생쥐 정소에서 C-terminal Src kinase (Csk) 발현과 Src kinase 활성의 조절

  • Protein tyrosine kinases는 표적단백질의 tyrosine 잔기를 인산화하는 효소로서 다양한 종류의 성장인자, peptide 호르몬, cytokine 수용체 하위의 세포 내 신호전달에 관여한다. Non-receptor tyrosine kinase의 일종인 c-Src는 세포막에서 발생한 ligand-receptor 상호작용 하위의 신호전달에서 중요한 역할을 하며 C-terminal Src kinase (Csk)는 Src kinase의 C-terminal tyrosine 잔기를 인산화시켜 Src kinase의 활성을 저해한다. 이러한 Src-Csk loop를 통한 세포 내 신호전달과정은 세포의 증식과 분화, 사멸 조절에 중요한 기능을 갖지만 정소의 발생과 분화 과정에서 Src-Csk loop의 발현 및 정자형성 과정에서의 기능은 밝혀지지 않았다. 본 연구에서는 생쥐 정소에서 출생 후 성적 성숙과정에서 Csk의 발현과 Src kinase 활성의 변동을 조사하였다. Csk mRNA 발현은 생 후 2주령 이하의 미성숙 정소에서 다량으로 발현되었고 사춘기 정소 이후에는 오히려 감소하였다. Csk 단백질의 발현 양상은 mRNA 발현양상과 일치하였다. c-Src kinase 활성은 생 후 2주에 급격히 증가하고 이 후 4주령에서 감소하다가 성체 (8주령)에서 다시 증가하여 가장 높았다. 성체 조직의 Csk 단백질 현존량이 미성숙 개체보다 적은 반면 Src kinase 활성은 가장 높아 Csk 발현의 감소는 Src kinase 활성을 증가하는 것으로 사료된다. 면역조직화학방법으로 정소 조직 내 Csk의 발현양상을 조사한 결과 Leydig cell, Sertoli cell, germ cell 등 도처에서 발현되었으며 Sertoli cell 에서의 발현은 세정관 상피의 구성에 따른 차이가 확인되었다. 성체의 세정관 내에서는 감수분열 이후의 정세포(spermatid)를 감싸고 있는 Sertoli cell의 강소측에서 강한 Csk 활성이 검출되어 생식세포의 분화과정 동안 세정관 상피의 조직재구성에 관여하는 것으로 사료된다. Leydig cell에서의 발현은 생후 1주령까지는 미미하였으나 이후 2주령 이후에는 다량으로 발현함이 확인되어 adult type Leydig cell에서 진행되는 steroidogenesis와의 관련성을 추측할 수 있다. 미성숙 정소로부터 분리한 Sertoli cell-enriched culture에 200 nM testosterone을 처리하였을 때 Csk mRNA의 발현의 증가를 확인할 수 있었으므로 androgen에 의한 Sertoli cell의 분화과정에 Csk가 관여하고 있음을 알 수 있다. 결론적으로 성적 성숙에 따른 생쥐 정소 내 Src-Csk loop의 발현과 Src kinase 활성의 변동은 정소 내 간충조직, 세정관 상피의 증식 및 기능적 분화 과정을 매개하는 생리적 활성분자 수용체 하위의 신호전달 과정에 Src-Csk loop에 의한 조절가능성을 확인할 수 있었다.

  • A great interest is emerging about green tea as a tool against human cancer proliferation or inflammation, as pointed out by recent reports describing the inhibitory action of epigallocatechin gallate (EGCG) on angiogenesis, urokinase, metalloproteinases, and induction of inducible nitric oxide synthase. We proposed that EGCG may regulate a multi target signaling having wider spectra of action than those actions of single enzymes. CSK (c-terminal Src kinase) protein is a non-receptor tyrosine kinase involved in the cross-talk and mediation of many signaling pathways that promote cell proliferation, adhesion, invasion, migration, and tumorigenesis. Based on the knowledge that CSK activation is important for cancer proliferation we hypothesized that CSK could be a target of EGCG. Here we showed that EGCG effectively suppressed the growth of CSK MEF cell when compare with CSK knockout MEF cell growth. These results indicate that EGCG could be used as a chemoprevention to modulate CSK signal pathway in inflammatory processes and tumor formation.

