Removal of Heavy Metals from Aqueous Solution by a Column Packed with Peat-Humin (Peat-Humin 충전 칼럼을 이용한 수용액 중의 중금속 제거)

  • Peat humin(p-Humin) extracted from Canadian Sphagnum peat moss was packed in a column and removal of heavy metal ions such as Cd, Cu and Pb from aqueous solution under flow conditions was studied. The metal ions were removed not only from single-element solutions but also from a multi-metal solution. Column kinetics for metal removal were described by the Thomas model. For single-component metal solutions, the maximum adsorption capacities of the p-Humin for Pb, Cu and Cd were 138.8, 44.66 and 41.61 mg/g, respectively. The results of multi-component competitive adsorption showed that adsorption affinity was in the order of Pb $\gg$ Cu > Cd. The adsorbed metal ions were easily deserted from the p-Humin with 0.05 N $HNO_3$ solution. It is apparent that 95% of the heavy metal ions were recovered from the saturated column. This investigation provides possibility to clean up heavy-metal contaminated waste waters by using the natural biomass, p-Humin as an environmentally friendly and cost-effective new biosorbents.

Characterization of the Nature of Cd(II) Ion Binding Sites on Poly(acrylic acid) Using$^{113}Cd$NMR (카드뮴-113 NMR을 이용한 폴리아크릴산의 카드뮴(Ⅱ) 이온에 대한 결합자리의 특성연구)

  • The cadmium(Ⅱ)-carboxylate complexes of monocarboxylates and poly(acrylic acid) (PAA) in an aqueous solution were investigated using 113Cd NMR spectroscopy. From these spectral data the chemical shifts of the monocarboxylate (e.g. acetate, benzoate and propanoate) complexes of Cd(Ⅱ) were evaluated (CdL+: -22 to -24 ppm range; CdL2: -39 to -40 ppm range; L: carboxylate). The chemical shift of cadmium(Ⅱ) bound on PAA changed in value from -36.6 to -38.2 ppm when the [PAA]/[Cd] ratios were varied from 12 to 118 eq/mol at a constant pH of 6. The 113Cd chemical shift was sensitive to the change in solution pH: the chemical shift changed from 1.6 to -37.6 ppm when the pH was rasied from 1 to 6 at a constant [PAA]/[Cd] ratio of 39 eq/mol. These results show that under these conditions, the dominant species formed in solution is 1 : 2 complex (CdL2), and 1 : 1 complex (CdL+) is only formed at low pH, when polyanion concentration is low.

  • Analyses for 6 metals (Cd, Zn, Cu, Ni, Pb, Mn) by Atomic absorption spectrophotometer were made on hair samples of healthy 135 in urban area and 130 in rural area who have not dealt with the above metals in their daily working life. Marksd variations were found. Samples of urban area contained more cadmium, copper, lead and manganese than those of rural area. No sexual difference in mean value could be observed and the amount of metals in hair did not increase with age. And no age dependency was found. There was a statistically significant correlation between the concentrations of cadmium and lead in both urban and rural people; Pb = 1.50 Cd + 6.69 (r = 0.213, p<0.05) in urban area Pb = 1.44 Cd + 3.67 (r = 0.327, p<0.001) in rural area

Effects of Danggwisayeokgaohsuyusaenggang-tang(當歸四逆加吳茱萸生薑湯) on Collagen-induced Arthritis in Mice (Collagen으로 유발된 생쥐의 관절염에 대한 당귀사역가오수유생강탕(當歸四逆加吳茱萸生薑湯)의 효과)

  • Objectives : This study was carried out to know the effects of Danggwisayeokgaohsuyusaenggang-tang(hereinafter referred to DST) on arthritis induced by collagen on DBA/1 OlaHsd mice. Methods : For this purpose, DST was orally administered to mouse with arthritis induced by collagen II. Cytotoxicity, high performance liquid chromatograph(HPLC) analysis, arthritis index, value of immunocyte in draining lymph node and paw joint, cytokine were measured in vivo. Results : 1. The cytotoxicity against human fibroblast cells(hFCs) was not measured in any concentration. 2. In HPLC analysis, There are high peak patterns at 8 minute(min), 12 min, 35 min, 45 min. 3. The arthritis index was decreased significantly. 4. The degree of arthritis induced damage of joint of DST group is slight compared with control group in histopathologic observation(Hematoxylin and eosin stain(H&E), Masson's trichrome(M-T) staining). 5. In total cell counts of draining lymph node(DLN) and paw joint, the cells in DLN decreased significantly on DST 200 mg/kg and the cells in paw joint decreased significantly on 200 mg/kg and 50 mg/kg. 6. In DLN, $CD4^+/CD25^+$, $CD3^+/CD69^+$, major histocompatibility complex(MHC), class-II/$CD11c^+$ cells decreased significantly on DST 200 mg/kg and 50 mg/kg $CD3^+/CD8^+$ cells decreased significantly on DST 200 200 mg/kg, $CD4^+$, $CD3^+/CD44^+$ cells decreased. 7. In paw joints, $CD4^+$, $CD11b^+/Gr-1^+$ cells decreased significantly on DST 200 mg/kg and 50 mg/kg. 8. In joints, levels of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, cyclo-oxygenase-2(COX-2), NOS-II were decreased on DST 200 mg/kg and DST 50 mg/kg. 9. In analysing of cytokine in CD3/CD28 activated spleen, IL-17 was decreased significantly, IL-4 was increased significantly $INF-{\gamma}$ was decreased on DST 200 mg/kg. 10. In analysing of cytokine in collagen activated spleen, IL-17 were decreased significantly, IL-4 was increased significantly. Conclusions : This results demonstrated that DST suppressed the inflammatory progression of collagen-induced arthritis(CIA) mice and supported further studies are required to survey continuously in looking for the effective substance and mechanism in the future.

