Induction of Unique STAT Heterodimers by IL-21 Provokes IL-1RI Expression on CD8+ T Cells, Resulting in Enhanced IL-1β Dependent Effector Function

  • IL-1β plays critical roles in the priming and effector phases of immune responses such as the differentiation, commitment, and memory formation of T cells. In this context, several reports have suggested that the IL-1β signal is crucial for CTL-mediated immune responses to viral infections and tumors. However, little is known regarding whether IL-1β acts directly on CD8+ T cells and what the molecular mechanisms underlying expression of IL-1 receptors (IL-1Rs) on CD8+ T cells and features of IL-1R+ CD8+ T cells are. Here, we provide evidence that the expression of IL-1R type I (IL-1RI), the functional receptor of IL-1β, is preferentially induced by IL-21 on TCR-stimulated CD8+ T cells. Further, IL-1β enhances the effector function of CD8+ T cells expressing IL-21-induced IL-1RI by increasing cytokine production and release of cytotoxic granules containing granzyme B. The IL-21-IL-1RI-IL-1β axis is involved in an augmented effector function through regulation of transcription factors BATF, Blimp-1, and IRF4. Moreover, this axis confers a unique effector function to CD8+ T cells compared to conventional type 1 cytotoxic T cells differentiated with IL-12. Chemical inhibitor and immunoprecipitation assay demonstrated that IL-21 induces a unique pattern of STAT activation with the formation of both STAT1:STAT3 and STAT3:STAT5 heterodimers, which are critical for the induction of IL-1RI on TCR-stimulated CD8+ T cells. Taken together, we propose that induction of a novel subset of IL-1RI-expressing CD8+ T cells by IL-21 may be beneficial to the protective immune response against viral infections and is therefore important to consider for vaccine design.

  • 카드뮴이 BALC/c 마우스의 세포성면역능에 미치는 영향을 평가하고자 여러 농도의 카드뮴이 첨가된 음료를 장기간 동안 자유급식하고, 비장내 T helper 및 T suppressor cell의 분포도, 복강대식세포의 탐식능 및 시험관내에서 Con-A 및 LPS에 대한 비장세포의 증식능을 조사하여 다음과 같은 결과를 얻었다. 1. 각군의 6주간 체중 증가율은 대조군이 27.0%이었으며, 25, 50, 100 및 200ppm $CdCl_2$, 투여군의 체중증가율은 각각 28.54%, 28.31%, 20.49% 및 18.04%로 나타났다. 2. 체중당 비장무게(mg/g)는 대조군이 $4.34{\pm}0.23$이었으며, 25, 50, 100 및 200ppm $CdCl_2$, 투여군은 각각 $4.20{\pm}0.54$, $4.80{\pm}0.87$, $4.25{\pm}0.32$$4.40{\pm}0.32$이었다. 또한 총비장세포수(${\times}10^7$)는 대조군($24.29{\pm}5.98$)에 비해 25, 50, 100 및 200ppm $CdCl_2$, 투여군에서 각각 14.1%, 35.7%, 16.6% 및 22.0%증가하였다. 3. 총 $CD_4{^+}$ 세포수(${\times}10^7$)는 대조군이 $9.15{\pm}2.24$, 25, 50, 100 및 200ppm $CdCl_2$, 투여군은 각각 $10.40{\pm}2.04$, $12.04{\pm}3.08$, $10.20{\pm}3.16$$10.80{\pm}1.48$ 이었으며, 총 $CD_8{^+}$ 세포수(${\times}10^7$)는 대조군이 $2.32{\pm}0.56$, 각 실험군의 총세포수는 $2.54{\pm}0.27$, $3.12{\pm}0.80$, $2.25{\pm}0.70$$2.24{\pm}0.28$ 이었다. 한편, $CD_4{^+}/CD_8{^+}$ 비율은 50ppm 투여군($3.88{\pm}0.01$)을 제외하고 모든 실험군에서 대조군($3.97{\pm}0.02$)에 비해 유의하게 증가하였다(p<0.001). 4. 카드뮴 처리군의 SRBC에 대한 복강대식세포의 탐식능은 25 및 50ppm 투여군에서는 대조군에 비하여 유의하게 높았으나(p<0.05 및 p<0.01), 100 및 200ppm 투여군에서는 대조군과 유사하였다. 5. 시험관에서의 Con-A 및 LPS에 대한 비장림프구 증식반응은 LPS의 경우 카드뮴 농도에 비례하여 억제되었으나 Con-A에 대한 증식반응은 저농도($10^{-7}-10^{-6}M$)에서 증가된 양상을 보였다. 6. $CdCl_2$에 대한 비장세포의 세독독성은 $10^{-4}M$에서 특히 높았다. 이상의 결과는 카드뮴이 세포성면역반응에서 중요한 역할을 하는 T세포 아군의 분포도를 변화시키므로써 면역반응에 영향을 줄 수 있음을 시사한다.

