Experimental research for the protective effect of Naoxingtong-containing serum on rat cerebral microvascular endothelial cells

  • The protective effect of Naoxingtong (NXT) on rat cerebral microvascular endothelial cell (rCMEC) was investigated. rCMEC was injured in vitro by incubating for 4 hours at 100% NO in a hypoxia chamber. After treated with NXT-containing serum, the cellular viability rate (90.3%) was significantly elevated when compared with that of control group and the inhibitive rate of lactic dehydrogenase activity (9.2%) was far lower than the control group with dose-dependent effect. The results indicate that NXT can increase viability of rCMEC, and protect cell membrane from injury during hypoxia.

  • Effects of Naoxintong (NXT, a formula of Chinese Materia Medica)-containing serum on Nitrogen monoxide (NO) and calcitonin gene related peptide (CGRP) in rat cerebral microvascular endothelial cells (rCMEC) was investigated, rCMEC was injured in vitro by incubating for 4 hours at 100% NO in a hypoxia chamber. The results indicated that NXT could antagonize the reduction of NO and CGRP secreted by rCMEC during hypoxia, the effect of which was dose-dependent. After treated with NXT-containing serum at dosage of 5.0 - 30 and 50 -1.1 g/kg/U respectively, the amount of NO and CGRP secreted by rCMEC were remarkably increased during hypoxia in vitro.

  • For the purpose of improving the fluidity of enteric-coated powders, various mineral silicates were added during spray drying process. Aqueous slurries of cimetidine, mineral silicates containing CMEC (carboxy methyl ethyl cellulose) were spray dried using a centrifugal wheel atomizer. The finely agglomerated powders obtained by this process were flowing as opposed to the original powders. The effect of four mineral silicates (colloidal silica, talc, bentonite, and kaolin) on the micromeritic properties and dissolution profiles of spray dried agglomerates were examined.

Proteomic Analysis of a Rat Cerebral Ischemic Injury Model after Human Cerebral Endothelial Cell Transplantation

  • Objective : Cerebral endothelial cells have unique biological features and are fascinating candidate cells for stroke therapy. Methods : In order to understand the molecular mechanisms of human cerebral endothelial cell (hCMEC/D3) transplantation in a rat stroke model, we performed proteomic analysis using 2-dimensional electrophoresis and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Protein expression was confirmed by quantitative real-time PCR and Western blot. Results : Several protein spots were identified by gel electrophoresis in the sham, cerebral ischemia (CI), and CI with hCMEC/D3 treatment cerebral ischemia with cell transplantation (CT) groups, and we identified 14 differentially expressed proteins in the CT group. Proteins involved in mitochondrial dysfunction (paraplegin matrix AAA peptidase subunit, SPG7), neuroinflammation (peroxiredoxin 6, PRDX6), and neuronal death (zinc finger protein 90, ZFP90) were markedly reduced in the CT group compared with the CI group. The expression of chloride intracellular channel 4 proteins involved in post-ischemic vasculogenesis was significantly decreased in the CI group but comparable to sham in the CT group. Conclusion : These results contribute to our understanding of the early phase processes that follow cerebral endothelial cell treatment in CI. Moreover, some of the identified proteins may present promising new targets for stroke therapy.

Enzymatic Hydrolysis of Egg White Protein Exerts a Hypotensive Effect in Spontaneously Hypertensive Rats

  • This study was conducted to investigate the hypotensive effect of egg white protein (EWP) hydrolysate (EWH) in spontaneously hypertensive rats (SHRs). The hydrolysis of EWP was effectively performed with a combination of 0.5% bromelain and 1% papain at 50℃ for 60 min. The resulting hydrolysate did not elicit an allergic reaction as confirmed by human mast cell activation test. The systolic and diastolic blood pressures of the SHRs fed the EWH diet were observed to be significantly or numerically lower than those of the other groups during the experimental period of 28 d. EWH treatment significantly (p<0.05) upregulated the nitric oxide levels in hCMEC/D3 cells and the plasma of the SHRs compared to those in the control. Moreover, EWH ingestion significantly (p<0.01) reduced the plasma angiotensin II level of the SHRs compared with that in the control. In conclusion, beyond its basic nutritional value, EWH prevents and manages hypertension, and thus can be an invaluable resource for functional food development.
