This study was designed to investigate the effects of aerobic and /or Ca supplementation for six months on lipid metabolism and blood pressure in postmenopausal women. The subjects were healthy 29 women aged from 60 to 70 years old. They were divided into four groups: placebo and Ca supplementation group with and without exercise. The amount of Ca supplemented was 500mg a day. The frequency of doing exercise was three times a week and each time, it took 40 minutes to complete all the course of aerobic dancing programmed for old women. During the experimental period, the subjects ate their usual diets and the use of druge as well as additional exercise was prohibited. The results were summarized as follows. 1. Percent body fat and BMI were significantly decreased and the body weight tended to decreased due to aerobic dancing. 2. Serum lipids content tended to show only a slihgt changes due to Ca supplementation: serum triglyceride, total-chol, LDL-chol, FFA content tended to decrease but serum HDL-chol / LDL-chol ratio tended to increase after the experiment in Ca group. On the contrary, serum lipids level was not changed by exercise. 3. Blood pressure did not show any significant changes by Ca supplementation and / or exercise for 6 months in old women. 4. Serum Ca level of Ca group was significantly increased after experiment in Ca group (p<0.05) but not in Ex-Ca group. In summarization, it appeared that the aerobic dancing was a definite way to reduce percent body fat and BMI in postmenopausal women. On the other hand, Ca supplementation seemed to be able to induce favorable changes in serum lipids. However, any synergistic metabolic effects of exercise and Ca supplementation was not seen in this study. Further study is needed to elucidate the relationship between exercise and/or Ca supplementation and the changes in blood lipids profile as well as blood pressure more clearly.
A 44-day metabolic study was conducted to compare the effects of soy protein and meat protein on N, Ca, P, and Mg metabolism in young Korean males. Meat protein diet(MPD) and soy protin diet(SPD) were provided for 3 weeks each. For MPD, meat protein Supplied 50.3% of total protein and mean daily suplies of Ca, P, and Mg were 33.28mg, 1013.01mg, 226.81mg respectively. For SPD, soy protein supplied 51.8% of total protein and mean contents of Ca, P and Mg were 737.86mg, 1261.76mg, and 451.3mg. Mean daily fecal and urinary excretions of Ca were 350.6mg and 113.3mg for MPD and 515.0mg and 118.3mg for SPD. Apparent absorption rates in MPD and SPD were 4.2mg in MPD and 700.4mg and 475.1mg SPD. Apparent absorption rates were 65.6% in MPD and 44.5% in SPD(p<0.01). For Mg, fecal and urinary excretions were 144.8mg and 81.2mg in MPD and 292.4mg and 65.5mg in SPD. Apparent absorption rates were 37.5% in MPD and 35.2% in SPD. The correlations between urinary Ca, P, and their balance were significant (p<0.001) in MPD, but not in SPD. Urinary Ca was significantly correlated with urinary N, P, Mg(p<0.001) in MPD, but not in SPD. From these results, meat protein seems to increase the excretions of major minerals compared to soy protein.
To study the effects of the age and the dietary protein content on Ca metabolism male rats of 1 month 6 month 12 month of age were fed experimental diets containing 5%, 15% or 50% casein for 4 weeks. Food and ca intake were higher in old rats and in high protein groups. The weight ash and Ca contents of femur and tibia were higher in old rats. The higher dietary protein level resulted in higher skeletal weigh ash and Ca contents. But high protein diet(50% casein) lead to reduced bone mineral density(ash/dry bone weight) and Ca density(Ca/dry bone weight) in 1 month old rats. Low protein diet(5% casein) on the other hand reduced the bone growth even though the bone density was higher in this group. The ill effect of low protein diet was not evident in 12 month old rats. Glomerular filteration rate(GFR) and urinary Ca excretionincreased with age and with dietary protein level especially in 12 month old rats. Serum immunoreactive parathyroid hormone(iPTH) level tended to be higher in aged rats but was not affected by dietary protein level except 1 month old rats where 50% protein group showed significantly higher value. This study showed that the dietary protein level seemed to have different effect on Ca metabo-lism in rats of different age., The low bone density in the high protein group of growing rats may be due to the higher iPTH level and increased urinary Ca. The dietary protein level however had no effects on the bone composition in aged rats even though the higher urinary Ca excretion. In conclusion this study suggests that high protein intake from young may lead to less peak bone mass and to increase the bone loss in later years, which would increase the risk for osteporosis.