태아 간세포의 거핵구 집락형성 (Megakaryocyte Colony Formation of Fetal Liver Cells)

  • 목 적 : 태아 간세포는 면역학적으로 미숙하여 이식에 따른 거부반응이 잘 일어나지 않으므로 조혈모세포 이식, 특히 자궁내 이식의 좋은 공여세포로 이용할 수 있으나 아직 국내에서는 이를 위한 기초연구가 미미한 실정이다. 연구자는 태아 간 단핵세포를 이용하여 면역조직학적 염색과 체외배양 결과를 분석하여 태아 간의 거핵구 집락형성과 관련된 기초자료를 마련하고자 이 연구를 시행하였다. 방 법: 임신중절시 수집한 태아 간조직과 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에서 단핵세포를 분리한 후 $MegaCult^{TM}-C$ 배지에서 12일간 배양 후 성장인자 첨가에 따른 거핵구 집락형성을 관찰하였고, 5일간의 Flask 배양 후 유세포 분석기를 이용하여 CD34, CD41 양성세포를 측정하였으며, methylcellulose 배지를 이용하여 $37^{\circ}C$ 5% $CO_2$ 배양기에서 배양한 후 제 7일과 14일에 성장인자 첨가에 따른 CFU-GM 집락 수를 조사하였다. 결 과 : 태아 간의 단핵세포 수는 재태주령 11주에 비해서 19주에서 더 많았으며 세포생존율은 $91.2{\pm}3.4%$였다. $MegaCult^{TM}-C$ kit를 이용하여 12일간 배양시 재태주령 11주의 태아 간이 19주에 비해서 거핵구 집락 수가 많았고, 4례 모두 성장인자의 첨가에 따른 집락증폭의 상승효과는 없었으며 배양 후 형성된 거핵구 집락 수는 태아 간, 특발성 혈소판 감소성 자반증 환아의 골수, 범혈구 감소증 환아의 골수 순이었다. 거핵구 집락 중 순수집락 수는 태아 간이 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에 비해서 유의하게 높았으며, 순수집락 중 large CFU-Mk의 비율은 태아 간이 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에 비해서 높았으나 유의성은 보이지 않았다. 태아 간 단핵세포를 5일간 flask 배양한 후 CD34 양성세포의 발현율은 증가하였으나 성장인자의 첨가에 따른 집락증폭의 상승효과는 TPO 투여군에서만 있었고, CD41 양성세포의 발현율 역시 배양 후 증가하였으나 성장인자 첨가군에 비해서 대조군의 발현율이 너무 높은 결과를 보였다. Methylcellulose 배지를 이용한 단핵세포 배양에서 CFU-GM 집락 수는 태아 간의 경우 배양 7일에 비하여 14일에 감소하였고, 성장인자의 첨가에 따른 집락증폭의 상승효과는 태아 간에서는 모든 군에서 보였으나 특발성 혈소판 감소성 자반증에서는 GM-CSF 투여군에서만 있었으며 범혈구 감소증 환아의 골수에서는 없었다. 결 론: $MegaCult^{TM}-C$ 배지를 이용하여 태아 간세포를 배양하고 면역조직학적 염색으로 거핵구 집락형성을 성공적으로 관찰하였으며, 태아 간에서 얻은 거핵구 집락이 특발성 혈소판 감소성 자반증 및 범혈구 감소증 환아의 골수에서 얻은 집락에 비하여 집락수나 순수집락의 분포, 집락의 크기 등에서 우수한 것으로 나타났으나 태아 간 조혈모세포 이식의 실제적인 임상적용을 위해서는 체계적인 더 많은 연구가 필요할 것으로 생각된다.

