CRISPR/Cas9-mediated knockout of CD47 causes hemolytic anemia with splenomegaly in C57BL/6 mice

  • CD47 (integrin-associated protein), a multi-spanning transmembrane protein expressed in all cells including red blood cells (RBCs) and leukocytes, interacts with signal regulatory protein ${\alpha}$ ($SIRP{\alpha}$) on macrophages and thereby inhibits phagocytosis of RBCs. Recently, we generated a novel C57BL/6J CD47 knockout ($CD47^{-/-}$ hereafter) mouse line by employing a CRISPR/Cas9 system at Center for Mouse Models of Human Disease, and here report their hematological phenotypes. On monitoring their birth and development, $CD47^{-/-}$ mice were born viable with a natural male-to-female sex ratio and normally developed from birth through puberty to adulthood without noticeable changes in growth, food/water intake compared to their age and sex-matched wild-type littermates up to 26 weeks. Hematological analysis revealed a mild but significant reduction of RBC counts and hemoglobin in 16 week-old male $CD47^{-/-}$ mice which were aggravated at the age of 26 weeks with increased reticulocyte counts and mean corpuscular volume (MCV), suggesting hemolytic anemia. Interestingly, anemia in female $CD47^{-/-}$ mice became evident at 26 weeks, but splenomegaly was identified in both genders of $CD47^{-/-}$ mice from the age of 16 weeks, consistent with development of hemolytic anemia. Additionally, helper and cytotoxic T cell populations were considerably reduced in the spleen, but not in thymus, of $CD47^{-/-}$ mice, suggesting a crucial role of CD47 in proliferation of T cells. Collectively, these findings indicate that our $CD47^{-/-}$ mice have progressive hemolytic anemia and splenic depletion of mature T cell populations and therefore may be useful as an in vivo model to study the function of CD47.

  • CD1 molecules belong to non-polymorphic MHC class I-like proteins and present lipid antigens to T cells. Five different CD1 genes (CD1a-e) have been identified and classified into two groups. Group 1 include CD1a-c and present pathogenic lipid antigens to ${\alpha}{\beta}$ T cells reminiscence of peptide antigen presentation by MHC-I molecules. CD1d is the only member of Group 2 and presents foreign and self lipid antigens to a specialized subset of ${\alpha}{\beta}$ T cells, NKT cells. NKT cells are involved in diverse immune responses through prompt and massive production of cytokines. CD1d-dependent NKT cells are categorized upon the usage of their T cell receptors. A major subtype of NKT cells (type I) is invariant NKT cells which utilize invariant $V{\alpha}14-J{\alpha}18$ TCR alpha chain in mouse. The remaining NKT cells (type II) utilize diverse TCR alpha chains. Engineered CD1d molecules with modified intracellular trafficking produce either type I or type II NKT cell-defects suggesting the lipid antigens for each subtypes of NKT cells are processed/generated in different intracellular compartments. Since the usage of TCR by a T cell is the result of antigen-driven selection, the intracellular metabolic pathways of lipid antigen are a key in forming the functional NKT cell repertoire.

Adsorption Characteristics of the Heavy Metals, Cd(II) and Pb(II) Ions, on the Si-immobilized Sargassum horneri (실리카고정 괭생이모자반에 대한 중금속 Cd(II), Pb(II) 이온의 흡착 특성)

  • Si-immobilized Sargassum horneri was used to study the adsorption characteristics along with maximum adsorption conditions of heavy metal ions, Cd(II) and Ph(II) on the Si-immobilized Sargassum horneri. More amount of Cd(II) and Pb(II) ions on the Si-immobilized Sargassum horneri than Sargassum horneri were adsorbed. And Pb(II) ions were more adsorbed in all algae than Cd(II) ions more effectively in alkaline than in acidity. Recovery ratios of Cd(II) and Pb(II) ions on the Sargassum horneri were 58.0-62.6%, 61.2-64.4% respectively, Si-immobilized Sargassum horneri 56.8-92.7%, 37.8-47.9%. Recovery ratio of Cd(II) ion was higher on the Si-immobilized Sargassum horneri but it of Pb(II) ion was lower on the Si-immobilized Sargassum horneri.

Improvement of cadmium tolerance and accumulation of Phragmites spp. Tabarka by ethyl methane sulfonate mutagenesis

  • Reed (Phragmites spp.) is a rhizomatous plant of the Poaceae family and is known as high tolerant plant to heavy metal contaminants. This plant is widely recognized as a Cd root-accumulator, but improved heavy metal tolerance and uptake capacity are still required for phytoremediation efficiency. To enhance capacity of hyperaccumulator plants, ethyl methane sulfonate (EMS) as chemical mutagen has been introduced and applied to remediation approaches. This study aimed to select EMS-mutagenized reeds representing high Cd resistance and large biomass and to investigate their ability of Cd accumulation. After 6 months cultivation of M2 mutant reeds under Cd stress conditions (up to 1,500 µM), we discovered seven mutant individuals that showed good performances like survivorship, vitality, and high accumulation of Cd, particularly in their roots. Compared to wild type (WT) reeds as control, on average, dry weight of mutant type (MT) reeds was larger by 2 and 1.5 times in roots and shoots, respectively. In addition, these mutant plants accumulated 6 times more Cd, mostly in the roots. In particular, MT8 reeds showed the greatest ability to accumulate Cd. These results suggest that EMS mutagenesis could generate hyperaccumulator plants with enhanced Cd tolerance and biomass, thereby contributing to improvement of phytoremediation efficiency in Cd-contaminated soil or wastewater. Further studies should focus on identifying Cd tolerance mechanisms of such EMS-mutagenized plants, developing techniques for its biomass production, and investigating the practical potential of the EMS mutants for phytoremediation.