Differentially Expressed Genes by Inhibition of C-terminal Src Kinase by siRNA in Human Vascular Smooth Muscle Cells and Their Association with Blood Pressure

  • C-terminal SRC kinase (CSK) is a ubiquitously expressed, cytosolic enzyme that phosphorylates and inactivates several SRC family protein tyrosine kinases. Recent genomewide association studies have implicated CSK in the regulation of blood pressure. The current study aim is to determine the blood pressure association of the genes regulated by CSK down-regulation. The CSK mRNA expression was downregulated in vascular smooth muscle cells using small interfering RNA (siRNA). CSK mRNA levels fell by 90% in cells that were treated with CSK siRNA; the RNA from these cells was examined by microarray using the Illumina HumanRef-8 v3 platform, which comprises 24,526 reference mRNA probes. On treatment with CSK siRNA, 19 genes were downregulated by more than 2-fold and 13 genes were upregulated by more than 2-fold. Three (CANX, SLC30A7, and HMOX1) of them revealed more than 3 fold differential expression. Interestingly, the HMOX1 SNPs were associated with diastolic blood pressure in the 7551 Koreans using Korea Association REsource data, and the result was supported by the other reports that HMOX1 linked to blood vessel maintenance. Among the remaining 29 differentially expressed genes, seven (SSBP1, CDH2, YWHAE, ME2, PFTK1, G3BP2, and TUFT1) revealed association with both systolic and diastolic blood pressures. The CDH2 gene was linked to blood pressures. Conclusively, we identified 32 differentially expressed genes which were regulated by CSK reduction, and two (HOMX1 and CDH2) of them might influence the blood pressure regulation through CSK pathway.

  • For the purpose of LAN or data transmission line design with high speed and reliability using power lines, in this paper, we present CSK/SS-DS signal transmission characteristics simulations with changing channel characteristics due to impedance fluctuation. And we also simulated the signal transmissions with a strong impulsive burst noise and with a gaussian random noise. We introduce a simple channel equalization filtering method that solves the synchronization problem of CSK/SS-DS with fluctuating channel characteristics or low S/N ratio.

  • The power line modem proposed in this paper transmits the remote control signal using CSK(Code Shift Keying) and DS/SS method. The CSK technique provides the increased capacity of transmission and robustness towards noise. Besides, the DS/SS technique provides protection against narrow-band Gaussian interference and multi-path interference. The modem supports full-duplex communication using FDD(Frequency Division Duplex) and the modem structure for forward link is same with that for reverse link. To switch each sub-controlled unit smoothly, 4/$\pi$-DQPSK is adopted for noncoherent demodulation. The PN code for spreading spectrum seues to divide each group which consists of sub-controlled units and Walsh code is used for the M-ary CSK technique. Each block is designed and verified with TMS320C5402 DSP. We show the superiority of the proposed method by analyzing numerically the system performance for the factors of the DS/SS and CSK method ullder additive white Gaussian noise and PBI.

  • CSK(Code Shift Keying) 방식은 전송 용량을 높이고, 잡음에 더욱 견고히 할 수 있다[1,2]. 전통적으로 다중 경로 페이딩, 예측 불가능한 위상과 진폭의 왜곡, 협대역 간섭 신호 및 예측 불가능한 시변 재밍 신호에 의한 간섭 등을 특징으로 하는 채널상에서는 DS-BPSK 방식이 사용되었지만 본 논문에서는 코드열의 반복적 특성을 적용한 CSK/SS(Code Shift Keying/Spread Spectrum) 방식을 제안하고 성능을 분석한다.

