To investigate the anti-inflammatory activity of natural products, we determined the anti-inflammatory activity of purified epigallocatechin (EGC) from Camellia sinensis leaves. In the present study, we found that EGC inhibited the production of proinflammatory mediators (IL-6, TNF-${\alpha}$, NO, and $PGE_2$) in lipopolysaccharide (LPS)-stimulated Raw 264.7 cells. Suppression of IL-6 seems to be at least partly attributable to the inhibitory effect of EGC. TNF-${\alpha}$ is a major cytokine produced by LPS-induced macrophages, and they have a wide variety of biological functions including regulation of inflammation. The inhibition of IL-6 and TNF-${\alpha}$ production by EGC may downregulate the acute-phase response to LPS, thereby reducing LPS-induced inflammation. In addition to IL-6 and TNF-${\alpha}$, EGC effectively reduced the production of other key inflammatory mediators, including NO and $PGE_2$. The inhibitory effect of EGC on NO and $PGE_2$ production was supported by the suppression of inducible nitric oxide synthase and COX-2 at protein levels. These results support the traditional use of EGC in the alleviation of various inflammation-associated diseases and suggest that EGC might be useful in the development of new functional foods for inflammatory diseases.
Lactobacillus casei 3260 (L. casei 3260) was evaluated in relation to the inflammatory response mediated by lipopolysaccharide (LPS)-induced nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and cyclooxygenase-2 (COX-2) expression in Raw264.7 macrophage cells. The treatment of Raw264.7 cells with L. casei 3260 significantly inhibited the secretion of tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and prostaglandins $E_{2}\;(PGE_{2})$, followed by suppression of COX-2. To clarify the molecular mechanism, the inhibitory effect of L. casei 3260 on the NF-${\kappa}B$ signaling pathway was examined based on the luciferase reporter activity. Although the treatment of Raw264.7 cells with L. casei 3260 did not affect the transcriptional activity of NF-${\kappa}B$, it did inhibit NF-${\kappa}B$ activation, as determined by the cytosolic p65 release and degradation of I-${\kappa}B{\alpha}$. Therefore, these findings suggest that the suppression of COX-2 through inhibiting the NF-${\kappa}B$ activation by LPS may be associated with the antiinflammatory effects of L. casei 3260 on Raw264.7 cells.
Water and ethanol extracts of 16 different Korean traditional oriental prescriptions used widely for prevention of degenerative arthritis were prepared and their total phenolic and total flavonoid contents were quantified. Additionally, antioxidant and anti-inflammatory activities of water, ethanol and enzyme-treated extracts were determined by in vitro assays. Total phenolic and total flavonoid contents of three extracts from the 16 different medicinal prescriptions varied from 10.03-78.03 and 0-16.24 mg/g, respectively. Among the three extracts of 16 different medicinal prescriptions tested, 'Mahangeuigamtang' showed the potent full term for DPPH ($RC_{50}$=71.26, 27.33, 63.00 ${\mu}g/mL$) and full term for ABTS ($RC_{50}$=21.11, 27.45, 152.11 ${\mu}g/mL$) radical scavenging activities, and its water and ethanol extracts exhibited significant cyclooxygenase-2 (COX-2) inhibitory activity (49.10 and 69.06%, respectively). Ethanol and enzyme-treated extracts of 'Euieuiintang' exerted the strongest COX-2 inhibitory activity (68.23 and 75.05%, respectively). 'Mahangeuigamtang' and 'Euieuiintang' may be useful as potential therapeutic agents for treatment of degenerative diseases, such as inflammation and aging.

