A Study on the Influence of Water Quality on the Phosphorus Fraction Properties from Reservoir Sediments (저수지 퇴적물로부터 인의 존재형태가 수질에 미치는 영향에 대한 연구)

  • The present study was attempted to find the effects of structural properties of phosphorus on the water quality of Gyehwa reservoir in Saemangeum. Relationship of phosphorus fractions between water and sediment properties was closely examined, and a few types of phosphorus were found from the sample sediment as : Saloid-P, Al-P, Fe-P, Ca-P, Red-P and Occd-P. Saloid-P (1.4%), Al-P (0.5%), Fe-P (39.8%), Ca-P (56.6%), Red-P (0.4%), Occd-P (1.3%) were extracted in a mass basis from the sediment of Gyehwa reservoir. Approximately more than 97% of phosphorus were calcium related phosphorus (Ca-P, 56%) and iron bound phosphorus (Fe-P, 39.8%). The Fe-P closely relates with water quality of T-N (r=0.761, p<0.05), $NO_3$-N (r=0.754, p<0.05), $NH_4$-N (r=0.728, p<0.05), T-P (r=0.774, p<0.05) and $PO_4$-P (r=0.767, p<0.05) while the Ca-P did not show any consistent dependency on the water quality. On the other hand, the correlation of Ca-P with $P_2O_5$ was high with r=0.783 (p<0.05) in the sediment. The Fe-P was affected significantly on the Ignition Loss (r=0.569, p<0.05), T-N (r=0.715, p<0.05) and T-P (r=0.983, p<0.01). In the research of correlation between phosphorus fraction and heavy metals in the sediment, Ca-P did not show any specific relationships with heavy metals. The Fe-P showed a significant correlation with As (r=0.817, p<0.01), Cu (r=0.793, p<0.05), Cd (r=0.786, p<0.05), Zn (r=0.738, p<0.05), so that it can be stated that the presence of Fe-P may implicate the volume of various metallic elements.

  • Nano-sized hydroxyapatite (HAp) powders were synthesized by a microwave-hydrothermal method using $H_3PO_4\;and\;Ca(OH)_2$ as starting materials. The applied microwave powers and mole ratio of Ca/P were served as powerful factors in the synthesis of calcium phosphate. In the case of relatively low microwave power of 450 Wand Ca/P ratio of 1.57, the mixed calcium phosphate compounds were detected in the synthesized powders. But in the case of running at 550 Wand 1.67(Ca/P), the synthesized powder showed the monophase of HAp having two kinds of morphologies. One was a needle shape with $5\~15\;nm$ in width and $20\~50\;nm$ in length, and the other was a spherical shape of $10\~40\;nm$ in diameter.

Purification of a Protease Produced by Bacillus subtilis PCA 20-3 Isolated from Korean Traditional Meju (전통 메주로부터 분리한 Bacillus subtilis PCA 20-3 유래 Protease 의 정제)

  • Bacillus subtilis PCA20-3 was isolated from meju and was found to produce a protease. The strain produced the maximum amount of enzyme in the medium containing soytone (0.2%), soluble starch (2%), $(NH_4)_2SO_4\;(0.1%),\;CaCl_2(0.1%),\;yeast\;extract\;(0.01%),\;K_2HPO_4\;(0.1%),\;and\;KH_2PO_4\;(0.1%)$. Protease was first concentrated by ammonium sulfate (80% saturation, w/v) precipitation of culture supernatant. Then the enzyme was purified by column chromatography using CM Sephadex C-50. The collected proteins were rechromatographed using Sephadex G-100 gel filtration column. The fraction with protease active from Sephadex G-100 gel chromatography was found to be pure when examined by SDS-polyacrylamide gel electrophoresis and YMC-pak reverse phase chromatography. Specific activity, yield and purity were 76 U/mg. 2.7%, and 7.6 fold, respectively. The molecular weight of the enzyme was estimated to be 31.5 kDa by SDS-PAGE. The number of amino acids calculated from molecular weight was evaluated about 321 residues. N-terminal sequence of the enzyme was $Val^1-Pro^2-Tyr^3-Gly^4-Val^5-Ser^6-Gln^7-Gly^8-Lys^9-Ala^{10}$.

