Microsatellite marker distribution pattern in rock bream iridovirus (RBIV) infected rock bream, Oplegnathus fasciatus

  • Rock bream (Oplegnathus fasciatus) is a highly valued aquaculture species in Korea. However, the aquaculture industry suffers huge economic losses due to rock bream iridovirus (RBIV) infection in summer. The objective of this study was to determine genetic diversity and relationships of DNAs isolated from two groups of rock bream after RBIV infection using five microsatellite (MS) markers. The first group of fish died early and the second group of fish died later after RBIV infection. In this experiment, 90 fish (5.1±1.0 cm and 4.1±1.3 g) were injected with 50 μl of RBIV (104 TCID50/ml) and maintained at 26℃ for 15 days. Genomic DNAs were extracted from fins of 20 fish that died earlier or later after RBIV infection. These DNAs were subjected to genotyping using five MS markers (CA-03, CA3-05, CA3-06, CA-10, and CA3-36). Of these markers, CA3-05 (early death group), CA3-06 (late death group), and CA3-36 (both early and late death groups) showed different alleles distribution rates. In-depth studies are needed to provide valuable information for selecting RBIV-resistant fish. In conclusion, microsatellite marker distribution pattern differences between early- and late- death groups of rock bream after RBIV infection showing different RBIV susceptibilities were determined using MS markers and genotyping. Results of this study suggest that MS markers could be used to facilitate the selection of RBIV resistant rock bream.

A Study on the Thermal Decomposition Characteristics of Waste PVC Wire Added with CaO (CaO를 첨가한 폐PVC전선의 열적분해 특성에 관한 연구)

  • The thermal decomposition characteristic of waste PVC wires has been studied by using TGA and fixed-bed reactor. The experimental conditions of decomposition temperatures, air flow rates and weight ratio of CaO/PVC were considered in this work. To verify the effectiveness of CaO addition to remove HCl and toxic gases generated from thermal decomposition of PVC wire, the gaseous products obtained from the thermal decomposition of PVC were analyzed by GC/MS(Gas Chromatograph and Mass Spectrometry). To investigate the effect of CaO in thermal decomposition of PVC, liquid products were also analyzed by GC/MS. And the effect of decomposition temperature, air flow rate and CaO/ PVC weight ratio on the yield of liquid, gas and residue fraction have been also studied. From this work, it was found that the removal amount of HCl generated from thermal decomposition of PVC increased with increase of CaO addition.

Simultaneous Determination of Anthraquinone, Flavonoids, and Phenolic Antidiabetic Compounds from Cassia auriculata Seeds by Validated UHPLC Based MS/MS Method

  • A systematic isolation and characterization study for Cassia auriculata (CA) seeds resulted in identifying antidiabetic compounds 1,3,8-trihydroxyanthraquinone and quercetin, quercetin-3-O-rutinoside, gallic acid, caffeic acid, ferulic acid, and ellagic acid. The ultra-high-performance liquid chromatography based triple quadrupole mass spectrometry methodology was developed and validated for simultaneous identification and confirmation of these compounds from CA seeds. Multiple reaction monitoring (MRM) based quantification method was developed with MRM optimizer software for MS1 and MS2 mass analysis. The method was optimized on precursor ions and product ions with the ion ratio of each compound. The calibration curves of seven bioactive analytes showed excellent linearity (r2 ≥ 0.99). The quantitation results found precise (RSD, < 10 %) with good recoveries (84.58 to 101.42%). The matrix effect and extraction recoveries were found within the range (91.66 to 102.11%) for the CA seeds. This is the first MS/MS-based methodology applied to quantifying seven antidiabetic compounds in CA seeds and its extract for quality control purposes.

Optical Properties of CaYAlO4:Tb3+/Eu3+/Ce3+ Phosphors (CaYAlO4:Tb3+/Eu3+/Ce3+형광체의 광학적 특성 분석)

  • $Tb^{3+}$ or $Eu^{3+}$ or $Ce^{3+}$-doped $CaYAlO_4$ phosphor were synthesized by solid-state method. $CaYAlO_4:Tb^{3+}$ is shown that the $Tb^{3+}$-doping concentration has a significant effect on the $^5D_4/^5D_3{\rightarrow}7F_J$ (J=6,...,0) emission intensity of $Tb^{3+}$. The $CaYAlO_4:Tb^{3+}$ phosphors show tunable photoluminescence from blue to yellow with the change of doping concentration of $Tb^{3+}$ ions. The $CaYAlO_4:Eu^{3+}$ phosphors exhibit a red-orange emission of $Eu^{3+}$ corresponding to $^5D_0$, $_{1,2}{\rightarrow}^7F_J$ (J=4,...,0) transitions. The $CaYAlO_4:Ce^{3+}$ phosphors show a blue emission due to $Ce^{3+}$ ions transitions from the 5d excited state to the $^2F_{5/2}$ and $^2F_{7/2}$ ground states. The decay time of $CaYAlO_4:Tb^{3+}$ phosphors decrease from 1.33 ms to 0.97 ms as $Tb^{3+}$ concentration increases from 0.1 mol% to 7 mol%. The decay time of $CaYAlO_4:Eu^{3+}$ phosphors increase from 0.94 ms to 1.17 ms as $Eu^{3+}$ concentration increases from 1 mol% to 9 mol%.

