EFFECT OF POTASSIUM ION ON INTRADENTAL NERVE ACITIVITY (칼륨이온이 치수내 신경활동에 미치는 영향)

  • The intradental nerve activity was recorded from single pulp nerve unit dissected from the inferior alveolar nerve in canine teeth of anesthetized cats. The effects of various test solutions on intradental nerve activity evoked by 4M NaCl applied to the deep dentinal cavities were investigated. 1. Total 15 single pulp nerve units were recorded. Of these 9 were Mi - fibers and 6 were C -fibers. The mean conduction velocity and electrical threshold of $A{\delta}$ - fiber were $6.3{\pm}3.7m/sec$, $1.2{\pm}0.7V$ and those of C - fiber were $1.0{\pm}0.2m/sec$, $2.3{\pm}1.3V$, respectively. The response to cold stimuli of $A{\delta}$ - fiber began immediately and that of C - fiber began after a latency. 2. When applied to deep dentinal cavity, 4M NaCl induced irregular bursts of action potential in 4 $A{\delta}$ - fibers and 3 C - fibers, which continued until the solution was washed away. 3. In the $A{\delta}$ - fiber, histamine failed to induce any nerve acitivity and did not produce an increase in intradental nerve activity evoked by 4M NaCl. However following the application of 1M KCl, the response to 4M NaCl was eliminated. 4. In the C - fiber, histamine generated some nerve activity and produced a significant increase in intradental nerve activity evoked by 4M NaCl, but 5M $CaCl_2$ did not abolish this enhandced response.

  • Ion transport of a polypyrrole/chloride (PPy/Cl) film and a polypyrrole/poly(styenesulfonate) (PPy/PSS) film as a function of applied dc potential was investigated by employing electrogravimetric impedance technique and electrochemical impedance technique. The cation and anion contribution to the whole charge capacitance and the diffusion coefficients of cation and anion in a PPy/PSS film were calculated by fitting the electrogravimetric impedance data with proposed model circuit. The diffusion coefficients of $Na^+$ in a 1 M $NaClO_4$ solution are over 1 order of magnitude larger than those of $ClO{_4}^-$, and $ClO{_4}^-$ contribution to charge compensation decreases as dc potential lowers. The charge compensation of a PPy/Cl film ir a 1 M CsCl solution is carried out largely by $Cl^-$ at 0.2 V vs. Ag/AgCl and by $Cs^+$ as well as $Cl^-$ at -0.4 V.

Application of the Central Composite Design and Response Surface Methodology to the Treatment of Dye Using Electrochemical Oxidation (전기화학적 산화를 이용한 염료 처리에 중심합성설계와 반응표면분석법의 적용)

  • The aim of this research was to apply experimental design methodology in the optimization condition of electrochemical oxidation of Rhodamine B(RhB). The reactions of electrochemical oxidation were mathematically described as a function of parameters amounts of current, NaCl dosage, pH and time being modeled by the use of the central composite design, which was used for fitting quadratic response surface model. The application of response surface methodology using central composite design(CCD) technique yielded the following regression equation, which is an empirical relationship between the removal efficiency of RhB and test variable in actual variables: RhB removal (%) = 3.977 + 23.279$\cdot$Current + 49.124$\cdot$NaCI - 5.539$\cdot$pH - 8.863$\cdot$time - 22.710$\cdot$Current$\cdot$NaCl + 5.409$\cdot$Current$\cdot$time + 2.390$\cdot$NaCl$\cdot$time + 1.061$\cdot$pH$\cdot$time - $0.570{\cdot}time^2$. The model predicted also agree with the experimentally observed result($R^2$ = 91.9%).

  • The nerve impulse pattern of Chorda tympani(CT) of cat was tested with Monosodium glutamate(MSG) solutions as well as some basic taste substances applied on the tongue of cat. The effect of MSG applied in the tongue prior to the stimulation of other taste substances was also investigated. The response impulse frequency of CT of cat was changed by the kind and concentration of taste substances. The response to citric acid was the highest among the tested substances, NaCl, KCl and MSG showed similar responses. When different concentrations of MSG were applied on the tongue prior to other substances, the response to NaCl increased with the maximum response at the MSG concentration of 0.02 M. The response to sucrose tended to be reduced, but the response to citric acid was distinctly supressed by the previous MSG stimulation. These results were well consistent with the sensory evaluation on the effect of MSG to some basic taste substances. previously reported by the authors.

