Genotoxicity study of Aralia elata extract in bacterial and mammalian cell system (두릅나무 추출물의 유전독성평가)

  • In order to investigate the safety of Aralia elata extract causing the reduction in the blood glucose level and oxidative stress in diabetes animals, these genotoxicity studies in bacterial and mammalian cell assay system such as Ames bacterial reverse mutation test and chromosomal aberration assay were performed. As results, in Ames bacterial reversion assay the extract in the range of 5,000-625 ug/plate did not induce mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535 and TA 1537 strains with and without metabolic activation of S-9 mixture. For chromosomal aberration assay, $IC_{50}$ (50% inhibition concentration of cell growth) of the extract were determined; 792 $\mu\textrm{g}$/$m\ell$ without and 524 $\mu\textrm{g}$/$m\ell$ with S-9 mixture in Chinese hamster lung (CHL) fibroblast cell culture. Any significant chromosomal aberration was not observed in CHL cells treated with the extract at the concentrations of 792, 396 and 198 $\mu\textrm{g}$/$m\ell$ or 524, 262 and 131 $\mu\textrm{g}$/$m\ell$ in the absence or presence of S-9 metabolic activation, respectively. From these results, Aralia elata extract did not induce any harmful effects on the gene in bacteria and mammalian cell system used in these experiments.

The Distribution and Standing Crop of Phytoplankton at the Estuaries of Galgok Stream and Incheon River in Jeollanam-do (전남 갈곡천과 인천강 하구역의 식물플랑크톤의 분포 및 현존량)

  • Species composition, standing crop and dominant species of phytoplankton were investigated at 6 sites of Galgok stream and Incheon river which run into Gomso Bay, Jeollanam-do in April, August and November 2007. Total of 210 taxa were found which were classified as 6 classes, 12 orders, 25 families, 66 genera, 177 species, 27 varieties, 5 forms and 1 unidentified species. These river and stream had lower concentration of T-N and T-P compared to that of others; however, the site 2 of Galgok stream in summer was hypertrophic in T-P and also near hypertrophic in TN, and the site 1 in Incheon river during fall appeared to be hypertrophic in T-N, and the site 3 in summer showed near hypertrophic level in T-P. Determining the trophic status of the water quality based on chlorophyll a (chl-a), the site 3 of Galgok stream in spring and the site 3 of Incheon river in fall were oligotrophic; moreover, 6 sites including the site 2 and 3 of Galgok stream in summer were mesotrophic, and 9 sites including all sites of Galgok stream in fall turned out to be eutrophic. Particularly, the site 1 of Galgok stream in summer was hypertrophic, having 58.19 mg chl-a m$^{-3}$. There was a conspicuous difference between two values of standing crops based on chl-a and cell counting. This discrepancy may have occurred because of abundant cyanophytes and exclusion of picoplankton cells in cell counting. In the study, 5 cyanophytes, Synechocystis aquatilis, Microcystis aeruginosa, M. flos-aquae, Oscillatoria angustissima, O. limnetica and 2 diatoms, Thalassiosira bramaputrae and Navicula viridula var. rostellata were abundant. Based on the T-N, T-P values, standing crops and cell counting in Galgok stream and Incheon river were between mesotrophic and eutrophic conditions.

  • The effect of ginseng glycosides and their aglycones on the thermodynamic characteristics of membranes from dipalmitoylphosphatidylcholine (DPPC) was investigated. Total saponins (TS) from Korean red ginseng, Panax ginseng C.A. Meyer, interacted with the Eel Phase of lipid in the Polar region and did not penetrate the deeper glycerol backbone of lipid molecule. From the all investigated components of TS (aglycons and ginsenosides), only 20-(S)-panaxadiol (PD) had an effect similar to TS. High concentration of TS penetrated in hydrophobic Cl-C8 region. The presence of cholesterol did not influence the interaction of TS with DPPC. An elimination of transition, however, took place at 10~100 $\mu\textrm{g}$/ml of TS. DPPC had a low ability to interact with cholesterol (CHL) as compared with other lecithins except ethanolamine. From our results, only TS and PD, at high concentrations (100 mol%), influenced the phase transition of mixture of DPPC:CHL.

  • To evaluate the genetic toxicity of NP-77A which is selected as the candidate of anti-HBV agent, we performed ames test, micronucleus test, and chromosome aberration test on the CHL cell in vitro. The Ames test was carried out with 5 fold diluted 5 concentrations from 25mg/plate using S. typhimurium and E.coli. After 48hrs incubation, revertant colony numbers was calculated with and without metabolic activation system. In vivo micronucleus test, we investigated the rate of the occurrence of micronucleus after I.P. administration to mice. Andalso, we observed the incidence rate of cells with chromosomal aberration by NP-77A treatment using CHL cell line.

  • CKD-712, named S-YS49 is a chiral compound derived from higenamine (one component of Aconite spp.) derivatives. To compare the cytotoxicity of CKD-712 between in the absence and in the presence of S9 metabolic activation system, we performed try pan blue dye exclusion assay in Chinese hamster lung (CHL) cell. In CHL cells, the cytotoxicity ($IC_{50}$) of CKD-712 was 92.9 ${\m}g$/$m\ell$ and 186.1 ${\m}g$/$m\ell$ in the absence and presence of S9 metabolic activation, respectively. (omitted)

Protective Effect of Yellow-Green Vegetable Juices on DNA Damage in Chinese Hamster Lung Cell Using Comet Assay (Comet Assay를 이용한 케일, 명일엽, 당근, 돌미나리 녹즙의 Chinese Hamster Lung 세포 DNA 손상 보호 효과)