Comparison of Dietary Habits and Nutrient Intakes in Subjects with Obesity or Hyperglycemia Classified Metabolic Syndrome (비만 또는 고혈당 증상 보유에 따른 대사성증후군의 식습관 및 영양상태 비교)

  • Metabolic syndrome (MS) was defined as condition in which the subjects have two or more abnormalities among obesity, hyperlipidemia, hypertension and hyperglycemia. To develop a nutritional education program for MS, this study was performed to compare the dietary habits and nutrients intake of complex symptoms of MS with obesity or hyper-glycemia. The participants in this study were 84 normal adults,62 MS with obesity, 33 MS with hyperglycemia and 54 MS with obesity and hyperglycemia (OB + HG). A dietary survey was conducted using 24-hour recall method. Total cholesterol level of MS with obesity group was significantly higher than other groups. WHR and systolic blood pressure showed no significant difference among MS with obesity, hyperglycemia and OB+HG groups. Dietary intakes of energy, Fe, Vit A, Vit $B_2$ and Ca were less than $75\%$ of 7th Korean RDA in the all groups. Especially, dietary intakes of Vit $B_2$, Vit A and Ca were less than $50\%$ of RDA in MS with hyperglycemia and OB+HG groups. The other nutrient intakes of each group were also below the RDA level except for P, Vit C. It appeared that most of the nutrient intakes in MS with hyperglycemia and OB + HG groups were significantly lower than normal group. In MS with obesity group, each consumption of sweet, organ meat and soup was higher than other groups. Each consumption of garlic and onion in MS with obesity, hyperglycemia and OB + HG groups was lower than normal group. Also, each consumption of soup in MS with hyperglycemia and OB + HG groups was higher than normal group. Indices of nutritional quality (INQ) for Ca, Vit A and Vit $B_2$ were below 1 in all the groups. Food composition group score of MS with hyperglycemia group was significantly lower than normal and MS with obesity groups. Our results indicated that nutritional education program for MS with obesity or hyperglycemia should include specific strategies to modify unsound dietary habits and inappropriate food intake for health.

Strength and Durability Properties of Concrete with Starch Admixture

  • This paper examines some properties of concrete, such as strength, oxygen permeability and sorptivity using starch [cassava (CA) and maize (MS)] as admixtures. Concrete cubes containing different percentages of the CA and MS by weight of cement (0, 0.5, 1.0, 1.5 and 2.0 %) were cast. Compressive strength tests were carried out after 3, 7, 14, 21, 28, 56, 90, 180, 270 and 365 days of curing. Oxygen permeability and sorptivity tests were carried out on another set of concrete specimens with the same percentages of starch at 7, 28, 90, 180, 270 and 365 days. Oxygen permeability and sorptivity tests data obtained were subjected to Kruskal-Wallis one-way analysis of variance by ranks. The strength increase after 1 year over the control for CA 0.5 and CA 1.0 are 2.7 and 3.8 % respectively, while MS 0.5 and MS 1.0 gave 1.5 % increase over control. These results showed a decrease in oxygen permeability and rates of sorptivity, with concretes containing starch as admixtures giving better performance than the control concretes.

The Properties of Na-Ca Exchange Current in Single Atrial Cells of ,The Rabbit (토끼 단일 심방근 세포에서 Na-Ca 교환전류의 특성에 관한 연구)

  • In single atrial cells isolated from the rabbit the properties of inward current of Na-Ca exchange were investigated using the whole cell voltage clamp technique. The current was recorded during repolarization following brief 2 ms depolarizing pulse to +40 mV from a holding potential of * 70 mV. Followings are the results obtained: 1. When stimulated every 30 seconds, the inward currents were activated and reached peak values 6-12 ms after the beginning of depolarizing pulse. The mean current amplitude was 342 pA/cell. 2. The current decayed spontaneously from the peak activation and the time course of the relaxation showed two different phases fast and slow phase. The time constants were 10-18 ms and 60-140 ms, respectively. 3. The recovery of inward current was tested by paired pulse of various intervals. The peak current recovered exponentially with time constant of 140 ms and 1 p M isoprenaline accelerated the recovery process. 4. Relaxation time course was also affected by pulse interval and time constant of the fast phase was reduced almost linearly according to the decrease of pulse interval between 30 sec and 1 sec. 5. The peak activation was increased in magnitude by long prepulse stimulation, 5 p M Bay K, 1 p M isoprenaline or internal and external application of c-AMP. 6. The relaxation time constant of the fast phase was prolonged by 5 p M Bay K or c-AMP, and shortened by isoprenaline. However the time course of the slow relaxation phase was not so much changed. From the above results, it could be concluded that increase of the calcium current by Bay K or c-AMP results in the potentiation and prolongation of intracellular calcium transient, and the facilitation of Ca uptake by SR might be a mechanism of shortening the time constant of current relaxation by short interval stimulation or isoprenaline.