아토피피부염 동물 병태모델에서의 형개연교탕(荊芥連翹湯)의 면역조절작용 (The effects of HYT on various immunological factors related to pathogenesis of allergic dermatitis in NC/Nga mice induced by Biostir AD)

  • Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of HYT on the recovery of dermatitic symptoms by how HYT influenced the immune related factors. The results are as below: 1. Compared to the control, HYT treated group showed recovery of atopic dermatitis by the naked eye observation, and significant reduction of dermatits index was observed after 14 weeks. 2. HYT treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and Gr-1+/CD11b+ immune cells in dorsal skin by 40.5%, 34.2%, and 48.1%, respectively. 3. HYT treated group showed increase in the ratio of CD19+ immune cells within PBMC by 10.8%, as well as decrease in CD3+, CD3+/CD69+, NKT+ ratios by 5.3%, 35.2%, and 44.9%, respectively. 4. HYT treated group showed increase in the expression of IFN-$\gamma$ in serum by 589.3%, whereas the expression of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 31.4%, 82.1%, 97.1%, 39.5%, 83.7%, 26.1%, 48.6%, respectively. A 47.2% decrease in IgE expression was also observed. The results above strongly supported the improvement of atopic dermatitis by HYT treatment through immune modulation. Further studies on the synergistic effect of each ingredients of HYT and therapeutic effects according to the dosage of each ingredient should be followed for clinical applications. 소풍활혈탕가미방이 Collagen으로 수발된 생쥐의 관절염 억제에 관한 연구 (A Study on the Effects of Sopunghwalhyeoltang-gamibang on the Inhibition of Arthritis Induced by Collagen on the Mouse)

  • This study was carried out to know the effects of sopunghwalhyeoltang-gamibang(hereinafter referred to SPHHT) on the inhibition of arthritis induced by collagen on the mouse. Various experimental were performed in vivo (in DBA/1 J mice which are experimental model of arthritis induced by collagen) to analyse the immunomodulatory effects of SPHHT. The results were obtained as follows: 1. The cytotoxicity against mLFC was not measured in all concentration. 2. The arthritis index, incidence, hind paw edema, delayed-type hypersensitivity reaction, spleen weight were reduced in SPHHT treated group. 3. The expression of CD3 ε +/CD4+, CD3 ε +/CD8+ and CD19+ in peripheral blood mononuclear were reduced in SPHHT treated group. 4. The production of pro-inflammatory cytokines such as IL-6 and TNF-α were significantly reduced in SPHHT treated group. 5. The formation of new bones and synivium were stimulated in SPHHT treated group. Comparison of the results for this study showed that Sopung-hwalhyeoltang-gamibang had immunomodulatory effects. So we expect that Sopunghwalhyeoltang-gamibang should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.

한국인 간장 및 신장피질에 함유된 카드뮴, 구리, 수은, 아연 함량과 메탈로치오나인과의 관계 (Relationships Between Cadmium, Copper, Mercury, Zinc Levels and Metallothionein in the Liver and Kidney Cortex of Korean)

  • In order to elucidate the relationships between cadmium, copper, mercury, zinc levels and metallothionein in the liver and kidney cortex of Korean, the levels of Cd, Zn, Hg, Cu and metallothionein (MT) were determined in the kidney cortex and liver of 50 subjects deceased in the period of January-November, 2001 in the area of Seoul and Gyeonggi Province of Korea. The mean age of the population studied was 36.3+/-12.3 years. The tissues were digested with microwave digestion system and the elements were determined by inductively coupled plasma atomic emission spectrometry. MT was determined by the Cd-hemoglobin affinity assay. The determined levels (mean+/-SD) were: 33.9+/-18.9 micrograms Cd/g wet weight; 47.5+/-12.6 micrograms Zn/g wet weight; 2.5+/-0.57 microgram Cu/g wet weight; 0.26+/-0.31 micrograms Hg/g wet weight, 4.0+/-3.1 mg MT/g wet weight in renal cortex and 2.5+/-1.9 micrograms Cd/g wet weight; 46.9+/-15.0 micrograms Zn/g wet weight; 6.2+/-2.5 micrograms Cu/g wet weight; 0.10+/-0.15 micrograms Hg/g wet weight, 0.92+/-0.57 mg MT/g wet weight in the liver. Positive relationships between Cd and MT, sum of four divalent metal and MT in the kidney cortex were observed. No other correlation was found between Cu and MT, Hg and MT, Zn and MT in either organs.