Human Immunodeficiency Virus-l Tat Positively Regulates the Human CD99 Gene via DNA Demethylation (Human Immunodeficiency Virus-1 Tat 단백에 의한 인간 CD99유전자의 조절기전에 대한 연구)

  • HIV affects many organ systems. Patients with HIV infection have substantially increased risk of developing various cancers, primarily by opportunistic infection with oncogenic viruses due to their immunocompromised status. However, extensive evidence also indicates that the viral protein, Tat itself, may playas a major factor in the development of AIDS-related neoplasms. The molecular mechanism underlying Tat's oncogenic activity may include deregulation of cellular genes. Therefore, in this study, we examined the effect of HIV-l Tat on CD99 as one of the target cellular genes, which is a well-known tumor marker in several cancers. By using established HeLa clones that are stably expressing Tat, we found that CD99 is upregulated by endogenous Tat, whereas STAT3 is down regulated. Upon the screening of genes differentially expressed between Tat-stable cells and the control cells by using the gene fishing technique, DEG, we detected 3 genes which expression is affected by the presence of Tat. Furthermore, the methylation specific PCR analysis of the stably Tat expressing cell lines revealed that the CD99 promoter is de methylated in the presence of Tat. Taken together, these results open a potential role of CD99 in AIDS-related oncogenesis via epigenetic regulation by HIV-1 Tat.

Studies on the Immunomodulatory Effect Using Combinations of Several Kinds of Anti-cancer Prescriptions (수종 항암 처방의 조합에 따른 면역조절 작용에 관한 연구)

  • In order to understand the anti-carcinogenic effects of Boo-jung-bae-bon-bang(扶正培本方)-B1), Hwal-hyul-hwa-eo-bang(活血化瘀方-B2), Cheong-youl-hae-dok-bang(淸熱解毒方-B3), prescriptions of individual B1, B2, and B3 treatments or combined treatments (B4; B1+B2, B5; B1+B3, B6; B1+B2+83, B7; B2+83) were applied to cultured cancer cell lines. The major findings on their anti-cardnogenic effects in terms of mechanism and synergistic interactions are summarized below. Results of cytokine gene expression analyses are summarized as follows; IL-2 gene expression was increased by B1 and B6 treatments, IFN-v gene expression increased by B3, B1, B6, and 85, and CD28 gene expression increased by B1 and B5. IL-4 expression was not affected by any treatments. In the FACS analysis, increases in numbers of immunoreactive CD3/sup +//CD25/sup +/ T cells by B1 and B5 treatment, CD3/sup +//CD69/sup +/ T cells by B1, B3, B5, and B6 treatments, CD19/sup +//CD44/sup +/ B cells by B1 and B6 treatments were observed when compared to the corresponding non-treated control groups.

Effects of Age and Gender on the Viability and Stem Cell Markers, mRNA, and Protein Expression of Bone Marrow-Derived Stem Cells Cultured in Growth Media

