Isolation and Purification of Berberine in Cortex Phellodendri by Centrifugal Partition Chromatography (Centrifugal Partition Chromatography에 의한 황백으로부터 Berberine의 분리 및 정제)

  • Cortex Phellodendri (CP) is derived from the dried bark of Phellodendron amurense. It has been widely used as a drug in traditional Korea medicine for treating diarrhea, jaundice, swelling pains in the knees and feet, urinary tract infections, and infections of the body surface. Many analytical methods have been used to study oriental herbal medicines, such as thin-layer chromatography, column liquid chromatography, and high performance liquid chromatography (HPLC). In this study, preparative centrifugal partition chromatography (CPC) was successfully carried out in order to separate pure compounds from a CP methanol extract. The optimum two-phase CPC solvent system was composed of n-butanol: acetic acid: water (4:1:5 v/v/v). The flow rate of the mobile phase was 3 mL/min in ascending mode with rotation at 1,000 rpm. The CPC-separated fraction and purification procedures were carried out by preparatory HPLC. The $^1H$ NMR spectrum revealed that the resonances at ${\delta}$ 4.10 and 4.20 ppm corresponded to three protons ($-OCH_3$), whereas those at ${\delta}$ 6.10 ppm corresponded to two protons ($-OCH_2O-$). Further, two aromatic protons (H-11 and H-12) conveys a doublet-doublet pattern. The H-11 doublet and H-12 doublet appear at ${\delta}$ 7.98 and 8.11, respectively. The $^{13}C$ NMR. spectrum showed a tetrasubstituted with a methylenedioxy group at C2 and C3, and two methoxy groups at C9 and C10. The chemical structure of the berberine was identified by $^1H$, $^{13}C$-nuclear magnetic resonance and electrospray ionization-mass spectroscopy spectral data analysis.

Interaction between odontoblast and bio-calcium phosphate cement reinforced with chitosan (치아모세포와 키토산으로 강화된 생체 적합 칼슘인산시멘트와의 상호작용)

  • Purpose: Calcium phosphate cement (CPC) is one of many useful materials for restoring tooth defects, periodontium and maxillofacial area. Chitosan is a biodegradable material that has been shown to promote the growth and differentiation of osteoblasts in culture. This study examined the interaction between odontoblasts and bio-calcium phosphate cement reinforced with chitosan. Materials and Methods: $5{\times}10^3$ odontoblastic cells were seeded into each well. Various concentrations of bio-calcium phosphate cement reinforced with chitosan (10, 20, 50, 100, 200, 500 ${\mu}g$/ml, 1, 2, 4 mg/ml) were diluted and added to the wells. The well was incubated for 24 h, 48 h and 72 h. After incubation, the number of cells was assessed to determine the cell viability. A cytokinesis-block micronucleus assay and chromosomal aberration test were carried out to estimate the extent of chromosomal abnormalities. Microscopic photographs and RT-PCR were performed to examine the adhesion potential of bio-calcium phosphate cement reinforced with chitosan. Results: Bio-CPC-reinforced chitosan did not show significant cytotoxicity. The number of damaged chromosomes in the cells treated with Bio-CPC-reinforced chitosan was similar to that in the control cells. There was no significant increase in the number of chromosomal aberrations in the Bio-CPC reinforced chitosan exposed cells. Microscopic photographs and RT-PCR confirmed the adhesive potential of bio-CPC reinforced chitosan to odontoblasts. Conclusion: Bio-CPC-reinforced chitosan did not affect the odontoblastic cell viability, and had no significant cytotoxic effect. Bio-CPC-reinforced chitosan showed adhesive potential to odontoblasts. These results are expected form the basis of future studies on the effectiveness of dental restorative materials in Bio-CPC reinforced with chitosan.