Investigations of Soil Chemical Properties in the Cultivation Fields of Rubus coreanus with Different Growth Ages (복분자 재배 기간에 따른 토양의 화학적 특성 변화)

  • This study was conducted to investigate the relations between soil chemical properties and Korean raspberry (Rubus coreanus Miq.) growth. Soils were collected from 54 sampling sites in 1 to 6 years old Korean raspberry cultivation fields (RCFs) at Gochang-gun, Jeollabuk-Do, Korea. In RCF with two to six years old plants, soil pH and exchangeable $Ca^{2+}$ and $Mg^{2+}$ contents were higher at the leaf-emergence time, but EC, available phosphorus content, CEC, and total nitrogen content were higher at the harvesting time; especially at the harvesting time, the content of available phosphorus in the RCF with 3~6 years old plants were at least three times higher than in the RCF with 2 years old plants. Water-soluble anions ($Cl^-$, ${NO_3}^-$, ${PO_4}^{2-}$, and ${SO_4}^{2-}$) in the RCF were also measured. The contents of ${NO_3}^-$ and ${SO_4}^{2-}$ were always higher in the RCF with older plants than in the RCF with younger plants, which were not affected by sampling time and sampling locations (rhizosphere and non-rhizosphere sites). However, soils collected from non-rhizosphere at the leaf-emergence time contained higher ${PO_4}^{2-}$ content, but $Cl^-$ content was higher in the soils collected from rhizosphere at the harvesting time. In general, soils in the RCFs contained excess amounts of inorganic nutrients such as available phosphorus and exchangeable $K^+$. Thus, optimal levels of soil properties for Korean raspberry cultivation should be reconsidered.

  • We investigated the effects of a highly saline solution (HS) containing hypochlorous acid, calcium chloride ($CaCl_2$), and phosphoric acid ($H_3PO_4$) on cell death and growth rate of laver Porphyra yezoensis and green laver Ulva intestinalis. Cell death rates of laver treated with HS and HS plus hypochlorous acid (HS + HOCl) in the harvesting stage were less than 0.5%, and there were no significant differences between the HS and HS + HOCl treatments. However, cell death of green laver treated with HS + HOCl in the harvesting stage was greater than 81.2%. These results indicate that the addition of HOCl is highly effective to eradicate noxious green laver without causing damage to laver. The addition of HOCl and $H_3PO_4$ to HS did not increase the area or weight of laver blades. A combination treatment of $CaCl_2$ and HS, however, significantly increased the area and weight of laver lades compared to controls (P<0.05).

  • This experiment was conducted to study on the productive conditions of cellulase by Aspergillus saitoi in the shaking culture medium. The results were as follows: 1. The production of enzyme required higher concentration of corn steep liquor than that of dextrin. 2. The concentration of 1.0% $NH_4H_2SO_4$ produced the enzyme excellently than 3.0%. 3. The cellulase was produced very slowly by adding $(NH_4)_2SO_4$, but the final concentration of the enzyme was higher than control. The production was suppressed by addition of $CaCO_3$. 4. The addition of $1.0{\sim}2.0%$ substrate caused an increase or stimulation in cellulase production.

Growth and Yield Response of Highbush Blueberry 'Duke' to Hydroponic Cultivation (블루베리 '듀크' 품종의 양액재배 시 수체 생장과 수량 반응)

  • The hydroponic culture for growing 'Duke' blueberry was evaluated in a protective greenhouse provided with similar environmental conditions to the conventional blueberry cultivations. One year old 'Duke' blueberry bushes planted in 180 L containers filled with 130 L peat moss and 40 L pearlite (v/v) were selected for the experiment. A nutrient solution consisted with NO3-N 4.6, NH4-N 3.4, PO4-P 3.3, K 3, Ca 4.6, and Mg 2.2 mmol-1 was supplied to the plants, comparing non-hydroponic treatment (provided with only underground water). Hydroponic culture increased number of shoot per bush by 18% and total shoot length by 24% compared with non-hydroponic culture. Total dry weight of a bush increased in the hydroponic with vigorous root growth 1.4-fold more than the non-hydroponic. Higher concentrations of inorganic elements and organic compounds were found in the hydroponic, indicating active nutrient absorption of the bush. The hydroponic produced high yield similar to adult bush from 4 years old age, maintaining the yield until 8 years old age. The findings of this study indicated that hydroponic cultivation systems will be useful for growing blueberry crop.

