  • The Purpose of this study was to isolate and identify fungi found in excavated clothes dating on the 16th century. As a result of microscoping with LPCB staining, seven isolated strains were classified-Acremonium strictum, Acremonium kiliense, Fonsecaea sp., Candida sp., Trichoderma sp., Cladosporium spp., Penicillium spp.. In the process of isolation and identification of fungi by the sort of textile, more fungi were found in cotton, cotton batt and silk than in ramie and hemp, suggesting that hemp has antibacterial characteristics due to the presence of lignin. In the Comparison washed samples with unwashed ones, there were more kinds of fungi in washed samples. In the cellulase activity test, Acremonium kiliense had the highest level cellulase activity, suggesting that Acremonium killiense has an important Influence on damage of textiles.

Biological Characterization of Periconicins, Bioactive Secondary Metabolites, Produced by Periconia sp. OBW-15

  • Periconicin A and B, two new fusicoccane diterpenes originally isolated from the cultures of endophytic fungus Periconia sp. OBW-15, were tested by several biological assays. Periconicin A was consistently more active than periconicin B. In an antifungal activity assay, periconicin A showed potent inhibitory activity against the agents of human mycoses, including Candida albicans, Trichophyton mentagrophytes, and T. rubrum, with minimum inhibitory concentration (MIC) in the range of 3.12- 6.25 $\mug$ /ml. In a plant growth regulatory activity assay, periconicins inhibited hypocotyl elongation and root growth of Brassica campestris L. and Raphanus sativus L. At concentrations below 1 μg/ml, however, both compounds accelerated root growth by 110- 135%. From these results, it is apparent that a methyl group positioned in a cyclopentane ring may play an important role in plant and fungal growth inhibitory activity.

  • In order to find some lead compounds for the treatment of opportunistic infections of malaria and pathogenic microbes, an undescribed Indonesian sponge Agelas sp. collected at Manado, Indonesian Waters, was suggested containing active compounds. Crude ethanolic extract of the sponge exhibited significant in vitro antimalarial and antimicrobial activity against Plasmodium falciparum (D6 colne) with $IC_{5O}$ values of $8\;{\mu}/ml$ and against pathogenic microbes such as Candida albicans $(150\;{\mu}/ml)$, Cryptococcus neoformans $(<20\;{\mu}/ml)$, Staphylococcus aureus $25\;{\mu}/ml$, methicillin-resistant Staphylococcus aureus $(<20\;{\mu}/ml)$, and Pseudomonas aeruginosa $(<20\;{\mu}/ml)$. Active compound (5.0 mg) was isolated from the ethanolic extracts of the sponge and purified by using silica gel and ODS column, successively. Active compound was elucidated as dibromosceptrin $(C_{22}H_{24}Br_2N_{10}O_2)$ by detailed analysis of FTESI-MS and comparison of $^1H,\;^{13}C$, DEPT and HMQC NMR spectral data with those reported.

Meroparamycin Production by Newly Isolated Streptomyces sp. Strain MAR01: Taxonomy, Fermentation, Purification and Structural Elucidation

  • Twelve actinomycete strains were isolated from Egyptian soil. The isolated actinomycete strains were then screened with regard to their potential to generate antibiotics. The most potent of the producer strains was selected and identified. The cultural and physiological characteristics of the strain identified. the strain as a member of the genus Streptomyces. The nucleotide sequence of the 16S rRNA gene (1.5kb) of the most potent strain evidenced a 99% similarity with Streptomyces spp. and S. aureofaciens 16S rRNA genes, and the isolated strain was ultimately identified as Streptomyces sp. MAR01. The extraction of the fermentation broth of this strain resulted in the isolation of one major compound, which was active in vitro against gram-positive, gram-negative representatives and Candida albicans. The chemical structure of this bioactive compound was elucidated based on the spectroscopic data obtained from the application of MS, IR, UV, $^1H$ NMR, $^{13}C$ NMR, and elemental analysis techniques. Via comparison to the reference data in the relevant literature and in the database search, this antibiotic, which had a molecular formula of $C_{19}H_{29}NO_2$ and a molecular weight of 303.44, was determined to differ from those produced by this genus as well as the available known antibiotics. Therefore, this antibiotic was designated Meroparamycin.

  • The glucoamylase of Candida tsuubaensis was purified to homogeneity form culture filtrate by means of ultrafiltration, Sephacryl S-200 gel filtration and Sp-Sephadex C-50 chromatography. The purified enzyme was a glycoprotein with a molecular mass of approximately 50 kDa, which was a monomeric protein. Km values were 5.8 mg/ml for soluble starch and 0.04 mM for maltose. Glucoamylase also released only glucose from both pullulan and isomaltose. The analysis of amino acid composition revealed that the enzyme contained a high content of acidic and polar amino acids. In addition, Western blotting analysis indicates that C. tsukubaensis glucoamylase is resistant to glucose repression.

  • Colonial morphology of the various yeasts often encountered in sputum or other clinical specimens was investigated on the corn meal-potato-yeast extract agar medium (GJCPY) containing orange-yellow pigments extracted from Gardenia jasminoides fruits in hopes of differential identification on primary cultures. The results obtained are as follows. 1) Cryptococcus neoformans which is a medically important yeast and whose colony showed brown to purple brown on GJCPY medium was distinguishable not only from buff colored Cr. laurentii after one week incubation but also from Candida spp. 2) Colony color of Candida albicans, a most common species in sputum specimens and of Ca. parapsilosis, a rare isolate, remained unchanged even after 15 days incubation. 3) Ca. tropicalis, second common isolate from sputums and Ca. krusei, a rare isolate, formed a characteristic rough and wrinkled colonies that permit to differentiate them from others. 4) Rare isolates, Ca. guilliermondii and Ca. lusitaniae, turned to prussian blue within three days of incubation. 5) Torulopsis sp. and Saccharomyces cerevisiae showed glossy grayish blue or light blue after one week incubation. The findings clearly showed that Ga. jasminoides pigments medium was useful to the morphological differentiation of medically important yeasts that were often encountered in sputum or other clinical specimens.

