Expression of Corazonin Gene by Developmental Stage of Scuttle Fly

  • The corazonin (Crz) gene showed two subtypes of different length at laval and pupal stage. The long subtype fade out in adult central nerve system (CNS) but the short one survive through all the life cycle from larva to adult. The short subtype has the same base sequences with mature Crz mRNA and detected in both brain and ventral nerve cord (VNC). The long one, on the contrary, was detected only in the brain tissue. As observed in above results, Crz neurons develop in different pattern in the CNS of scuttle fly and the Crz gene expresses two different subtypes. These results suggest that this neurotransmitter may perform differential neurophysiological functions in the scuttle fly. Variation in the amino acid composition of the final active undecapeptide supports in strong those possibilities. We expect further studies on the relationship between neurophysiological functions of Crz and behavioral characteristics of the scuttle fly.

Identification and Characterization of Calcineurin Targets in Cryptococcus neoformans

  • Calcineurin governs stress survival, sexual differentiation, and virulence of the human fungal pathogen Cryptococcus neoformans. Herein, we identified and characterized calcineurin substrates in C. neoformans by employing phosphoproteomic $TiO_2$ enrichment and quantitative mass spectrometry. The identified targets include the zinc finger transcription factor Crz1 and proteins whose functions are linked to P-bodies/stress granules (PBs/SGs) and mRNA translation and decay, such as Pbp1 and Puf4. We show that Crz1 is a bona fide calcineurin substrate, and localization and transcriptional activity of Crz1 are controlled by calcineurin. Several of the calcineurin targets localized to PBs/SGs, including Puf4 and Pbp1, and are required for survival at high temperature and for virulence. Genetic epistasis analysis revealed that Crz1 and the novel targets Lhp1, Puf4, and Pbp1 function in a branched calcineurin pathway that orchestrates stress survival and virulence. These findings propose that calcineurin controls thermal stress and virulence at the transcriptional level via Crz1 and post-transcriptionally by regulating target factors involved in mRNA metabolism.

Calcineurin-Responsive Transcription Factor CgCrzA Is Required for Cell Wall Integrity and Infection-Related Morphogenesis in Colletotrichum gloeosporioides

  • The ascomycete fungus Colletotrichum gloeosporioides infects a wide range of plant hosts and causes enormous economic losses in the world. The transcription factors (TFs) play an important role in development and pathogenicity of many organisms. In this study, we found that the C2H2 TF CgCrzA is localized in both cytoplasm and nucleus under standard condition, and it translocated from cytoplasm to nucleus in a calcineurin-dependent manner. Moreover, the ΔCgCrzA was hypersensitive to cell wall perturbing agents and showed severe cell wall integrity defects. Deletion of the CgCRZA inhibited the development of invasive structures and lost pathogenicity to plant hosts. Our results suggested that calcineurin-responsive TF CgCrzA was not only involved in regulating cell wall integrity, but also in morphogenesis and virulence in C. gloeosporioides.

The Expression of Corazonin Neurons in Pupa and Adult Stage of Scuttle Fly

  • The scuttle fly is a fly species in the Phoridae family. Scuttle fly which moves abruptly after standing for a while and stop suddenly to rush off again. These characteristic behaviors of the scuttle fly seem to be related to muscular and nervous system or neurotransmitters. Thus, we focused at the neurotransmitter, corazonin (Crz) that is known to be related to resistance to stress and investigated the developmental process of the neurons in the scuttle fly. In a previous studies, we found that there are three groups of corazoninergic neurons in the larval CNS of the scuttle. Larva has 3 pairs of Crz neurons at the dorsolateral area of the brain, 1 pair at the dorsomedial brain and 8 pairs at the ventral nerve cord. In this studies, among these neurons, 1 pair of dorsomedial brain and 8 pairs of ventral nerve cord disappear in early pupal stage after metamorphosis. Only the 3 pairs of dorsolateral brain persist expression of Crz gene through all the period of pupa stage. This group of neurons converge gradually to frontal center of the brain and situated at the medial region. These pairs of corazoninergic neurons keep their number and location in adult stage. In the future, we expect further studies on the histological characteristics of corazonin-expressing cells and the expression of corazonin gene.

This species like rotten organic materials and it is known to proliferate even in the industrial materials including organic solvents. These characteristic behaviors of the scuttle fly seem to be related to muscular and nervous system or neurotransmitters. Thus, we focused at the neurotransmitter, corazonin (Crz) that is known to be related to resistance to stress and investigated the developmental process of the neurons in the scuttle fly. Corazonin is a neuropeptide being expressed in the central nervous system (CNS) and is known to control mainly physiological functions and behaviors. Its many functions that have been proposed are still in controversy. In this studies, we found that there are three groups of corazoninergic neurons in the larval CNS of the scuttle fly and these neurons undergo distinguishable changes through metamorphic process compared to different fly species. Larva has 3 pairs of Crz neurons at the dorsolateral area of the brain, 1 pair at the dorsomedial brain and 8 pairs at the ventral nerve cord.