Changes of Vascular Contractility of isolated Rat Aorta treated with Salt Stress (Salt 스트레스에 의한 흰쥐 적출대동맥의 수축력 변화양상)

  • To examine whether salt stress would alter or not contractility of isolated rat aorta, under anesthesia with sodium pentobarbital(50 mg kg-1 i.p.), male Sprague Dawley rats(300-330 g) were subjected to 0, 50, and 150 mM of sodium chloride at 37$^{\circ}C$ for 60 min. where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsp) by Western blotting. One, three and eight hours later, we measured vascular contractility of isolated rat aorta treated with KCI, phenylephrine from organ bath study. The dose-vascular responses of potassium chloride and phenylephrine showed a little augmentation by NaCl concentration in the strips exposed to NaCl for 8 hours. And the response of relaxation induced by nitroprusside and acetylcholine was not influenced by NaCl stress in isolated aorta ring for 8 hours, respectively. Expression pattern of Hsp 70 of vascular muscle in isolated rat aorta showed a little increase in 150 mM NaCl group at 8 hours after NaCl treatment but not at 3 hours, and Hsp 60 expression of rat aorta was markedly increased in 50 mM NaCl group at 8 hours after NaCl treatment. Taken together, NaCl induced dose-and time dependent accumulation of the Hsp but not affected contraction of rat aorta. These data suggest that short term high salt stress was not sufficient to induce hypertension of rat aorta.

Effects of N-acetylcysteine (NAC) on chemiluminescence response as a non-specific immune parameter in Far Eastern catfish Silurus asotus (항산화물질 N-acetylcysteine (NAC)이 메기에서 비특이적 면역지표인 화학발광 반응에 미치는 영향)

  • It has been reported that various anti-oxidant substances stimulate non-specific immune responses in fishes. In this study it was examined whether N-acetylcysteine (NAC), a precusor for anti-oxidant glutathione (GSH) synthesis, can modulate non-specific immune function in Far Eastern catfish Silurus asotus. Immune functions were assessed using the respiratory burst activity monitored by chemiluminescence (CL) responses in isolated leucocyte. NAC stimulated CL responses with doses of 10 or 100 mg/kg, but not with 1 mg/kg after 48 hr injection. It was observed with 10 mg/kg NAC that CL activity continued to elevate from 24 hr through 96 hr post-dosing, and returned to the near preinjection level by 10 days. To understand whether NAC can also activate CL activity in vitro, NAC was directly added to isolated catfish leucocytes. It was observed, however, that NAC can not stimulate CL at reasonable concentration ranges in vitro. As NAC is a precursor for the strong anti-oxidant glutathione (GSH), a putative immune stimulator, it was assessed whether GSH can also stimulate CL responses. Observed results show that GSH activated CL both in vivo and in vitro. The data obtained collectively support the proposition that NAC indirectly stimulates non-specific immune functions in catfish by enhancing GSH biosynthesis, but not by direct action of NAC. Such effects may have beneficial significance in aquaculture for practical utilization.

Changes in the metabolic profile and nutritional composition of rice in response to NaCl stress

  • Salinity is a major abiotic stress that adversely affects crop productivity and quality. In this study, the metabolic profile and nutritional composition of rice in response to NaCl were analyzed. The plants were exposed to stressed or unstressed conditions, and their metabolic changes were examined in the shoots, roots, and grains collected at different growth stages. The levels of nutrients and anti-nutrients, including proximates, amino acids, fatty acids, minerals, vitamins, and phytic acid, were also determined for the grains. Application of NaCl significantly decreased the shoot and root growth and induced metabolic alterations at the tillering stage. During the heading stage, only the root metabolites were influenced by NaCl, and no metabolic variations related to salinity were found in the shoot, roots, and grains at the ripening stage. Nutritional analysis of the grain samples revealed that the amounts of linolenic acid and tricosanoic acid were significantly reduced while those of copper, sodium, and phytic acid were enhanced in response to stress. However, except for sodium, those differences were not great. Our results suggest that although NaCl-salinity influences the phenotypic and metabolic profiles of rice shoots and roots at the tillering stage, this impact becomes negligible as tissue development proceeds. This is especially true for the grains. Compositional analysis of the grains indicated that salinity induces some changes in fatty acids, minerals, and anti-nutrients.

