마우스 모델을 이용한 사이프러스 오일의 알러지성 천식 억제 효과 (Anti-asthmatic activities of Cypress oil in a mouse model of allergic asthma)

  • 본 연구는 천식 생쥐 모델을 이용하여 사이프러스 오일(CS)이 알러지성 천식에 미치는 영향을 평가하기 위하여 수행되었다. Ovalbumin으로 천식을 유발시킨 천식 생쥐 모델을 사용하여 실험군에 0.3% CS를 3 주간, 1주 3회, 1회 30분간 분무기를 이용하여 흡입시켰다. 기도과민성, 백혈구 중 호산구 수, 폐 세포 내 면역세포와 Th2 싸이토카인의 변화를 관찰한 결과 CS를 처리한 실험군에서 기도과민성, 호산구 수 및 폐 세포 내 IL-5,IL-13 수치, 혈청내 IgE 분비량, 폐 세포 내 CCR3, CD3, CD4 세포의 수 등 모든 항목에서 현저한 감소효과를 보였다. 이러한 결과는 CS가 천식 반응의 주요인자인 Th2 싸이토카인과 호산구에 긍정적인 영향력이 있음을 시사한다. 따라서 CS는 천식 치료제로서 가능성이 있을 것으로 사료된다.

가미청심연자탕(加味淸心蓮子場)이 NC/Nga mice의 아토피양(樣) 피부염에 미치는 영향 (Effect of Kami-chungsimyeunjatang on atopic dermatitis-like skin lesions induced in NC/Nga mice by mite antigen stimulation)

  • Objectives : The purpose of this study is to examine of the effect of Kami-chungsimyeunjatang(KCSYJT) medicine on the atopy eruption control. Methods : The expression of IgE, IL-4, IL-6, $TNF-{\alpha}$, IgG2b, IgM, IgG2a and IgG1 level in serum, and $IFN-{\gamma}$ production by KCSYJT were analyzed. CD3e+/CD69+, CD4+/CD25+, B220+/IgE+ and B220+/CD23+ positive cells by flow cytometry in splenocytes were assayed and the revelation of CD3e+/CD69+, CD4+/CD8+ and CD4+/CD25+ marker in PBMC, spleen and DLN were observed. The outturn of IL-4, eotaxin 2, CCR3, TARC mRNA in splenocytes werw observed. We also analyzed NC/Nga mice's ear, DLN and neck-back skin after biopy and dye by H&E, and toluidine staining (mast cells marker) method, measured about epidermis and dermis part in comparison with control group. Results : NC/Nga mice suffered from dermatitis very similar to human AD with IgE hyperproduction. Specially, result that measure IgE content in serum on 8 weeks, 12 weeks, 16 weeks decreased remarkably than control group. After experiment end, result that observe revelation CD3e+/CD69+, CD4+/CD8+ and CD4+/CD25+ marker in PBMC, spleen and DLN establishment observed recover as normal with political background. And decreased than result control group which measure IL-4, IL-6, $TNF-{\alpha}$, IgG2b, IgM, IgG2a, IgG1 level in serum, and $IFN-{\gamma}$ production secreted in Th1 cell displayed increase by KCSYJT medicines. Ear thickness decrease than control group in result that observe effect that get in ear of a NC/Nga mouse. Course inflammation immunocyte etc.. permeated of result that effect that KCSYJT medicines get to NC/Nga mouse's skin establishment analyzes ear, DLN and neck-back skin after biopy, and dye by H&E, and toluidine staining (mast cells marker) method decreased about epidermis. and inflammation of dermis part remarkably than control group. Immunohistochemical examination of the skin lesion showed decrease by KCSYJT medicines on numbers of mast cells (CCR3) and CD4+ T cells containing IL-4 necessary for IgE. Conclusions : Th1 cell and Th2 cell was observed to be shift by secretion amount of IL-4 and $IFN-{\gamma}$ by KCSYJT medicines. Therefore, the KCSYJT medicine turned out to be useful in allergy autoimmune disease.

  • Objectives : The aim of this study was to investigate the effect of Asthma-depression and Immunoregulation with PR-HAS(Herbal-acupuncture with Platycodi Radix infusion solution) injection at Joksamni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). The experimental group(OVA-PR-HA) wase treated with concentrations(1%) of PR-HAS at Joksamni(St36) for the later 8 weeks(3times/week). The second experimental group(OVA-Needle prick) was treated with Needle-Prick at Joksamni(St36) for the later 8 weeks(3times/week). Results : 1. The weight and total cells in the mice lung treated with PR-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with PR-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with PR-HA decreased significantly compared with those of control group. 4. The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. 5. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5,IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. Conclusion : The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. These result suggests that Platycodi Radix Herbal-acupuncture at Joksamni(St36) in C57BL/6mice may be an effictive part to OVA-induced asthma in C57BL/6 mice.

