Cadmium, Lead, and Zinc Accumulation in Rice Grown at Paddy Soils near Old Zinc-Mining Sites (아연광산(亞鉛鑛山) 인근답(隣近沓)의 토양중(土壤中) 중금속함량(重金屬含量)과 현미중(玄米中) 함량(含量)과의 관계(關係))

  • Effect of Cd, Pb, and Zn content in soil on their accumulation in rice was studied by analyzing brown rice(93) and soil samples(180) collected from paddy soils near old zinc-mining sites in 1979 and 1980. Ratio of Cd, Pb, and Zn in brown rice to soil decreased with the increase of their contents in soil and found to be linear function of the inversed values of their contents in soil. Contents of Cd, Pb, and Zn in brown rice harvested in 1980 were lower than those in 1979. The significant difference in contents of Pb and Zn between two years might be attributed to weather. Air temperature and duration of sunshine in 1980 were significantly lower than those in 1979.

Effect of Soyangin-Hyeongbangpaedok-san on Anti-CD3 Stimulated Mouse T Cells In Vivo and In Vitro (소양인 형방패독산(荊防敗毒散)의 마우스 T 세포 활성에 대한 실험적 연구)

  • Soyangin-Hyeongbangpaedok-san(SHBPDS) is a herbal formula used for the common cold or upper respiratory illness. In order to investigate the effect of SHBPDS, mice were orally administered with SHBPDS alcohol extract for 7 days followed by intravenous anti-CD3 injection. In addition, splenocytes and CD4 T cells were cultured with SHBPDS in response to anti-CD3 in vitro and cytokines and transcription factors were evaluated. In vivo treatment with SHBPDS significantly augmented the expressions of the percentage of CD4 T cells and CD 69, an indicator of early T cell activation. Serum levels of IL-4 were significantly increased but those of IFN-${\gamma}$ and IL-2 did not reach statistical significance. The expressions of IFN-${\gamma}$ and T-bet mRNA were significantly downregulated in SHBPDS treated mice while those of IL-4 and C-Maf were significantly upregulated. In vitro stimulation of splenocytes and CD4 T cells by SHBPDS resulted in a reduction in IFN-${\gamma}$ secretion and STAT4 activity. The IL-4 releases from both cells were slightly reduced, but STAT6 activity was rather increased. In conclusion, SHBPDS exerted an inhibition in the expression of IFN-${\gamma}$, T-bet and STAT4 while IL-4, C-Maf and STAT6 were increased. Further studies are required to examine its pharmacological effects using more appropriate animal experiments.

Inhibition of Human $CD8^+$ Cytotoxic T Lymphocyte (CTL) -mediated Cytotoxicity in Porcine Fetal Fibroblast Cells by Overexpression of Human Cytomegalovirus Glycoprotein Unique Short (US) 2 Gene

  • Xenotransplantation of pig organs into humans is a potential solution for the shortage of donor organs for transplantation. However, multiple immune barriers preclude its clinical application. In particular, the initial type of rejection in xenotransplantation is an acute cellular rejection by host $CD8^+$ cytotoxic T lymphocyte (CTL) cells that react to donor major histocompatibility complex (MHC) class I. The human cytomegalovirus (HCMV) glycoprotein Unique Short (US) 2 specifically targets MHC class I heavy chains to relocate them from the endoplasmic reticulum (ER) membrane to the cytosol, where they are degraded by the proteasome. In this study we transfected the US2 gene into minipig fetal fibroblasts and established four US2 clonal cell lines. The integration of US2 into transgenic fetal cells was confirmed using PCR and Southern blot assay. The reduction of Swine Leukocyte Antigen (SLA)-I by US2 was also detected using Flow cytometry assay (FACS). The FACS analysis of the US2 clonal cell lines demonstrated a substantial reduction in SLA-I surface expression. The level (44% to 76%) of SLA-I expression in US2 clonal cell lines was decreased relative to the control. In cytotoxicity assay the rate of $CD8^+$ T cell-mediated cytotoxicity was significantly reduced to 23.8${\pm}$15.1% compared to the control (59.8${\pm}$8.4%, p<0.05). In conclusion, US2 can directly protect against $CD8^+$-mediated cell lysis. These results indicate that the expression of US2 in pig cells may provide a new approach to overcome the CTL-mediated immune rejection in xenotransplantation.

