Role of CXCR7 and Effects on CXCL12 in SiHa Cells and Upregulation in Cervical Squamous Cell Carcinomas in Uighur Women

  • CXCR7 is involved in tumor development and metastasis in multiple malignancies. However, the function and molecular mechanisms of action of CXCR7 in human cervical cancer are still unclear. In the present study a loss of-function approach was used to observe the effects of recombinant CXCR7 specific small interfering RNA pBSilence1.1 plasmids on biological behavior including proliferative activity and invasive potential, as indicated by MTT assays with the cervical cancer SiHa cell line in vitro. Reverse transcription polymerase chain reaction and Western blotting revealed that CXCR7 was downregulated in transfected compared with control cells, associated with inhibited cell growth, invasiveness and migration. The expression of CXCR7 and CXCL12 was also determined immunohistochemically in 152 paraffin-embedded, cervical squamous cell carcinoma (CSCC) and cervical intraepithelial neoplasia (CIN), or normal cervical epithelial to assess clinico-pathological pattern and CXCR7 status with respect to cell differentiation and lymph node metastasis in Uighur patients with CSCC. CXCR7 and CXCL12 expression was higher in cervical cancer than CIN and normal cervical mucosa, especially in those with higher stage and lymph node metastasis. CXCL12 appeared to be positively regulated by CXCR7 at the post-transcriptional level in CSCC. We propose that aberrant expression of CXCR7 plays a role in carcinogenesis, differentiation and metastasis of CSCC, implying its use as a potential target for clinical biomarkers in differentiation and lymph node metastasis.

Does Cervical Screening in Young Women Aged 20-25 Years Lead to Unnecessary and Harmful Interventions?

  • Background: Cervical human papillomavirus (HPV) infection among young women (20-25 years of age) is common and normally transient. There are growing concerns that referral to a colposcopy clinic may lead to unnecessary treatment with an increased risk of obstetric complications. Therefore, the purpose of this study was to determine the level of intervention for cervical abnormalities in this age group of the Northern Ireland population. Materials and Methods: A review of all serial new patients under 25 years of age, who were referred to colposcopy clinics in Northern Ireland between January 1, 2009 to June 30, 2009 formed the basis of this study. Results: During the study period, a total of 4,767 women under 25 years of age were screened. Two-hundred-and-thirty-four (4.9%) cases were referred to the colposcopy clinics. The cervical cytology results were: high-grade abnormality in 35%, and low-grade abnormality in 31% of these cases. One-hundred-and-seventy-eight (76%) of the referred women received at least one treatment. One-hundred-and-twenty-one of 234 (51.5%) women underwent an excisional treatment with histology showing the presence of high-grade abnormalities (CIN2-3) in 52%, CIN1 in 28%, and Koilocytosis or normal tissue in 20% of this sub-group of cases. Conclusions: Screening women under the age of 25 years cause unnecessary referral for colposcopy. This may also result in considerable anxiety and psychosexual morbidity. It leads to an over-treatment with a potential of negative impact on the future pregnancy outcomes (including pre-term delivery, low birth weight, and pre-term premature rupture of membranes).

Colposcopy Requirement of Papanicolaou Smear after Atypical Squamous Cells of Undetermined Significance (ASC-US) by Follow-up Protocol in an Urban Gynaecology Clinic, a Retrospective Study in Thailand

  • Background: ASC-US cases are managed according to the current American Society for Colposcopy and Cervical Pathology (ASCCP) guideline in which a human papillomavirus (HPV) test and repeat Pap smear are performed in the next 1 year. Colposcopy in cases of positive high risk HPV and persistent ASC-US or more in subsequent Pap smear is recommended. The HPV test is more expensive and still not currently a routine practice in Thailand. Objective: To identify the risk factors of persisted abnormal Pap smear and the colposcopic requirement rate in women with ASC-US. Materials and Methods: During 2008-2013, this study was conducted in Prapokklao Hospital, Chanthaburi, Thailand. Participants were women who attended gynaecology clinic for cervical cancer screening. Women who had cytological reports with ASC-US were recruited. During the study period, 503 cases were enrolled. Colposcopic requirement was defined as those who were detected with an ASC-US or more in subsequent Pap smears up to 1 year follow-up. Results: The colposcopic referral rate was 23.2 (85/365) percent at 12 months. Prevalence of cervical intraepithelial neoplasia (CIN) 2/3 was 3.3 (12/365) percent. Loss follow-up rate of subsequent Pap smear and colposcopic appointment were 27.4 (138/503) and 48.2 (41/85) percent, respectively. There was no invasive cancer. High risk factors for persisted abnormal Pap smears in subsequent test were premenopausal status, HIV infected patients and non-oral contraceptive pills (COC) users. Conclusions: Referral rate for colposcopy in women with ASC-US reports was rather high. Loss to follow-up rate was the major limitation. Immediate colposcopy should be offered for women who had high risk for silent CIN.