Regional Amyloid Burden Differences Evaluated Using Quantitative Cardiac MRI in Patients with Cardiac Amyloidosis

  • Objective: This study aimed to investigate the regional amyloid burden and myocardial deformation using T1 mapping and strain values in patients with cardiac amyloidosis (CA) according to late gadolinium enhancement (LGE) patterns. Materials and Methods: Forty patients with CA were divided into 2 groups per LGE pattern, and 15 healthy subjects were enrolled. Global and regional native T1 and T2 mapping, extracellular volume (ECV), and cardiac magnetic resonance (CMR)-feature tracking strain values were compared in an intergroup and interregional manner. Results: Of the patients with CA, 32 had diffuse global LGE (group 2), and 8 had focal patchy or no LGE (group 1). Global native T1, T2, and ECV were significantly higher in groups 1 and 2 than in the control group (native T1: 1384.4 ms vs. 1466.8 ms vs. 1230.5 ms; T2: 53.8 ms vs. 54.2 ms vs. 48.9 ms; and ECV: 36.9% vs. 51.4% vs. 26.0%, respectively; all, p < 0.001). Basal ECV (53.7%) was significantly higher than the mid and apical ECVs (50.1% and 50.0%, respectively; p < 0.001) in group 2. Basal and mid peak radial strains (PRSs) and peak circumferential strains (PCSs) were significantly lower than the apical PRS and PCS, respectively (PRS, 15.6% vs. 16.7% vs. 26.9%; and PCS, -9.7% vs. -10.9% vs. -15.0%; all, p < 0.001). Basal ECV and basal strain (2-dimensional PRS) in group 2 showed a significant negative correlation (r = -0.623, p < 0.001). Group 1 showed no regional ECV differences (basal, 37.0%; mid, 35.9%; and apical, 38.3%; p = 0.184). Conclusion: Quantitative T1 mapping parameters such as native T1 and ECV may help diagnose early CA. ECV, in particular, can reflect regional differences in the amyloid deposition in patients with advanced CA, and increased basal ECV is related to decreased basal strain. Therefore, quantitative CMR parameters may help diagnose CA and determine its severity in patients with or without LGE.

The Fermentation Characteristics of the Interspecific Protoplast Fusant of S. peucetius and S. platensis (Streptomyces peucetius subsp. caesius와 S. platensis사이의 원형질체 융합균주의 발효특성)

  • An interspecific fusant strain, Streptomyces MS1 was obtained by protoplast fusion between S. peucetius subsp. caesius and S. platensis. We studied on the fermentation characteristics of the fusant strain. The fermentation products of the fusant MS1 was identical with S. peucetius, but its production of anthracycline was more stable than S. peucetius under various fermentation conditions in regard to acidogenesis of fermentation broth. The optimal medium composition for anthracycline production by fusant MS1 as follows: sucrose 2.0%, glucose 1.0%, soytone 0.7%, $CaCO_3$ 0.2%, $KH_2PO_4$ 0.013%, casamino acids 0.01%, $K_2SO_4$ 0.025%, $MaCl_2\;6H_2O$ 1.024%, 5M $CaCl_2\;5H_2O$ 0.4%, 1N NaOH 0.7%, 20% L-proline 1.5%. In this condition, the productivity of anthracycline was $80{\sim}100\;{\mu}g/ml$.

Proteomic Analysis of Serum of Women with Elevated Ca-125 to Differentiate Malignant from Benign Ovarian Tumors

  • Clinically, elevated cancer antigen 125 (CA-125) in blood predicts tumor burden in a woman's body, especially in the ovary, but cannot differentiate between malignant or benign. We here used intensive modern proteomic approaches to identify predictive proteins in the serum of women with elevated CA-125 to differentiate malignant from benign ovarian tumors. We identified differentially expressed proteins in serum samples of ovarian cancer (OC) patients, benign ovarian tumor (BT) patients, and healthy control women using mass spectrometry-based quantitative proteomics. Both the OC and BT patients had elevated CA-125. Quantitation was achieved using isobaric tags for relative and absolute quantitation. We obtained 124 quantified differential serum proteins in OC compared with BT. Two proteins, apolipoprotein A-4 (APOA4) and natural resistance-associated macrophage 1, were verified using Western blotting. Proteome profiling applied to OC cases identified several differential serum proteins in the serum of women with elevated CA-125. A novel protein, APOA4, has the potential to be a marker for malignant tumor differentiation in the serum of women with elevated CA-125.
