Effects of Feeding Different Protein Supplements on Digestibility, Nitrogen Balance and Calcium and Phosphorus Utilization in Sheep

  • Two metabolism trials were conducted with 24 wether lambs to investigate the effects of feeding crab meal and other protein supplements on N utilization, digestibility and Ca and P balance in sheep. The lambs (avg. BW, 25 kg) were randomly allotted to eight diets in each of two trials. The supplements were: i) none, negative control (NC); ii) soybean meal (SBM), control; iii) supplement based on industrial byproducts of both plant and animal origin (IPA); iv) experimental supplement based on byproducts of animal origin (ESA); v) hydrolyzed supplement No 4. (HESA); vi) commercial supplement based on animal protein (CS), $Pro-Lak^{(R)}$ vii) crab meal (CM); and viii) urea (U). The supplements supplied 33% of the total dietary N (CP, 9.8%; DM basis). Lambs fed the NC diet had lower (p<0.05) DM and OM digestibility. Lower (p<0.05) apparent absorption of N was recorded for the lambs fed the HESA and NC diets. Sheep fed CM had lower Ca absorption compared to SBM. Highest (p<0.05) P absorption was observed for lambs fed CS and CM and lowest for U and NC diets. Sheep fed CM had higher (p<0.05) total VFA concentration (65.7 ${\mu}mol/ml$), compared to those fed ESA, CS, and NC diets (47.3, 49.8, and 49.5 ${\mu}mol/ml$, respectively). Highest (p<0.05) ruminal $NH_3$ N (29.6 mg/dl) was observed in lambs fed the U diet, while those fed the NC diet had the lowest (p<0.05) average value (7.66 mg/dl). Lambs fed the U diet had the highest (p<0.05) blood urea N (10.67 mg/dl). The present study showed that N utilization of diets supplemented with experimental supplements based on feather meal and blood meal; commercial supplement based on animal protein, $Prolak^{(R)}$ supplement based on plant protein and blood meal; and crab meal are comparable with that of soybean meal.

Effect of Boron on the Development of Adventitious Roots in Sunflower Seedlings (해바라기 유식물의 부정근 발달에 미치는 붕소의 효과)

  • Three-day-old sunfower(Helianthus annuus L.) seedlings were do-rooted and incubated in the nutrient solutions that contained either deficient or sufficient boron supply, and exposure to various pH and temperatures. In the absence of boron, no adventitious roots were formed in the majority of the seedlings. Boron caused the development of numerous adventitious roots in the lower part of the hypocotyl. The low pH damage was ameliorated by the simultaneous application of calcium, implying the involvement of calcium-requiring process in overcoming proton toxicity. Borate showed a strong ameliorative effect with $Ca^{2+}$. These results suggest that the primary target of proton toxicity may be linked to a disturbance of the stability in the pectic polysaccharide network, where calcium plays a key role in plant roots. Maximum temperature for the growth of adventitious roots was $25^{\circ}C$. Although cessation of growth is the most apparent symptom of boron deficiency, exogenous ascorbate improved adventitious root growth in plants in the absence of boron. From the results it is suggested that the inhibition of adventitious root resulting from boron deficiency, low pH and temperature damage may be a consequence of disrupted ascorbate metabolism.

The Effect of Dietary Sodium on Calcium Metabolism in Healthy Young Adult Women (한국 성인 여자에 있어서, 나트륨 섭취 수준이 체내 칼슘대사에 미치는 영향)

  • The present study was conducted to examine the effect of an increased level of dietary sodium on calcium excretion in 8 health young adult Korean women on a controlled diet. After adaptation period of 2 days, each subject received 2811.8$\pm$68.1 mgNa(day during the initial period of 5 days (low sodium period) and 6417.1$\pm$248.6mgNa(day during the following period of 5 days (high sodium period). Calcium intake was 593.7$\pm$15.7 mg Ca/day during the low sodium period of 596.1$\pm$25.1 Ca/day duing the high sodium period. When the low sodium period is compared with the high soidum period, the results were as following. 1) Mean urinary sodium excretion was significantly higher during the high sodium period (5760.1$\pm$156.5mg0 than during the low sodium period (2272.2$\pm$108.6mg)(P<0.001). Fecal sodium excretion of the high sodium period was also significantly higher than that of the low sodium period(P<0.001). Mean value of sodium balance during the high sodium period was higher than that of the low sodium period . However, the difference was not significant. 2) Mean urinary calcium excretion was significantly higher during the high sodium period than during the low sodium period ; mean value of the low sodium period was 124.7$\pm$11.3mg and that of the high sodium period was 202.6$\pm$17.2mg)P<0.001). Fecal calcium excretion was higher during the high sodium period (284.9$\pm$31.0mg) than during the low sodium period (253.9$\pm$15.3mg0, but there was no significance. Mean value of calcium balance during the high sodium period was significantly lower than that of the low sodium period(P<0.001). The above results show that high sodium intake increases calcium excretion as well as sodium excretion.

