Chlorsulfuron 과 Imazaquin에 대한 옥수수의 생리적 반응 (Physiological Responses of Corn Plants to Chlorsulfuron and Imazaquin)

  • Chlorslulfuron 및 imazaquin의 옥수수에 대한 저해양상의 차이 및 혼합효과를 생리학적인 측면에서 조사한 결과, 1. 수경재배시 배양액에 약제를 처리하였을 경우 CHL은 약제처리 6시간 후부터 생장저해 증상이 관측되었으나, IMA의 경우에는 36시간 이후에 관측되었고, 처리농도가 증가되면 생장저해증상의 발현 속도가 증대되었다. 2. 세포분열은 CHL의 경우 약제처리 3시간 후부터 저해되기 시작하였으나, IMA의 경우에는 세포분열의 저해율과 저해속도가 모두 낮았다. 3. CHL 및 IMA에 의한 세포신장에 대한 영향은 나타나지 않았다. 4_ 근부의 ALS에 대한 영향은 세포분열의 저해시간보다 빠른 CHL의 처리 l시간 이후부터 저해되었다. 5. CHL과 IMA을 경엽에 동시 혼합처리를 하면 생장저해효과가 상가적으로 나타났으나, 상호 순차적 체계처리는 상승적인 효과를 나타내었다. 6. ALS 에 대한 in vitro실험에서도 두 약제의 동시처리시 상가적인 효과가 발현되었다.

Rice 7-Hydroxymethyl Chlorophyll a Reductase Is Involved in the Promotion of Chlorophyll Degradation and Modulates Cell Death Signaling

  • The loss of green coloration via chlorophyll (Chl) degradation typically occurs during leaf senescence. To date, many Chl catabolic enzymes have been identified and shown to interact with light harvesting complex II to form a Chl degradation complex in senescing chloroplasts; this complex might metabolically channel phototoxic Chl catabolic intermediates to prevent oxidative damage to cells. The Chl catabolic enzyme 7-hydroxymethyl Chl a reductase (HCAR) converts 7-hydroxymethyl Chl a (7-HMC a) to Chl a. The rice (Oryza sativa) genome contains a single HCAR homolog (OsHCAR), but its exact role remains unknown. Here, we show that an oshcar knockout mutant exhibits persistent green leaves during both dark-induced and natural senescence, and accumulates 7-HMC a and pheophorbide a (Pheo a) in green leaf blades. Interestingly, both rice and Arabidopsis hcar mutants exhibit severe cell death at the vegetative stage; this cell death largely occurs in a light intensity-dependent manner. In addition, 7-HMC a treatment led to the generation of singlet oxygen ($^1O_2$) in Arabidopsis and rice protoplasts in the light. Under herbicide-induced oxidative stress conditions, leaf necrosis was more severe in hcar plants than in wild type, and HCAR-overexpressing plants were more tolerant to reactive oxygen species than wild type. Therefore, in addition to functioning in the conversion of 7-HMC a to Chl a in senescent leaves, HCAR may play a critical role in protecting plants from high light-induced damage by preventing the accumulation of 7-HMC a and Pheo a in developing and mature leaves at the vegetative stage.

  • It was reported that the digestion ratio of chlorella was low because it had a low metabolic rate in body. Generally, the thickness of a cell membrane of it is $200-250\;{\AA}$, the weight of it is approximatly 13% of the total weight of a dry cell. And it is composed of protein, lipid, hemicellulose and ash etc. So, in order to elevate the digestion ratio of chlorella in body, we experimented the crude treatment methods of chlorella. The results obtained in this experiment are summarized as follows : 1. The digestion ratios calculated from ordinary N- balance method were 83.05% for 10% chl. (b) plus diastase group ; 81.25% for 10% chl. (b) plus amylase group, and 79.23% for 10% chl. (b), 58.55% for 10% chl. (a). 2. Biological values from this method were 80.25% for 10% chl. (b) plus diastase group, and 60% for 10% for chlorella(a).

천수만 일차생산자의 생물량 -식물플랑크톤 탄소량과 세포개체수 및 클로로필과의 관계- (Biomass of Primary Producer in the Ch$\check{o}$nsu Bay -Relationships between Phytoplankton Carbon, Cell Number and chlorophyll-)