  • The present study was attempted to investigate the antioxidant capacity of popular yellow-green vegetable juices (kale, Angelica keishei, carrot, small water dropwort) and to investigate the effect of vegetable juices on protecting oxidative damage to DNA in cultured Chinese hamster lung (CHL) cells. Antioxidant capacity was analyzed by TRAP assay (Total radical-trapping antioxidant potential). Cellular DNA dmamage was measured by SCGE (single-cell gel electrophoresis, also known as comet assay. Cells incubated in medium with PBS (negative control) or with various concentration of the freeze dried green juices (25, 50, 100, 250 $\mu\textrm{g}$/$m\ell$) resuspended in PBS were treated with $H_2O_2$ (200 ${\mu}{\textrm}{m}$) as an oxidative stimulus for 5 min at 4$^{\circ}C$. The physiological function of each vegetable juice on oxidative DNA damage was analyzed and expressed as tail moment (tail length X percentage migrated DNA in tail) . Kale juice had the highest TRAP value suggesting that kale has the highest antioxidant capacity followed by Angelica keishei, small water dropwort and carrot. Cells treated with $H_2O_2$ had extensive DNA damage compared with cells treated with PBS or pre-treated with vegetable juice extracts. All green juices inhibited $H_2O_2$-induced DNA damage with kale being the most effective juice among the tested juices. These results indicate that green juice supplementation to CHL cells followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species. Investigation of AI-based dual-model strategy for monitoring cyanobacterial blooms from Sentinel-3 in Korean inland waters

  • The frequent occurrence of cyanobacterial harmful algal blooms (CHABs) in inland waters under climate change seriously damages the ecosystem and human health and is becoming a big problem in South Korea. Satellite remote sensing is suggested for effective monitoring CHABs at a larger scale of water bodies since the traditional method based on sparse in-situ networks is limited in space. However, utilizing a standalone variable of satellite reflectances in common CHABs dual-models, which relies on both chlorophyll-a (Chl-a) and phycocyanin or cyanobacteria cells (Cyano-cell), is not fully beneficial because their seasonal variation is highly impacted by surrounding meteorological and bio-environmental factors. Along with the development of Artificial Intelligence (AI), monitoring CHABs from space with analyzing the effects of environmental factors is accessible. This study aimed to investigate the potential application of AI in the dual-model strategy (Chl-a and Cyano-cell are output parameters) for monitoring seasonal dynamics of CHABs from satellites over Korean inland waters. The Sentinel-3 satellite was selected in this study due to the variety of spectral bands and its unique band (620 nm), which is sensitive to cyanobacteria. Via the AI-based feature selection, we analyzed the relationships between two output parameters and major parameters (satellite water-leaving reflectances at different spectral bands), together with auxiliary (meteorological and bio-environmental) parameters, to select the most important ones. Several AI models were then employed for modelling Chl-a and Cyano-cell concentration from those selected important parameters. Performance evaluation of the AI models and their comparison to traditional semi-analytical models were conducted to demonstrate whether AI models (using water-leaving reflectances and environmental variables) outperform traditional models (using water-leaving reflectances only) and which AI models are superior for monitoring CHABs from Sentinel-3 satellite over a Korean inland water body.

Genotoxicity Study of Sophoricoside, a Constituent of Sophora japonica, in Bacterial and Mammalian Cell System

  • Sophoricoside was isolated as the inhibitor of IL-5 bioactivity from Sophora japonica (Leguminosae). It has been reported to has an anti-inflammatory effect on rat paw edema model. To develope as an anti-allergic drug, genotoxicity of sophoricoside was investigated in bacterial and mammalian cell system such as Ames bacterial reversion test, chromosomal aberration assay and single cell gel electrophoresis (Comet) assay. As results, in the range of 1,250~40 $\mu\textrm{g}$/plate sophoricoside concentrations was not shown significant mutagenic effects in Salmonella typhimurium TA 98, TA 100, TA 1535 and TA 1537 strains in Ames test. The 80% cell growth inhibition concentration (IC/SUB 80/) of sophoricoside was determined as above 5,000 $\mu\textrm{g}$/$m\ell$ in Chinese hamster lung (CHL) fibroblast cell and L5178Y mouse lymphoma cell line for the chromosomal aberration and comet assay, respectively. Sophoricoside was not induced chromosomal aberration in CHL fibroblast cell at concentrations of 700, 350 and 175 $\mu\textrm{g}$/$m\ell$ or 600, 300 and 150 $\mu\textrm{g}$/$m\ell$ in the absence or presence of S-9 metabolic activation system, respectively. Also, in the comet assay, the induction of DNA damage was not observed in L5178Y mouse lymphoma cell line both in the absence or presence of S-9 metabolic activation system. From these results, no genotoxic effects of sophoricoside were observed in bacterial and mammalian cell systems used in these experiments.

  • This study was conducted to investigate the inhibitory effects of daidzein against H. pylori and its cholesterol $\alpha$-glucosyltransferase ($CHL{\alpha}GcT$). $CHL{\alpha}GcT$ is responsible for the production of $\alpha$-glucosyl cholesterol which constitutes more than 25% of cell wall lipids in H. pylori, and it has been suggested that it is essential for H. pylori viability. $CHL{\alpha}GcT$ was inhibited by daidzein, in a dose-dependant manner, of which $IC_{50}$ value was $128.5\;{\mu}M$. Daidzein also showed the inhibitory effect toward H. pylori growth by paper disc diffusion assay. Therefore, it is thought that the inhibition of daidzein on $CHL{\alpha}GcT$ was related to its anti-Helicobacter activity.