Effect of Fermented Sea Tangle on the Alcohol Dehydrogenase and Acetaldehyde Dehydrogenase in Saccharomyces cerevisiae

  • Sea tangle, a kind of brown seaweed, was fermented with Lactobacillus brevis BJ-20. The gamma-aminobutyric acid (GABA) content in fermented sea tangle (FST) was 5.56% (w/w) and GABA in total free amino acid of FST was 49.5%. The effect of FST on the enzyme activities and mRNA protein expression of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) involved in alcohol metabolism in Saccharomyces cerevisiae was investigated. Yeast was cultured in YPD medium supplemented with different concentrations of FST powder [0, 0.4, 0.8, and 1.0% (w/v)] for 18 h. FST had no cytotoxic effect on the yeast growth. The highest activities and protein expressions of ADH and ALDH from the cell-free extracts of S. cerevisiae were evident with the 0.4% and 0.8% (w/v) FST-supplemented concentrations, respectively. The highest concentrations of GABA as well as minerals (Zn, Ca, and Mg) were found in the cell-free extracts of S. cerevisiae cultured in medium supplemented with 0.4% (w/v) FST. The levels of GABA, Zn, Ca, and Mg in S. cerevisiae were strongly correlated with the enzyme activities of ADH and ALDH in yeast. These results indicate that FST can enhance the enzyme activities and protein expression of ADH and ALDH in S. cerevisiae.

Effects of Red-Yeast-Rice Supplementation on Bone Mineral Density and Bone Mineral Content in Overiectomized Rats (홍국의 첨가가 난소절제 쥐에서 골밀도 및 골함량에 미치는 영향)

  • This study was performed to evaluate the effect of red-yeast-rice on bone metabolism in overiectomized (OVX) rats. Forty female Sprague-Dawley rats (body weight 210 $\pm$ 5 g, 9 weeks old age) were divided into two groups. One group were OVX, and the other group received sham operation (SHAM), and received either control diet (20% casein) or a red-yeast-rice power supplemented diet (0.1%) for 9 weeks. And then each rat group was further divided into control diet (casein 20%) and red-yeast-rice powder supplemented (0.1%) diet group. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin, USA) in spine and femur on 5, 9 weeks after feeding. The serum and urine concentrations of Ca and P were determined. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, IGF-l and calcitonin were analyzed using radioimmunoassay kits. Urinary Ca and P excretion were not significantly different among the groups. Within the OVX group, the red-yeast-rice group had a lower crosslinks value than the casein group. Therefore the red-yeast-rice supplemented groups had a lower bone resorption ratio than the casein group in the ovariectomized rats. And, the red-yeast-rice group had significantly higher IGF-l hormone than casein group in ovariectomized rats. The red-yeast-rice group had higher spine bone mineral content than those of control group within the OVX groups. This study was an important first step in establishing that the observed beneficial effects of red-yeast-rice on bone, and this study also established the need for a study on the long-term effect of this supplement in a human.

