UPLC-DAD-ESI(+)-QToF/MS를 이용한 국내산 두릅나무 및 음나무 순 내 페놀산 특성 비교 (Comparison of Phenolic Acid from Shoots of Aralia elata and Kalopanax pictus Cultivated in Korea Using UPLC-DAD-ESI(+)-QToF/MS)

  • UPLC-DAD-ESI(+)-QToF/MS를 사용하여 국내산 두릅나무 및 음나무 순의 페놀산을 분석한 결과 두릅나무 순에서 총 13종, 음나무 순에서 총 8종의 페놀산 성분을 동정하였으며, 내부표준물질을 이용하여 이들 개별 성분을 정량하였다. 개별 페놀산 성분 구조 동정 시 모분자에 $Na^+$ 또는 $K^+$가 결합되는 특징적인 단편이온들이 관찰되어 모분자의 분자량을 쉽게 판단할 수 있었다. 전반적인 구조 동정은 선행연구 및 표준품의 UV, MS 분석 결과를 참고하여 비교하였으며, 특히 이성질체의 효율적인 동정을 위해 SIM mode 방법을 적용하였다. 따라서 두릅나무 및 음나무 순에서 10종(3,4-dihydroxybenzoic acid, 3-O-caffeoylquinic acid, caffeic acid, 4-O-caffeoylquinic acid, 5-O-p-coumaroylquinic acid, 5-O-feruloylquinic acid, 5-O-caffeoylquinic acid methyl ester, 4,5-di-O-caffeoylquinic acid, 3-O-feruloyl-5-O-caffeoylquinic acid, 3-O-caffeoyl-5-O-feruloylquinic acid)의 페놀산이 처음으로 확인되었다. 총 페놀산 함량은 두릅나무 순, 음나무 순이 각각 754.8, 845.3 mg/100g으로 확인하였다. 주요 성분인 5-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid는 두릅나무 순에서 각각 총 페놀산 함량 중 49, 44%로 서로 비슷한 함량을 보인 것이 비해 음나무 순에서는 각각 91, 3%로 확인되었다. 따라서 음나무 순에 비해 두릅나무 순에서 dicaffeoylquinic acid류의 생합성이 활발하게 이루어졌다는 것을 짐작할 수 있었으며, 본 연구 결과는 이들의 생합성에 관여하는 효소, 유전 인자 등을 규명하는데 중요한 기초 자료로 활용될 것으로 사료된다.

비파엽 열수 추출물에서 분리한 Caffeoylquinic Acid 3 종의 Nitric Oxide 생성 억제 효과 (Nitric Oxide Production Inhibitory Effects of Three Caffeoylquinic Acids Isolated from Hot Water Extract of Eriobotrya japonica L. Leaves)

  • Background: Research on hot water extracts of medicinal plants that are easily applicable in the clinical setting is essential. To confirm the anti-inflammatory-related active compounds present in the hot water extract of Eriobotrya japonica leaves, ability to inhibit nitric oxide (NO) production was measured and active compounds isolated from the extract were analyzed. Methods and Results: Sovent fractionation by solvent was performed to identify the active compounds present in the hot water extract, and the ability of the extract and the fractions obtained to inhibit NO production was measured. Subsequently, based on the results of liquid chromatography (LC) profile analysis of the n-butanol fraction that had a relatively high inhibitory ability of NO production, six subfractions were separated around the main peak. Among the separated subfractions spectra from mass spectroscopy (MS) were analyzed and standard comparisons were performed on the compounds of the three main peaks on the chromatogram. NO production inhibitory activity of subfraction 2 identified as neochlorogenic acid was the highest with an IC50 of 18.49 ㎍/㎖ followed by that of subfraction 5 identified as cryptochlorogenic acid with IC50 of 25.82 ㎍/㎖. Conclusions: Our result, it was confirmed that several caffeoylquinic acids, including neochlorogenic acid and cryptochlorogenic acid present in the hot water extract of E. japonica leaves have an important role as compounds exhibiting anti-inflammatory activity.