This study was carried out to find out the physiological activity effect of Sicyos angulatus L. extract which is known as an ecosystem disturbance plant and confirm the availability as a functional cosmetic material. Total polyphenol and flavonoid contents were measured, and DPPH radical scavenging, intracellular ROS, and its inhibitory effect on the expression Nitric oxide of and COX-2 were evaluated. The content of polyphenol and flavonoid was found to be 3.079 mg(CA)/100g and 72 mg(Q)/100g of Sicyos angulatus L. extract and antioxidant activity through high radical scavenging activity was confirmed. ~Significant cytotoxicity was not observed up to a concentration of $100{\mu}g/mL$ in RAW 264.7 cells and HDF cells and concentration-dependent inhibition of ROS production in HDF cells, inhibition of high NO production and inhibition of COX-2 protein expression in RAW 264.7 cells were confirmed. Through these results, we found the possibility of use as a functional cosmetic material with excellent antioxidant effect and antioxidant and anti-inflammatory effects through inhibition of intracellular ROS generation, inhibition of NO generation, and inhibition of COX-2 protein expression.

This study was carried out to investigate the effects of herbal extracts on the skin inflammatory reactions which use the makeup preparations. In experiment 1, among the herbal ingredients of herbal extracts, ethanol extracts and 1,3-BG(Butylene Glycol) extracts of Galla Rhois showed potent radical scavenging activity, more than 91% at all concentrations, tested by DPPH (1,1-diphenyl-2-picryl-hyrazyl) method. In experiment 2, ethanol extracts of Chrysanthemi Flos, Gardenias Flos, Galla Rhois showed potent inhibitory activity of the lipopolysaccharide-induced nitric oxide(NO) production, more than 87% at $10{\mu}g/m{\ell}$, by the macrophage RAW 246.7 cells. And 1,3-BG extracts of Taraxaci Herbs, Corm Fructus, Galla Rhois showed potent inhibitory activity of nitric oxide production, more than 89% at $10{\mu}g/m{\ell}$. In experiment 3, ethanol extracts of Chrysanthemi Flos, Gardeniae Flos, Galla Rhois showed potent inhibitory effects of cyclooxygenase-II activity, more than 78% at $10{\mu}g/m{\ell}$, by using ELISA kit. And 1,3-BG extracts of Galla Rhois, Carthami Flos, Chrysanthemi Flos, Taraxaci Herba, Corm Fructus showed potent inhibitory effects of cyclooxygenase-II(COX-II) activity, more than 80% at $10{\mu}g/m{\ell}$. Therefore, 1 expect that herbal extracts, especially Galla Rhois may be used as a drug for treatment on skin inflammation and a material of the makeup preparations.
Objectives This study was designed to investigate whether the Gagamtongsoon-San (GT) has an inhibitory effect and its mechanisms are associated with the iNOS and COX-2. Methods Cytotoxic activity of GT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Inflammatory condition was induced by LPS. NO production was measured using Griess reagent system. The expressions of iNOS and COX-2 mRNA and protein were determined by realtime PCR. The concentrations of PGE2 were measured by an enzyme immunoassay (EIA). Results The GT does not impair the cell viability in tested concentration $500{\mu}g/ml$ or below. GT significantly reduced the NO production in a dose-dependent manner. GT $500{\mu}g/ml$ also suppressed LPS-induced mRNA expressions of iNOS and COX-2. GT $500{\mu}g/ml$ reduced the PGE2 secretion in LPS induced RAW264.7 cells. Conclusions These outcomes show that GT extract has an anti-inflammatory activities. And also this conclusion can be the data that supports the GT's anti-inflammatory effect objectively.