  • 일차생산자의 생물량을 연구하기 위해 천수만에서 1985년 9월부터 1986년 8월까지 월별로 채집된 식물플랑크톤의 체적으로부터 계산한 식물플랑크톤의 탄소량은 $26.7-960.7{\mu}gC/l$의 분포를 보이며 월 평균 탄소량은 $58.6-684.7{\mu}gC/l$(연 평균 $208.5{\mu}gC/l$) 범위의 변화를 보인다. 식물플랑크톤 탄소량과 세포 개체수 및 클로로필 농도의 상호 관계를 분석한 결과, net plankton(> $20{\mu}m$)에서는 탄소량, 개체수 및 클로로필 상호간의 상관 관계가 밀접하게 나타나 이들 상호 관계가 주로 net plankton에 의해 좌우됨을 보여주고 있으며, nanoplankton(< $20{\mu}m$)의 경우, 상관 관계가 낮게 나타났는데 이는 nano-fraction이 제거하지 않은 picoplankton(< $2{\mu}m$)을 상당량 포함하고 있기 때문으로 사료된다. 중회귀 분석 결과 또한 식물플랑크톤 탄소량, 개체수 및 클로로필 상관 관계가 주로 net plankton에 의해 좌우되고 있음을 나타낸다. C/Chl-a 비율은 9.1-100.5의 범위로 변화폭이 큰데 net plankton의 C/Chl-a 평균 비율은 111, nanoplankton의 비율은 6.4로 나타났다. Net plankton의 비율은 높은 시기에는 C/Chl-a값이 높은 반면 nanoplankton비율이 높은 시기에는 기 값이 낮게 나타나는 것으로 보아 C/Chl-a 비율을 변화시키는 주요인은 식물플랑크톤의 크기인 것으로 사료된다. 본 연구 결과 C/Chl-a값의 변화 폭이 크기 때문에 클로로필에 일정 비율을 적용하여 탄소량을 추정하는 방법은 연안생태계에서 적절치 못한 것으로 판단된다.

Evaluation of the Genetic Toxicity of Synthetic Chemicals (XIII) - Single Cell Gel Electrophoresis of Benzoyl Chloride, 2-Propyn-1-ol, and 2-Phenoxyethanol in Chinese Hamster lung Fibroblast -

  • Three synthetic chemicals, benzoyl chloride, 2-propyn-l-ol, and 2-phenoxy ethanol were selected for genotoxicity testing, based on production quantity and available genotoxic data. In our previous report, benzoyl chloride induced chromosomal aberrations in Chinese hamster lung (CHL) fibroblast in vitro with and without metabolic activation, while 2-propyn-l-ol and 2-phenoxy ethanol induced only with metabolic activation. To compare the genotoxicity of chromosome aberration assay, the single cell gel electrophoresis (comet) assay subjected using CHL cells. As a result, statistically significant differences of tail moment values of benzoyl chloride, 2-propyn-1-ol, and 2-phenoxy ethanol were observed compared with control values on almost all concentrations with S9 or without S9 metabolic activation system. This results suggest that genotoxic results of the comet assay and the chromosome aberration assay show correlationship of genotoxicity in the CHL fibroblast. In summary, the positive result of chromosome aberration of benzoyl chloride, 2-propyn-l-ol, and 2-phenoxy ethanol was also induced DNA damages in comet assay with same cell line. Consequently, comet assay will be useful and more accurate tool to detect and to confirm the genotoxicity especially DNA damages in CHL fibroblast.

Deposition Behavior and Photoelectrochemical Characteristics of Chlorophyll a Langmuir-Blodgett Films

  • The deposition behavior and photoelectric response characteristics of chlorophyll a(Chl a) monolayers and multilayers were investigated under various film fabrication conditions. Chl a LB films were deposited onto quartz and pretreated ITO glass substrates under several fabrication conditions, including surface pressure and number of layers. The absorption spectra of Chl a in a solution state and solid-like state (LB films) were fairly consistent with each other, and two absorption peaks were found at 678 and 438nm, respectively. The prepared Chl a LB films were set into an electrochemistry cell equipped with a Pt plate as the counter electrode, and the photoelectric response characteristics were obtained and analyzed relative to the light illumination. By considering the resulting photocurrents, the optimal fabrication conditions for Chl a LB films were determined as 20mN/m of surface pressure and 20 layers. The action spectrum of the Chl a LB films was obtained in the visible region, and was found to be in good agreement with the absorption spectrum. The possible application of the proposed system as a constituent of an artificial color recognition device was suggested based on combining with the photoelectric conversion property of another light-sensitive biological pigment.

초음파 장치의 주파수 변화에 따른 Microcystis aeruginosa의 성장억제 평가 (Evaluation of Growth Inhibition for Microcystis aeruginosa with Different Frequency of Ultrasonic Devices)

