japanese apricot in Korea is caused by Pseudomonas syringae pv. morsprunorum.

Kiwifruit (Actinidia spp.) is an economically important crop and a bacterial canker disease, caused by Pseudomonas syringae pv. actinidiae (Psa), is the most destructive disease in kiwifruit production. Therefore, prevent and control of the disease is a critical issue in kiwifruit industry worldwide. Unfortunately, there is no reliable control methods have been developed. Recently, interest in disease control using microbial agents is growing. However, kiwifruit microbiota and their roles in the disease control is mainly remaining unknown. In this study, we secured bacterial libraries from kiwifruit ecospheres (rhizosphere, endospere, and phyllosphere) and screened reliable biocontrol strains against Psa. As the results, Streptomyces racemochromogenes W1SF4, Streptomyces sp. W3SF9 and S. parvulus KPB2 were selected as anti-Psa agents from the libraries. The strains showed forcible antibacterial activity as well as exceptional colonization ability on rhizosphere or phyllosphere of kiwifruit. Genome analyses of the strains suggested that the strains may produce several anti-Psa secondary metabolites. Our results will contribute to develop biocontrol strains against the kiwifruit canker pathogen and the disease management strategies.
Plantation forests are established by planting Eucalyptus tree species to provide timber and pulp for the construction industries and to meet the energy needs in Ethiopia. Besides the extensive Eucalyptus plantations in the country, fungal pathogen-related diseases are the main challenges to successful production and management. The disease survey was conducted in the Eucalyptus growing areas of Ethiopia during 2019/2020 and 2020/2021. The objective of this research was to assess the diseases associated with Eucalyptus plant species and identify the causal fungal species. Plants of E. camaldulensis were the dominant species in the survey fields and were severely associated with stem and branch canker diseases. Diseased samples were collected and fungal species were identified as Didymellaceae according to culture morphology and affirmed by internal transcribed spacer sequence analysis. In phylogeny, isolates in the study and a reference strain formed supportive monophyletic clades with strong 90% and 95% support with Didymella coffeae-arabicae and Didymella pinodella respectively. Pathogenicity tests revealed that Didymellaceae could infect E. camaldulensis. The findings are the first reports of Eucalyptus stem canker disease caused by Didymellaceae in Ethiopia.
Citrus canker disease is caused by bacteria Xanthomonas axonopodis .pv. Citri. Shiranuhi cultivar, a hybrid of Kiyomi tangor and Nakano No.3 ponkan was evaluated for resistance to citrus canker based on initiation of disease, percent area of lesion infected and growth rate of bacteria in the leaf under growth chamber condition. Significant differences between susceptible plant and resistant plants were observed in these assays. Resistant plants showed delayed disease symptoms compared to the susceptible plants after spray inoculation of the pathogen. The resistant verities, satsuma, yuzu, and Shiranuhi showed symptoms after six days where as susceptible, mexican lime showed the symptoms just after three days of inoculation. 18 days after inoculation, percent area of lesions developed on leaf and disease severity differed significantly in susceptible and resistant plants, and were ranked as follows: mexican lime > early satsuma =Shiranuhi =yuzu (P <, 0.01). However, 30 days after inoculation, percent area of lesion was further differentiated into resistant and highly resistant plants. That was ranked as follows: sweet orange> early satsuma =Shiranuhi =Kiyomi > yuzu (P < 0.01). These results indicate that host reaction to the bacterial was more distinct when the disease developed for a longer period. Growth rates of a citrus canker bacterium during 16 40 h also were distinct after infiltration into leaves of susceptible and resistant plants, and were ranked as follows: sweet orange> early satsuma =Shiranuhi =Kiyomi =yuzu (P < 0.01). Based on these results, we concluded that Shiranuhi is resistant to citrus canker as compared to Kiyomi, early satsuma, and yuzu.
Northern stem canker caused by $Diaporthe$$phaseolorum$ var. $caulivora$ ($Dpc$) has become a serious disease in soybean. The objectives of this study were to survey the existence of $Dpc$ on soybean in Korea, and to examine the potential pathogenicity of $Dpc$ in seed decay. One such isolate, SSLP-4, isolated from a field-grown plant of the Korean soybean cultivar Danbaekkong, was identified as $Dpc$, based on its morphological and molecular characteristics by sequences of internal transcribed spacer (ITS), translation elongation factor (TEF) 1-${\alpha}$ and ${\beta}$-tubulin regions, as well as pathogenic analyses. Moreover, morphological and molecular analyses revealed that isolate SSLP-4 was nearly identical to $Dpc$ strains from the United States. Pathogenicity tests on hypocotyls of soybean seedlings and detached leaves resulted in typical symptoms of soybean northern stem canker and inoculation on plants at R5-R7 stage caused seed decay. All results suggest that the $Dpc$ strain SSLP-4 can cause both stem canker and seed decay on soybean. Thus, the SSLP-4 isolate has the potential to contribute greatly to understanding of host plant resistance mechanisms, both at vegetative and reproductive growth stages in soybean.