In vitro Culture Response to NaCl of Korean Ginseng (Panax ginseng C.A. Meyer) Tissues (기내배양을 통한 고려인삼(Panax ginseng C.A. Meyer)조직의 NaCl에 대한 반응)

  • High salt concentrations in the ginseng nursery soil environment of Korea is one of important reducing factors for the stable production of quality ginseng. These studies were accomplished for check the response on germination of ginseng seed, somatic embryogenesis of zygotic embryo, and biosynthesis of ginsenoside from ginseng hairy root against NaCl. Ratio of germination was at the $3\%\;and\;84.5\%$ on the basic media with 0.1M and free of NaCl repectedly, but $0\%$ at the upper of 0.2M NaCl. Somatic embryogenesis from zygotic embryo were the highest when immatured embryo was cultured on free of NaCl concentration, and which was intend to decrease at treatment of NaCl. However, in case of using the matured embryo, treatment of 0.05M NaCl resulted in better embryogenesis than NaCl free media. Red pigment was synthesized from ginseng hairy root cultured on the medium with various NaCl concentration(from 0.04 to 0.08M) and its pigment was analyzed as spectrum of anthocyane by spectrophoto- meter scanning. This cell line biosynthesized lots of crude saponin and total ginsenoside than other cell lines, also had 2 times of panaxadiol than panaxatriol.

Germination and Growth Response of Spergularia marina Griseb by Salt Concentration (갯개미자리(Spergularia marina Griseb)의 염농도에 따른 발아 및 생장 반응)

  • This study was conducted to investigate the germination and growth response by Salinities of Spergularia marina Griseb grown in the western coastal region in South Korea. The germination was investigated for 10 days at temperature $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$ in order to examine the germination of Spergularia marina Griseb by NaCl concentration 0.0, 0.3, 0.5, 1.0, 2.0%. The germination of NaCl concentration 0.0~1.0% was 90% over at $15^{\circ}C$ treatment, but the germination of NaCl concentration 2.0% was 0% at all temperature treatments. To identify the growth response of Spergularia marina Griseb according to the salinity, Spergularia marina Griseb was cultivated for 8 weeks in Hoagland culture medium and sea water. In nutrient solution culture, growth was best in NaCl 50mM among 0~400 mM concentrations and in 0, 12.5, 25, 50, 100% of sea water, growth was best in 12.5% (dilution ratio with fresh water) treatment. Spergularia marina Griseb's inorganic component contents according to salinity showed that $Na^+$ content increased, but contents of $K^+$, $Ca^+$, $Mg^+$ decreased. As a result, appropriate condition for Spergularia marina Griseb's germination is considered to be maintained at $15^{\circ}C$ and in less than NaCl 1.0% of salinity. When nutrient solution culture, NaCl 0.3% of treatment level is considered to be the optimum salinity.

Optimization of the Turbidity Removal Conditions from TiO2 Solution Using a Response Surface Methodology in the Electrocoagulation/Flotation Process (전기응집/부상 공정에서 반응표면분석법을 이용한 TiO2 수용액의 탁도 처리조건 최적화)

  • The removal of turbidity from $TiO_2$ wastewater by an electrocoagulation/flotation process was studied in a batch reactor. The response surface methodology (RSM) was applied to evaluate the simple and combined effects of the three main independent parameters, current, NaCl dosage and initial pH of the $TiO_2$ solution on the turbidity removal efficiency, and to optimize the operating conditions of the treatment process. The reaction of electrocoagulation/flotation was modeled by use of the Box-Behnken method, which was used for the fitting of a 2nd order response surface model. The application of RSM yielded the following regression equation, which is an empirical relationship between the turbidity removal efficiency of $TiO_2$ wastewater and test variables in uncoded unit: Turbidity removal (%)=69.76+59.76Current+11.98NaCl+4.67pH+5.00Current${\times}$pH-160.11$Current^2-0.34pH^2$. The optimum current, NaCl dosage and pH of the $TiO_2$ solution to reach maximum removal rates were found to be 0.186 A, 0.161 g/l and 7.599, respectively. This study clearly showed that response surface methodology was one of the most suitable method to optimize the operating conditions for maximizing the turbidity removal. Graphical response surface and contour plots were used to locate the optimum point.