Sound Enhancement of low Sample rate Audio Using LMS in DWT Domain (DWT영역에서 LMS를 이용한 저 샘플링 비율 오디오 신호의 음질 향상)

  • In order to mitigate the problems in storage space and network bandwidth for the full CD quality audio, current digital audio is always restricted by sampling rate and bandwidth. This restriction normally results in low sample rate audio or calls for the data compression scheme such as MP3. However, they can only reproduce a lower frequency range than a regular CD quality because of the Nyquist sampling theory. Consequently they lose rich spatial information embedded in high frequency. The propose of this paper is to propose efficient high frequency enhancement of low sample rate audio using n adaptive filtering and DWT analysis and synthesis. The proposed algorithm uses the LMS adaptive algorithm to estimate the missing high frequency contents in DWT domain and it then reconstructs the spectrally enhanced audio by using the DWT synthesis procedure. Several experiments with real speech and audio are performed and compared with other algorithm. From the experimental results of spectrogram and sonic test, we confirm that the proposed algorithm outperforms the other algorithm and reasonably works well for the most of audio cases.

Improving the Electrical and Optical Properties of Blue Polymer Light Emitting Diodes by Introducing TPBI Electron Transport Layer (TPBI 전자 수송층을 이용한 청색 고분자 유기발광다이오드의 전기·광학적 특성 향상)

  • In this study, we fabricated a polymer light emitting diode (PLED) and investigated its electrical and optical characteristics in order to examine the effects of the PFO [poly(9,9-dioctylfluorene-2-7-diyl) end capped with N,N-bis(4-methylphenyl)-4-aniline] concentrations in the emission layer (EML). The PFO polymer was dissolved in toluene ranging from 0.2 to 1.2 wt%, and then spin-coated. To verify the influence of the TPBI [2,2',2"-(1,3,5-Benzinetriyl)-tris(1-phenyl-1-H-benzimidazole)]electron transport layer, TPBI small molecules were deposited by thermal evaporation. The current density, luminance, wavelength and current efficiency characteristics of the prepared PLED devices with and without TPBI layer at various PFO concentrations were measured and compared. The luminance and current efficiency of the PLED devices without TPBI layer were increased, from 117 to $553\;cd/m^2$ and from 0.015 to 0.110 cd/A, as the PFO concentration increased from 0.2 to 1.0 wt%. For the PLED devices with TPBI layer, the luminance and current efficiency were $1724\;cd/m^2$ and 0.501 cd/A at 1.0 wt% PFO concentration. The CIE color coordinators of the PLED device with TPBI layer at 1.0 wt% PFO concentration showed a more pure blue color compared with the one without TPBI, and the CIE values varied from (x, y) = (0.21, 0.23) to (x, y) = (0.16, 0.11).

A Comparative Study of Douchi Hataedock versus Omega-3 on the Effect of Anti-inflammation in the Colonic Muscle through Th2 Skewed Condition Control (Th2 skewed condition 조절을 통한 대장점막 내 염증개선효과에 대한 두시 하태독법과 오메가3 비교 연구)

  • Objectives Hataedock is an orally-administered herbal extract that used to treat toxic heat and help excrete meconium accumulates in the fetus from pregnancy. This study compared the effects of Hataedock using the extract of Douchi and omega-3 to control inflammation in the colonic mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), FGT group (given the extract of Douchi), and O3T group (given omega-3). The 3-week-old mouse from each group were given the extract of Douchi and omega-3. After 2 weeks, the colonic mucosa tissues of the mice were observed via Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF in the intestinal mucosa. Results In both FGT and O3T group, IL-4, IL-13, STAT-6 levels and CD40-positive reactions were decreased. However, EGF-positive responses were increased. FGT group showed 35% decrease in IL-4, 45% decrease in IL-13, 48% decrease in CD40, 50% decrease in $Fc{\varepsilon}RI$, 54% decrease in $p-I{\kappa}B$, 52% increase in EGF compared to the control group. O3T group also showed 14% decrease in IL-4, 23% decrease in IL-13, 22% decrease in CD40, 29% decrease in $Fc{\varepsilon}RI$, 41% decrease in $p-I{\kappa}B$, 39% increase in EGF compared to the control group. Conclusions Both early administration of Douchi extract and omega-3 were effective in regulating Th2, relieving inflammation and developing colonic mucosal tissues. Hataedock with Douchi extract may be more effective in modulating immune system in colonic mucosa than omega-3.

Anti-asthmatic Effects of Samjajihwang-tang in OVA-induced Mice (삼자지황탕(三子地黃湯)의 생쥐 모델에 대한 항천식 효과)