Isolation and Identification of bakkenolides and caffeoylquinic acids from the aerial parts of Petasites japonicus

  • The major aim of this work is the research of secondary metabolites isolated from the aerial parts of Petasites japonicus. The plant material is extracted with a polar solvent, which is 95% by volume methanol at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAC and n-BuOH fraction, two bakkenolides and two caffoylquinic acid were isolated using the Diaion HP-20, silica gel, ODS-A, and Sephadex LH-20 column chromatographies. According to the results of the results of physico-chemical and spectroscopic data including NMR, MS and UV. The chemical structures of the compounds were respectively determined as bakkenolide B (1), bakkenolide D (2), 1,5-dicaffeoylquinic acid (3), and 5-O-caffeoylquinic acid (4). These results suggest that the compounds isolated from the aerial parts of this plant were almost identical with known components of Petasites japonicus. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest area and time.

국화과 17종 식물 30% 에탄올 추출물의 페놀성 화합물 분석 및 Peroxynitrite 소거효과 (Phytochemical Analysis of Phenolic Compounds in 30% Ethanolic Extracts from the Compositae Plants and Peroxynitrite-scavenging Effect)

  • Seventeen 30% EtOH extracts from the Compositae plants collected in Gangwon-do, Korea during autumn season were analyzed by HPLC using three standard caffeoylquinic acids (chlorogenic acid, 3,5-di-O-caffeoylquinic acid, 3,5-di-Omuco-quinic acid) and six flavonoids (rutin, isoquercitrin, astragalin, quercitrin, quercetin and kaempferol) to find the composition of phenolic compounds and also assayed to evaluate the peroxynitrite (ONOO$^-$) scavenging effect. The extracts with $IC_{50}$ values less than 2.0 ${\mu}g/ml$ were as follows: Aster tartaricus ($IC_{50}$, $1.26{\pm}0.10\;{\mu}g/ml$), A. maaki ($1.45{\pm}0.03\;{\mu}g/ml$), Solidago virga-aurea, ($1.45{\pm}0.03\;{\mu}g/ml$), Picris hierraciodes var. glabrescens ($1.45{\pm}0.04 \;{\mu}g/ml$), Lactuca triangulata ($1.50{\pm}0.09\;{\mu}g/ml$), Chrysanthemum zawadskii ssp. acutilobum, ($1.79{\pm}0.14\;{\mu}g/ml$). Particularly, the proportion of total phenolic compounds measured in the extract of L. triangulata was highest as the value 54.51%.

Antioxidant and Neuronal Cell Protective Effects of Columbia Arabica Coffee with Different Roasting Conditions

  • In vitro antioxidant activities and neuronal cell protective effects of ethanol extract from roasted coffee beans were investigated. Colombia arabica coffee (Coffea arabica) green beans were roasted to give medium ($230^{\circ}C$, 10 min), city ($230^{\circ}C$, 12 min) and french ($230^{\circ}C$, 15 min) coffee beans. Total phenolics in raw green beans, medium, city and french-roasted beans were $8.81{\pm}0.05$, $9.77{\pm}0.03$, $9.92{\pm}0.04$ and $7.76{\pm}0.01$ mg of GAE/g, respectively. The content of 5-O-caffeoylquinic acid, the predominant phenolic, was detected higher in medium-roasted beans than others. In addition, we found that extracts from medium-roasted beans particularly showed the highest in vitro antioxidant activity on ABTS radical scavenging activity and FRAP assays. To determine cell viability using the MTT assay, extracts from medium- roasted beans showed higher protection against $H_2O_2$-induced neurotoxicity than others. Lactate dehydrogenase (LDH) leakage was also inhibited by the extracts due to prevention of lipid peroxidation using the malondialdehyde (MDA) assay from mouse whole brain homogenates. These data suggest that the medium-roasting condition to making tasty coffee from Columbia arabica green beans may be more helpful to human health by providing the most physiological phenolics, including 5-O-caffeoylquinic acids.

