Morphological and Genetic Characteristics of Colletotrichum gloeosporioides Isolated from Newly Emerging Static-Symptom Anthracnose in Apple

  • Filamentous fungi of the genus Colletotrichum (teleomorph, Glomerella) are considered major plant pathogens worldwide. Cereals, legumes, vegetables, and fruit trees may be seriously affected by this pathogen (1). Colletotrichum species cause typical disease symptoms known as anthracnoses, characterized by sunken necrotic tissue, where orange conidial masses are produced. Anthracnose appears in both developing and mature plant tissues (2). We investigated disease occurrence in apple orchards from 2013 to 2014 in northern Gyeongbuk province, Korea. Typical anthracnose with advanced symptoms was observed in all apple orchards studied. Of late, static fruit spot symptoms are being observed in apple orchards. A small lesion, which does not expand further and remains static until the harvesting season, is observed at the beginning of fruit growth period. In our study, static symptoms, together with the typical symptoms, were observed on apples. The isolated fungus was tested for pathogenicity on cv. 'Fuji apple' (fully ripe fruits, unripe fruits, and cross-section of fruits) by inoculating the fruits with a conidial suspension ($10^5$ conidia/ml). In apple inoculated with typical anthracnose fungus, the anthracnose symptoms progressed, and dark lesions with salmon-colored masses of conidia were observed on fruit, which were also soft and sunken. However, in apple inoculated with fungi causing static symptoms, the size of the spots did not increase. Interestingly, the shape and size of the conidia and the shape of the appressoria of both types of fungi were found to be similar. The conidia of the two types of fungi were straight and cylindrical, with an obtuse apex. The culture and morphological characteristics of the conidia were similar to those of C. gloeosporioides (5). The conidia of C. gloeosporioides germinate and form appressoria in response to chemical signals such as host surface wax and the fruitripening hormone ethylene (3). In this study, the spores started to germinate 4 h after incubation with an ethephon suspension. Then, the germ tubes began to swell, and subsequently, differentiation into appressoria with dark thick walls was completed by 8 h. In advanced symptoms, fungal spores of virtually all the appressoria formed primary hyphae within 16 h. However, in the static-symptom fungus spores, no primary hyphae formed by 16 h. The two types of isolates exhibited different growth rates on medium containing apple pectin, Na polypectate, or glucose as the sole carbon. Static-symptom fungi had a >10% reduction in growth (apple pectin, 14.9%; Na polypectate, 27.7%; glucose, 10.4%). The fungal isolates were also genetically characterized by sequencing. ITS regions of rDNA, chitin synthase 1 (CHS1), actin (ACT), and ${\beta}$-tubulin (${\beta}t$) were amplified from isolates using primer pairs ITS 1 and ITS 4 (4), CHS-79F and CHS-354R, ACT-512F and ACT-783R, and T1 and ${\beta}t2$ (5), respectively. The resulting sequences showed 100% identity with sequences of C. gloeosporioides at KC493156, and the sequence of the ${\beta}$t gene showed 100% identity with C. gloeosporioides at JX009557.1. Therefore, sequence data from the four loci studied proves that the isolated pathogen is C. gloeosporioides. We also performed random amplified polymorphic DNA-PCR, which showed clearly differentiated subgroups of C. gloeosporioides genotypes. The clustering of these groups was highly related to the symptom types of the individual strains.

  • Objective : The purpose of the present study was to evaluate the effect of acupuncture therapy on the premenstrual syndrome. Methods : Volunteers of twenty subjects were employed using DRSP questionnaire. Subjects were divided into two groups including active-acupuncture treatment group, placebo acupuncture treatment group. In the control group, subjects were needled at $SI_5$, $ST_{40}$ points and in the acupuncture group, subjects were stimulated at two basic points, $SP_6$ and $CV_6$, and several points were inserted additionally depending the symptoms including $LR_2$, $LR_3$, $SP_10$, $LI_4$, and $ST_3$. A total of 13 acupuncture sessions were performed for each patient depending on the individual menstruation cycle over 8 weeks. Concentration of blood progesterone was examined and Digital Infrared Thermal Imaging (DITI) was taken before and after acupuncture treatment.

