The Effect of Temperature and Concentration of Setting Solution on the Rheological Properties of Injectable Calcium Phosphate (경화액의 농도와 온도가 인산칼슘시멘트의 유변학적 성질에 미치는 영향에 관한 연구)

  • Injectable calcium phosphate cement (CPC) has been used as bone substitute successfully due to good biocompatibility and osteoconductivity. One of the important mechanical characteristics of CPC is flowablility, which can be evaluated by measuring rheological parameters. However, there have been few studies that measured rheological properties of CPC. The purpose of this study was to evaluate the effects of temperature and concentrations of 2 kinds of setting solutions, hydroxyprophyl methylcellulose (HPMC) and polyacrylic acid (PAA), on rheological properties of CPC. The CPC used was dicalcium phosphate dihydrate (DCPD). Rheological properties of CPC paste were measured using rheometer. The effect of concentrations of each solution (2% and 1% HPMC and 35% and 17.5% PAA) was evaluated. The effect of temperature ($25^{\circ}C$ and $37^{\circ}C$) on the rheological properties of CPC was also investigated. The statistical analysis was carried out with Mann-whitney test with Bonferronis collection. CPC with both setting solutions showed shear thinning behavior. Higher concentrations of setting solution (2% HPMC and 35% PAA) produced significantly higher viscosity than lower concentrations of setting solution (1% HPMC and 17.5% PAA). CPC with HPMC showed significantly higher viscosity at $37^{\circ}C$ that at $25^{\circ}C$. CPC with PAA showed lower viscosity at $37^{\circ}C$ than at $25^{\circ}C$, although the difference was not statistically significant. The results showed that CPC with HPMC or PAA solutions are pseudoplastic and the concentrations of setting solutions and temperature may have an effect on the rheological properties of CPC paste. These results showed that the flowability of injectable CPC could be improved by use of increasing frequency of oscillation. In clinical practice, the use of ultrasonic vibration would be helpful in application of injectable CPC. CPC with HPMC could be more easily applicated at $25^{\circ}C$ than $37^{\circ}C$. The use of lower concentrations of HPMC and PAA solution would be beneficial in terms of flowability.

  • Immobilization isotherms for methyl tort-buty1 ether (MTBE) in sodium dodecy1 sulfate(SDS) and cetylpyridinium chloride (CPC) were investigated for application to micellar enhanced remediation. Headspace solid-phase microextraction was used to analyze immobilization isotherms. Maximum partitioning coefficients of MTBE were 48 L/mol and 9 L/mol for SDS and CPC, respectively, The values decreased gradually as the MTBE mole fraction in the micelles increased.

Inhibitory Effects of Yongbu-tang on Osteoclast Differentiation and Bone Resorption (용부탕의 파골세포 분화 억제와 골 흡수 억제효과)

  • Objectives : This study was performed to evaluate the effects of water extract of Cervi Parvum Cornu(CPC), Aconiti Lateralis Radix Preparata(ALR), and Yongbu-tang(YBT) on suppression of the receptor activator of nuclear factor kappa-B ligand(RANKL)-induced osteoclast differentiation and bone resorption. Methods : The effects of CPC, ALR, YBT extracts on osteoclast differentiation were determined by culture of bone marrow macrophage(BMM). The mRNA expression levels of the nuclear factor of activated T-cells cytoplasmic 1(NFATc1), c-Fos and tartrate-resistant acid phosphatase(TRAP) in BMMs were analyzed by reverse transcriptase polymerase chain reaction(RT-PCR). Similarly, the protein expression levels of NFATc1, c-Fos, mitogen-activated protein kinase(MAPK)s and ${\beta}$-actin in cell lysates were measured by western blotting. In addition, effects of CPC, ALR and YBT extracts were determined by means of Lipopolysaccharide(LPS)-induced bone-loss with mice. Results : CPC, ALR and YBT extracts showed remarkable inhibition on RANKL-induced osteoclast differentiation without cytotoxicity. CPC and ALR extracts significantly reduced the protein expression level of NFATc1. YBT extract significantly reduced the mRNA expression levels of c-Fos, NFATc1 and the protein expression levels of c-Fos, NFATc1, AKT, p38, c-Jun N-terminal kinase(JNK). Further, YBT extract suppressed degradation of$ I-{\kappa}B$. And ALR extract significantly restored the bone erosion by LPS treatment in mice. Conclusions : YBT extract showed more remarkable inhibition on osteoclast differentiation than CPC and ALR extracts in vitro. ALR extract showed remarkable inhibition on bone resorption in vivo. Thus, YBT extract can be a useful treatment for bone-loss diseases such as osteoporosis.