Antioxidant and anti-inflammatory activities of eugenol and its derivatives from clove (Eugenia caryophyllata Thunb.) were evaluated using in vitro assay systems by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, cyclooxygenase-2 (COX-2), and 15-lipoxygenase (15-LOX). Among eight different crude medicinal drugs tested, volatile extracts of clove extracted by steam distillation extraction (SDE) showed potent DPPH radical scavenging activity ($IC_{50}$=8.85 ${\mu}g/mL$) as well as strong inhibitory activity against COX-2 (58.15%) and 15-LOX (86.15%) at 10 ${\mu}g/mL$ and 25 ${\mu}g/mL$, respectively. Major volatile components of clove were identified as eugenol, trans-caryophyllene, and acetyleugenol by GC-MS analysis. Out of three eugenol derivatives, eugenol, methyl eugenol, and acetyl eugenol, eugenol showed the strongest DPPH radical scavenging activity and COX-2 inhibitory activity, whereas methyl eugenol exhibited the strongest 15-LOX inhibitory activity. Finally, the contents of the three eugenol derivatives in clove were quantified by analytical HPLC. Contents of eugenol and acetyl eugenol in clove were 6.95% and 1.85% per dry weight, respectively. These results suggest that eugenol and its derivatives in steam distilled extract of clove may be useful as potential antioxidant and anti-inflammatory agents.
This research was undertaken to find the in vitro inflammatory action of the essenetial oil (CS-oil) extracted from Chrysanthemum sibiricum (Compositae) herbs. We investigated the effects of the CS-oil not only on the formation NO, $PGE_2$, and $TNF-{\alpha}$ but also on inducible nitric oxide synthase and cyclooxygenase-2 (COX-2) in lipopolysaccharide (LPS)-induced murine macrophage RAW 264.7 cells. The data obtained were consistent with the modulation of iNOS enzyme expression. A similar fashion was also observed when LPS-induced $PGE_2$ release and COX-2 expression were tested. The significant inhibitory effects were shown in concentration-dependent manners. In addition, CS-oil also mildly but significantly reduced the formation of TNF-a. These findings support the application of CS-oil as an antiinflammatory essential oil.

Objectives : The various grape extracts derived from grape pulp, seed and skin, containing various types of polyphenols and flavonoids, have been known to have anti-inflammatory, antioxidant and improve cardiovascular condition as well as sun's damaging effects. However, there have been rare reports of various beneficial effects of grape fruit stem extract (GFSE), one of the waste products of grapes. We investigated anti-inflammatory and melanogenesis inhibitory effects of GFSE. Methods : One-hundred gram of grape fruit stem was extracted with 80% ethanol at room temperature for 3 days. After filtration, the ethanol was removed using vacuum evaporator, then lyophilized to obtain the dry extract which was stored at $-20^{\circ}C$ until used. NO levels were measured by using Greiss reagent. Prostaglandin $E_2$ ($PGE_2$) production was measured by ELISA assay. The expression levels of iNOS, COX-2, TRP-1 and TRP-2 were evaluated by western blot analysis. Results : GFSE reduced the level of nitric oxide and prostaglandin $E_2$ ($PGE_2$) production in a dose-dependent manner, compared to control. Expressions of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) protein were also effectively inhibited by the GFSE. In a tyrosinase inhibitory activity, GFSE significantly reduced the tyrosinase activity and melanin content in a dose dependent manner, compared to control. GFSE also decreased the expression of tyrosinase related protein-1 (TRP-1) and tyrosinase related protein-2 (TRP-2), known as a melanocyte-specific gene product involved in melanin synthesis. Conclusions : Therefore, these results indicated that GFSE had powerful anti-inflammatory and anti-melanogenic effects.
This experiment was conducted to enhance the colored potatoes utilization and to determine the biological activity of colored potato extracts. In order to understand the factors responsible for the potent anti-oxidant ability of colored potatoes, it has been evaluated for anti-oxidative activity using oxygen radical absorbance capacity (ORAC) assay. 'Hongyoung' extract was significant anti-oxidant activities in ORAC assay. About two-fold higher radical absorbance capacity was found in 'Hongyoung' compared to that in 'Jayoung'. The ability of 80% ethanol extracts from colored potatoes to influence the inhibitory activity of nitric oxide (NO) and nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) has also been investigated. The various therapeutic benefit claims in the new functional medicinal usage of colored potatoes ascribed to the phenolic compounds and anthocyanin. This result revealed that the extracts of colored potatoes are expected to be good candidate for development into sources of free radical scavenger or COX-2 inhibiting agents.