  • Purpose: Bone marrow has long been a source of primary cells. This study was performed to evaluate the effects of age and sex on the cellular viability and expression of stem cell markers of mRNA and on the protein expression of bone marrow stem cells (BMSCs) derived from healthy donors. Materials and Methods: Stem cells were isolated from human bone marrow and plated in culture plates. The shape of the BMSCs was observed under inverted microscope. Quantitative cellular viability was evaluated using a Cell-Counting Kit-8 assay. The expression of stem cell surface markers was tested and a series of quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot was performed to evaluate the expression in each group. Result: The shapes of the cells at 20s, 30s, and 50s were similar to each other. No significant changes in cellular viability were noted among different age groups or sex groups. The BMSCs expressed CD44, CD73, and CD90 surface markers but did not express CD14 and CD34. There were no noticeable differences in CD surface markers among the different age groups. The expressions of CD surface markers were similar between men and women. No significant differences in the secretion of vascular endothelial growth factors (VEGFs) were noted at Day 3 between different age groups. qRT-PCR regarding the expression showed differences between the age groups. However, Western blot analysis showed a decrease in expression but did not reach statistical significance (P>0.05). Conclusion: This study clearly showed no significant differences in shape, cell viability, expression of stem cell surface markers, or secretion of human VEGF among different age groups. However, western blot analysis showed a tendency of age-related decrease which did not reach statistical significance. Collectively, autologous or allogeneic BMSCs should be meticulously applied to obtain optimal results regarding age and sex.

Production and Breeding of Transgenic Cloned Pigs Expressing Human CD73

  • One of the reasons to causing blood coagulation in the tissue of xenografted organs was known to incompatibility of the blood coagulation and anti-coagulation regulatory system between TG pigs and primates. Thus, overexpression of human CD73 (hCD73) in the pig endothelial cells is considered as a method to reduce coagulopathy after pig-to-non-human-primate xenotransplantation. This study was performed to produce and breed transgenic pigs expressing hCD73 for the studies immune rejection responses and could provide a successful application of xenotransplantation. The transgenic cells were constructed an hCD73 expression vector under control porcine Icam2 promoter (pIcam2-hCD73) and established donor cell lines expressing hCD73. The numbers of transferred reconstructed embryos were $127{\pm}18.9$. The pregnancy and delivery rate of surrogates were 8/18 (44%) and 3/18 (16%). The total number of delivered cloned pigs were 10 (2 alive, 7 mummy, and 1 died after birth). Among them, three live hCD73-pigs were successfully delivered by Caesarean section, but one was dead after birth. The two hCD73 TG cloned pigs had normal reproductive ability. They mated with wild type (WT) MGH (Massachusetts General Hospital) female sows and produced totally 16 piglets. Among them, 5 piglets were identified as hCD73 TG pigs. In conclusion, we successfully generated the hCD73 transgenic cloned pigs and produced their litters by natural mating. It can be possible to use a mate for the production of multiple transgenic pigs such as ${\alpha}-1,3-galactosyltransferase$ knock-out /hCD46 for xenotransplantation.

Assessment of Surface Water Quality in Suburban Golf Courses in Korea

  • In the current study, we examined the quality of surface water in ponds at two golf courses, located in southern (Country Club P) and eastern (Country Club B) areas of Korea respectively. Seasonal measurements were made of following physical parameters;pH, dissolved oxygen, biological oxygen demand, chloride, alkalinity, hardness, and nitrogen compounds [$NH_3$-N, $NO_2$-N, $NO_3$-N] and the concentrations of four heavy metals (Cd, Cu, Pb, and Zn) throughout the courses. The pH values were within the alkaline range (7.3 to 9.0), and the biological oxygen demand was generally between 0.5 and 3.1 mg/L. The alkalinity and hardness ranged from 19.1 to 68.5mg/L and 16.1 to 63.6 mg/L, respectively. Nitrogen as ammonium $(NH_3-N)$ was detected in all samples, and the samples had low concentrations of $NO_2-N$ and $NO_3-N$. Cd concentrations were relatively high (to 22.44 mg/L); the highest Cd concentration was observed in the pond in Country Club P. Other metal concentrations were low compared to the Quebec guidelines.

Evaluation of Cd and Pb Intake and Exposure Routes in Some Korean Women (우리나라 일부 여성의 카드뮴과 납 섭취량 및 노출경로 평가)

  • The publications on dietary intake, respiratory intake and blood concentration of cadmium and lead in some Korean women are reviewed. Reported values of dietary Cd intake as geometric mean were $17.1{\mu}g/day$ in 1986, $21.2{\mu}g/day$ in 1994, $16.7{\mu}g/day$ in 2000, and blood concentration were $1.45{\mu}g$ in 1986, $1.27{\mu}g/l$ in 1994 and $2.74{\mu}g/l$ in 2000, respectively. In case of Pb, the dietary intake were $33.1{\mu}g/day$ in 1986, $20.5{\mu}g/day$ in 1994, and $18.4{\mu}g/day$ in 2000 and the blood concentration were $51.5{\mu}g/l$ in 1986, $44.3{\mu}g/l$ in 1994, $37.3{\mu}g/l$ in 2000, respectively. Dietary intake is an almost exclusive route of Cd exposure, however respiratory intake in case of Pb is thought to affective exposure route in Korean women. When compared with the values reported in the literature, both of dietary Cd and Pb intake levels appear to be similar to or somewhat higher than the levels in east and south-east Asia.