  • The contents of Pb and CD measured with the barks of Pinus densiflora in Seoul area, Yangjaedong area near the Kyungbu-highway, Mt. Keryong and Mt. Seolak area, from Sep. to Ock. in 1988. Pb contents in Seoul area were 17.8 ppm at Changkyungkung, 17.1 ppm at Chongamdong, 13.2 ppm at Mt. Kwanak, 7.6 ppm at Mt. Dobong and 3.7 ppm at Mt. Surak. Except Mt. Kwanak, the difference between areas of central and mountains in Seoul were high significant. Mt Seolak area also showed variations in Pb contents from 16.8 ppm to 32.1 ppm according to studied sites. Cd contents in Seoul area were 0.47 ppm at Changkyungkung, 0.65 ppm at Chongamdong, 0.61 ppm at Mt. Kwanak, 0.52 ppm at Mt. Dobong, 0.47 ppm at Mt. Surak. Therefore, Cd contents in Seoul area exhibited more variations between sampling plots than between the areas of central and mountains in Seoul. Mt Seolak area also showed variations from 0.20 ppm to 0.34 ppm in Cd contents, and it was the lowest in all the studied area.

  • Compared with organic fluorophores, semiconductor quantum dots (QDs) have the better properties such as photostability, narrow emission spectra coupled to tunable photoluminescent emissions and exceptional resistance to both photo bleaching and chemical degradation. In this work, CdSe/ZnS QDs nanobeads were prepared by the incorporation of CdSe/ZnS QDs with mesoporous silica to use as the optical probe for detecting toxic and bio- materials with high sensitivity, CdSe/ZnS core/shell QDs were synthesized from the precursors such as CdO and zinc stearate with the lower toxicity than pyrotic precursors. The QD-nanobeads were characterized by transmission electron microscopy, FL microscopy, UV-Vis and PL spectroscopy, respectively.

Korean Red Ginseng modulates immune function by upregulating CD4+CD8+ T cells and NK cell activities on porcine

  • In the present study, we investigated whether treatment with KRG improve the parameters of immune activity such as the cytotoxicity, populations of CD4+ CD8+T cell, CD3-CD172-CD8+ NK cell and CD172+ monocyte as well as natural cytotoxicity receptors such as Nkp46, Nkp44, Nkp30. In results, KRG significantly increased these immune activities. These results indicate that KRG has distinct immuneenhancing effects by increasing the roles of T cells and NK cell in porcine.

Cadmium Accumulation and Tolerance of Iris pseudacorus and Acorus calamus as Aquatic Plants Native to Korea (자생 수생식물 노랑꽃창포와 창포의 카드뮴 축적 및 내성)

  • This study was conducted to find out the cadmium (Cd) accumulation and tolerance of Iris pseudacorus and Acorus calamus as aquatic plants native to Korea for Cd removal in water. In the range of Cd concentration from $10{\mu}M$ to $130{\mu}M$, the Cd lethal dose 50 ($LD_{50}$) was $78.5{\mu}M$ in I. pseudacorus and $47.6{\mu}M$ in A. calamus. In I. pseudacorus, superoxide dismutase and peroxidase as antioxidants were relatively effective against oxidative stress caused by Cd, while catalase, superoxide dismutase, and polyphenolics were effective in A. calamus. The polyphenolics known as typical antioxidants were not detected in I. pseudacorus. In both species, the Cd accumulation in plants increased with the higher Cd concentration and the longer processing period. Also, the absorbed Cd was accumulated mainly in the roots. The amount of Cd accumulated in the shoot part was maximally $548.1mg{\cdot}kg^{-1}$ (82.1% to Cd accumulated in the root part) in I. pseudacorus and $121.4mg{\cdot}kg^{-1}$ (13.7%) in A. calamus, which implied that both species all were enough evaluated as Cd hyper-accumulators based on 0.01% or more Cd accumulation in the shoot. Especially I. pseudacorus showed outstanding ability to move well Cd into the shoots from the roots and high tolerance to Cd stress.

Tertiary Structure of PreSl(21-47) of Hepatitis B Virus Studied by NMR Spectroscopy

  • To design more efficient peptide antagonist against the HBV, preSl(21-47) which carries the HBV receptor binding site for hepatocytes was synthesized and the solution structure of preSl(21-47) was investigated using CD spectroscopy and NMR spectroscopy in membrane-mimicking environments. According to CD spectra, preSl(21-47) has a random structure in aqueous solution, while conformational change was induced by addition of TFE and SDS micelle. Tertiary structures as determined by NMR spectroscopy shows that preSl(21-47) has a very flexible structure even in SDS micelle.

카드뮴 중독자 검색을 위한 메탈로치오네인 분석 - 동물실험을 중심으로 -

  • The purpose of this study was to investigate the metallothionein of acute cadmium poisoning mice as a Cadmium index. Forty male ICR mice were injected with cadmium chloride solution from 1/8LD$\sub$50/ to 1/2 LD$\sub$50/ dose. At 24 hours after exposed Cd, I examined Cd and metallothionein (MT) in tissues (liver and kidney) and fluids (whole blood and urine) and also measured low molecular proteins, N-acety1-${\beta}$-D-glucosaminidase (NAG) and ${\beta}$$\sub$2/- microglobuline (${\beta}$$\sub$2 /-MG) in urine. The concentration of Cd and MT of liver, kidney whole blood and urine were increased with dose dependent manner. Urinary Cd and urinary MT had very good significance (p<0.01) and urinary MT had good significance with kidney Cd and NAG but not ${\beta}$$\sub$2/-MG. Conclusionally MT in urine was very correlated with kidney Cd and urine Cd. So MT maybe useful as a Cd poisoning index.