전북지방 치과기공실 공기중 및 치과기공사의 뇨중중금속 함량에 관한 연구 (Study on Heavy Metal Contents in Air of Den Tal Laboratories and Urine of Dental Laboratory Technicians in CHONBUK Area)

  • For this study, I used the urine of 100 dental laboratroy technicians and 24 students in Chonbuk region. The purposes of this study were to investigate concentrations of cadmium (Cd), nickel (Ni) and chromium (Cr) in the air of dental laboratories. The results were as follows: 1. The concentrations of Cd is 0.0115mg/m$^{3}$ in the porcelain part, 0.0090mg/m$^{3}$ in the polishing part, that of Ni is 0.2864mg/m$^{3}$ in the porcelain part, 0.3188mg/m$^{3}$ in the polishing part, that of Cr is 0.0448mg/m$^{3}$ in the porcelain part, 0.1032mg/m$^{3}$ in the polishing part. 2. The concentrations of Cd, Ni and Cr in the technicians urine are 3.06 $\mu$g/l, 44.55 $\mu$g/l and 14.58 $\mu$g/l. Those of students, urine as the control group are 1.93 $\mu$g/l, 19.05 $\mu$g/l and 7.48 $\mu$g/l. There is significant difference between experimental group and control group (P < 0.01). 3. Watching for age and working place, 31-40 years age group reveals the highest group in the concentrations of Cd. Over 41 year age group represents the highest group in the concentrations of Ni and Cr (P < 0.01) The concentration of Cd turns up the highest in the partial part and that of Ni and Cr appears the highest in the polishing part (P < 0.01). Looking into working age, over 7 year group is the highest group in the concentrations of all investigated (P < 0.05).

Bone Marrow-derived Side Population Cells are Capable of Functional Cardiomyogenic Differentiation

  • It has been reported that bone marrow (BM)-side population (SP) cells, with hematopoietic stem cell activity, can transdifferentiate into cardiomyocytes and contribute to myocardial repair. However, this has been questioned by recent studies showing that hematopoietic stem cells (HSCs) adopt a hematopoietic cell lineage in the ischemic myocardium. The present study was designed to investigate whether BM-SP cells can in fact transdifferentiate into functional cardiomyocytes. Phenotypically, BM-SP cells were $19.59%{\pm}9.00\;CD14^+$, $8.22%{\pm}2.72\;CD34^+$, $92.93%{\pm}2.68\;CD44^+$, $91.86%{\pm}4.07\;CD45^+$, $28.48%{\pm}2.24\;c-kit^+$, $71.09%{\pm}3.67\;Sca-1^+$. Expression of endothelial cell markers (CD31, Flk-1, Tie-2 and VEGF-A) was higher in BM-SP cells than whole BM cells. After five days of co-culture with neonatal cardiomyocytes, $7.2%{\pm}1.2$ of the BM-SP cells expressed sarcomeric ${\alpha}$-actinin as measured by flow cytometry. Moreover, BM-SP cells co-cultured on neonatal cardiomyocytes fixed to inhibit cell fusion also expressed sarcomeric ${\alpha}$-actinin. The co-cultured BM-SP cells showed neonatal cardiomyocyte-like action potentials of relatively long duration and shallow resting membrane potential. They also generated calcium transients with amplitude and duration similar to those of neonatal cardiomyocytes. These results show that BM-SP cells are capable of functional cardiomyogenic differentiation when co-cultured with neonatal cardiomyocytes.

플라스틱 기판상에 적색 OLED 제작과 특성 연구 (Fabrication and Characterization of Red OLED on the Plastic Substrate)

  • A high efficient organic red light emitting device with structure of DNTPD/TAPC/$Bebq_2$ :[$(pq)_2Ir(acac)$, SFC-411]/SFC-137 was fabricated on the plastic substrate, which can be applied in the fields of flexible display and illumination. In the device structure, N,N'-diphenyl-N,N'-bis-[4-(phenyl-m-tolylamino)-phenyl]-biphenyl-4,4'-diamine[DNTPD] as a hole injection layer and 1,1-bis-(di-4-tolylaminophenyl) cyclohexane [TAPC] as a hole transport were used. Bis(10-hydroxybenzo[h]quinolinato) beryllium complex [$Bebq_2$] was used as a light emitting host material. The host material, $Bebq_2$ was doubly doped with volume ratio of 7% iridium(III)bis-(2-phenylquinoline)acetylacetonate[$(pq)_2$Ir(acac)] and 3% SFC-411[red phosphor dye coded by the proprietary company]. And then, SFC-137 was used as an electron transport layer. The luminous intensity and current efficiency of the fabricated device were $22,780\;cd/m^2$ at 9V and 17.3 cd/A under $10,000\;cd/m^2$, respectively. The maximum current efficiency of the device was 22.4cd/A under $580\;cd/m^2$.