The effects of dietary calcium levels on the blood pressure and calcium metabolism were investigated. Nine normotensive female college students having hypertention family history were participated in 4-week dietary expeiments. They were provided with either high Ca diet (HCa, average 797mg/day) or low Ca diet(LCa, average 225mg/day) during two weeks, each, consecutively. Sodium amounts of the body diets were 3566~4022mg/day, which were ordinary sodium intake levels in Korea. After the HCa, systolic blood pressures(SBR) in both seated and isogrip-seated postitions were decreased by about 2.5mgHg, comparing with those after the LCa(p<.05). Diastoilc blood pressures(DBP) were not changed by dietary calcium levels. Serum total Ca, ionized Ca, Mg and P levels and Ca/Mg ratio were not different between the HCa and the LCa. Serum parathyroid hormone(PTH) levels were similar between two diets, but individually in seven of nine subjects, the slightly lower values of PTH were observed after the HCa than after the LCa. Urinary excretion of Ca(p<.01), Mg(p<.05) and P(p<.1) were increased after the HCa comparing with the LCa, but Ca/Mg ratio were not different between the two diets. SBP was in positive correlations with boty urinary excretion of Ca(supine, r=.7356, p<.05) and urinary Ca/Mg ratio(isogrip-seated, r=.7483, p<.05). SBP was also negatively correlated with serum P level(supine, r=-.6930, p<.05) and DBP was in negative correlation with urinary P excretion(seated, r=-.8586, p<.01). Serum total and ionized Ca, Mg, Ca/Mg ratio were not significantly correlated with blood pressures.
TRPA1 and TRPV1 are members of the TRP superfamily of structurally related, nonselective cation channels. TRPA1 and TRPV1 are often co-expressed in sensory neurons and play an important role in somatosense such as cold, pain, and irritants. The first leaves of Kalopanax pictus Nakai (Araliaceae) have long been used as a culinary ingredient in Korea because of their unique chemesthetic flavor. In this study, we observed the intracellular $Ca^{2+}$ response to cultured cells expressing human TRPA1 (hTRPA1) and human TRPV1 (hTRPV1) by $Ca^{2+}$ imaging analysis to investigate the ability of the first leaves of K. pictus to activate the hTRPA1 and hTRPV1. An 80% ethanol extract of K. pictus (KPEx) increased intracellular $Ca^{2+}$ influx in a response time- and concentration-dependent manner via either hTRPA1 or hTRPV1. KPEx-induced response to hTRPA1 was markedly attenuated by ruthenium red, a general blocker of TRP channels, and HC-030031, a specific antagonist of TRPA1. In addition, the intracellular $Ca^{2+}$ influx attained with KPEx to hTRPV1 was mostly blocked by ruthenium red, and capsazepine, a specific antagonist of TRPV1. These results indicate that KPEx selectively activates both hTRPA1 and hTRPV1, which may provide evidence that the first leaves of K. pictus primarily activate TRPA1 and TRPV1 to induce their unique chemesthetic sense.
This study was performed to investigate the effects of dietary protein and calcium levels on calcium metabolism in eight healthy Korean adult females. The 2-day metabolic study consisted of a 2 day adaptation period and three 6-day experimental periods. Three experimental diets were low protein low calcium(LPLCa : protein 44g, Ca 422mg), higher protein low calcium(HPLCa : protein 85g, Ca 365mg), and high protein high calcium (HPHCa : protein 84g, Ca 727mg). The apparent calcium absorption was likely to be affected by the calcium intake rather than by the protein intake. Average calcium absorption rate was about 23-29% of calcium intake. The calcium balance was -21.44mg for LPCa, -25.02mg for HPLCa, and -3.22mg for HPHCa. Avergae urinary calcium excretion was 127.7mg for LPLCa, 108.6mg for HPLCa, and 215.4mg for HPHCa. Urinary calcium excretion was more closely related to the changes of calcium intake rather than of protein intake. These results seemed to be due to the interactions between the high phosphours contained in the high protein diet and the little discrepancy of protein intake levels.