Influence of Processing Method on Ileal Digestibility of Nutrients from Soybeans in Growing and Finishing Pigs

  • Eight crossbred barrows (four growing and four finishing pigs with average initial BW of 40 and 82 kg, respectively) were fitted with T-cannulas at the distal ileum and used in a 36 d metabolism experiment ($4{\times}4$ Latin squares) to determine the effects of roasting and extruding full-fat soybeans on nutrient utilization. Treatments were: 1) soybean meal; 2) roasted soybeans; 3) extruded soybeans; and 4) soybeans extruded with an extrusion enhancer (sodium sulfite). The control diet was corn starch-based with 0.90% lysine, 0.65% Ca and 0.55% P for the growing pigs and 0.75% lysine, 0.55% Ca and 0.45% P for the finishing pigs. For the growing pigs, apparent total tract digestibilities of DM (p<0.04) and GE (p<0.008) were greater for soybean meal than full-fat soy products. However, ileal digestibilities of DM, GE, N and most amino acids were, in general, greatest for extruded soybeans and lowest for roasted soybeans, with soybean meal intermediate. For finishing pigs, trends in digestibilities of nutrients were very similar to those for the growing pigs. Total tract digestibilities of DM (p<0.03) and GE (p<0.001) for soybean meal were greater than for the full-fat soy products and ileal digestibilities of DM, GE, N and most amino acids were greater for the extruded soybeans than for the roasted soybeans. In conclusion, nutrient digestibilities and availabilities of indispensable amino acids tended to be greatest in extruded soybeans, intermediate in soybean meal and lowest in roasted soybeans for growing and finishing pigs.

Influence of Inclusion of Salicornia Biomass in Diets for Rams on Digestion and Mineral Balance

  • A metabolism trial was conducted with 28 Najdi rams allocated into seven dietary groups to evaluate the effect of dietary inclusion of Salicornia bigelovii Torr biomass on nutrient digestibility, rumina I fluid metabolites and nitrogen and mineral balances. Either the stems (ST) or spikes (SP) of this seawater-irrigated halophyte were incorporated into complete diets at rates of 0, 10, 20 and 30% levels, replacing equal amounts of rhodesgrass hay in a ground mixed control diet. Digestibility of DM, OM, EE, NFE and fecal and urinary nitrogen were not affected by increased level of ST in the diet. As level of ST increased from 0 to 20% in the diets, CP digestibility and nitrogen retention approached their maximum (p<0.01), whereas CF digestibility reached its minimum (p<0.01). On the other hand, except for EE, digestion of all nutrients and nitrogen retention were linearly depressed (p<0.01) as SP increased in the diets from 10 to 30% level. Concentration of ammonia-N, total VFA and pH values in the rumen fluid were lower (p<0.01) with the ST- or SP-fed diets than with the control diet. Increasing level of ST or SP in the diet was associated with an increase (p<0.01) in the proportion of acetate and a decline (p<0.01) in molar percentage of propionate in the ruminal fluid. Sodium absorption increased (p<0.01) with increased ST and SP in the diets up to the 10 and 20% level, respectively, followed by constant absorption values up to the 30% level. When the level of ST in the diet gradually increased to 30%, a concomitant increased (p<0:01) in Ca and P absorption were obvious; whereas, increased level of SP in the diets from 0 to 30% resulted in noticeable (p<0.01) depression in Ca and P apparent absorption.

NMR Signal Assignments of Human Adenylate Kinase 1 (hAK1) and its R138A Mutant (hAK1R138A)

  • Adenylate kinase (AK) enzyme which acts as the catalyst of reversible high energy phosphorylation reaction between ATP and AMP which associate with energetic metabolism and nucleic acid synthesis and signal transmission. This enzyme has three distinct domains: Core, AMP binding domain (AMPbd) and Lid domain (LID). The primary role of AMPbd and LID is associated with conformational changes due to flexibility of two domains. Three dimensional structure of human AK1 has not been confirmed and various mutation experiments have been done to determine the active sites. In this study, AK1R138A which is changed arginine[138] of LID domain with alanine[138] was made and conducted with NMR experiments, backbone dynamics analysis and mo-lecular docking dynamic simulation to find the cause of structural change and substrate binding site. Synthetic human muscle type adenylate kinase 1 (hAK1) and its mutant (AK1R138A) were re-combinded with E. coli and expressed in M9 cell. Expressed proteins were purified and finally gained at 0.520 mM hAK1 and 0.252 mM AK1R138A. Multinuclear multidimensional NMR experiments including HNCA, HN(CO)CA, were conducted for amino acid sequence analysis and signal assignments of $^1H-^{15}N$ HSQC spectrum. Our chemical shift perturbation data is shown LID domain residues and around alanine[138] and per-turbation value(0.22ppm) of valine[179] is consid-ered as inter-communication effect with LID domain and the structural change between hAK1 and AK1R138A.