Characterization of Triterpenoids, Flavonoids and Phenolic Acids in Eclipta prostrata by High-performance Liquid Chromatography/diode-array Detector/electrospray Ionization with Multi-stage Tandem Mass Spectroscopy

  • High-performance liquid chromatographic method with diode-array detector and electrospray ionization with multi-stage tandem mass spectroscopy (HPLC/DAD/ESI-$MS^n$) was used to identify the major constituents in a methanolic extract of Eclipta prostrata. The chromatographic separation was performed on a C18 column. Acetonitrile-water was used as a mobile phase. HPLC/DAD/ESI-$MS^n$ allowed the characterization of constituents of E. prostrata, mainly triterpenoids (eclalbasaponin I, II, III, IV, VI), flavonoids (luteolin 7-O-glucoside, demethylwedelolactone, wedelolactone, luteolin, demetylwedelolactone sulfate, luteolin sulfate, apigenin sulfate) and phenolic acids (5-O-caffeoylquinic acid, 3, 4-O-dicaffeoylquinic acid, 3, 5-O-dicaffeoylquinic acid, 4, 5-Odicaffeoylquinic acid).

Chemical Composition of Artemisia argyi Extract (RW0117) and Protective Effects against Gastric Lesions in vivo

  • In this study, we investigated the chemical profile and effects of RW0117 (Artemisia argyi 65 .5 % ethanol extract) on gastric lesions in rats. We optimized and validated a method to obtain the chemical profile of RW0117. We then investigated the antioxidant and anti-inflammatory effects in vitro, and the protective effects on gastric lesions in vivo. The IC50 of 2,2-diphenyl-1-picrylhydrazyl free radical scavenging considering the antioxidant effects of RW0117 was 166.55 ㎍/mL, and the IC50 of nitric oxide scavenging considering the anti-inflammatory effects was 41.16 ㎍/mL. Oral administration of RW0117 at lower concentrations (25, 50, 100 mg/kg) had similar or greater effects than the daily intake conversion concentration (115mg/kg) of a health functional food (Avexol®) in the acetic acid-induced ulcer and the ethanol-induced gastric injury rat models. In addition, oral administration of RW0117 increased the expression of prostaglandin E2, which enhances the protective effect in the gastric mucosa in the ethanol-induced gastric injury rat model. These results suggest that RW0117 may have potential therapeutic uses in the protection of the gastric mucosa.

Inhibition of Glycation End Products Formation and Antioxidant Activities of Ilex paraguariensis: comparative study of fruit and leaves extracts

  • Objectives: Ilex paraguariensis (Aquifoleaceae) is cultivated to produce "yerba mate". Due to its nutritional, energizing, hypoglycemic and antioxidant effects, it is used in the elaboration of food, pharmaceuticals, and cosmetics. The oxidative stress related to protein glycation and production of advanced glycation end products (AGEs) leads to the development of several diseases. The objective of this work was to compare the antioxidant and anti-AGEs activity of a decoction of fruits (F) and leaves (L). Methods: The antioxidant activity was assayed by the DPPH assay and the inhibition of egg yolk lipid peroxidation (ILP), and anti-AGEs activity, through the inhibition of the formation of fructosamine (IF), β-amyloid (Iβ), protein carbonylation (IC) and AGEs (IA). Polyphenols were quantified by HPLC. Results: Maximum response ± SEM: For F 0.01 ㎍/mL: IF = 42 ± 4%, IC = 17 ± 2% and for 10 ㎍/mL: IA = 38 ± 4%, Iβ = 67 ± 7%. For L 0.1 ㎍/mL: IF = 35 ± 2%, IC = 19 ± 2% and for 100 ㎍/mL: IA = 26 ± 3%, Iβ = 63.04 ± 2%. The DPPH IC50 = 134.8 ± 14 ㎍/mL for F and 34.67 ± 3 ㎍/mL for L. The ILP IC50 = 512.86 ± 50 ㎍/mL for F and 154.8 ± 15 ㎍/mL for L. By HPLC L presented the highest amounts of flavonoids and caffeoylquinic acids. F and L showed strong anti-AGEs activity, affecting the early stages of glycation at low concentrations and the late stages of glycation at high concentrations. The highest activity for both F and L was seen in the IF and Iβ. F presented the highest anti-AGEs potency. L presented the highest antioxidant potency, which was related to the highest content of polyphenols. Conclusion: The fruits of I. paraguariensis could be a source of antioxidant and anti-AGEs compounds to be used with medicinal purposes or as functional food.