Value of PAX1 Methylation Analysis by MS-HRM in the Triage of Atypical Squamous Cells of Undetermined Significance

  • Background: Detection of cervical high grade lesions in patients with atypical squamous cells of undetermined significance (ASCUS) is still a challenge. Our study tested the efficacy of the paired boxed gene 1 (PAX1) methylation analysis by methylation-sensitive high-resolution melting (MS-HRM) in the detection of high grade lesions in ASCUS and compared performance with the hybrid capture 2 (HC2) human papillomavirus (HPV) test. Materials and Methods: A total of 463 consecutive ASCUS women from primary screening were selected. Their cervical scrapings were collected and assessed by PAX1 methylation analysis (MS-HRM) and high-risk HPV-DNA test (HC2). All patients with ASCUS were admitted to colposcopy and cervical biopsies. The Chisquare test was used to test the differences of PAX1 methylation or HPV infection between groups. Results: The specificity, sensitivity, and accuracy for detecting CIN2 + lesions were: 95.6%, 82.4%, and 94.6%, respectively, for the PAX1 MS-HRM test; and 59.7%, 64.7%, and 60.0% for the HC2 HPV test. Conclusions: The PAX1 methylation analysis by MS-HRM demonstrated a better performance than the high-risk HPV-DNA test for the detection of high grade lesions (CIN2 +) in ASCUS cases. This approach could screen out the majority of low grade cases of ASCUS, and thus reduce the referral rate to colposcopy.

Thermodynamic Characteristics Associated with Localized Torrential Rainfall Events in the Middle West Region of Korean Peninsula (한반도 중서부 국지성 집중호우와 관련된 열역학적 특성)

  • Thermodynamic conditions related with localized torrential rainfall in the middle west region of Korean peninsula are examined using radar rain rate and radiosonde observational data. Localized torrential rainfall events in this study are defined by three criteria base on 1) any one of Automated Synoptic Observing System (ASOS) hourly rainfall exceeds $30mmhr^{-1}$ around Osan, 2) the rain (> $1mmhr^{-1}$) area estimated from radar reflectivity is less than $20,000km^2$, and 3) the rain (> $10mmhr^{-1}$) cell is detected clearly and duration is short than 24 hr. As a result, 13 cases were selected during the summer season of 10 years (2004-13). It was found that the duration, the maximum rain area, and the maximum volumetric rain rate of convective cells (> $30mmhr^{-1}$) are less than 9hr, smaller than $1,000km^2$, and $15,000{\sim}60,000m^3s^{-1}$ in these cases. And a majority of cases shows the following thermodynamic characteristics: 1) Convective Available Potential Energy (CAPE) > $800Jkg^{-1}$, 2) Convective Inhibition (CIN) < $40Jkg^{-1}$, 3) Total Precipitable Water (TPW) ${\approx}$ 55 mm, and 4) Storm Relative Helicity (SRH) < $120m^2s^{-2}$. These cases mostly occurred in the afternoon. These thermodynamic conditions indicated that these cases were caused by strong atmospheric instability, lifting to overcome CIN, and sufficient moisture. The localized torrential rainfall occurred with deep moisture convection result from the instability caused by convective heating.