Antioxidant Activity from Different Root Parts of 6-year-old Panax ginseng C.A. Meyer (Yun-poong) (6년생 인삼(연풍)의 뿌리부위별 항산화 활성)

  • The objective of this study was to evaluate the contents of reducing sugar and total polyphenol and the antioxidant activity of freeze-dried ginseng (Panax ginseng C.A. meyer cv Yun-poong). Ginseng root consists of the main root (MR), lateral root (LR) and fine root (FR). These roots were sorted into 3~4 groups (MR 1~4 groups, LR 1~4 groups, FR 1~3 groups) depending on the diameter of center region. The reducing sugar content has no influence on the diameter, but the reducing sugar content was decreased by subdivision. Total polyphenol contents of ginseng were the highest in FR-3 among all others. The antioxidant activity was measured using DPPH radical scavenging activity and ABTS radical scavenging activity. The $IC_{50}$ (50% inhibitory concentration) value of the hydrogen donating activity was the highest in FR-3 as 7.03 mg/mL and the lowest in MR-2 as 37.02 mg/mL. Overall, hydrogen donating activity of FR was higher than the main root and lateral root significantly (p<0.05). The ABTS radical scavenging activity in FR-3 showed the highest radical scavenging activity as 25.49%. This study's findings suggest that the total polyphenol contents and antioxidant activity of ginseng were the highest in FR-3, and that polyphenol contents and antioxidant activity of ginseng were related to root area and diameter.

Trend of Acupuncture Treatment Study for Functional Dyspepsia (기능성 소화불량증의 침치료 연구 동향 : Pubmed를 중심으로)

  • Objectives: This study was conducted to help clinical studies for treating Functional Dyspepsia with acupuncture. We inspected tendency of studies of acupuncture treatment for functional dyspepsia and analyzed the assessing instrument for it by scrutinizing Pubmed recent publications. Materials and Methods: We inspected 8 theses and scrutinized their objectives, periods, participants, materials and methods, the instrument of assessment for the studies, and criteria and symptom score for questionaire, results and JADAD score. Results: 1. Most studies were published by oriental doctors except one which was published by an American doctor. More and more studies are being published in America and written in English recently. So it is considered that western doctors think that the acupuncture treatment is useful as unorthodox medicine for functional dyspepsia. 2. Among the 4 clinical studies, 2 studies were RCT and 1 study was not RCT and 1 study with vagueness. All 4 clinical studies were not double-blinded because they were about comparison between acupuncture treatment and medicine. 3. ST36, PC6, CV12, BL21 were chiefly used, and auricular point, ST40, SP6 were also used. Manual acupuncture were used in 2 studies, and low frequency pulse and electroacupuncture were used in other studies respectively. 4. 4 clinical studies used 0~3 points symptom score as the instrument of assessment. Additional assessments were on gastric emptying time, EGG and plasma level of motilin. All thesis were not verified of validity and reliability. 5. Among the 4 reviewed thesis, 2 theses were about acupuncture as one of the various treatment of functional dyspepsia, and the other 2 theses were about efficacy of acupuncture treatment. And the result generally showed effectivity of acupuncture treatment. Conclusion: Acupuncture treatment for functional dyspepsia is considered useful as in unorthodox medicine by western doctors, and it is necessary to provide objective instrument for assessing acupuncture treatment for functional dyspepsia.

Physiological Response and Cadmium Accumulation of MuS1 Transgenic Tobacco Exposed to High Concentration of Cd in Soil: Implication to Phytoremediation of Metal Contaminated Soil (토양 중 고농도 카드뮴에 노출된 MuS1 형질전환 담배 (Nicotiana tabacum cv. Xanthi)의 생리적 반응 및 카드뮴 축적: 식물학적 오염토양정화를 위한 형질전환 식물 탐색)