  • Factors affecting isolation and fusion of protoplasts of three Quercus species were investigated and procedures for isolation, purification and fusion of protoplasts of the three species were also established. Unhardened leaves and rapidly growing callus cultures were good source of viable protoplasts. The optimum composition of enzyme mixture for rapid isolation of protoplasts from leaf mesophyll tissues and calli was Cellulase Onozuka R-10 (20g/l, Macerozyme R-10(10g/l), Pectinase(250 units/l, $CaCl_2$, $2H_2O$(14mM), $MgSO_4{\cdot}7H_2O$(1.8mM), $KNO_3$(1.0mM), $H_3BO_3$(1.0mM), $KH_2PO_4$(0.2mM), KI($1.0{\mu}M$), 1,4-dithiothreitol (0.1mM) and mannitol (0.6M). Optimum density of protoplasts for maximum fusion was $2{\times}10^5/ml$ which was the highest protoplast density given in this study. Optimum concentration and duration of PEG 1450 treatment for inducing fusion appeared to be 29%(W/V) final PEG 1450 concentration and 5-10 minutes, respectively.

Phosphorus removal by lime-natural mineral dissolved solutions

  • In previous studies, solely ferric (Fe3+) and calcium (Ca2+) ions were commonly used for removal of PO4-P (considered as T-P in this study) in wastewater via chemical precipitation. Herein, the removal of total phosphorus (T-P) in wastewater was performed using various mineral and lime dissolved solutions. The dissolution kinetics of different minerals (feldspar, olivine, elvan, illite, sericite, and zeolite) and lime was compared and used their solutions for T-P removal of real wastewater. The highest T-P removal (almost 90%) was obtained by the lime dissolved solution and followed by zeolite, illite, feldspar, and others. We observed a significant co-relationship (R of 0.96) between the amount of initial Ca2+ and T-P removal. This was induced by formation of hydroxyapatite-like mineral via Ca-P precipitation reaction at high pH solution. Furthermore, additional removal of suspended solid (SS) and chemical oxygen demand (COD) was achieved by only lime dissolved solution. Finally, the lime-feldspar dissolved solutions were prepared at different ratios (10-50%), which showed a successive T-P removal up to two times by samples of 40 and 50%.

Production and Purification of Acetylcholinesterase Inhibitor from Pseudomonas sp960903 (Pseudomonase sp. 960903에 의한 acetylcholinesterase 억제제의 생산 및 정제)

  • To screen agent for the treat-ment of Alzhimers Disease several strains of bacteria producing acetylcholinesterase inhibitor ware isolated from soil. Strain 960903 showed strong acetylcholinesteras inhibitory activity and low butyrylcholinesterse inhibitory activity. The strain 960903 was identified as Pseudomonas sp. Acetylcholinesterase inhibitor ws highly achieved in fermentation medium containing soluble starch 3.0%, glycerol 1.0%, pharmamedia 0.5%, KCI 0.3%, $CaCO_3$ 0.2%, MgS $O_4$..$7H_2$O 0.05%, $KH_2$$PO_4$ 0.05%(pH6.5) at $30^{\circ}C$ for 4 days. Acetylcholinesterase inhibitor was purified by Diaion WA-30($OH^{-}$) column charomatography and cellulose column chromatography. Acetylcholinesterase inhibi-tor showd the maximum wavelength at 205 nm and was soluble in water, acetic acid, ethanol, methanol and dime-thyl sulfoxide. The concentration of 50% inhibition($IC_{50}$) of inhibitor against acetylcholinesterase was 25$\mu\textrm{g}$/ml. The inhibitor was inactivated on heating ar $100^{\circ}C$ fro 15 min and more stable in acidic region than alkaline region.n.