Prediction of Vapor Pressure of the Inert Gases (비활성 기체의 증기압 예측)

  • Experimental vapor pressure measurements available in the literature for the inert gases have been rigorously analyzed and used to evaluate the constants A, B, C, D, and exponent of the following equation in the form of reduced vapor pressure and reduced temperature : $InP_r=A+{\frac{B}{T_r}+CInT_r+DT_n^r}$ According to varying exponent n all four constants have been obtained for the inert gases by the error analysis. This has provided us the best n and four constants for each of the inert gases ; Argon, krypton, xenon, helium, and neon. In order to obtain the calculated vapor pressure by the above equation, only the normal boiling point and the critical pressure and critical temperature are necessary to get the vapor pressure for an overall average deviation of 0.31 % for 406 experimental vapor pressure points consisting of five gases available in the literature. The average deviation for argon, krypton, and xenon is 0.24%, 0.09%, and 0.22%, respectively, for neon 1.31% and for helium 0.61%. These results are not unexpected in view of the significant quantum effects associated with helium and to a lesser degree with neon.

Understanding centrosome amplification in cancer: A pathway toward precision-targeted cancer drug development (암의 중심체 증폭 이해를 통한 표적 항암제 개발)

  • Cell division is an essential process for the survival and development of living organisms. It is critical that duplicated chromosomes are properly segregated into daughter cells during mitosis. The centrosome is the core organelle that forms the microtubule-organizing center (MTOC), which generates the microtubules that make up the mitotic spindle during cell division. The centrosome is also involved in cell signaling and motility. In normal cells, there is one centrosome in G1 that replicates into two in the S phase and matures through G2. During the M phase, duplicated centrosomes move to both ends of the cell, and spindle microtubules that are generated from MTOC move the chromosome to both ends. The cells then split into two to complete the cell division. However, a phenomenon called centrosome amplification (CA), in which the number of centrosomes is higher than normal, is common in cancer cells and can lead to chromosome instability (CIN). This paper discusses the process of centrosome replication and the role of PLK4 in this process. The possible consequences of centrosome amplification and how the PLK4 inhibitor may be able to treat certain types of cancer cells, such as breast cancer and neuroblastoma, will also be discussed.

A Study on the Development of ZVS Half-Bridge Converter for PDP Drive (PDP 구동을 위한 ZVS Half-Bridge 컨버터 개발에 관한 연구)

  • In this paper, we have developed a ZVS half-bridge converter with a clamping circuit for the DC power source of PDP circuit. The clamping circuit in the developed converter reduces the oscillating current of the switch by resonant inductor and parasitic parameters in output rectifier diodes. Finally, comparing the experimental results of the developed converter to the conventional ZVS half-bridge converter, it is clarified that the developed converter is more efficient and lower noise than the conventional one.

Analysis of the aromatic components of the forest bathing (삼림욕 향기 성분 분석)

  • As the suitable place of the forest bathing, Two places of the needle-leaf trees, which are located in the Oh-Saek mineral spring near place from the Sorak mountain, were selected. And then, Headspace gas trapping apparatus were setted in that two places and the aromatic components of the forest were adsorbed by Tenax-TA column for 24 hours. And Tenax-TA column were analyzed by the GC SE GC-MS. The analyzed components were found to contain up to between 70-80% of pollutants, which are Toluene, Methyl Chloride, Hexane, p-Xylene, Benzene, ... etc. On the other hand, the aromatic components of the forest, which give aromatheraphitical effectness, are as follow: alpha-Pinene, Limonene, 1, 8-Cin-eol, Benzaldehyde, . . . etc.

Early History of Korean Restaurants in Manhattan, NY - Focused on 1960's~1970's - (뉴욕 맨해튼 한국 음식점의 초기 역사 - 1960년대~1970년대를 중심으로 -)

  • The purpose of this paper was to research the history of Korean restaurants in Manhattan, NY in the 1960's-1970's. These Korean restaurants were the pioneers in the globalization of Korean food. It is assumed that 'Mi Cin' was the first Korean restaurant in Manhattan and opened on March $1^{st}$, 1960. In the 1960's, it is estimated that there were four Korean restaurants in Manhattan. In the 1970's, the number of Korean restaurants increased to more than 18, and their main menu items were divided into three types: Korean fusion menu such as 'Lunch Special' for American customers, beef barbecue menu for American and Korean customers, and Korean traditional menu for increasing Korean immigrants.