Effect of Trisodium Phoshate and Cetylpyridinium chloride on E. coli O157:H7 and Listerial monocytogenes (Trisodium Phoshate와 Cetylpyridinium chloride의 E. coli O157:H7 및 Listerial monocytogenes 에 대한 살균 작용)

  • The in vitro effects of trisodium phosphate and cetylpyridinium chloride on E. coli 0157:H7 and L. monocytogenes were investigated. The trisodium phosphate and cetylpyridinium chloride was bactericidal toward E. coli 0157:H7 and L. monocytogenes. The killing effects of the $1{\times}10^{-2}\;M$ trisodium phosphate on E. coli 0157:H7 and L. monocytogenes were 30~40%, 40~50%, respectively. The killing effects of the $5{\times}10^{-7}\;M$ cetylpyridinium chloride on E. coli 0157:H7 and L. monocytogenes were 90~95%, 95~99%, respectively. The killing effects of the trisodium phosphate was $10^5$ times that of the cetylpyridinium chloride. Factors effecting the bactericidal action of trisodium phosphate and cetylpyridinium chloride were investigated and the action depended on temperature and pH.

A Study on Cold Damage(傷寒) in the Compendium of Prescription from the Countryside(鄕藥集成方) - Focusing on citation, medical theory, prescription, medicinal herbs - (조선 의서 『향약집성방』 중에 실린 상한(傷寒) 논의 연구 - 인용 문헌, 의론(醫論), 처방, 본초 등을 중심으로 -)

  • The purpose of this paper is to derive the features of cold damage clinical medicine during the early days of the Chosun(朝鮮) period by analyzing discussions on cold damage published in the official medical book of the Chosun period, Compendium of Prescription from the Countryside(鄕藥集成方, CPC). Cold damage was one of the typical diseases in East Asia where there was constant seeking of the utilization of prescriptions, ways of preparations, and awareness regarding cold damage as shown in Zhang, Zhongjing(張仲景)'s Treatise on Cold Damage Disease(傷寒論, TCDD) below. Traditional Korean medicine which possessed the medical universality of East Asia also was no exception and through an analysis of the part on cold damage in CPC, it is expected that medical features of cold damage in Korea passed down from the Koryo(高麗) Dynasty to the early Chosun period will be revealed. For this, first there needs to be an organization of past discussions on cold damage surrounding the existence of infection and after checking the issues, exploring which of the writings related to TCDD and editions are being utilized through an analysis on citing literature of Cold Damage Disease Literature(傷寒門) and Heat Pathogen Disease Literature(熱病門) which have developed discussions on cold damage in CPC. In addition, by comparing Peaceful Holy Benevolent Prescription(太平聖惠方, PHBP) and Complete Record of Sacred Benevolence(聖濟總錄, CRSB), known to have greatly influenced CPC and Cold Damage Literature and Heat Pathogen Disease Literature, features of form and content used by CPC were analyzed. Features of form were examined through pattern of organization and number of citing literature were examined and for features of content, cold damage infection, classification, syndrome differentiation method, and utilization of materia medica among prescriptions were examined. Discussions on cold damage as being uninfectious as stated in Treatise on the Pathogenesis and Manifestations of All Diseases(諸病源候論) unlike pestilence, epidemic pathogen(時氣), warm pathogen disease(溫病), and heat pathogen disease were excluded in PHBP. PHBP opened the possibility of cold damage infection and later writings, CRSB and CPC also follow this. As a result of analyzing citing literature of the part on cold damage in CPC, it is uncertain which edition of TCDD is being utilized; however, the most distinctive feature was that Classified Emergency Materia Medica(證類本草) and not writings specializing in cold damage are in use. In general, although CPC in terms of form is similar to CRSB, content creation predominantly depended on PHBP. More specifically; first, in terms of the existence of cold damage infection, arguments of PHBP and CRSB are maintained. Second, in terms of cold damage classification, although CRSB is followed, heat pathogen disease is classified separately developing PHBP as is. Third, in terms of method, as Book of Keep Healthy(南陽活人書) and CRSB compiled in later times are cited, it is deemed that arguments were raised to a certain extent regarding six-meridian syndrome differentiation(六經辨證). Fourth, although the majority of utilized materia medica among cold damage prescriptions utilize Materia Medica from the Countryside(鄕藥本草) in CPC and materia medica from Korean Peninsula, this is due to the desire for the compilation performance of CPC to be propagated to ordinary citizens and not the ruling class. CPC as the official medical book compiled in the early days of the Chosun period was greatly influenced by the Song(宋) Dynasty's medical books, PHBP and CRSB shows that cold damage medicine in the early Chosun Period indeed possesses the medical universality of East Asia. Furthermore, the features of published medical theory and prescriptions reveal the existence of the cold damage medical tradition of the Chosun period serving as clues for cold damage research tradition among Korea's medical history.