LES Studies on the Characteristics of Turbulent Premixed Flame with the Configurations of Burner Exit (버너 출구의 형상변화에 따른 난류 예혼합 화염의 특성에 관한 LES 연구)

  • In the present paper, the effects of combustion instability on flow structure and flame dynamic with the configurations of burner exit in a model gas turbine combustor are investigated using large eddy simulation(LES). A G-equation flamelet model is employed to simulate the unsteady flame behavior. As a result of mean flow field, the change of divergent half angle(${\alpha}$) at burner exit results in variations in the size and shape of the central toroidal recirculation(CTRZ) as well as flame length by changing corner recirculation zone(CRZ). The case of ${\alpha}=45^{\circ}$ show smaller size and upstream location of CTRZ than that of $90^{\circ}$ and $30^{\circ}$ by the development of higher swirl velocity. The flame length in the case of ${\alpha}=45^{\circ}$ is the most shortest, while that in the case of ${\alpha}=30^{\circ}$ is the longest by the decrease of effective reactive area with the absence of CRZ. Through the analysis of pressure fluctuation, it is identified that the case of ${\alpha}=45^{\circ}$ shows the most largest damping effect of pressure oscillation in all configurations and brings in the noise reduction of 2.97dB, comparing with that of ${\alpha}=30^{\circ}$ having the largest pressure oscillation. These reasons are discussed in detail through the analysis of unsteady phenomena about recirculation zone and flame surface. Finally the effects of flame-acoustic interaction are evaluated using local Rayleigh parameter.

In Vitro Antifungal Activity and Mode of Action of 2',4'-Dihydroxychalcone against Aspergillus fumigatus

  • 2',4'-Dihydroxychalcone (2',4'-DHC) was identified from a heat shock protein 90 (Hsp90)-targeting library as a compound with Hsp90 inhibitory and antifungal effects. In the presence of 2',4'-DHC ($8{\mu}g/mL$), radial growth of Aspergillus fumigatus was inhibited 20% compared to the control, and green pigmentation was completely blocked. The expression of the conidiation-associated genes abaA, brlA, and wetA was significantly decreased (approximately 3- to 5-fold) by treatment with 2',4'-DHC. The expression of calcineurin signaling components, cnaA and crzA, was also significantly reduced. The inhibitory effects of 2',4'-DHC on metabolic activity and mycelial growth were significantly enhanced by combination treatment with itraconazole and caspofungin. Docking studies indicated that 2',4'-DHC bind to the ATPase domain of Hsp90. These results suggest that 2',4'-DHC act as an Hsp90-calcinurin pathway inhibitor.

Effect of Flare Angle in Counter-Rotating Swirler on Swirling Flow (동축 반전 스월러의 플레어 각도변화가 스월러 유동에 미치는 영향 연구)

  • Swirler generates the overall swirling flow in the combustion chamber and this swirling flow governs the flame stability and enhances fuel atomization. This paper deals with the flare angle effects on flow streamlines, recirculation zone, Central Toroidal Recirculation Zone(CTRZ) and Corner Recirculation Zone(CRZ) in the model combustion chamber using counter-rotating swirler. 2D PIV system was employed to obtain the velocity components and test condition was obtained using Reynolds Analogy equivalent to air test. We observed transitional flow patterns of flare angle increased. The obtained results show that the flare angle controls the behavior of Recirculation zone, Central Toroidal Recirculation Zone and Corner Recirculation Zone.

Morphological characteristics of Neural Tissue and Corazonin Neurons of Central Nervous System in Larval Stage of Scuttle Fly

  • Through previous studies, the central nervous system (CNS) was collected by dividing the scuttle fly into larval, pupa, and adult stages by developmental stage, and the morphological characteristics were observed. In situ hybridization (ISH) using the collected central nervous system, it was possible to confirm the location and extent of expression of the neurotransmitter corazonin (Crz) at each stage of development. In this study, paraffin specimens were prepared using central nervous system tissues of 3rd instar larval stage of scuttle fly, which had completed in situ hybridization, and general histochemical staining (hematoxylin-eosin, H-E) and special histochemical staining (luxol fast blue-cresyl violet) was performed to observe the histological and cytological morphology characteristics of corazonin neurons. As a result, a variety of nerve cell body existed between many myelin sheath. The corazonin neurons compose cortex of central nervous system with other neurons congregating in this tissue and show larger cell body relatively in neurohistochemical analysis.