Materials and Methods : Atopic dermatitis (AD) usually develops in patients with an individual or family history of allergic diseases, and is characterized by chronic relapsing inflammation seen specially in childhood, association with IgE hyperproduction and precipitation by environmental factors. However, the exact etiology of AD has been unclear. To further explore the pathogenesis and treatment of AD, a suitable animal model is required. We found that skin lesions, which were chnically and histologically very simlar to human AD, mite antigen-induced dermatitis on the face, neck, ears and dorsal skin of inbred NC/Nga mice. Results and Conclusion : Kami-KangHwalSan medicines controlled CD3+/CD69+, CD4+/CD25+, B220+/IgE+, and B220+/CD23+ revelation that an experiment that motive allergy immune reponse because an in vitro experiment stimulates splenocytes of a NC/Nga mouse same t1me by PWM, and interleukin-4, eotaxin 2, CCR3, TARC mRNA outturn that bear in splenocytes decreased remarkably by Kami-KangHwalSan medicines. Th1 cell and Th2 cell observe to be shifted by secretion amount of IL-4 and IFN-$\gamma$ by Kami-KangHwalSan medicines could know that Kami-KangHwalSan medicines can use usefully in allergy autoimmnune diease.

  • Objectives : The aim of this study was to investigate the asthma-suppressive and immuno-regulatory effect of NR-HA(Notopterygii Rhizoma Herbal-acupuncture) at Pyesu(BL13) on OVA(ovalbumin)-induced asthma in mice. Methods : C57BL/6 mice out of all the experimental sloops, except the Normal group and the NR-HA group, were sensitized and challenged with OVA. The mice in the NR-HA group and the OVA-NR-HA group were treated with NR-HA(1%) at Pyesu(BL13). The mice in the OVA-Saline group were injected with saline at Pyesu(BL13). The mice in the OVA-Needle-prick group were treated with a single prick with an injection needle at Pyesu(BL13). NR-HA, saline injection and needle prick were administered for 8 weeks, three times a week Results : in vitro 1. The populations of granulocytes, $CD3e^-/CCR3^+$cells, $CD69^+/CD3e^+$ cells, $CD4^+\;cells\;and\;CD23^+/B220^+$ cells in the OVA-induced asthmatic mouse lungs decreased significantly by NR-HAS(Notopterygii Rhizoma Herbal-acupuncture solution). 2. The lung weight and total cells in lung of the OVA-NR-HA group decreased significantly compared with those of the OVA-Control group. 3. Total leukocytes and eosinophils in BALF of the OVA-NR-HA group decreas ed significantly compared with those of the OVA-Control group. 4. The collagen accumulation in the lung sections of the OVA-NR-HA group decreased significantly compared with that of the OVA-control group. 5. The concentrations of IL-4, IL-5, IL-13, IgE in BALF and serum of the OVA-NR-HA group decreased significantly compared with those of the OVA- Control group. 6. The numbers of $Gr-1^+/CD11b^+,\;CCR3^+,\;CD3e^+, \;CD19^+,\;CD3e^+/CD69^+$ cells in the OVA-NR-HA group decreased significantly compared with those of the OVA-Control group. 7. The mRNA expressions of $TNF-{\alpha}$, IL-5, IL-4 and IL-13 in lung of the OVA-NR-HA group decreased significantly compared with those of the OVA- Control group. 8. The NR-HA group did not show my considerable difference from the Normal group. The OVA-saline group and the OVA-Needle prick group showed suppressive effects on OVA-induced asthma however they were not statistically significant. Conclusion : These results suggest that NR-HA at Pyesu(BL13) is considered to be effective in treating asthma and to be put to practical use in the future asthma clinic.

가미패독산(加味敗毒散) 경구 투여에 의한 Nc/Nga 생쥐의 아토피 피부염 억제 작용 (Suppression of Spontaneous Dermatitis in Nc/Nga Atopic Model by Gamipaidok-san, a Traditional Herbal Medicine)