Effects of the C31G, Listerine and CPC as a adjunctives to the mechanical plaque control on the early periodontitis (기계적 치태조절 보조제로서 C31G, Listerine, CPC의 초기 치주염에 대한 임상적 효과)

  • The purpose of this study was to access the clinical effects of C31G, Listerine and CPC on the earlyx periodontitis when they were used as a adjunctives to the mechanical plaque control. Each groups were composed of 12 patients and in three test groups, C31G(Exp 1 group), Listerine(Exp 2 group) and CPC mouthrinse(Exp 3 group) were used three times a day. and as a control, placebo solution was used. Plaque index, gingival index, bleeding index, pocket depth and loss of attachment were measured as clinical parameters. After scaling and oral hygiene instruction, root planing is done two weeks later. During the eight weeks of experimental period, mouth gargling is done by all groups. The changes in the clinical parameters of the all sites were monitored every two weeks. The results were as follows : 1. The plaque index showed a siginificant difference in Exp 1 group compared with test 2 group after 2 weeks use of mouth rinse(P<0.05). 2. The gingival index showed a significant difference in Exp 1 group compared with baseline value(P<0.05) but there was no significant difference between the groups after 2 weeks use of mouthrinse (P<0.05). 3. The plaque index, gingival index and bleeding index showed a significant difference in all groups compared with baseline value after 4 weeks of root planing and mouthrinse use(P<0.05). but there was no significant difference between the groups(P<0.05). 4. Periodontal pocket depth showed a significant difference in all groups compared with baseline value after 4 weeks of root planing and mouthrinse use(P<0.05). and there was significant difference between the Exp 1 group and control group(P<0.05). 5. Loss of attachment showed a significant difference in all Exp groups compared with baseline value after 4 weeks of root planing and mouthrinse use(P<0.05). and there was significant difference between the Exp 1 group and control group(P<0.05). The results suggest that C31G, Listerine and CPC were effective for early periodontitis as a adjunctive to mechanical plaqe control.

Characteristics of Nitrate Removal Using Micellar-enhanced Ultrafiltration (MEUF에 의한 질산성 질소 제거에 관한 연구)

  • Feasibility of micellar-enhanced ultrafiltration far the removal of nitrate was investigated using cationic surfactants, cetylpyridinium chloride and octadecylamine acetate. The removal of nitrate increased as the molar ratio of surfactant increased. With the molar ratio of 3, at least 80% of nitrate was removed, while > 98% of nitrate was removed at the surfactant molar ratio of 10. Octadecylamine acetate showed higher removal efficiency of nitrate and higher rejection of surfactant than cetylpyridinium chloride because of the accessibility of nitrate to surfactant micelles due to head group of surfactant. Octadecylamine acetate turned out to be a better surfactant than cetylpyridinium chloride for micellar-enhanced ultrafiltration to remove nitrate from groundwater.

  • The purpose of this paper is to give information on e-CPC implementation in the manufacturing industry by studying and analyzing a small and medium sized manufacturing company. After analyzing Various factors on J's e-CPC implementation such as environmental factors, process management factor, and result factors, success factors on J's e-CPC implementation are analyzed. The results of this paper can be summarized as follows. First, the most leading role on J's ERP implementation is the will and support of CEO and team leaders. They led the implementation to be stability successfully by settling the different opinions among teams, showing their ability on process decision and communication with consultants. Second, J tried to prompt system stability by mapping out and conducting the long term change management strategy. Third, J organized ERP project team to implement ERP quickly, and it turned out to be successful Finally, J tried to get outside information from consultants and collaborative companies that have experience in implementing ERP. Especially J made full use of overseas consultants. In respect that this paper giver lots of information on e-CPC implementation in manufacturing industry by a case study of a small and medium sized manufacturing company which has not been carried out so far, it would be useful enough.