  • 초음파의 고주파 (1.6 MHz), 저주파 (23 kHz) 조사를 통해 Microcystis aeruginosa (M. aeruginosa)의 성장 억제 효과 (growth inhibition effect)를 대용량 (7.2 L) 조류 시료를 활용해 실험실 규모 (lab-scale) 실험을 통해 검증하였다. 6시간 고주파 조사 후 chl-a 농도와 M. aeruginosa 개체수는 점진적인 감소 경향이 관측된 반면, 6시간 저주파 조사 후 chl-a 농도 및 M. aeruginosa 개체수가 고주파 대비 급격하게 감소하는 것을 확인하였다. 또한, 초음파 조사기간 보다 조사종료 이후의 일차분해상수(k)가 큰 것으로 확인되었으며, 이러한 결과는 기존 연구에 비해 비교적 낮은 단위부피당 초음파 에너지가 조류 세포막과 내부 기관에 미치는 영향이 지연되어 일정 시간 이후 점진적인 조류 성장 억제 효과가 관측되는 것으로 사료된다. 초음파 조사 후 chl-a 농도와 M. aeruginosa 개체수 변화, 성장률과 일차분해율을 종합적으로 비교한 결과, 저주파에서 조류 성장 억제 효과가 우수한 것으로 확인되었으며, 저주파에서 M. aeruginosa 세포막으로 에너지 투과 효율이 우수해 기낭 등 내부기관에 더 큰 손상을 유도하기 때문인 것으로 사료된다. SEM과 TEM image 관측을 통해서도 고주파 보다 저주파에서의 M. aeruginosa의 세포 표면 및 세포막의 손상이 명확하게 관측되었다. 마지막으로 초음파에 의한 M. aeruginosa의 기낭 파괴 및 세포막의 기능 손상을 통해 용출되는 독성물질인 microcystin-LR의 수중 유출은 검출한계 (0.1 ㎍ L-1) 미만으로 용출되어 수생태계에 미치는 유해성은 미미한 것으로 판단된다.

Long-term Environmental Changes: Interpretations from a Marine Benthic Ecologist's Perspective (II) -Eutrophication and Substratum Properties

  • Chemical oxygen demand (COD), phytoplankton cell number and chlorophyll-a concentration (Chl-a), sediment mean grain size and ignition loss were studied to determine their temporal trends in the study area. Historical data of COD, cell number and Chl-a were gathered from the late 1960s or early 1980s to 1997, and trends in temporal domain were obtained from a simple regression. Sediments for grain size and ignition loss (as organic contents in sediments) were sampled from the Chokchon macrotidal flat bimonthly from September 1990 to November 1996, and were analyzed using the decomposition method of time series analysis. In general, the first three data showed increasing trends based on regression analysis. The trends of sediment grain size fluctuated in a neutral pathway while those of ignition loss yielded no increasing pattern. In contrast with the suggestions from Ahn and Choi (1998) who reported a coarsening variation in sediment grain size to be a cause of the directional and remarkable changes of macrofaunal communities in this area, we could not find such a corresponding variation pattern from our samples. In diagnosing eutrophication, a paradoxical phenomenon was encountered between the trends in water column (COD, cell number and Chl-a) and sediment (ignition loss) data. In this paper, we inferred the possible processes that produce the discrepancy. Some explanations and biological responses to eutrophication were predicted and discussed.

제주도 어승생 저수지 색도 원인 분석 (Analysis of chromaticity cause in Jeju Eoseungsaeng Lake)

  • Jeju Eoseungsaeng lake which is a main water resource of Eoseungsaeng water treatment plant($Q=15,000m^3/d$) experienced high chromaticity(40 CU) and pH(9.46) in 2013. This could decline customer's confidence on drinking water quality unless proper identification and removal of chromaticity were implemented. To find cause of chromaticity, water monitoring on various water parameters including TOC, algal cell count, Chl-a, turbidity, SS, conductivity, etc. were implemented. Iron and manganese were excluded from the cause of chromaticity due to its low concentration (i.e., < 0.02 mg/L). Correlation among water parameters showed that relationship between algal cell count and chromaticity was the highest(R=0.43), which suggested that presumably the main reason of chromaticity occurrence in Jeju Eoseungsaeng lake was algae.

황기와 Chlorambucil 병용(倂用)이 P388D1 백혈병(白血病) 세포(細胞) 및 담암(擔癌)생쥐에 미치는 영향(影響) (Combined application of Astragali Radix and chlorambucil on anticancerous effect and host safety in Lukemia Cell(P388D1 cell) and its bearing mice)

  • Objective : Anticancerous and host safety effects of the combination of Astragali Radix and chlorambucil are studied when it is applied to leukemia cell(P388D1 cell) related disease. Methods : After 5 groups of mice are treated by respective procedures (HG, CHL, HG+CHL, etc), the quantitative analyses (cell proliferation assay, mutagenesis test, survival rate, weight shift observation, tissue and blood analyses, etc) are conducted. Results : While sole injection of Astragali Radix extends the survival period and deter the liver and marrow function deterioration, combined injection (HG+CHL) shows more strong anticancerous effect than sole injection of Astragali Radix or Cholrambucil, and also does not cause any mutation on the normal cells and virulence on marrow and liver caused by leukemia. Conclusions : The combined application of Astragali Radix and chlorambucil has more effective anticancerous and host safety effects than independent use of them.