  • A high concentration of inorganic phosphate (above 25 mM), which was suboptimal for vegetative growth in the minimal production medium, suppressed cephalosporin C (CPC) production in Cephalosporium acremonium. Results from the determination of intracellular concentrations of ATP, ADP and AMP with phosphate-starved resting cells indicated that phosphate exerted its effect indirectly by regulating the ratio of adenylated nucleotides, the so-called adenylated energy charge. It was also found that the type of phosphate regulation of CPC biosynthesis was not a repression effect but an inhibition effect.

Stimulation of Cephalosporin C Production by Acremonium chrysogenum M35 with Fatty Acids

  • Supplementation with rice oil and its major components (oleic acid and linoleic acid) was found to have a significant influence on cephalosporin C (CPC) production and cell growth by A. chrysogenum M35 in shake flask cultures. Five percent (v/v) rice oil had the most robust effect and 5% (v/v) oleic acid was the second most efficient on cell growth, whereas 3% (v/v) linoleic acid was found to be optimal for CPC production. Rice oil, oleic acid, and linoleic acid also significantly improved the rates of glucose consumption. When glucose was almost consumed, CPC production was initiated and, on the addition of rice oil, lipase activity increased steadily to 1.56 U/ml for 4 days. These results suggest that rice oil and fatty acids are used as carbon source to produce CPC by A. chrysogenum M35. Moreover, a mixture, composed of 40% (v/v) oleic acid and 60% (v/v) linoleic acid, had the strongest stimulatory effect on CPC production, due to a synergistic effect of the two fatty acids. Consequently, the maximum CPC titer (7.44 g/l) was improved about 4.5-fold.

노심보호/감시계통 운전성 시험 방법론의 개선

  • 영광 3,4호기 노심보호연산기(CPC)와 노심감시계통(COLSS)의 운전성 시험은 실시간 (on-line)으로 측정된 노심보호연산기 및 노심감시계통의 결과들(DNBR, LPD, DNBR 출력제한치, LHR 출력제한치)을 노심보호 연산기 및 노심 감시계통 각각의 시뮬레이터 시스템(CEDIPS/CPC FORTRAN, COLSS FORTRAN)의 계산 결과와 비교하여 그 타당성을 검증하는 것이다. 그러나 기존의 노심보호연산기 및 노심감시계통의 운전성 시험자료 취득 방법은 관측(CPC)과 상세 보고서(COLSS)에 의한 것으로 정확한 자료 취득에 어려움이 있고 많은 시간이 소요되며 또한 자료 취득시 실수유발 가능성이 높다. 따라서 본 연구에서는 발전소 전산계통(Plant Computer System)으로부터 발전소 운전 상황을 순간적으로 기록한 자료(Snapshot)를 취하여 노심보호 연산기 및 노심감시계통 운전성 시험에 필요한 자료를 자동으로 얻는 방법을 고안하였다. 또한 발전소 Snapshot으로부터 필요한 자료를 자동으로 얻어내기 위하여 컴퓨터 프로그램(CODAP)도 개발하였다. 본 연구에서 고안된 방법은 영광 3호기 1주기 CPC/COLSS 운전성 시험에서 검증이 되어 영광4호기 1주기에 적용하였고, 이후의 후속기에도 적용할 예정이다.

Effect of Humic Substances on the Simultaneous Removal of Nitrate and Phosphate in a Micellar-Enhanced Ultrafiltration (MEUF) (미셀 한외여과(MEUF)를 이용한 질산성 질소와 인산의 동시제거 시 휴믹산의 영향)

  • The effect of humic acid on the simultaneous removal of nitrate and phosphate was investigated in a micellar-enhanced ultrafiltration (MEUF). At the low molar ratio of cetylpyridinium chloride (CPC) to contaminants, the removal of nitrate was lower to 50% by 100 ppm of humic acid due to the competition for binding on micelles. At the molar ratio higher than 3, however, the removal of nitrate was over 80%. Phosphate was removed over 80% at the molar ratio higher than 1. The CPC and humic acid were rejected over 99 % by UF membrane. The flux did not decrease by 100 ppm of humic acid but rather slightly increased since the humic acid adsorbed on the membrane made the membrane more hydrophilic. As a result, humic acid did not diminish the performance of MEUF in the simultaneous removal of nitrate and phosphate.