Comparison of consumption behaviors and development needs for the home meal replacement among Chinese college students studying abroad in Korea, Chinese college students in China, and Korean college students in Korea

  • BACKGROUND/OBJECTIVES: The consumption of home meal replacement (HMR) is increasing among college students in Korea and China. In particular, Chinese college students studying abroad in Korea (CSK) show changes in their dietary behavior after migration, and HMR consumption for meal substitution is also increasing. This study was conducted to compare the HMR consumption behaviors and HMR development needs of CSK, Chinese college students in China (CSC), and Korean college students in Korea (KSK). SUBJECTS/METHODS: The subjects were 570 college students (180 CSK, 200 CSC, and 190 KSK) who had experience of HMR consumption. Data were collected by face-to-face survey in 2019 and analyzed using SPSS 25.0. RESULTS: The majority of the subjects purchased HMR to 'saving time' and 'preventing meal skipping'. Average purchase price per HMR was about 5,000 won for the CSK and KSK, and about 3,000 won for the CSC. The most important attributes when selecting HMR for the CSK and CSC were hygiene, freshness, and taste in that order, while for the KSK were taste, price, and hygiene. Rice was preferred by the KSK while grilled and fried dishes were preferred by the CSK and CSC. In terms of development needs, dessert and meat-based side dishes were highest in all three groups. The preferred food materials for more than 50% of the subjects of all groups were beef, chicken, pork, shrimp, and squid, and spinach and Chinese cabbage in the CSK, and onion in the KSK. CONCLUSIONS: This study shows more effort is needed to develop the healthy customized HMR for college students studying in Korea and China, and that focuses are placed by CSK on hygiene and freshness, by CSC on meat side dishes, hygiene, and price, and by KSK on snacks (as meal substitutes), taste, and price.

  • For the purpose of high speed and reliable data transmission through the frequency response variation channel by load fluctuation, we present CSK signal transmission characteristics by computer simulation. Also using the simple structure of Roll-off filter at the CSK receiving system, the exact synchronization tracking and data detection without error at the worst case of S/I=2 and S/N=2 were achieved.

  • Visible light communication is a converging communication by technologies that enable solutions using the LED Lighting. There is a need to research about it in order to improve the performance of the wireless communication by using a Color LED and improve the efficiency of data. In this paper, Assume the General indoor communication environment and LED communication systems based on CSK-OFDM applied to a MIMO communication system, We studied about transmitting method of more efficient communication. After performing the simulation based on IEEE 802.15.7 Standardization, as a result of the analysis of performance, it was confirmed the there is a significant impact on the performance of SNR and BER performance by the transmitting method based on MIMO and external noise using the LED communication system based on CSK-OFDM.

Role of Extracellular Signal-Regulated Kinase 1/2 and Reactive Oxygen Species in Toll-Like Receptor 2-Mediated Dual-Specificity Phosphatase 4 Expression (Toll-Like Receptor 2 매개 Dual-Specificity Phosphatase 4 발현에서 Extracellular Signal-Regulated Kinase 1/2와 활성산소의 역할)

  • Background: Toll-like receptors (TLRs) are well-known pattern recognition receptors. Among the 13 TLRs, TLR2 is the most known receptor for immune response. It activates mitogen-activated protein kinases (MAPKs), which are counterbalanced by MAPK phosphatases [MKPs or dual-specificity phosphatases (DUSPs)]. However, the regulatory mechanism of DUSPs is still unclear. In this study, the effect of a TLR2 ligand (TLR2L, Pam3CSK4) on DUSP4 expression in Raw264.7 cells was demonstrated. Methods: A Raw264.7 mouse macrophage cell line was cultured in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum and 1% antibiotics (100 U/mL penicillin and 100 g/mL streptomycin) at $37^{\circ}C$ in 5% $CO_2$. TLR2L (Pam3CSK4)-mediated DUSP4 expressions were confirmed with RT-PCR and western blot analysis. In addition, the detection of reactive oxygen species (ROS) was measured with lucigenin assay. Results: Pam3CSK4 induced the expression of DUSP1, 2, 4, 5 and 16. The DUSP4 expression was also increased by TLR4 and 9 agonists (lipopolysaccharide and CpG ODN, respectively). Pam3CSK4 also induced ERK1/2 phosphorylation and ROS production, and the Pam3CSK4-induced DUSP4 expression was decreased by ERK1/2 (U0126) and ROS (DPI) inhibitors. U0126 suppressed the ROS production by Pam3CSK4. Conclusion: Pam3CSK4-mediated DUSP4 expression is regulated by ERK1/2 and ROS. This finding suggests the physiological importance of DUSP4 in TLR2-mediated immune response.