Screening and Characterization of Flocculent Yeast, Candida sp. HY200, for the Production of Xylitol from D-Xylose

  • On the basis of high osmotic tolerance and xylitol production, a novel yeast strain was screened from soils of rice farming. The isolated strain HY200 was systematically characterized by using general approaches of Biolog Microlog$^{TM}$ and 18S rRNA sequence analyses, and consequently was designated as Candida tropicalis HY200. Under formulated culture conditions, relatively high xylitol yield ($77\%$) and productivity (2.57 g/l$\codt$h) were obtained, in practice, when 200 g/l of xylose was supplemented. In the utilization of nitrogen, inorganic compounds could not serve as nitrogen sources. As a promising phenotype, HY200 steadily flocculated during and/or after growing in the formulated medium. The extent of flocculation was partly affected by nitrogen sources. However, regardless of the kinds of carbon source fed, the flocculent cells were always observed at the end of the exponential growth phase. These observations strongly suggest that the strain HY200 could effectively be used as a potential candidate for the production of xylitol from xylose, especially in repeated batch mode, because of its flocculation ability and tolerance to high substrate concentrations.

Prevalence and Distribution of Some Dermatophytosis in Relation to Age and Sex of Patients in Minia, Egypt

  • Common dermatophytosis in upper Egypt were studied at three hospitals in Minia city. Tinea corporis (ringworm of glabrous skin) was diagnosed in 127 patients (84 males and 53 females). Trichophyton was the most common genus and was represented by 6 species among which T. violaceum was the commonest fungus. Microsporum was represented by 4 species and M. canis was the prevalent species. Tinea pedis (ringworm of the foot) was noticed in 22 cases (13 males and 9 females) and also Trichophyton and Microsporum were the causal organisms. Three species of Trichophyton namely: T. violaceum, T. kuryngei and Trichophyton sp. were isolated. Tinea cruis (ringworm of the groin) was diagnosed in 21 patients (16 males and 5 females). Epidermophyton floccosum was the most prevalent fungus associated with tinea cruris whereas Trichophyton and Candida were less frequent. Tinea unguium (ringworm of the nail) was diagnosed in 9 patients, 8 were females and one was male and the age incidence ranged between 5 and 55 years. M. canis occurred in 2 cases and T. mentagrophytes in one case. Out of 86 cases of tinea versicolor, 71 were males and 15 were females. Malassezia furfur was the main causal agent. All patients of candidiasis and paronychia were females and Candida was the most prevalent causal agent.

Xylose 발효효모의 분리 및 성질 (Isolation and Identification of Xylose fermenting Yeast)

  • Xylose 발효효모를 토양에서 분리하였으며 (X-6-41) 이는 Lodder씨 분류기준에 따라 Candida 속으로 동정되었다. 삼각플라스크 정치배양법에 의한 xylose 발효시 2% xylose, N원 asparagine, pH 4에서 발효율 약 80%로 나타났고 Mini-Jar for mentor를 이용하여 glucose와 xylose를 1 : 1로 혼합한 총당 5% 기질에서 발효를 시켜 본 바 발효율은 총당에 대해 약 72.3%(alcohol 2.3%(v/v)였다. 본 균주에 의한 xylose 발효는 일반적으로 보고된 바와 같이 호기적 발효과정이였으며 glucose와 xylose 혼합기질의 경우 glucose가 먼저 이용되고 xylose가 나중에 이용되는 것으로 나타났다. 5% xylose를 기질로 하여 Mini-Jar fermentor에서 발효시켰을 때 최대 발효율 69%(alcohol 2.2%(v/v)로 나타났으며 발효중에 xylitol이 생성되었다.

Dual Role of Acidic Diacetate Sophorolipid as Biostabilizer for ZnO Nanoparticle Synthesis and Biofunctionalizing Agent Against Salmonella enterica and Candida albicans

  • In the present study, a yeast species isolated from CETP, Vellore, Tamilnadu was identified as Cryptococcus sp. VITGBN2 based on molecular techniques and was found to be a potent producer of acidic diacetate sophorolipid in mineral salt media containing vegetable oil as additional carbon source. The chemical structure of the purified biosurfactant was identified as acidic diacetate sophorolipid through GC-MS analysis. This sophorolipid was used as a stabilizer for synthesis of zinc oxide nanoparticles (ZON). The formation of biofunctionalized ZON was characterized using UV-visible spectroscopy, XRD, scanning electron microscopy (SEM), and Fourier transform infrared spectroscopy. The antimicrobial activities of naked ZON and sophorolipid functionalized ZON were tested based on the diameter of inhibition zone in agar well diffusion assay, microbial growth rate determination, protein leakage analysis, and lactate dehydrogenase assay. Bacterial pathogen Salmonella enterica and fungal pathogen Candida albicans showed more sensitivity to sophorolipid biofunctionalized ZON compared with naked ZON. Among the two pathogens, S. enterica showed higher sensitivity towards sophorolipid biofunctionalized ZON. SEM analysis showed that cell damage occurred through cell elongation in the case of S. enterica, whereas cell rupture was found to occur predominantly in the case of C. albicans. This is the first report on the dual role of yeast-mediated sophorolipid used as a biostabilizer for ZON synthesis as well as a novel functionalizing agent showing antimicrobial property.