  • The objective of this study was to understand the physiological response and cadmium accumulation of MuS1 transgenic tobacco exposed to high concentration of Cd in soil. For this, a pot experiment was carried out in a greenhouse for a month, with two lines of MuS1 transgenic tobaccos (S4 and S6) and non-transgenic tobacco cultivated in the soils spiked at three different Cd concentrations (0, 60 and 180 mg $kg^{-1}$). Both transgenic and non-transgenic tobacco showed visible toxic symptoms such as chlorosis and leaf roll as treated concentration increased. The net photosynthetic rates of MuS1 plants (S4 and S6) exposed at 180 mg $kg^{-1}$ Cd were 6.3 and $7.7{\mu}mol\;m^{-2}s^{-1}$, being higher than those of the non-transgenic plant ($4.8{\mu}mol\;m^{-2}s^{-1}$). Values of stomatal conductance of MuS1 transgenic plants (0.05 and 0.008 mmol $H_2O\;m^{-2}s^{-1}$) were also higher than those of non-transgenic plant (0.03 mmol $H_2O\;m^{-2}s^{-1}$). In addition, fresh and dry weights of MuS1 transgenic plants were heavier than those of non-transgenic plant. Likewise, MuS1 transgenic plants appeared to be better physiological performance than non-transgenic tobacco when exposed at high concentration of Cd in soil. With regard to metal accumulation, MuS1 transgenic tobaccos accumulated more Cd in their roots than non-transgenic tobacco implying that MuS1 transgenic tobacco is suggested to be used for phytostabilization of heavy metals.

Changes in Onion (Allium cepa L.) Bulb Quality during Storage at Room Temperature and Greenhouse (비닐하우스를 이용한 양파의 상온 저장 중 품질 변화)

  • This study was conducted to investigate the possibility of changes in onion (cv. 'Turbo' and 'Changnyongdaego') bulb quality during storage at room and cold temperature. Increased weight loss was found in the onion bulb stored at both cold and room temperature conditions. 4% weight loss was found in bulbs stored at room temperature and 2% at cold storage as compared to fresh cultivated onion bulb; after three months. Initially, there was no significant difference in the firmness of bulbs between cultivars and storage conditions. Hunter a value of bulb stored at room temperature was higher than the value of bulb stored at cold temperature. 27% and 7.9% decay was found after three months in 'Changnyongdaego' and 'Turbo' cultivars stored under room temperature condition respectively. In contrast, only 2% decay was noticed in both cultivars stored at cold storage condition. Hunter L value of onion bulb gradually decreased in both room and cold temperature storage conditions. In conclusion, quality of onion bulb can be maintained until three months. For long term storage and quality maintenance, under room temperature conditions a resistance cultivar should be selected.

Composition of Buckwheat (Fagopyrum esculentum Moench) Cultivars from Korea (한국산 메밀의 성분)

  • To clarify the values and varieties of the buckwheats as a dietary source of nutritional and functional components, thirteen different samples of buckwheat were analyzed for this investigation. Six developed seeds were given by RDA, Korea or RDA branch of Kangwondo, and seven land race seeds were collected from a farmhouse. Amino acid analysis showed that glutamate, arginine and asparagine were major amino acids, whereas tryptophan, methionine and cysteine were minor ones of buckwheat. In addition, tryptophan content of buckwheat cultivars from Korea was 195 mg% on average. The content of rutin tended to be higher in developed cultivars than land races. On the other hand, the contents of phytic acid in buckwheats were in the range of 7.0 to 13.6 mg/g. In the tocopherol homologues of the buckwheats analyzed by HPLC, mean ${\gamma}-tocopherol$ contents were 6.16 mg/100 g with the actual range of $4.67{\sim}8.58\;mg/100g$, whereas ${\beta}-form$ was very low or zero. There were a big variations in the iron content of the buckwheats of the minerals. SDS-PAGE showed that total proteins from buckwheats exhibited a relatively similar electrophoretic patterns on the whole. The results show that CV Suwon 1 has good quality, judged from the distribution of the components of buckwheats analyzed.

Effects of the Limited Nutrient Supply at the Pollination Stage on the Growth and Nutrient Uptake of Muskmelon Grown in Rockwool (온실멜론의 암면재배에 있어서 수분기의 양분공급제한이 생육 및 양분흡수에 미치는 영향)