To investigate the effect of levels of dietary protein and age on bone metabolism 40% and 5% casein were fed to the rats of 2 & 13 months of age for 12 weeks. High protein groups showed higher bone weight and Ca content than low protein groups and urinary Ca loss was increased in high protein groups but the difference disappeared gradually. A significant increase in urinary hydroxyproline excretion was noted in high protein groups of both age. Another short term study was undertaken to study if the above effect was related with renal function or PTH. Extremely high and low protein diets(60%, 6%) were fed to the rats of different ages(6wks, 6mos.) for 2 weeks, Urinary Ca excretion was significantly increased in high protein groups of young and aged rats and GFR was increased as well. There was no difference in serum iPTH levels between low and high protein groups, but it was elevated in aged rats. Alkaline phosphatase activity was higher in young rats, reflecting faster bone formation. The observed hypercalciuria in high protein groups, especially in aged rats, seems to be related to higher GFR, and PTH dose not appear to be a major mediator.
Acetone-butanol-ethanol (ABE) fermentation by the anaerobic bacterium Clostridium acetobutylicum has been considered a promising process of industrial biofuel production. Phosphotransbutyrylase (phosphate butyryltransferase, PTB) plays a crucial role in butyrate metabolism by catalyzing the reversible conversion of butyryl-CoA into butyryl phosphate. Here, we report the crystal structure of PTB from the Clostridial host for ABE fermentation, C. acetobutylicum, (CaPTB) at a 2.9 Å resolution. The overall structure of the CaPTB monomer is quite similar to those of other acyltransferases, with some regional structural differences. The monomeric structure of CaPTB consists of two distinct domains, the N- and C-terminal domains. The active site cleft was formed at the interface between the two domains. Interestingly, the crystal structure of CaPTB contained eight molecules per asymmetric unit, forming an octamer, and the size-exclusion chromatography experiment also suggested that the enzyme exists as an octamer in solution. The structural analysis of CaPTB identifies the substrate binding mode of the enzyme and comparisons with other acyltransferase structures lead us to speculate that the enzyme undergoes a conformational change upon binding of its substrate.
The effects of dietary Ca and Na levels on lipid metabolism in hyper lipidemic/hypercholesterolemic rats were examined. In Expt. 1, normal rats were divided into six groups and fed high fat(15%, w/w)/cholesterol(1%, w/w) diet containing two levels of Na, low (0.05) or high(1.5%) and three levels of Ca, low(0.1%), normal (0.5%), or high(1.5%) for 8 weeks. In Expt. 2, hyperlipidemia / hypercholesterolemia rats were induced by feeding high fat / cholesterol diet for 4 weeks. They were divided into four groups and fed the high fat / cholesterol diet, containing two levels of Na, low or high and two levels of Ca, low or high for 4 weeks. In Expt. 1, total lipid and total cholesterol contents in serum and liver were significantly lower in rats fed high Ca diet than in rats fed normal or low Ca diet regardless of dietary Na levels. Serum TG was the highest in rats fed low Ca and low Na diet. In Expt. 2, Serum total lipid, TG, and total cholesterol levels decreased by 24, 35, 26% respectively in rats fed high Ca diet regardless of dietary Na levels. Serum total lipid level tended to increased in rats fed low Na diet. The total lipid and TG contents in liver slightly decreased in rats fed high Ca diet. Another observation was that high Ca intake significantly faciliated the fecal lipid and cholesterol excretion regardless of dietary Na levels. There results suggest that the hypolipidemidc/hypocholesterolemic effects of high Ca diet could be partly due to increase in lipid and cholesterol excretion and these effects may be independent of dietary Na levels.