Pseudomonas syringae pv. actinidiae, the causal agent of bacterial canker, is currently causing severe economic losses to kiwifruit production worldwide. The pathogen has affected green-fleshed kiwifruit cutlivars and yellow-fleshed kiwifruit cultivars since 1988 and 2006 in Korea, respectively. In recent years, the biovar 3 strains of P. syringae pv. actinidiae were introduced through imported contaminated pollens and have rapidly spread to neighboring kiwiruit orchards by secondary infection, leading to outbreaks of bacterial canker and tremendous damages on yellow- and red-fleshed kiwifruit cultivars. In this review, we summarize the various management practices of bacterial canker of kiwifruit such as disease escaping, cultural practices, blocking of dissemination, early diagnosis, eradication of inoculum sources, chemical control, and trunk injection on the basis of our research works and field experiences and important research products conducted during the last three decades in the world. Finally, we propose a manual for the efficient management of the disease that can be practically utilized at the farmers' orchards in order to keep kiwifruit vines healthy in the future.
Undiluted neoasozin solution (6.5% a.i.), an arsenic fungicide, was sprayed on 169 cankers of apple trees from early March to September in 1987 twice at intervals of one week without scraping off the affected barks. Among the treated cankers, 79.9% ceased to grow within 1∼7 weeks, 13.0% showed partial development, and 7.1% grew continuously to girdle the branches. The partially developed cankers, however, could also be cured by an additional spray after slightly piercing at the edge of cankers to facilitate the penetration of the chemical. When the canker growth was blocked, cankers were encircled by cracks developed at the marginal area of the cankers. If the cracks developed once, very few cankers grew beyond them. The above results suggest that the crack development may be the consequence of the host defense activity to wall off the pathogen. In addition to the curative efficacy, the neoasozin solution inhibited sporulation of the pathogenic fungus almost completely. However, the pathogen survived for more than three months in some cankers that externally appeared to be cured, suggesting that an indirect mode of action of the chemical against apple Valsa canker seems to be still more persuasive than the direct fungicidal effect. In the final examination conducted in the mid April of the next year, 72.7% of the cankers were completely cured by the two successive neoasozin treatments. Moreover the cure rate became 83.1% if that of partially developed cankers which were also completely cured by an additional treatment was also taken into account. Since 1989 when this method was widely applied in apple orchards in Korea, apple Valsa canker has been effectively controlled to reach a tolerable level.

A bacterial pathogen, Pseudomonas syringae pv. actinidiae (Psa), is a causal agent of kiwifruit bacterial canker worldwide. Psa biovar 3 (Psa3) was first detected in 2011 at an orchard in Dodeok-myeon, Goheung-gun, Jeonnam Province in Korea. In this study, we present the results of an epidemiological study regarding Psa3 occurrence on kiwifruit orchards in Korea for the period of 2013 to 2015. Since the first detection of Psa3 in 2011, there was no further case reported by 2013. However, Psa3 was rapidly spreading to 33 orchards in 2014; except for three orchards in Sacheon-si, Gyeongnam Province, most cases were reported in Jeju Island. Entering 2015, bacterial canker by Psa3 became a pandemic in Korea, spreading to 72 orchards in Jeju Island, Jeonnam, and Gyeongnam Provinces. Our epidemiological study indicated that the first Psa3 incidence in 2011 might result from an introduction of Psa3 through imported seedlings from China in 2006. Apart from this, it was estimated that most Psa3 outbreaks from 2014 to 2015 were caused by pollens imported from New Zealand and China for artificial pollination. Most kiwifruit cultivars growing in Korea were infected with Psa3; yellow-fleshed cultivars (Yellow-king, Hort16A, Enza-gold, Zecy-gold, and Haegeum), red-fleshed cultivars (Hongyang and Enza-Red), green-fleshed cultivars (Hayward and Daeheung), and even a kiwiberry (Skinny-green). However, susceptibility to canker differed among cultivars; yellow- and red-fleshed cultivars showed much more severe symptoms compared to the green-fleshed cultivars of kiwifruit and a kiwiberry.
Bacterial canker caused by Clavibacter michiganensis is considered to be one of the most serious diseases, leading to economic damage to tomato worldwide. Diagnosis of the bacterial canker on tomato is known to be difficult because the causal pathogen is slow-growing on artificial media as well as causes latent infection in tomato. In this study, as a less time-consuming method, a specific primer set was newly designed for rapid detection of C. michiganensis. The method presented here is so simple, easy, and fast that it can be useful and practical in direct detection of the bacterial canker pathogen from tomato plants.