  • The Rkp1/CPC2, a receptor for activated protein kinase C of Schizosaccharomyces pombe, contains seven WD motifs found in the G-protein $\beta$-subunit. A 110-kDa protein was identified to interact with Rkp1/CPC2 by immunoprecipitation and following in vitro binding assay. To examine its kinase activity and binding ability to Rkp1, the $pck1^+$, a PKC homolog of S. pombe, was cloned. Pckl phosphorylated myelin basic protein (MBP) and histone Hl in a phospholipid-dependent and $Ca^{2+}$-independent manner. It was demonstrated that the N-terminal region of Pck1 was responsible for the binding to Rkp1 , thus suggesting that Rkp1 interacted with Pckl to regulate Pckl-mediated signaling in S. pombe.

Study of Degradation of Bisphenol A with $TiO_2$ Powder in CPC System (CPC (Compound Parabolic Collector) 내 이산화티탄을 이용한 비스페놀 A (Bisphenol A)의 분해에 관한 연구)

  • In this study, photocatalytic degradation and mineralization of bisphenol A (BPA), which has been listed as one of endocrine disruptors, were carried out in the CPC system using $Tio_2$ slurry and UVA irradiation. The degradation efficiency has been investigated under the controlled parameters including initial concentration (5, 10, 20 mg/L), dosage of $Tio_2$ (0.1, 0.5, 1.0 g/L), UVA power (0, 80, 120 W) and temperature (0, 20, 30). At 10mg/L of initial concentration, BPA was degraded above 80% after 10min, BPA were degraded 97% and 49% at 20 mg/L and 30 mg/L, respectively. At $Tio_2$ dosage was 0.1 and 0.5 g/L, the degradations of BPA showed similar trend and were about 70% after 1 hr, and the degradation of BPA was above 80% after 30 min at 1 g/L of $Tio_2$ dosage. The increase of degradation seem to be due to the increase in the total surface area, namely number of active sites, available for the photocatalytic reaction as the dosage of photocatalyst increased. When the UVA power was 120 W, BPA was degraded rapidly above 60% after 10min of reaction time. To investigate the effect of temperature, carried out experiment controlled temperature, there were no significant differences depending on the temperature. After 1hr, the degradation of BPA were 46%, 67%, and 69% at 10, 20 and $30^{\circ}C$.

Isolation of Eckol from Ecklonia cava via Centrifugal Partition Chromatography (CPC) and Characterization of it's Anti-inflammatory Activity (고속원심분배 크로마토그래피를 이용한 감태(Ecklonia cava)로부터 Eckol의 분리 및 항염증 활성)

  • Phlorotannins and marine algal polyphenols, including dieckol, 6,6-bieckol, phloroglucinol, phlorofucofuroeckol-A, and eckol, were isolated from brown seaweeds. These compounds have beneficial bioactivities, and Ecklonia cava has become widely used for the extraction and isolation of phlorotannins. Eckol, in particular, has been to shown to have antioxidant, anti-inflammatory, anticoagulatory, and photoprotective properties. However, due to its low abundance in weaweed, the isolation and purification of eckol are difficult. Its limited availability renders the isolation and purification of eckol labor-intensive processes. Centrifugal partition chromatography (CPC) is an efficient technique for the isolation and purification of eckol. In this study, eckol was isolated from the ethyl acetate fraction of the 70% ethanol extract of E. cava using CPC with a two-phase solvent system of a n-hexane:EtOAc:methanol:water (2:8:3:7, v/v) solution. The purity and anti-inflammatory activity of the isolated eckol were verified by high-performance liquid chromatography and by assaying lipopolysaccharide-induced inflammatory responses in an immortalized murine BV2 microglial cell line, respectively. In conclusion, CPC is a useful technique for simple and efficient isolation of eckol from E. cava.
