Chemopreventive Effects of Hydatid Disease on Experimental Breast Cancer

  • Breast cancer is one of the most common and letal cancers in all over the world. Since there have been significant improvements in treatment of breast cancer, there is still a big need for alternative approaches. In this study, we aimed to investigate protective role of hydatid disease against breast cancer. Twenty Wistar rats were divided into two groups of 10 rats each Group I (control) and Group II. In Group II intraperitoneal hydatidosis was performed. Then DMBA was applied to mammary tissues of all rats. Immunohistochemistry studies for Ki-67 and S-100 in the tumoral tissue sections of DMBA induced mammary tumor in rats were performed. TUNEL Assay was used to detect apoptotic cells of tumoral tissue. In vivo anticancer activity testing was carried out by preventing the tumorigenesis by DMBA in mammary tissue of rats. The expressions of the Ki-67 and S-100 protein decreased in rats who had Hydatid Disease (HD) (Group II), compared with the control rats (Group I). TUNEL positive cells were higher in rats with HD (Group II), compared with the control rats (Group I). In vivo studies showed that HD prevented the tumorigenesis by DMBA in mammary tissue of rats with 50 percent.In the light of the evidence the present study showed that HD may have chemopreventive effects on DMBA induced breast cancer.

인간 대장암 세포주에서 파이토케미칼 처리에 의한 유전자 발현 변화 (Global Gene Expression Changes by Several Phytochemicals in Human Colorectal Cancer Cell)

  • 파이토케미칼은 식물유래의 비 영양 성분으로서 항염증, 항산화, 및 화학적 암 예방 등의 생리활성을 가지고 있는 물질이다. 본 연구에서, 우리는 다섯 가지의 다른 파이토케미칼(resveratrol, genistein, epicatechin gallate, diallyl disulfide, caffeic acid phenethyl ester)이 대장암 세포주의 성장과 유전자 발현에 미치는 영향을 연구하였다. 세포 생존율 연구결과, 처리한 ECG를 제외한 모든 파이토케미칼에 의해 농도의존적으로 세포생존율이 감소함을 확인하였다. 또한, oligo DNA microarray 실험을 통해 다섯 종류의 파이토케미칼에 의해 공통적으로 증가 되는 유전자 6개와 공통적으로 발현이 감소되는 유전자 7개를 선별하였다. 공통적으로 발현이 증가되는 유전자를 선택하여 RT-PCR 방법을 통해 발현을 증명하였다. 또한, 파이토케미칼에 의한 NAG-1 단백질의 발현 증가도 확인하였다. 이러한 연구결과는 파이토케미칼에 의해 중재되어 지는 화학적 암 예방법의 일반적인 분자 기전을 이해하는데 도움을 줄 것으로 생각된다.

Antioxidant Properties of Rajgira (Amaranthus paniculatus) Leaves and Potential Synergy in Chemoprevention

  • In recent years there has been a substantial increase in the use of functional foods for disease control. Fruits and vegetables produce phytochemicals such as flavonoids and antioxidants which can lower oxidative stress and reduce the risk of chronic ailments like cancer. The aim of the present study was to investigate the antioxidant capacity and the possible protective effects of Amaranthus paniculatus leaves on the antioxidant defense system in Ehrlich's ascites carcinoma (EAC)-treated mice. Oral administration of the leaf extract at different doses caused a significant decrease in tumor volume, viable cell count and tumor weight and elevated the life span of EAC bearing mice. It also showed an improved antioxidant potential as evidenced by a significant increase in the cellular antioxidant defense system such as catalase, superoxide dismutase and reduced glutathione and also significantly reduced the levels of TBARS. The levels of RBC, hemoglobin and lymphocyte count were altered in EAC bearing mice and were reverted back to near normal levels after the treatment with the leaf extracts. Their adequate content of total phenolics and flavonoids, DPPH scavenging activity which further suggests that the extracts exert a significant protection against oxidative stress conditions.

석류 추출성분이 암세포 증식 억제와 Quinone Reductase 유도활성에 미치는 효과 (Effects of Punica granatum L. Fractions on Quinone Reductase Induction and Growth Inhibition on Serveral Cancer Cells)

  • Various lines of evidence suggest that dietary components protect the initiation step of carcinogenesis. In this study, the ethylacetate (PGMEA), ethylether (PGMEE), butanol (PGMB) and aqueous (PGMA) soluble fractions of Punica granatum L. (PG) were screened for their growth inhibition using the MTT assay on HepG2, HeLa, C6, MCF-7 and HT-29 cells and for their activity to induce quinone reductase (QR) in HepG2 cells. Among various fractions of Punica granatum L., the PGMEE showed the strongest growth inhibition at 500 $\mu\textrm{g}$/mL which resulted 92.5% on Hela cell lines and 97.8% on C6 cell lines. The PGMEA and PGMB also showed significant growth inhibition. The assay of QR induction on HepG2 cells, grown in the presence of PGMEE at the concentration of 50$\mu\textrm{g}$/mL, was 1.4 times more effective compared with the control value of1.0. These results suggested that useful cancer chemoprevention materials could be isolated from PGMEE fraction of Punica granatum L.