  • This experiment was carried out to investigate the effects of limited nutrient supply during 21 days before and after pollination stage on the growth, fruit quality and nutrient uptake of muskmelon in rockwool culture. Muskmelon, cv. Earl's Favorite seeds sowed on rockwool cube and transplanted on rockwool slab($90\times15\times7.5cm$) when 2 to 3 true leaf appeared on Sep. 6, 1991. Three kinds of nutrient composition recommended by Shizuoka university, combinated with the composition of Otsuka house A and composition Shizuoka III. One half of calcium nitrate(Ca(NO$_3$)$_2$.4$H_2O$) for limiting nitrogen supply during 21 days was treated and then fertigated the nutrient composition recommended by Shizuoka university up to harvest time. Trickling nozzles(Netafim Co. Israel) were used for fertigation of nutrient solution and noncirculating system was employed. Temperature was maintained $18^{\circ}C$ in night but 23 to $25^{\circ}C$ for 10 days after pollination for softening the fruit. The drainage ratio of nutrient solution was adjusted 20 to 30 percent. Fertigated and drained amount, and the pH and EC of nutrient solution were recorded. The concentrations of mineral elements including N, P, K, Ca, and Mg were analyzed and compared among treatments. In both autumn and winter cultivation, the limitation of nutrient supply by adjustment of nutrient composition(NO$_3$-N : 8meㆍ$\ell^{-1}$) caused the nutrient deficiency in muskmelon plant due to the limited nutrient supply. After pollination nutrient limitation by the lowering the nitrate retarded the over thickening of upper leaves of muskmelon but plant height and fresh weight of fruit were higher in the plot of nonlimited nutrient supply. The phenomena were attributed to the differences of the amount of nutrient uptake due to the limited time of nutrient solution, duration of nutrient supply and concentration of nutrient solution. These results suggested that increasing nutrient supply in the pollination stage was favorable for better appearance of fruit and improving fruit quality. Further trials would be required for the incre-ment of sugar degree of muskmelon grown in rockwool.

Rapid Nutrient Diagnosis of Cucumber by Test Strip and Chlorophyll Meter (Test Strip과 Chlorophyll Meter를 이용한 오이의 신속한 영양진단)

  • This study was performed to develop a more rapid and simple nutrient diagnosis method of plants than the conventional leaf analysis method. Cucumber (Cucumis sativus L. cv. jangil banbaek) was planted in the mixed media produced by mixing perlite and rock wool at 1:1 (v/v) ratio. The Yamazakki nutrient solution for cucumber was supplied to the media using micro-drip irrigation system. Experimental plots were consisted of no fertilization, deficient fertilization, adequate fertilization, and surplus fertilization for N, P and K. Specific color difference sensor value (SCDSV) measured by chlorophyll meter was closely related to total-N concentration in leaves measured by the conventional method. Nitrate, $PO_4$ and K concentration in petiole sap measured by test strips showed a significant relationship with total-N, P and K concentration in leaves. Linear regression equations between $NO_3$, $PO_4$ and K concentration in petiole sap and total-N, P and K concentration in the leaves were prepared. Optimum levels of $NO_{3}$, $PO_{4}$ and K in petiole sap were obtained by plugging the optimum concentrations of total-N, P and K in the leaves by other researchers into the equations. In conclusion, the SCDSV measured by chlorophyll meter and the concentration of $NO_3$, P and K in petiole sap measured by the test strips would be suitable for rapid estimation of plant nutrient status.

Growth of Creeping Bentgrass on Bottom Ash and Dredged up Sand with Four Organic Matter Amendment Rates Under Saline Irrigation Condition (염해 조건에서 유기물이 첨가된 준설모래와 석탄회 토양이 크리핑 벤트그래스의 생육에 미치는 영향)

  • This study was carried out to check the possibility of substituting bottom ash from the Seosan power plant for sand as growing media for creeping bentgrass (Agrostis stolonifera L.) under saline irrigation condition. Characteristics of growing media were evaluated by using column and leaching method. Creeping bentgrass cv. Pen-A1 was grown in pots with dredged up sand (DS) and bottom ash (BA) media those were amended using 1%, 2%, and 3 % OM rates in a green house. The plants were irrigated with 1.5 $dSm^{-1}$ saline water. Results showed that visual quality, plant height and shoot dry weight from DS treatment were higher than those of BA treatment. Even though BA contained more salts, repeated leaching could decrease ECe efficiently. In case of no OM amendment, the visual quality, plant height and shoot dry weight were similar between in BA and DS. Amendment of 2% OM increased the height of creeping bentgrass in DS, while decreased the plant growth in BA.