  • This study aimed to evaluate the anti-asthmatic effects of Samjajihwang-tang (SJT) using OVA-induced asthmatic mice model. Asthmatic mice model was conducted by repeated challenge of OVA using C57BL/6 mice. Each group was treated with distilled water, SJT (400 mg/kg and 200 mg/kg) extract or cyclosporin A (10 mg/kg) for the later 8 weeks, Penh (plethysmography and enhanced pause), immune cells subpopulation, eotaxin, IL-5, TNF-${\alpha}$, Anti-OVA-lgE in BALF (bronchoalveolar lavage), and lung tissue was analyzed, No cytotoxicity of SJT was shown on hFCs (human fibroblast cells). Administration of SJT significantly decreased Penh levels comparing to control group. SJT treatment significantly ameliorated the increase of total cells number and eosinophil including of immune cell subpopulation of $CD3^+/CD69^+$, $CCR3^+$, $B220^+/CD22^+$, $B220^+/CD45^+$, and $B220^+/lgE^+$ cells in BALF comparing to control group. Eotaxin, IL-5, TNF-${\alpha}$, and Anti-OVA-lgE level in BALF were significantly decreased by SJT treatment too. Histopathological finding verified the improvement of infiltration of inflammatory cells and collagen tissue in the SJT groups comparing to control group. These results strongly suggest that SJT would be a effective candidate for herbal-originated anti-asthmatic drug. However, this drug should be further studied for characterization of the accurate action and underlying mechanisms using variant disease model in the future.

Effects of Seonpye-tang on OVA-Induced Asthmatic Mice (천식모델생쥐에서 선폐탕(宣肺湯)의 면역조절 효과)

  • This study aimed to evaluate the anti-asthmatic effects of Seonpye-tang (SPT) using OVA-induced asthmatic mice model. Scavenging activity of SPT on DPPH free radical and SOD-like activity of SPT were measured at final concentration 62.5, 125, 250, 500 (${\mu}g/m{\ell}$), RBL-2H3 cells were treated with DNP IgE for 24hr, and treated with SPT (1, 10, 100 ${\mu}g/m{\ell}$) for 1hr, followed by treatment with DNP-HSA for 1hr at $37^{\circ}C$. The level of IL-4 and TNF-${\alpha}$ were measured by ELISA. Asthmatic mice model was conducted by repeated challenge of OVA using C57BL/6 mice. Immune Responses Induced by HSP60 DNA Vaccine against Toxoplasma gondii Infection in Kunming Mice

  • Toxoplasma gondii can infect all the vertebrates including human, and leads to serious toxoplasmosis and considerable veterinary problems. T. gondii heat shock protein 60 (HSP60) is associated with the activation of antigen presenting cells by inducing initial immune responses and releasing inflammatory cytokines. It might be a potential DNA vaccine candidate for this parasite. A pVAX-HSP60 DNA vaccine was constructed and immune responses was evaluated in Kunming mice in this study. Our data indicated that the innate and adaptive immune responses was elicited by successive immunizations with pVAX-HSP60 DNA, showing apparent increases of CD3e+CD4+ and CD3e+CD8a+ T cells in spleen tissues of the HSP60 DNA-immunized mice ($24.70{\pm}1.23%$ and $10.90{\pm}0.89%$, P<0.05) and higher levels of specific antibodies in sera. Furthermore, the survival period of the immunized mice ($10.53{\pm}4.78day$) were significantly prolonged during the acute T. gondii infection. Decrease of brain cysts was significant in the experimental group during the chronic infection (P<0.01). Taken together, TgHSP60 DNA can be as a vaccine candidate to prevent the acute and chronic T. gondii infections.

The Concentrations of Heavy Metals in Sediment Seawater and Oyster (Crassostrea gigas) in Coastal Region of Industrial Complex in Korea (한국 임해 공단 연안에서 퇴적물 해수 및 굴 (Crassostrea gigas)의 중금속 함량)

  • To investigate the degree of contamination from Korean coastal region, the concentrations of Cu, Pb, Zn and Cd in sediments, seawater and oyster (Crassostrea gigas) at Masan Bay, Onsan Bay, Daesan industrial complex and their control areas were analysed. Values for sediments, seawater and oyster in the industrial complex coastal region were higher than those in the control area except for seawater in Daesan. The values for dissolved phasc of Cu, Pb, Zn and Cd in seawater showed 0.3~1.75 ${\mu}g/l$, 0.013~0.12 ${\mu}g/l$, 0.20~6.14 ${\mu}g/l$ and 0.007~0.021 ${\mu}g/l$, respectively. The concentrations of Cu, Pb, Zn and Cd in sediments werd 12.0~47.8 ${\mu}g/g$, 6.16~59.5 ${\mu}g/g$, 43.0~230 ${\mu}g/g$ and 0.52~11.2 ${\mu}g/g$, respectively. The concentrations of Cu, Pb, Zn and Cd in oyster showed 12.1~85.6 ${\mu}g/g$, 0.267~1.48 ${\mu}g/g$, 1, 070~3, 250 ${\mu}g/g$ and 3.23~7.71 ${\mu}g/g$, respectively. The contents of heavy metals in oysters at industrial complex coastal region showed that they were not seriously contaminated compared with those of Mussel Watch (1992).