Suppression of Human Fibrosarcoma Cell Metastasis by Phyllanthus emblica Extract in Vitro

  • Phyllanthus emblica (PE) is known to exhibit various pharmacological properties. This study aimed to evaluate the antimetastatic potential of a PE aqueous extract. Cytotoxicity to human fibrosarcoma cells, HT1080, was determined by viability assay using the 3-(4,5-dimethylthiazol,2-yl)-2,5-diphenyltetrazolium bromide (MTT) reagent. Cell migration and invasion were investigated using chemotaxis chambers containing membranes precoated with collagen IV and Matrigel, respectively. Cell attachment onto normal surfaces of cell culture plates was tested to determine the cell-adhesion capability. The molecular mechanism of antimetastatic activity was assessed by measuring the gene expression of matrix metalloproteinases, MMP2, and MMP9, using reverse transcription-polymerase chain reaction (RT-PCR) assay. The mRNA levels of both genes were significantly down-regulated after pretreatment with PE extract for 5 days. Our findings show the antimetastatic function of PE extract in reducing cell proliferation, migration, invasion, and adhesion in both dose- and time-dependent manners, especially growth arrest with low $IC_{50}$ value. A decrease in the expression of both MMP2 and MMP9 seems to be the cellular mechanism for antimetastasis in this case. There is a high potential to use PE extracts clinically as an optional adjuvant therapeutic drug for therapeutic intervention strategies in cancer therapy or chemoprevention.

녹차성분 EGCG의 CSK 단백질 조절을 통한 암예방 효과 (Cancer Prevention Effect of Epigallocatechin-3-gallate through Regulate in C-terminal Src Kinase (CSK) Signaling Pathway)

  • A great interest is emerging about green tea as a tool against human cancer proliferation or inflammation, as pointed out by recent reports describing the inhibitory action of epigallocatechin gallate (EGCG) on angiogenesis, urokinase, metalloproteinases, and induction of inducible nitric oxide synthase. We proposed that EGCG may regulate a multi target signaling having wider spectra of action than those actions of single enzymes. CSK (c-terminal Src kinase) protein is a non-receptor tyrosine kinase involved in the cross-talk and mediation of many signaling pathways that promote cell proliferation, adhesion, invasion, migration, and tumorigenesis. Based on the knowledge that CSK activation is important for cancer proliferation we hypothesized that CSK could be a target of EGCG. Here we showed that EGCG effectively suppressed the growth of CSK MEF cell when compare with CSK knockout MEF cell growth. These results indicate that EGCG could be used as a chemoprevention to modulate CSK signal pathway in inflammatory processes and tumor formation.

Chemopreventive Potential of Coumarin in 7,12-dimethylbenz[a] anthracene Induced Hamster Buccal Pouch Carcinogenesis

  • The aim of the present study was to investigate the chemopreventive effect of coumarin against 7, 12-dimethylbenz[a]anthracene (DMBA)-induced hamster buccal pouch carcinogenesis by monitoring tumor incidence and histopathological changes as well as by analyzing the status of biochemical markers (lipid peroxidation, enzymatic and non-enzymatic antioxidants, phase I and phase II detoxification enzymes). Oral squamous cell carcinomas were induced in the buccal pouch of Syrian golden hamsters by painting with 0.5% DMBA in liquid paraffin three times a week for 14 weeks. We noted 100% tumor formation with marked abnormalities in the biomarkers status in hamsters treated with DMBA alone. Oral administration of coumarin at a dose of 100 mg/kg body weight (bw) to DMBA treated hamsters completely prevented the tumor formation as well as restored the staus of biochemical variables. The results of the present study thus suggest that the chemopreventive effect of coumarin is probably due to its anti-lipid peroxidative potential and modulating effect on carcinogen detoxification agents in favor of the excretion of ultimate carcinogenic metabolites of DMBA during DMBA-induced hamster buccal pouch carcinogenesis.

Effect of the Geijibokryunghwan on human hepatocarcinoma cells

  • We invesgated the GBH water extracts can be used as a potential cancer chemopreventive agent in humans, especially in hepatological cancer cell lines. The GBH was found to act as an potent inhibitor of COX-I only, but not as COX-2 inhibitor. Furthermore, the extract mediated anti-inflammatory effects and inhibited COX-associated hydroperoxidase functions(antipromotion activity). Inhibitory effect of the GBH water extracts on the growth of cancer cell lines such as HepG2 cell and Hep3B cell was shown.

Effects of the Geijibokryunghwan on Carrageenan-induced Inflammation and COX-2 in Hepatoma Cells

  • In oriental medicine, Geijibokryunghwan(GBH) was used to improvement various symptoms created by the thrombosis. We investigated the effects of an oriental medicinal prescriptions, Geijibokryunghwan (GBH) consisting of herbs of Cinnamomi Ramufus (Geiji; 桂枝), Poria cocos (Bokrung; 茯?), Moutan Cortex Radicis(Modanpi; 牧丹皮), Paeoniae Radix (Jakyak; 芍藥) and Persicae Semen (Doin; 桃仁) on tumor growth-inhibitory activity and cancer chempreventive activity in assays representing three maior stages of carcinogenesis. Cancer chempreventive agents include nonsteroidal anti-inflammatory drugs (NSAIDS) such as indomethacin, aspirin, piroxicam, and sulindac, all of which inhibit cyclooxygenase (COX). Effects of the GBH extracts on carrageenan-induced edema Inflammation using female (C57BL/6XC3H) Fl (B6C3Fl ) mice and tumorigenesis were examined. Finally, cyclooxygenase metabolites were determined after extracts treatment. These data suggest that GBH extracts merits investigation as a potential cancer chempreventive agent in humans.