Response of Odontoblast to the Bio-Calcium Phosphate Cement

  • Purpose: If the tooth structure is damaged, then it is impossible to regenerate the tooth. The materials used to restore the tooth structure are not related to the composition of the tooth. The materials used to restore the structure can't replace the natural tooth because they just fill the defective structure. Calcium phosphate cement remineralizes the dentin and almost replaces the natural tooth, but there are some disadvantages. We conducted basic tests with Biomimetic CPC (Bio-CPC) to make sure of the possibility of the biomaterial to remineralize the defective tooth structure. Methods: In this study, the bioactivity and biocompatibility of Bio-CPC were evaluated for its potential value as the bio-material for regeneration of damaged tooth structure by conducting a cell toxicity assay (WST-1 assay), a cytokinesis-block micronucleus assay, a chromosomal aberration test, total RNA extraction and RT-PCR on MDPC-23 mouse odontoblast-like cells. Results: The in vitro cytotoxicity test showed that the Bio-CPC was fairly cytocompatible for the MDPC-23 mouse odontoblast-like cells. Conclusion: Bio-CPC has a possibility to be a new biomaterial and further study of Bio-CPC is needed.

Studies on the Perception on Clinical Pharmaceutical Care and Clinical Pharmaceutical Sciences (임상약학, 임상약사, 임상약료와 임상약과학에 대한 인식도 조사 연구)

  • Background: There exist some different perceptions on clinical pharmacy between Korean and western societies. Since the pharmacists who received the 6-year pharmacy education join the pharmacy profession soon, it appears imperative to know whether the western-style clinical pharmacy is adaptable to Korean-style pharmacy education and profession. Methods: The authors surveyed 54 professors in the membership directory of the Korean College of Clinical Pharmacy (KCCP) on their perceptions on clinical pharmacy and clinical pharmacists, and their willingness for adapting clinical pharmaceutical care (CPC) and clinical pharmaceutical sciences (CPS) into clinical pharmacy education. The survey consists of 47 questions including dichotomized and Likert scale questionnaires in the areas of respondent demographics, clinical pharmacy, clinical pharmacists, CPC and CPS. Results: Of the 54 KCCP members surveyed, 29 completed the questionnaires in full and one in part. It appears that most KCCP members acknowledge the existence and importance of the two major fields of clinical pharmacy, CPC and CPS. Twenty-eight (96.6%) and seventeen members (68.0%) agreed to introduce CPC and CPS into the clinical pharmacy education in Korea, respectively. Seventeen (63.0%) answered CPC would be successfully adapted in Korea. Twenty (74.1%) agreed that it is desirable for clinical pharmacists to perform CPC and CPS simultaneously. Conclusion: Based on this survey, the authors suggest that pharmacy schools provide their students with the education opportunities on CPC to nurture clinical pharmacists through a master's program and CPS to foster clinical pharmaceutical scientists through a Ph.D. program.

Efficacy of Using Sequential Primary Circulating Prostate Cell Detection for Initial Prostate Biopsy in Men Suspected of Prostate Cancer

  • Background: Sequential use of circulating prostate cell (CPC) detection has been reported to potentially decrease the number of unnecessary prostate biopsies in men suspected of prostate cancer. In order to determine the real world effectiveness of the test, we present a prospective study of men referred to two hospitals from primary care physicians, one using CPC detection to determine the necessity of prostate biopsy the other not doing so. Materials and Methods: Men with a suspicion of prostate cancer because of elevated PSA >4.0ng/ml or abnormal DRE were referred to Hospitals A or B. In Hospital A all underwent 12 core TRUS biopsy, in Hospital B only men CPC (+), with mononuclear cells obtained by differential gel centrifugation identified using double immunomarking with anti-PSA and anti-P504S, were recommended to undergo TRUS biopsy. Biopsies were classifed as cancer or no-cancer. Diagnostic yields were calculated, including the number of posible biopsies that could be avoided and the number of clinically significant cancers that would be missed. Results: Totals of 649 men attended Hospital A, and 552 men attended Hospital B; there were no significant differences in age or serum PSA levels. In Hospital A, 228 (35.1%) men had prostate cancer detected, CPC detection had a sensitivity of 80.7%, a specificity of 88.6%, and a negative predictive value of 89.5%. Some 39/44 men CPC negative with a positive biopsy had low grade small volume tumors. In Hospital B, 316 (57.2%) underwent biopsy. There were no significant differences between populations in terms of CPC and biopsy results. The reduction in the number of biopsies was 40%. Conclusions: The use of sequential CPC testing in the real world gives a clear decision structure for patient management and can reduce the number of biopsies considerably.