  • Atopic dermitiis(AD) is a chronic inflammatory skin disease, which requires safe and effective medicinal therapy. Over production of Th2 cytokines and chemokines as well as IgE, which are mediated by highly activated immune cells, have been considered as pathologic factors in this disease. We found that Gamipaidok-san(GPDS), which is a traditional herbal medicine clinically prescribing for atopic dermitis patients in the hospital, has suppressive effects on the development of DNC8 induced dermatitis in Nc/Nga atopic model. Oral administration of GPDS at the concentration of 250 mg/Kg for 12 weeks significantly suppressed the clinical severity of the dermatitis including pruities, edema, eczematous and dryness. Histological examination revealed that thickness of dermis and epidermis were considerably reduced, and the number of infiltrated inflammatory immune cells including mast cells, CCR3+, and CD4+ T cells were decreased in the affected skin and ear, and consistantly, the number of CD3+/CCR3+ cells in Iymph nodes were decreased. The levels of Th2 cytokines produced by activated splenocyte from atopic mice were also down-regulated by GPDS. Furthermore, the serum levels of IgE were considerably reduced, which accompanied by a decrease in the number of B220+IgE+ B cells in the Iymph nodes. Taken together, these results suggested that oral administration of GPDS, a traditional herbal medicine, has suppressive effects on atopic dermitis of Nc/Nga mouse by the modulation of the immune system, therefore GPDS has potential as a natural therapeutic for treatment of atopic dermatitis.

황금이 천식모델 생쥐의 폐조직내 면역세포에 미치는 영향 (Effect of Scutellariae Radix extract on immune cells in OVA-induced asthmatic mice lung tissue)

  • Objective : The purpose of this research is to examine the effects of Scutellariae Radix(SR) extract on immune cells in ovalbumin(OVA)-induced asthmatic mice. Methods : In vitro, C57BL/6 mice were exposed to OVA for 6 weeks. The mice lungs were taken out, chopped, grinded with collagenase and activated by rIL-3/rIL-5. The lung cells were treated with SR extract and analyzed by a flow cytometer. Results : The population of granulocytes/lymphocytes, CD3e-/CCR3+, CD69+/CD3e+, CD23+/B220+, CD4+ cells in the SR group in vitro decreased and population of CD8+ cells in the 100 and $10{\mu}g/m{\ell}$ SR group in vitro decreased, compared with that of control group. Conclusion : The results of this study support a role for SR as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions.

DA-6034 ameliorates hepatic steatosis and inflammation in high fat diet-induced obese mice

  • Background: Nonalcoholic fatty liver disease (NAFLD) is characterized by an increase in hepatic triglyceride content and increased inflammatory macrophage infiltration through the C-C motif chemokine receptor (CCR) 5 pathway in the liver. DA-6034 (7-carboxymethyloxy-3',4',5-trimethoxy flavone), is a synthetic derivative of eupatilin that exhibits anti-inflammatory activity in inflammatory bowel disease. However, the effect of DA-6034 on the inflammatory response in NAFLD is not well elucidated. Therefore, we aimed to determine the effect of DA-6034 on hepatic steatosis and inflammation. Methods: Forty male C57BL/6J mice were divided into the following four groups: (1) regular diet (RD), (2) RD with DA-6034, (3) high fat diet (HFD), and (4) HFD with DA-6034. All mice were sacrificed 12 weeks after the start of the experiment. The effects of DA-6034 on macrophages were assessed using RAW 264.7 cells. Results: DA-6034 not only reduced hepatic triglyceride levels and lipid accumulation but also macrophage infiltration and proinflammatory cytokines in HFD-fed mice. According to fluorescence-activated cell sorter analysis, DA-6034 reduced the CD8+ T cell fraction in the liver of HFD-fed mice. DA-6034 also reduced CCR5 expression and the migration of liver macrophages in HFD-fed mice and inhibited CCR2 ligand and CCR4 ligand, which stimulated the migration of macrophages. Conclusion: Overall, DA-6034 attenuates hepatic steatosis and inflammation in obesity by regulating CCR5 expression in macrophages.

유세포 형광 분석기를 통한 아로마 요법의 알러지 천식 억제 효과 탐색 (The Search for Inhibitory Effect of Aroma Therapy on Allergic Asthma by Flow cytometer)

  • The purpose of this study was to investigate the inhibitory effect of the aroma therapy of three kinds of aroma oil mixtures on asthma. 1. The percentage of granulocytes/lymphocytes population in mouse OVA-induced asthma lung cells was decreased significantly compared with those of control group. 2. The number of CCR3+ cells, CD4+ cells, CD8+ cells, CD23 and CD3e+/CD69+ in lungs of the mice group treated with M1 were decreased significantly compared with those of control group. 3. The number of IgE+/B220+ cells in the lungs of the mice group treated with M1 decreased significantly compared with those of control group. But the number of B220+ cells in the lunes of the mice group treated with M1 didn't show significant difference compared with those of control group. 4. The number of Gr-1+/CD11b+ cells in lungs of the mice group treated with M1 didn't show significant difference compared with those of control group. But the number of CD11b+ cells in lungs of the mice group treated with M1 decreased significantly compared with those of control group.