The Effect of Woohwangcheongsim-won on Circulatory Disturbance in Diabetes (우황청심원이 당뇨병 Rat의 혈액순환장애에 미치는 영향)

  • Object: Death rate due to hypertension, atherosclerosis, ischemic heart disease and cerebral infarction induced by Westernized diet and increased average life span is on the rise. Decrease in blood circulation, activation of thrombus generation and intravascular lipid accumulation, cited as the principal causes of the above mentioned diseases in recent studies, result in circulatory disturbance and blood vessel obstruction leading to ischemic cell death of heart, brain and peripheral vessels. Method: We investigated the biochemical changes in microvascular permeability, aggregation of platelet and the intravascular lipid accumulation in induced-diabetic rat using Streptozotocin. We also studied the effects of Woohwangcheongsirn-won after oral administration on blood circulation, platelet function and lipid metabolism. The results are as follows: I. Woohwangcheongsim-won increased blood circulation in microvessels. 2. Woohwangcheongsim-won increased the reduced erythrocyte deformability in diabetes. 3. Woohwangcheongsim-won induced the reduction of contents of 2, 3-DPG, but failed to affect the reduced contents of ATP in erythrocyte in diabetes. 4. Woohwangcheongsim-won reduced the activity of Ca/sup 2+/-ATPase in the membrane of erythrocyte. 5. Woohwangcheongsim-won reduced the platelet aggregation evoked by platelet agglutinin factor. 6. Woohwangcheongsim-won reduced the production of platelet-derived granules. 7. Woohwangcheongsim-won reduced the production of metabolites of arachidonic acid in diabetes, and also reduced the production of increased thromboxane B2. 8. Woohwangcheongsim-won reduced the synthesis of oxidized LDL-cholesterol. In conclusion, Woohwangcheongsim-won enhanced blood circulation in microvesseles, erythrocyte deformability and inhibited the increased platelet aggregation and the synthesis of oxidized LDL-cholesterol in diabetes. Therefore Woohwangcheongsim-won is believed to positively affect blood circulation (J Korean Oriental Med 2002;23(2):164-179)

Effects of Sagungsan on the Hemostasis, Intracranial Pressure, Blood Pressure and Cardiovascular System in Expreimental Animals (사궁산(莎芎散)이 실험동물(實驗動物)의 지혈(止血), 뇌압(腦壓), 혈압(血壓) 및 심혈관계(心血管系)에 미치는 영향(影響))

  • According to the original documents, Sagungsan is considered as an effective drug for controlling the hypertensive epistaxis induced by tension of autonomic nerve and it's hyperfunction. The present experiment was designed to understand the effect of Sagungsan extract on the hemostatic action, intracranial pressure, blood pressure and cardiovascular system in experimental animals. And thus the bleeding time, prothrombin time, capillary dilation, blood pressure, Intracranial pressure, and enzymatic analysis of the ATPase activities were studied. The result obtained here were as followings: 1. Sagungsan water extract reduced the bleeding time in mouse, and prolonged the prothrombin time in rabbits. 2. The drug extract increased the tail volume by capillary dilation in rats. 3. The drug extract inhibited the increase of intracranial pressure and arterial blood pressure in rabbits. 4. At the early time, the increase of arterial blood pressure by the drug extract significantly inhibited by pretreated atropin and regitine in rabbits. 5. The drug extract relaxed the smooth muscle by stimulating the Mg2+-Ca2+-ATPase activities of gastric sarcoplasmic reticulum isolated from rabbit stomach. 6. The drug extract stimulated the heart contraction by inhibiting the $Mg^{2+}-Ca^{2+}-ATPase$ activities of cardiac sarcoplasmic reticulum isolated from rabbit heart. The inhibitory mechanism was reversible and noncompatitive. 7. The drug extract increased the hepatic blood volume by stimulating the hepatic total ATPase activities and hepatic metabolism. 8. The drug extract acted as a tranquilizer by inhibiting the neural Na+-K+-ATPase activity. According to the results, Sagungsan water extract dilated the capillaries, stimulated the heart beat, and thus increased the blood flow with decreasing the intracranial pressure and blood pressure. These effects stanches the epistaxis collectively.
